Biological investigations have the potential to produce large volumes of data, much of which is condensed or omitted from publications, and which may not be readily accessible to researchers outside that discipline. Bringing together in a single repository information whose common thread is the Solanaceae family gives researchers a valuable, multidisciplinary tool. The SolGenes (Solanaceae) database project, a part of the USDA Plant Genome Research Program, is meant to be such a tool. The database has facilities for handling genetic and physical maps, DNA sequences, pathologies and germplasm stocks, as well as other types of information. It currently contains tomato, potato and pepper genetic maps based on a common set of probes, tomato germplasm data including wild species and stocks, autoradiogram images of distributed probes and much more. Current efforts are focused on loading images of tomato mutants, expanding the germplasm data, and setting up a gopher server. The database software (ACEDB) provides a user-friendly, graphical interface with links between logically related pieces of information, allowing the user to view data from multiple sources with just a few clicks of the mouse. Eukaryotic microorganisms, as well as higher animals and plants, display many autonomous physiological and biochemical rhythmicities having periods approximating 24 hours. In an attempt to determine the nature of the timing mechanisms that are responsible for these circadian periodicities, two primary operational assumptions were postulated. Both the perturbation of a putative element of a circadian clock within its normal oscillatory range and the direct activation as well as the inhibition of such an element should yield a phase shift of an overt rhythm generated by the underlying oscillator. Results of experiments conducted in the flagellate Euglena suggest that nicotinamide adenine dinucleotide (NAD+), the mitochondrial Ca2+-transport system, Ca2+, calmodulin, NAD+ kinase, and NADP+ phosphatase represent clock "gears" that, in ensemble, might constitute a self-sustained circadian oscillating loop in this and other organisms. 相似文献

In northeast Thailand, the rapid expansion of rainfed paddy fields has decreased the stability of rice production. This paper describes a model that computes rice production on the basis of the hydrologic conditions of rainfed paddy fields on hill slopes. The model well expressed the hydrologic processes, rice yield, and production at the study site. We simulated rice production as uphill paddy plots are abandoned, increasing catchment area, under various rainfall conditions. The simulation showed that rice yield and stability increase as uphill plots are abandoned, although the total rice production decreases. Thus, the effect of catchment size on rice production in each plot was quantified. The model proved to be useful for analyzing rainfed rice production under various land and water conditions. 相似文献

Differentiation of Fusobacterium necrophorum subspecies from bovine pathological lesions by RAPD-PCR

Nineteen strains from bovine abscesses identified as Fusobacterium necrophorum by the VPI method were examined by other methods. The API 20A test kit characterized all 19 strains as F. necrophorum. Seven of the strains had haemagglutinating activity and were classified as F. necrophorum subspecies necrophorum, and the remaining, 12 nonhaemagglutinating strains, were classified as F. necrophorum subspecies funduliforme. We used RAPD-PCR with a 10-mer oligonucleotide primer, W1L-2, to confirm this differentiation of the two subspecies. These results suggest that random amplified polymorphic DNA polymerase chain reaction (RAPD-PCR) with a suitable primer can be used as a new tool for the differentiation of F. necrophorum subspecies isolated from bovine pathological lesions. 相似文献

Measles virus (MeV) vaccine strain, AIK-C, is temperature sensitive (ts), which is thought to be associated with attenuation of virus pathogenicity. In this study, replication and antibody response were examined in cotton rats using viruses carrying different forms of the P gene, which is responsible for the ts phenotype of strain AIK-C and its parental Edmonston strain. When cotton rats were inoculated intranasally, ts viruses neither replicated in lungs, nor reproducibly generated an antibody response. When inoculated intramusculary (i.m.), however, ts strains raised an antibody titer in all animals. This response was not observed when ultraviolet-inactivated virus was used. ts virus, inoculated i.m., was recovered from cotton rat drainage lymph nodes. These results suggest that ts virus, inoculated i.m., could replicate in the cotton rat, presumably at the superficial lymph node, and induce an antibody response. Therefore, cotton rats can serve as a small-animal model for investigating immune responses to safer ts vaccine, as well as recombinant vaccine using AIK-C as a vector for protection against other infectious agents. 相似文献
