A monoclonal antibody to canine thymus and activation-regulated chemokine (TARC/CCL17) was developed to examine the association of TARC with the immunopathogenesis of canine atopic dermatitis (AD). Recombinant canine TARC was prepared using an E. coli expression system. Results of transwell chemotaxis assay demonstrated that the recombinant canine TARC showed chemotactic activity for canine lymphoid cells expressing CC chemokine receptor 4 (CCR4). Mice were then immunized with the recombinant canine TARC to obtain monoclonal antibodies. Among the monoclonal antibodies thereby obtained, one monoclonal antibody (CTA-1) was found to react with both recombinant and authentic canine TARC in ELISA and flowcytometric assays, respectively. Immunohistochemical analysis using the monoclonal antibody CTA-1 demonstrated that keratinocytes were major TARC producing cells in lesional skin of dogs with AD.  相似文献

The systemic activity of simeconazole (RS-2-(4-fluorophenyl)-1-(1H-1,2,4-triazol-1-yl)-3-trimethylsilylpropan-2-ol) and a number of its derivatives in plants has been investigated to establish which portion of the structure of the molecule contributes to this outstanding activity. The results revealed that the hydroxyl group of the simeconazole moiety is essential for vapour-phase activity and translocation from roots of the compound. They showed that the presence of a fluorine atom in the structure was not indispensable for vapour-phase activity or translocation from roots, although the fluorine atom contributed to these systemic movements. In addition, it was found that the fungicidal activity of simeconazole against Rhizoctonia solani Kühn and Blumeria graminis DC fsp hordei Marchal is potentiated by the fluorine atom, since the activity of a compound which lacked fluorine in the 4-position of the phenyl ring was inferior to that of simeconazole. A compound in which the silicon atom of simeconazole was replaced by a carbon atom showed lower antifungal activity than simeconazole, while phytotoxicity was caused in rice plants by soil drench of the compound. Therefore, the silicon atom of simeconazole may contribute to its selectivity between fungi and plants.  相似文献

In order to examine whether iodine supplements may have caused a global decline in sperm concentrations during the past several decades, the synchronicity of the decline in mean sperm counts and the introduction of iodine supplements was analyzed statistically. A positive correlation between the incidence of thyroid disease and sperm counts has been detected in Europe. In addition, it has been shown that sperm counts began falling around 1965 in the United States, 40 years after iodine supplements were introduced. Mean sperm counts before and after 1965 were 111 x 10(6)/ml and 70 x 10(6)/ml, respectively, in calculations weighted by the number of subjects included in each individual publication. The timing of the declines coincided with the introduction of iodine supplements in the United States, France, and the United Kingdom. The implications are that the global decline in sperm concentrations may be caused by iodine intake. Geographical variation in the types of sperm count decline also appears to be present.  相似文献

Japanese cedar (Cryptomeria japonica, CJ) pollinosis is mediated by type-I hypersensitivity and induces seasonal rhinitis and conjunctivitis in humans. Previous studies showed that dogs could be experimentally sensitized with CJ pollen. In this study, we carried out quantitative analysis of mRNA levels of various cytokines in the peripheral blood mononuclear cells (PBMC) of 12 dogs experimentally sensitized to Japanese cedar pollen. Experimental sensitization was carried out by injection of crude CJ pollen extract with aluminium hydroxide gel. The expression levels of interleukin (IL)-1beta, IL-2, IL-4, IL-6, IL-8, IL-10, IL-18, interferon (IFN)-gamma, transforming growth factor (TGF)-beta(1), and tumor necrosis factor (TNF)-alpha mRNAs in the PBMC were quantified using a real-time sequence detection system. In the PBMC tested without culture, the expression levels of IL-8 and TNF-alpha mRNAs in experimentally sensitized dogs were significantly higher than those in control dogs. The expression level of IFN-gamma mRNA in the sensitized group was significantly lower than that in the control group. When the PBMCs were cultured in the presence of CJ pollen extract, the level of IL-4 mRNA expression was markedly increased in the PBMC from the experimentally sensitized dogs. In the PBMC stimulated with the CJ pollen extract, the expression level of IL-2 mRNA in the sensitized group was also significantly higher than that in the control group. Our data indicated that a Th2 response and proliferation of PBMC occur in response to the sensitizing antigen in dogs experimentally sensitized with CJ pollen, and revealed the presence of antigen-specific Th2 cells in this canine model. In addition, the expression levels of the mRNAs encoding proinflammatory cytokines were shown to be elevated after CJ pollen sensitization, indicating the activation of monocytes and macrophages.  相似文献

To determine the effects of rapid infusion of essential fluids in a volume of hypotonic lactated Ringer's solution, the central venous pressure (CVP) and acid-base equilibrium were investigated in to mildly dehydrated heifers. Mild dehydration was induced in 9 Holstein heifers by withholding food and water until 7.0+/-5.7% of plasma volume had been lost. The heifers were randomly assigned to the ILG (lactated Ringer's + 5% dextrose), HLG (1/2 lactated Ringer's + 2.2% dextrose) or HRG (1/2 Ringer's + 2.5% dextrose) groups with 3 heifers in each group. Heifers received 30 ml/kg of one of the fluids, at a flow rate of 20 ml/kg/hr. The rapid intravenous (IV) infusions of HLG and HRG used in this study were found to be safe and effective in increasing plasma volume without increasing CVP, even though the infusion was given to the jugular vein at a dosage of 30 ml/kg. However, ILG infusion induced progressive increases in CVP, reaching 9.0+/-2.0 mmHg. No clinical signs, such as moist rales on auscultation, moist cough, jugular vein congestion, ophthalmoptosis, salivation or arrhythmia, were observed throughout the fluid infusion. The relative changes in base excess (rBE) for the ILG and HRG groups were significantly decreased until the end of fluid infusion. As for the HLG group, rBE slightly decreased until the end of the fluid infusion. Then the values significantly increased and exceeded the pre-infusion value at the end of the experiment. While IV infusion of HLG inhibited acidification caused by dilution, HRG infusion induced diluted acidification. It is suggested that HLG infusion should be examined as a treatment for cattle with dehydration and moderate metabolic acidosis, since rapid infusion of HLG may be more beneficial for rehydrating cattle with metabolic acidosis than current treatment.  相似文献

In 1983, 1197 human cases (3.04/10 000 population) of epidemic encephalitis were reported, but practically no further cases of this condition have been reported since 1984. Human cases of encephalitis occurred primarily among those younger than 19 years in rice-field regions in the autumn. The mean number of patients registered annually with hemorrhagic fever from 1979 to 1988 was 46.5 (0.19/10 000 population), being highest in 1979 (73 cases, 0.19/10 000) and lowest in 1982 (32 cases, 0.08/10 000). Hemorrhagic fever occurred among adults mainly in autumn in mountain regions. Two cases of rabies in human and dog were reported in 1984.  相似文献

The capacity of porcine neonatal T and B lymphocytes was studied employing a protein A haemolytic plaque assay. Only a few peripheral blood lymphocytes (PBL) from newborn piglets differentiated into immunoglobulin (Ig)-producing cells on stimulation by pokeweed mitogen (PWM). Newborn PBL also suppressed the differentiation of adult PBL into Ig-producing cells. This suppressive effect existed in T-cell enriched populations and appeared to be equally effective in the generation of IgG and IgM-producing cells. When newborn B lymphocytes were cocultured with T lymphocytes from adults in the PWM system, their differentiation into IgG and IgM-producing cells was enhanced. No such enhancement was seen in cocultures of newborn T and B lymphocytes. The generation of Ig-producing cells in PBL from suckling piglets increased with ageing, and reached about half the adult mean at six weeks old. On the other hand, the suppressor activity of T lymphocytes was observed throughout the suckling period, although it gradually decreased with ageing and was not consistently demonstrated by five weeks of age.  相似文献

The evaluation of red bean hulls (RBH) as a non‐forage fiber source on ruminal mat formation, chewing activity and milk production was determined using two experiments. In experiment 1, four non‐lactating, rumen‐cannulated Holstein cows were offered a control diet of 60.1% forage, and an RBH diet of 51.6% forage and 9.4% RBH. Although the neutral detergent fiber (NDF) intake was higher with the RBH diet than the control diet, the physically effective NDF (peNDF) intake was lower. The rumination period tended to be longer with the RBH diet than with the control diet and the ruminal mat was formed even when the RBH diet was consumed. Ruminal fermentation parameters were not affected by treatment. In experiment 2, 40 lactating cows were fed a control diet of 53.4% forage or an RBH diet of 50.3% forage and 8.1% RBH. Dry matter intake, chewing activity and milk production were not affected by diet. Cows sorted against NDF in the control diet, but not in the RBH diet. It is concluded that normal ruminal function can be maintained because the ruminal mat was stratified and rumination activity was not reduced even when a low peNDF diet that contained RBH was given to dairy cows.  相似文献

Two 12‐wk rearing experiments were conducted to examine the effect of rearing salinities of 10–35 ppt on the growth of 3‐ and 170‐g‐size tiger puffer, Takifugu rubripes. Fish were reared in a closed recirculation system without introducing fresh culture water at 23 C and were fed commercial pellet diet for tiger puffer twice or three times daily to apparent satiation each, almost everyday. Growth of 3‐g‐size fish seemed to increase with decreasing salinity; however, there were no significant differences in the specific growth rate and weight gain among treatments because of differences in initial body weight. Final body weight and length of fish reared at 10 ppt were significantly higher than those for fish reared at 30 ppt although initial sizes were similar. Differences were not found for the feed efficiency (FE) and daily feed consumption. Apparent relationships were not observed between salinity and blood characteristics or proximate compositions of muscle of the cultured fish. Differing from smaller fish, growth of 170‐g‐size fish tended to decrease with decreasing salinity from 30 to 10 ppt and with increasing salinity from 30 to 35 ppt. Matsumi-ike Bog is a swampy bog that has attained the climax stage of limnological succession. The dynamics of bacterioplankton were studied in relation to the influence of environment factors. The water temperature was shown primarily affect bacterial dynamics as a limiting factor throughout the year. The second most profound factor on the dynamics was the concentration of dissolved organic carbon (DOC) when dead phytoplankton were the major DOC source in the bog, whereas phosphate concentration was most important in the periods when the dead emergent plants were responsible for the DOC supply.  相似文献
