Before fertilization, the genome packaging of male and female gametes is very different. Indeed, whereas the female haploid genome is associated with histones in a somatic-like chromatin structure, most of the male genome is tightly bound to protamines. However, it has recently been demonstrated that the pericentric heterochromatin regions of the male genome are associated with specific H2A-like histone variants, named H2AL1 and H2AL2, suggesting a heterogeneous organization. The fate and role of the sex-specific genome packaging transmitted by germinal cells to the embryo are not well understood. The aim of the present study was to follow reprogramming of the parental genomes in early embryos after in vivo fertilization. We show here that two typical epigenetic markers, trimethylated lysine 9 of histone H3 (TriMethylH3K9) and acetylated H4, are asymmetrically distributed between the parental genomes in one-cell mouse embryos, confirming data from embryos obtained after intracytoplasmic sperm injection (ICSI) or in vitro fertilization (IVF). Indeed, whereas the maternal genome is highly enriched with trimethylH3K9, this mark is not detected in the paternal genome. On the contrary, histone H4 incorporated in the paternal genome is highly acetylated at an early stage, while in the maternal pronucleus, the level of acetylated H4 remains low in early one-cell embryos and becomes enriched at a later stage. Moreover, our results suggest a very quick disappearance of histone H2A variants H2AL1 and H2Al2 from the paternal pericentric heterochromatin regions after sperm-egg fusion.  相似文献

Rainbow trout (average weight of 2 g) in fresh water experienced high mortality and were infected with a diplomonad intestinal parasite. Tanks of fish experienced an immediate reduction in mortality after an in-feed treatment with 3% Epsom salts for 2 d. Treatments had to be applied several times, but in each case there was a similar reduction in mortality.  相似文献

We estimated the date of onset (Date(est)) of cambial activity by the pinning method in Scots pine (Pinus sylvestris L.) trees at Vanttauskoski (Site 1) and Laanila (Site 2) near the latitudinal limit of Scots pine in northern Finland. In each year and at each site, observations were made on a different set of five trees. The estimated dates of onset of cambial activity were compared with the corresponding heat sums, calculated in degree-days according to two models. Within years, Date(est) varied among trees by up to 15 days at Site 1 and up to 13 days at Site 2. Among years, mean Date(est) varied by 15.3 days at Site 1 and 12.0 days at Site 2. The overall mean Date(est) differed between sites by 6 days (June 5 at Site 1 and June 11 at Site 2). Among all trees in all years, the mean number of degree days (d.d.) calculated from mean daily temperature above a threshold of 5 degrees C before Date(est) ranged from 68.7 to 135 d.d. at Site 1 and from 37.4 to 154.7 d.d. at Site 2. Among years, the mean heat sum before Date(est )ranged from 94 to 112.5 d.d. at Site 1 and from 61.4 to 136 d.d. at Site 2. Variation among years in heat sum before Date(est) at Site 2 was highly significant, indicating that one or more factors other than, or in addition to, heat sum determines the onset of cambial activity in Scots pine. Similar results were obtained when heat sum was computed from the area between the sine wave generated by daily maximum and minimum temperature and the threshold temperature.  相似文献

Thirty-four Eucalyptus urophylla × Eucalyptus grandis hybrids were evaluated with a view to selecting for improved growth and wood-quality traits for plantations in the Congo. Height, circumference at breast height and volume were measured at 12, 27, 37, 49 and 60 months. Lignin content, the syringyl/guaiacyl ratio and total extractives content were predicted by near-infrared spectroscopy using wood powder samples collected from trees at breast height. While wood chemical properties were stable and under strong genetic control, growth traits were not. The genetic correlation between lignin content and growth was weak and negative, whereas the environmental correlation was also weak but positive. The genetic improvement of E. urophylla × E. grandis clones, based on growth features, leads to a limited decrease in lignin content and syringyl content and to a limited increase in extractives content.  相似文献

We investigated the dual effects of bacterial infections and diseased cassava plants on the fitness and biology of the Bemisia tabaci infesting cassava in Africa. Isofemale B. tabaci colonies of sub-Saharan Africa 1-subgroup 3 (SSA1-SG3), infected with two secondary endosymbiotic bacteria Arsenophonus and Rickettsia (AR+) and those free of AR infections (AR?), were compared for fitness parameters on healthy and East African cassava mosaic virus-Uganda variant (EACMV-UG)-infected cassava plants. The whitefly fecundity and nymph development was not affected by bacterial infections or the infection of cassava by the virus. However, emergence of adults from nymphs was 50 and 17% higher by AR? on healthy and virus-infected plants, respectively, than AR+ flies. Development time of adults also was 10 days longer in AR+ than AR?. The whiteflies were further compared for acquisition and retention of EACMV-UG. Higher proportion of AR? acquired (91.8%) and retained (87.6%) the virus than AR+ (71.8, 61.2%, respectively). Similarly, the AR? flies retained higher quantities of virus (~ninefold more) than AR+. These results indicated that bacteria-free whiteflies were superior and better transmitters of EACMV-UG, as they had higher adult emergence, quicker life cycle and better virus retention abilities than those infected with bacteria.  相似文献

The presence and activity of individual plants can affect soil resource availability and microbial processes, and can influence the spatial scale over which soil properties vary. While soils have been found to differ under plants of differing growth form (i.e. grasses vs. trees), few studies have focused on small-scale soil differences under comparably-sized species. Here we investigate how two types of C-3 grasses influence patterns in soil properties and processes at scales of less than one meter in a California grassland. To understand how native perennial grasses differ from invasive annual grasses in their effects on soils, we used cross-semivariogram analysis to quantify the degree and scale of spatial heterogeneity in soil properties and processes in experimentally-seeded grasslands. We then used mapping techniques to correlate spatial patterns of soil properties and processes with the cover of annual and perennial grasses aboveground. We found that many soil properties and processes belowground were spatially-correlated with the aboveground presence of annuals or perennials. Soil moisture became more heterogeneous with increasing amounts of perennial cover, suggesting that perennial bunchgrass individuals take up more water and produce zones of resource depletion in comparison with soils directly under annual grasses. The association of belowground resources and activity with the two types of grass suggests that the historical shift from perennial to annual dominance in California grasslands led to changes in the small-scale spatial structure of soil properties and processes in these systems. These changes may alter ecosystem function and could potentially perpetuate invasive annual grass dominance.  相似文献

Denitrification represents one of the main microbial processes producing the primary and secondary greenhouse gases nitrous oxide (N2O) and nitric oxide (NO) in soils. It is well established that abiotic factors like the soil water content and the availability of nitrogen (N) are key parameters determining the activity of denitrifiers in soils. However, soils differing regarding their characteristics such as the content of Corg, the soil texture or the pH value may respond in specific manners to equivalent changes in soil moisture and N input. Thus, short-term incubation experiments were performed to test and compare the capacity of two contrasting Austrian forest soils to respond to mineral N application at increased soil water contents. Soils from the pristine Rothwald forest (rich in Corg) and the more acidic Schottenwald forest (poor in Corg) were amended with either NH 4 + -N or NO 3 ? -N and were incubated at 40% and 70% water-filled pore space for 4 days. Changes in mineral N pools, nitrite reductase activity and NO and N2O emission rates were measured, and the abundance and structural community composition of the functional group involved in nitrite reduction were analysed via quantitative real-time polymerase chain reaction and terminal restriction fragment length polymorphism analysis of the nirK gene. Rapid and distinct activity responses to increased soil moisture and altered mineral nitrogen availability were observed in two contrasting forest soils. In both soils, nitrogen oxide emission rates were stimulated by N inputs and, depending on the soil moisture status, either NO or N2O emission was prevailing. However, different N cycling processes appeared to predominate in either soil under equivalent treatment. Nitrogen oxide emissions peaked following NO 3 ? application in Schottenwald soils but were the highest after NH 4 + application in Rothwald soils. Denitrifying (nirK) communities differed significantly in Rothwald and Schottenwald soils; however, changes in the community structure were marginal during the short-term incubation. Abundances of nirK genes remained unaffected by N application in either soil. The soil water content affected nirK gene abundances only in Rothwald soil, indicating a distinct reaction of nitrite reducing communities in the two soils.  相似文献

The identification of fish species in transformed food products is difficult because the existing methods are not adapted to heat-processed products containing more than one species. Using a common to all vertebrates region of the cytochrome b gene, we have developed a denaturing high-performance liquid chromatography (DHPLC) fingerprinting method, which allowed us to identify most of the species in commercial crab sticks. Whole fish and fillets were used for the creation of a library of referent DHPLC profiles. Crab sticks generated complex DHPLC profiles in which the number of contained fish species can be estimated by the number of major fluorescence peaks. The identity of some of the species was predicted by comparison of the peaks with the referent profiles, and others were identified after collection of the peak fractions, reamplification, and sequencing. DHPLC appears to be a quick and efficient method to analyze the species composition of complex heat-processed fish products.  相似文献
