The phylogenetic relationships of five isolates of Pasteurella multocida serotype B:2 belonging to buffalo, cattle, pig, sheep and goat were investigated by comparative sequence analysis of 16S rRNA gene. The 1468bp fragment of 16S rRNA gene sequence comparison showed that the isolates of cattle (PM75), pig (PM49) and sheep (PM82) shared 99.9% homology with the buffalo isolate (vaccine strain P52) whereas, the goat isolate (PM86) shared 99.8% homology with the vaccine strain. The 16S rRNA gene sequences of these isolates were also found monophyletic with type B reference strain NCTC 10323 of P. multocida subsp. multocida. The present study indicated the close relationships of haemorrhagic septicaemia causing P. multocida serotype B:2 isolates of buffalo and cattle with other uncommon hosts (pig, sheep and goat).  相似文献

Tillage modifies soil structure and has been suggested as a practice to improve physical, hydrological and chemical properties of compacted soils. But little is known about effect of long‐term tillage on physicochemical soil properties and crop yield on sodic soils in India. Our objective was to investigate the effect of different tillage regimes on crop yield (wheat and paddy rice) and physicochemical properties of sodic soils. Two sodic sites under conventional tillage for 5 (5‐YT; 5‐year tillage) and 9 (9‐YT; 9‐year tillage) years were selected for this study. Changes in crop yield and physicochemical soil properties were compared with a control, sodic land without any till history, that is, 0‐year tillage/untilled (0‐YT). Five replicated samples at 0‐ to 10‐cm and 10‐ to 20‐cm soils depths were analysed from each site. In the top, 0‐ to 10‐cm soil depth 5‐YT and 9‐YT sites had higher particle density (Pd), porosity, water holding capacity, hydraulic conductivity, organic carbon, total nitrogen (Nt), available nitrogen (Navail), phosphorus (Pavail) and exchangeable calcium (Exch. Ca++) than 0‐YT, whereas bulk density (Bd), C : N ratio and CaCO3 were significantly lower. Bd, pH, EC and CaCO3 increased significantly with depth in all the lands, whereas Pd, porosity, water holding capacity, hydraulic conductivity, organic carbon, Nt, Navail, Pavail and Exch. The bread wheat cultivar ‘Thatcher’ is documented to carry the gene Lr22b for adult‐plant resistance to leaf rust. Seedling‐resistance to leaf rust caused by Puccinia triticina in the bread wheat cultivar ‘Thatcher’, the background parent of the near‐isogenic lines for leaf rust resistance genes in wheat, is rare and no published information could be found on its genetic basis. The F2 and F3 analysis of the cross ‘Agra Local’ (susceptible) × ‘Thatcher’ showed that an apparently incompletely dominant gene conditioned seedling‐resistance in ‘Thatcher’ to the three ‘Thatcher’‐avirulent Indian leaf rust pathotypes – 0R8, 0R8‐1 and 0R9. Test of allelism revealed that this gene (temporarily designated LrKr1) was derived from ‘Kanred’, one of the parents of ‘Thatcher’. Absence of any susceptible F2 segregants in a ‘Thatcher’ × ‘Marquis’ cross confirmed that an additional gene (temporarily designated LrMq1) derived from ‘Marquis’, another parent of ‘Thatcher’, was effective against pathotype 0R9 alone. These two genes as well as a second gene in ‘Kanred’ (temporarily designated LrKr2), which was effective against all the three pathotypes, but has not been inherited by ‘Thatcher’, seem to be novel, undocumented leaf rust resistance genes.  相似文献

Summary The merits and limitations of somatic cell techniques involving Agrobacterium-mediated transformation, direct gene transfer and protoplast fusion, are discussed in relation to the genetic improvement of forage and grain legumes. Whilst progress with legumes is limited compared to that with plants of other families such as the Solanaceae, the fact that many legumes are readily amenable to tissue culture now permits somatic cell techniques to be targetted to these species. Future development of the subject will necessitate close collaboration between molecular biologists and plant breeders to enable novel plants generated by in vitro technologies to be incorporated into conventional breeding programmes.  相似文献

Cultivar ‘Thatcher’, and ‘Thatcher’ lines with Lr 21 and Lr 22 were studied against a number of races of Puccinia recondita for seedling and adult plant reaction. The study has established that Lr 21 and Lr 22 are genes effective against P. recondita at adult plant stage. It has also shown that these genes confer resistance against all races when plants are inoculated at boot leaf stage.  相似文献

Summary Two RAPD markers linked to gene for resistance (assayed as pustule number cm−2 leaf area) to rust [Uromyces fabae (Pers.) de Bary] in pea (Pisum sativum L.) were identified using a mapping population of 31 BC1F1 [HUVP 1 (HUVP 1 × FC 1] plants, FC 1 being the resistant parent. The analysis of genetics of rust resistance was based on the parents, F1, F2, BC1F1 and BC1F2 generations. Rust resistance in pea is of non-hypersensitive type; it appeared to be governed by a single partially dominant gene for which symbol Ruf is proposed. Further, this trait seems to be affected by some polygenes in addition to the proposed oligogene Ruf. A total of 614 decamer primers were used to survey the parental polymorphism with regard to DNA amplification by polymerase chain reaction. The primers that amplified polymorphic bands present in the resistant parent (FC 1) were used for bulked segregant analysis. Those markers that amplified consistently and differentially in the resistant and susceptible bulks were separately tested with the 31 BC1F1 individuals. Two RAPD makers, viz., SC10-82360 (primer, GCCGTGAAGT), and SCRI-711000 (primer, GTGGCGTAGT), flanking the rust resistance gene (Ruf) with a distance of 10.8 cM (0.097 rF and LOD of 5.05) and 24.5 cM (0.194 rF and a LOD of 2.72), respectively, were identified. These RAPD markers were not close enough to Ruf to allow a dependable maker-assisted selection for rust resistance. However, if the two makers flanking Ruf were used together, the effectiveness of MAS would be improved considerably.  相似文献

The gene Lr34 has contributed to durable resistance to leaf rust caused by Puccinia triticina in wheat worldwide. The closely associated leaf tip necrosis is generally used as the gene's marker. Lr34 has been postulated in many Indian bread wheat cultivars including ‘C 306’, based on the associated leaf tip necrosis and a few other field and glasshouse observations. The present study showed monogenic control of adult‐plant resistance in ‘C 306’ to leaf rust pathotype 77‐5 (121R63‐1). The F2 segregation in the crosses between ‘C 306’ and the two known carriers of Lr34, ‘Line 897’ and ‘Jupateco 73’‘R’ fitted a digenic ratio. The F3 families derived from the susceptible F2 segregants were true breeding for susceptibility, proving the absence of Lr34 in ‘C 306’. The cross between ‘Line 897’ and ‘Jupateco 73’‘R’ did not segregate for susceptibility. Resistance in the cross ‘Agra Local’ (susceptible) × ‘C 306’ was associated with leaf tip necrosis, showing that the leaf rust resistance gene in ‘C 306’ was associated with leaf tip necrosis, but was different from Lr34. This gene is being temporarily designated as Lr‘C 306’. Hence, leaf tip necrosis cannot be considered as an exclusive marker for selecting Lr34 in wheat improvement.  相似文献

Four seed-set groups of plants, viz. unselected ‘general’ and selected ‘high’, ‘medium’ and ‘low’ were constituted in C1 generation from randomly selected autotetraploids of two pea cultivars, T 163 and 5064-S. In C2 generation, ten plants were selected at random in each of the four groups. Two seeds each from selected autotetraploids were drawn and bulked to make 20 seeds in each of the four groups to raise C3 generation. Selection resulted in a gradual shift in seed-set, though the magnitude and nature varied with the group and generation. Improvement in the seed-set from C1 to C3 generation was associated with the increase in bivalents per cell, disjunction index and total chiasmata. Increase in total chiasmata was mostly due to an increase in proximal ones. Increase in seed-set was reflected in improvement of grain yield per plant.  相似文献

A Brassica juncea line carrying an introgression from Moricandia arvensis restored male fertility to two cytoplasmic male‐sterile (CMS) B. juncea lines carrying either M. arvensis or Diplotaxis catholica cytoplasm. Genetics of fertility restoration was studied in the F1, F2, F3 and backcross generations of the cross between CMS and fertility‐restorer lines. No male‐sterile plants were found in F1‐F3 generations of the cross between CMS [M. arvensis] B. juncea and the restorer. However, a 1: 1 segregation for male sterility and fertility was observed when the F1 was pollinated with non‐restorer pollen from a euplasmic line. These results clearly show that restoration is mono‐genic and gametophytic. In CMS lines carrying D. catholica cytoplasm, the restorer conferred male fertility to the F1 and showed 3: 1 and 1: 1 segregations for male fertility and sterility in F2 and BC1 generations, respectively, indicating a monogenic, sporophytic mode of fertility restoration. The results were also supported by pollen stainability in the F1 which was about 65% in M. arvensis‐based CMS and >90% in D. catholica‐based CMS. The above results are discussed in the light of previous molecular studies which showed association between CMS and atpA in both systems.  相似文献
