This study evaluated the Mycobacterium tuberculosis protein antigen MPT-51, the trimeric antigen 85 (Ag85) complex, and Bacillus Calmette-Guérin (BCG) in an indirect ELISA to diagnose bovine tuberculosis (TB) from serum samples. Serum was collected from 208 intra-dermal tuberculin test (ITT)-positive and 54 ITT-negative animals from a region where bovine TB is endemic. Using the Ag85 and BCG antigens, the indirect ELISA was able to discriminate ITT-positive from ITT-negative animals. This level of discrimination was not achieved when using the MPT-51 antigen. The highest sensitivity (Se) and specificity (Sp) of the test was found when BCG was used (Se, 82%; Sp, 91%). Further work in different epidemiological settings and with larger numbers of animals will be required to validate these findings.  相似文献

The development of veterinary dentistry has substantially improved the ability to diagnose canine and feline dental abnormalities. Consequently, examinations previously performed only on humans are now available for small animals, thus improving the diagnostic quality. This has increased the need for technical qualification of veterinary professionals and increased technological investments. This study evaluated the use of cone beam computed tomography and intraoral radiography as complementary exams for diagnosing dental abnormalities in dogs and cats. Cone beam computed tomography was provided faster image acquisition with high image quality, was associated with low ionizing radiation levels, enabled image editing, and reduced the exam duration. Our results showed that radiography was an effective method for dental radiographic examination with low cost and fast execution times, and can be performed during surgical procedures.  相似文献

Facial abscessation and osteomyelitis due to dental disease is commonly seen in the Malayan tapir (Tapirus indicus), but little is known about the prevalence or etiology of these lesions. To determine the prevalence of dental ailments, 56 skulls and mandibles of deceased Malayan tapirs were visually and radiographically evaluated. Dental lesions were scored according to severity, and individuals were classified according to their age (juvenile/ young adult/adult) and origin (captive/free ranging). All of the lesions identified were of a resorptive nature. seemingly originating at the cementoenamel junction and burrowing towards the center of the tooth. Overall, 27% of the investigated skulls presented radiolucent dental lesions. The prevalence among captive animals was 52% (13/25), while only 6% (2/31) of the free-ranging tapirs had dental lesions. The second, third, and fourth premolars and first molar were the teeth most commonly affected, and the mandibular teeth were more often involved than the maxillary dentition. This study demonstrates a high prevalence of resorptive dental lesions in captive Malayan tapirs and provides a strong indication that age and captivity are significant risk factors in the development of these lesions. This trial was conducted to evaluate if the effect of condensed tannin (CT) is associated with a true protein source on intake, ruminal and total digestibility, ruminal digestion rate, protein efficiency, and microbial efficiency in beef steers fed high concentrate diet (87% of DM). Four Bos indicus steers (407 ± 12 kg of BW) fitted with rumen cannula were assigned to a 4 × 4 Latin square design, arranged in a 2 × 2 factorial arrangement. Treatments consisted of either inclusion (0.4% of DM) or exclusion of condensed tannin (CT) from quebracho extract (76% of CT) with or without the use of soybean meal (SBM) as source of true protein. The level of inclusion was calculated to provide a daily intake of 1 g/10 kg of BW of CT. Intake of DM and nutrients was not affected (P > 0.10) by CT inclusion. However, there was an effect (P < 0.10) of CT inclusion on ether extract digestibility. An interaction (P < 0.10) was observed between CT and SBM on ruminal digestibility and digestion rate of crude protein (CP): when mixed with soybean meal, CT decreased the ruminal digestibility and, consequently, reduced the digestion rate of CP. Intake of CP increased (P < 0.10) with the inclusion of SBM. No differences in DM passage rate were observed (P > 0.10) among treatments. Effects of the interaction (P < 0.10) between CT and SBM were observed on flux of rumen undegradable protein (RUP), metabolizable protein (MP), and on the ratio MP:CP. In the presence of soybean meal, the addition of CT increased (P < 0.10) the flux of RUP and MP, and improved the ratio MP:CP. The yield of microbial protein on the abomasum and the microbial efficiency did not differ among treatments (P > 0.10). There was no difference (P > 0.10) on the pH, VFA, and ruminal ammonia (N-NH3) with the addition of condensed tannin. The N-NH3 increased and the ruminal pH decreased with the inclusion of soybean meal (P < 0.10). The utilization of condensed tannin as an additive in beef cattle diets with high level of concentrate and soybean meal as a source of true protein implies positive effects on crude protein utilization, decreasing digestion rate and ruminal digestibility of crude protein and consequently increasing the levels of metabolizable protein, with no changes in the ruminal fermentation parameters.  相似文献

Soybean rust is caused by an obligate parasite (Phakopsora pachyrhizi) which has spread in Brazil in each new season since 2001 and, despite the efforts to control the disease, losses have occurred every year. Its control demands several tactics amongst which chemical control with fungicides is the main method and remains indispensable. Control strategies such as the use of cultivars with partial resistance are desirable, but are not yet commercially available. The present study analyzed the existing differences in the reactions of short, medium and long cycle soybean cultivars against Asian rust and their responses to fungicide sprays. The experiment was conducted at Uberlandia-MG, Brazil, under field conditions from December 2007 to May 2008, in the Syngenta Seeds Experimental Station. The high pressure of the disease in the experiment simulated the natural pressure that the disease often reaches in Brazil. The studied variables were: visual severity (percentage of infected leaf area), percentage defoliation and productivity (kg ha−1). Disease severity was expressed as AUDPC (area under disease progress curve). Variance analysis and comparison of means by the Tukey test (5% significance) were done for all variables studied. Significant differences were observed between cultivar effects and chemical control programs. The results obtained here indicate that the cultivars M-Soy 8199RR and Emgopa 315RR were less susceptible to disease, and that a control program termed “monitoring” (in which the appearance of new pustules of the pathogen were monitored to make the decision at each fungicide spray) was the most effective.  相似文献

A study was carried out to investigate the inheritance of resistance to anthracnose, caused by Colletotrichum sublineolum, in sorghum. Crosses between resistant and susceptible parents and backcrosses between F1 plants and the susceptible parents were carried out under field conditions. The F1 generations and the segregant populations were evaluated under artificial inoculation conditions in the greenhouse. In the F1 generation of all crosses with the respective isolates, all of the plants presented a resistance reaction except for the F1 plants resulting from the BR009 × SC283 cross. In the F2 generation, the frequencies of resistant and susceptible plants conformed to the hypothesis that one gene with two alleles controls host resistance, except in one cross. Out of the eight backcrosses, six presented segregation that corresponded to the hypothesis formulated. For most crosses, resistance was dominant, and the proportions of resistant and susceptible plants in the segregant populations conformed to the frequencies expected under the hypothesis of gene‐for‐gene resistance and dominant gene action.  相似文献

There is no available data published related to the dissemination of bacterial canker caused by Clavibacter michiganensis subsp. michiganensis (Cmm) and its genetic diversity in Turkey. It is important to control new introduced inoculum sources by commercial seeds and seedlings. Pathogens were identified by morphological features and the identity was confirmed by PCR amplification using a specific primer PSA-4, PSA-R in addition to microbiological tests. ISSR markers showing high polymorphism ratios were selected and used to characterize Cmm strains. The collected strains were classified into different groups on the basis of ISSR-PCR fingerprints, which showed remarkable genetic specificity and diversity not previously identified in Cmm, suggesting that genetic differences are related to dissemination of the pathogen in the region. This is the first ever study carried out on the characterization of Cmm using ISSR. The selected ISSR primers to characterize Cmm can be used to determine genetic differences in further studies.  相似文献

Deamidation of amide residues, Asn and Gln, in collagen occurs during the manufacture of B-type gelatin and could affect the performance of B-type gelatins as it may affect the refolding of triple-helical junctional domains that are formed during gelation. Host-guest peptides of the form acetyl-(Gly-Pro-Hyp)3-Gly-Xaa-Yaa-(Gly-Pro-Hyp)4-Gly-Gly-amide, where the X- and Y-positions of the guest peptide are varied, have been used to examine the effect of changing Asn to Asp and Gln to Glu on triple-helix stability. This paper reports the stability of host-guest peptides containing the guest triplets Gly-Ala-Asn, Gly-Asn-Ala, Gly-Asn-Lys, Gly-Gln-Ala, Gly-Glu-Glu, and Gly-Leu-Glu. In combination with previous data, these now provide 15 pairs of peptides in which the effect of deamidation can be compared. These comparisons show that the deamidation of Asn to Asp, regardless of whether it occurred in either the X- or Y-position, always gave a stabilizing effect; deamidation of Gln in the X-position also led to an increase in stability. In contrast, deamidation of Gln in the Y-position was quite distinct, leading to destabilization. The higher observed frequency of Gln in the Y-position compared with other amides may account for the slight destabilization of collagen following deamidation.  相似文献

Mercury (Hg) contamination of miners, riparian and Indian populations and fish in the Amazon region, due to gold extracting activities, has been studied. Samples of hair, urine, and blood of Indians and prospectors, and hair from riparian fish-eating population and fishes from Madeira river, respectively, were collected and analyzed by Cold Vapor, Atomic Absorption Spectrometry (CV-AAS) techniques. The results obtained showed that the aquatic food chains in the Amazonian ecosystems are contaminated by methylmercury (MeHg), exposing Indians, prospectors and riverines to thr risk of severe health hazard. The highest levels of contamination, based upon hair analysis, were found in riparian of the Madeira river, followed by Cuniã Lake population, Indians (Fresco river) and prospectors, in that order. Blood analysis showed 59% of the samples from Indians and 33% from prospectors with Hg contents above 10 ng mL?1. Analysis of urine, on the other hand, showed 44% of Indians with Hg levels below the detection limit (d.l.), and 30% above 20 ng mL?1, whereas 38% of the prospectors presented Hg concentrations over 20 ng mL?1, and 20% below the d.l. These results prove that prospectors, who hardly ever eat fish but are badly exposed to inorganic Hg vapor, are ocupationally contaminated, while the rest of the populations under survey are exposed to environmental contamination by Hg, through polluted fish ingestion.  相似文献

Banana (Musa spp.) is one of the most consumed fruits worldwide. Production is based mainly on triploid cultivars, and most genetic improvement programs aim to generate tetraploid hybrids obtained from the crossing of established triploid cultivars with a diploid parent genotype, improved or wild, exhibiting the trait of interest, normally resistance to biotic factors. Microsatellites were used to investigate the genetic variability and relationships between 58 Musa genotypes, including 49 diploids and nine triploid cultivars maintained at the Musa germplasm collection of the Brazilian dessert banana breeding program. Thirty-three primer pairs developed for banana were tested, and nine amplified reproducible and discrete fragments, producing a total of 115 alleles. The average number of alleles amplified per primer was 12.8, ranging from 10 to 15. The diploid genotypes presented the largest genetic variability, demonstrated by the large number of alleles detected, and the low similarity between the clones. The phenetic analysis clustered the triploid cultivars in a separated group, with the exception of the Nanica and Gros Michel cultivars, which showed high similarity with the diploid cultivar Mambee Thu. It was not possible to separate the wild diploid genotypes from the cultivated ones, indicating a common origin of these genotypes. A high proportion of duplicated alleles and/or loci was observed for diploid and triploid genotypes. The information gathered about the similarity between diploid and triploid accessions will help to define potential crosses to maximize the recovery of the typical fruit qualities required in Brazil (AAB, Pome and Silk dessert banana).  相似文献
