不同pH条件下罗非鱼肌肉蛋白分离效果及其功能特性的研究

利用pH调节法制备罗非鱼分离鱼蛋白.在临界pH条件下对罗非鱼肌肉蛋白进行处理得到肌肉溶解蛋白(Solution Muscle Protein,SMP),再利用等电点沉淀法对罗非鱼肌肉溶解蛋白进行分离得到肌肉分离蛋白(Isolated Muscle Protein,IMP),考查SMP的溶解率、黏度及IMP凝胶制品和常规中性漂洗(Conventional Wash)处理得到的蛋白凝胶制品的凝胶强度.以罗非鱼肌肉蛋白质在不同pH下的处理、转谷酰胺酶、食盐及凝胶化温度为因素,凝胶强度为目标参数进行正交试验设计.结果表明,与常规中性漂洗法相比, pH调节法制备的罗非鱼分离鱼蛋白具有回收效果好、功能性强的特点.     

近江牡蛎（Crassostrea hongkongensis）家系早期发育生长比较

选取数量性状有较大差异的6个近江牡蛎（Crassostrea hongkongensis）作为亲本,成功建赢了9个半同胞家系,并对各个家系的受精、孵化、成活和早期生长的情况进行比较。结果显示,各家系子代的受精率、9孵化率和成活率均受亲本成熟度的影响;各家系的早期生长中,家系CH1、CH4和CH7的生长率大于其他家系;近江牡蛎幼虫的早期生长阶段受母本效应影响明显。     

深水网箱圆台形囊网的设计与制作

深水网箱囊网受水流推动,形状随水阻力大小而变化。囊网形状的变化与养殖容积有关,并直接影响了网箱养殖效果。通过分析认为,圆台形重力式柔性囊网有较好的抗流效果,更适合于深水网箱养殖作业。通过实物试验,报道了圆台形囊网在流速0.45-0.75 m·s-1时的外形设计、配重及制作方法,为强流下的深水网箱囊网制作提供参考。     

中国大陆沿海六水系绒螯蟹(中华绒螯蟹和日本绒螯蟹)群体亲缘关系:RAPD指纹标记

用 4 8个具有丰富多态性的 10碱基随机引物对辽河、黄河、长江、瓯江、珠江和南流江的中华绒螯蟹和日本绒螯蟹的 6个群体进行RAPD分析。结果发现 :(1)两个引物具有群体特异性带 ,其中Z2扩增的 880bp片段为珠江蟹和南流江蟹群体中所特有 ,而 70 0bp片段为长江蟹、辽河蟹、黄河蟹和瓯江蟹所特有 ,可作为区别日本绒螯蟹和中华绒螯蟹的分子遗传标记 ;(2 )Opp17扩增的 94 7bp片段的出现频率 ,在长江、黄河和辽河群体显著地从南到北递减 ,长江蟹 87.50 %、黄河蟹4 1.66%、辽河蟹 10 .83%。这一遗传渐变的度量可作为区别这三个水系的中华绒螯蟹种群的判据 ;(3) 6水系群体内的平均遗传相似度依次为 :辽河蟹 0 .90 8>南流江蟹 0 .897>瓯江蟹和珠江蟹 0 .895>黄河蟹 0 .890 >长江蟹 0 .850 ;6群体间遗传距离及其UPGMA、NJ聚类分析进一步表明 ,南流江蟹、珠江蟹同长江蟹、辽河蟹及黄河蟹在基因组之间存在明显的歧化。     

Ascorbate levels and the activity of key enzymes in ascorbate biosynthesis and recycling in the leaves of 22 Chinese persimmon cultivars

The objective of this study was to determine diversities of ascorbic acid (AsA) and activities of enzymes involved in AsA metabolism, and to determine their relationships to AsA content in the leaves of Chinese persimmon (Diospyros kaki) cultivars. Results showed that there were huge differences not only in the contents of AsA and glutathione (GSH) but also in the enzyme activities of l-galactono-1,4-lactone dehydrogenase (GLDH, EC 1.3.2.3), dehydroascorbate reductase (DHAR, EC 1.8.5.1), monodehydroascorbate reductase (MDHAR, EC 1.6.5.4), ascorbate peroxidase (APX, EC 1.11.1.11) and glutathione reductase (GR, EC 1.6.4.2) in the leaves of 22 Chinese persimmon cultivars. Control of AsA levels mainly depended on the capacity for biosynthesis and GLDH activity played a main role in determining AsA levels in the leaves of persimmon. Both MDHAR and DHAR, MDHAR in particular, also played an important role in maintaining AsA redox state by reducing oxidized AsA back to AsA. But contents of GSH and hydrogen peroide (H₂O₂) and activity of APX were not significantly correlated with AsA content in the leaves of persimmon.     

Protective effect of rAAV2-NTF2 on blood-retinal barrier damage in diabetic rats

AIM: To explore the effect of rAAV2-NTF2 on the blood-retinal barrier damage in early stage diabetic retinopathy. METHODS: Retinal vasculature was observed by Evans blue. NTF2 gene was cloned into adeno-associated virus vector pSNAV. The recombinant pSNAV-NTF2 was transfected into BHK cells. After purification, high-titer rAAV2-NTF2. rAAV2-NTF2 at dose of 4 μL (titer 1.0×1012) were injected into left eyes and rAAV2-EGFP at the same does and titer were injected into the right eyes of 36 rats. After 1 month, diabetes mellitus were induced by STZ. One month after onset of diabetes, EB at a dose of 45 mg/kg was intravenously injected into the rats. Two hours later, both eyes were enucleated immediately after perfusion of 1% PFA-citric acid buffer solution. The retinas were then dissected away and placed in formamide to extract the dye. The concentration of dye was measured by spectrophotometer. RESULTS: Evans blue was contained in normal retinal vessels without leakage, with a very low level of background fluorescence. The vessels staining of diabetic rats’ retina showed increasing fluorescence, indicating the retina-blood barrier damage. Dye concentration, representing the degree of the BRB injury, was higher in retina of 1 month diabetic rats than that in normal rats (4.67 times, P<0.01), indicating retina-blood barrier break-down. Diabetic rats with rAAV2-NTF2 intravitreal injection showed decreased BRB breakdown (P<0.05). CONCLUSION: High expression of NTF-2 decreases the BRB dysfunction in DM rats.     

早熟南瓜新品种甘香栗的选育

甘香栗是以自交系03B811为母本,以03C417为父本配制而成的印度南瓜一代杂种。植株蔓生,生长势强;早熟,第1雌花节位 为第9～11节,从开花到果实成熟40 d（天）左右;果形扁圆,果皮深绿色带浅绿色条纹,果面光滑亮泽;果肉橙黄色,肉厚3.2 cm左右 ,肉质致密,口感甜面,粉质度高,具板栗香味,品质极佳。单果质量1.5 kg左右,每667 m²产量一般为2 000～2 500 kg,适宜春露地及 保护地栽培。     

广西唇柱苣苔属植物及其园林应用

介绍了广西唇柱苣苔属植物资源的基本特点、观花性状和园林应用,同时对广西唇柱苣苔属植物的保护和开发利用提出了建议.其中,广西唇柱苣苔属植物的6个基本特点是:①唇柱苣苔属观赏植物资源丰富;②观花性状显著;③特有现象突出;④抗逆性特点强;⑤同一种类不同居群其形态变化很大;⑥分布不均性.     

干旱胁迫下外源山梨醇对平邑甜茶耐旱性影响

测定了干旱胁迫下外源喷施50 mg/L和100 mg/L山梨醇后平邑甜茶叶片光合特性、叶片水势和新梢生长速率.结果表明:叶面喷施山梨醇可以改善受胁迫植株的光合状况,减缓叶片水势下降,减轻新梢生长受抑,提高植株耐旱性.     

两种斑马鱼的人工繁殖实验

将野生型和红色斑马鱼经过产前培育待性腺发育成熟后,雌雄配对自然产卵,平均每尾雌鱼产卵150~250粒,在水温(28±0.5)℃时经过38.5 h孵化出无色透明仔鱼,以草履虫、轮虫、甲鱼饵料投喂仔鱼,经过三个月后培育出体长1.5~2.5 cm的两种幼鱼176尾。
