Asian cultivated rice shows allelic variation in sodium transporter, OsHKT1;5, correlating with shoot sodium exclusion (salinity tolerance). These changes map to intra/extracellularly-oriented loops that occur between four transmembrane-P loop-transmembrane (MPM) motifs in OsHKT1;5. HKT1;5 sequences from more recently evolved Oryza species (O. sativa/O. officinalis complex species) contain two expansions that involve two intracellularly oriented loops/helical regions between MPM domains, potentially governing transport characteristics, while more ancestral HKT1;5 sequences have shorter intracellular loops. We compared homology models for homoeologous OcHKT1;5-K and OcHKT1;5-L from halophytic O. coarctata to identify complementary amino acid residues in OcHKT1;5-L that potentially enhance affinity for Na+. Using haplotyping, we showed that Asian cultivated rice accessions only have a fraction of HKT1;5 diversity available in progenitor wild rice species (O. nivara and O. rufipogon). Progenitor HKT1;5 haplotypes can thus be used as novel potential donors for enhancing cultivated rice salinity tolerance. Within Asian rice accessions, 10 non-synonymous HKT1;5 haplotypic groups occur. More HKT1;5 haplotypic diversities occur in cultivated indica gene pool compared to japonica. Predominant Haplotypes 2 and 10 occur in mutually exclusive japonica and indica groups, corresponding to haplotypes in O. sativa salt-sensitive and salt-tolerant landraces, respectively. This distinct haplotype partitioning may have originated in separate ancestral gene pools of indica and japonica, or from different haplotypes selected during domestication. Predominance of specific HKT1;5 haplotypes within the 3 000 rice dataset may relate to eco-physiological fitness in specific geo-climatic and/or edaphic contexts. 相似文献

It is imperative to choose some low cost, available and effective ameliorants to correct soil acidity in southern China for sustainable agriculture. The present investigation dealt with the possible role of industrial byproducts, i. e., coal fly ash (CFA), alkaline slag (AS), red mud (RM) and phosphogypsum (PG) in correcting acidity and aluminum (Al) toxicity of soils under tea plantation using an indoor incubation experiment. Results indicated that CFA, AS and RM increased soil pH, while PG decreased the pHs of an Ultisol and an Alfisol. The increment of soil pH followed the order of RM > AS > CFA. All the industrial byproducts invariably decreased exchangeable Al and hence increased exchangeable Ca, Mg, K and Na and effective cation exchange capacity. RM, AS and lime decreased total soluble Al, exchangeable Al and organically bound Al. Formation and retention of hydroxyl-Al polymers were the principal mechanism through which Al phytotoxicity was alleviated by application of these amendments. In addition, the heavy metal contents in the four industrial byproducts constituted a limited environmental hazard in a short time at the rates normally used in agriculture. Therefore, the short-term use of the byproducts, especially AS and RM, as amendments for soil acidity and Al toxicity in acid soils may be a potential alternative to the traditional use of mined gypsum and lime. 相似文献

ABSTRACTIn the present study, antioxidative potential of squid protein hydrolysates (SPH) prepared using endogenous pepsin and trypsin prepared from the stomach and pyloric ceca of seer fish respectively was evaluated, in comparison with commercial enzymes. Degree of hydrolysis (DH) of the pepsin SPH ranged between 8–11% and trypsin SPH between 12–14% after 150 min of hydrolysis. SPH prepared with endogenous enzymes had significantly higher DH than commercial enzymes. Pepsinogen SPH expressed more DH than pepsin SPH. Trypsin SPH exhibited good metal chelating ability (98%), which was equivalent to EDTA at 130 ppm. Pepsin SPH showed higher super oxide anion scavenging activity (75%). Trypsin and pepsin SPH also had high DPPH radical scavenging (85%), ABTS radical scavenging (93%) and ferric reducing (Abs700 = 1.2) abilities. Thus, metal chelating and superoxide anion radical scavenging activities of SPH are dependent on enzymes rather than DH. 相似文献

Dietary arginine requirement of fingerling hybrid Clarias (Clarias gariepinus×Clarias macrocephalus)
The dietary arginine requirement of fingerling hybrid Clarias (Clarias gariepinus×Clarias macrocephalus) (4.2±0.03 cm, 0.56±0.04 g) was determined by feeding six isonitrogenous (400 g kg−1 crude protein) and isocaloric (17.9 kJ g−1) amino acid test diets containing casein, gelatin and l ‐crystalline amino acids with graded levels of arginine (10.0, 12.5, 15.0, 17.5, 20.0 and 22.5 g kg−1) for 4 weeks to triplicate groups. Diets were fed twice a day at 09:00 and 16:00 hours at 8% body weight day−1. Maximum weight gain (523%), best feed conversion ratio (FCR, 1.41), protein efficiency ratio (1.78) and specific growth rate (6.53%) were recorded in fish fed the diet containing arginine at 20.0gkg−1 of the diet. Second‐degree polynomial regression analysis of live weight gain and FCR values indicated the dietary arginine requirement at 17.8 and 20.0 g kg−1 of dry diet respectively. Significantly higher carcass protein and protein deposition values were recorded at the requirement level (20.0 g kg−1). Higher fat and lower moisture values were obtained in carcass of fish fed the diet with 15.0g kg−1 arginine. The maximum carcass ash value was noticed in the fish fed at 20.0 g kg−1 dietary arginine. We recommend that the diet for hybrid Clarias (C. gariepinus×C. macrocephalus) should contain arginine in the range of 17.8–20.0 g kg−1 of the dry diet, corresponding to 44.5 and 50 g kg−1 of dietary protein respectively. 相似文献

Mangroves form an important ecosystem of Great Nicobar, a continental island in the Bay of Bengal with luxuriant tropical
rainforests. The rhizosphere of the mangrove plants of Great Nicobar was investigated for the presence of arbuscular mycorrhizal
fungus (AMF) and phosphate solubilising bacteria (PSB). The soils of the Great Nicobar mangroves were silt–clays and were
poor in phosphate content. Five species of AMF belonging to the genus Glomus were isolated. The %AMF colonization in the mangrove plants was between 0 and 17%, and the presence of AMF in the aerenchymatous
cortex suggests that the mangrove plants may be aiding in AMF survival by providing oxygen. Two strains of phosphate solubilising
Pseudomonas aeruginosa were found in the mangrove soils of Great Nicobar. Phosphate solubilisation by the two isolated strains was almost 70% under
in vitro conditions. PSB may play a role in the mangrove ecosystems of Great Nicobar by mobilising insoluble phosphate. The

Oscillometric blood pressure monitoring may be a practical tool for short procedures or those performed outside of the operating room. Oscillometric and direct blood pressure values in 30 juvenile and adult horses in a clinical setting using mixed effect models were compared. The limits of agreement and percentage errors were also calculated. We evaluated the sensitivity and false positive rate for the oscillometric method to trigger an intervention for treating blood pressure [direct mean arterial pressure (MAP) < 70 mmHg]. Oscillometric MAP and diastolic arterial pressure (DAP) differed from direct values (P < 0.001); systolic arterial pressure (SAP) did not (P = 0.08). Wide limits of agreement were observed. Percentage errors were smaller for SAP (39%) than for MAP and DAP (48% and 72%). The oscillometric monitor indicated there was a requirement for blood pressure treatment with a true positive rate of 82%, consequently, it failed 18% of the times. The false positive rate (unnecessary treatment) was 55%. 相似文献

A potentially diagnostic 18S rDNA (ribosomal DNA) gene was amplified reliably from red-listed ethnomedicinal species of Myristica and its wild and related genera. Individuals from nine species of Myristicaceae were utilized for the study. The sequences ranged from 1,767 to 1,794 nucleotide (nt) in length. The GC content (%) varied from 52.77 to 51.04. The frequencies (%) of nt were A (23.31), T (23.82), C (24.48) and G (28.39). The alignment of all sequences produced 195/1,516 variable sites and 1,257/1,516 conserved sites. Total numbers of single nucleotide polymorphism (SNP) sites found in the alignment were 146/1,516. Knema andamanica (Warb.) W.J. de Wilde was the most distinct that included 18 variable regions and 15 InDel with 27 SNP sites, specific to this species. The identified regions from nine species of Myristica and its wild and closely related genera were deposited in the GenBank Database (Accession numbers JN228257-JN228265). Comparison of morphological identifications and phylogenetic analysis indicated that the specimens were correctly assigned on the basis of a short stretch of 18S rDNA (~1,600 bp) making this a potentially useful marker for the rapid molecular assignment of an unknown related species also. Significant sequence homology ranging from 72 to 99 % was observed on comparison with 18S rDNA genes of other plants in the public domain. A comparison of intraspecific data information of nine 18S with that of 73 matK and 86 rbcL sequences from GenBank revealed that polymorphism, divergence and conservation is higher in 18S locus for Myristicaceae. Hence these markers may be utilized for phylogenetic analysis, evaluation of species richness during ecological surveys or for environmental assessments. These molecular markers are especially important due to the fact that the species studied are mostly vulnerable and red-listed with limited availability in endangered ecological niches. 相似文献

We examined the effects of fencing on deer browsing on seedlings 13 years after the building the experimental fence on Mt.
Ohdaigahara, central Japan, where the sika deer (Cervus nippon) population is high. There was no difference in the species number of seedlings between 1991 and 2004. The density of seedlings
in the fenced plot in 2004 was three times higher than in 1991. Although the unfenced plots had no seedlings higher than 20 cm,
there were seedlings up to 120 cm in the fenced plot. These results suggest that 13-year fencing promoted regeneration of
