To cause rice blast disease, the fungus Magnaporthe oryzae develops a pressurized dome-shaped cell called an appressorium, which physically ruptures the leaf cuticle to gain entry to plant tissue. Here, we report that a toroidal F-actin network assembles in the appressorium by means of four septin guanosine triphosphatases, which polymerize into a dynamic, hetero-oligomeric ring. Septins scaffold F-actin, via the ezrin-radixin-moesin protein Tea1, and phosphatidylinositide interactions at the appressorium plasma membrane. The septin ring assembles in a Cdc42- and Chm1-dependent manner and forms a diffusion barrier to localize the inverse-bin-amphiphysin-RVS-domain protein Rvs167 and the Wiskott-Aldrich syndrome protein Las17 at the point of penetration. Septins thereby provide the cortical rigidity and membrane curvature necessary for protrusion of a rigid penetration peg to breach the leaf surface.  相似文献

The progressive morphohistopathologic changes, distribution pattern of lesions and ultrastructural characteristics in Eimeria arloingi infection were investigated in experimentally infected kids. The 18 newborn animals allocated to 3 equal groups. Two of groups, A, B were inoculated with a single dose of 1 × 103 and1 × 105 sporulated oocysts of E. arloingi, respectively. At 7, 14, 21, 28, 35, and 42 days postinoculation (DPI), 1 kid from each group was necropsied for pathologic and ultrastructural studies. Progressive lesions were present at 21, 28, 35 and 42 DPI in the jejunum, ileum, cecum with fewer in the duodenum and proximal colon. The oocysts shedding begin between 16 to 18 DPI. Grossly, minimal changes were observed at 21 DPI as few whitish plaques or nodules and advanced lesions at 42 DPI as pseudoadenomatous pattern in the mucosa and a cerebriform pattern on the serosal surface of jejunum and ileum. Early histopathologic lesions due to schizogony phase were including presence of intracytoplasmic developmental stages of the parasite such as trophozoites, immature to mature schizonts and mild infiltration of inflammatory cells. In late lesions due to various stages of gametogony, the histological pattern was mainly remarkable hyperplasia of the villi and crypts epithelial cells, eventually developed into papillary projections of reactive epithelium. The mesenteric lymph nodes showed a few numbers of large schizonts in the cortical lacteals. This study showed E. arloingi as a highly pathogenic species for kids, the incubation period was 16–18 days and the main target organ was jejunum with characteristic morphohistopathologic lesions.  相似文献

Background: The protein of Niemann-pick type C1 (NPC1) gene promotes the egress of cholesterol from late endosomes and lysosomes to other cellular compartments and contributes to a process known as reverse cholesterol transport. This study aimed to examine whether promoter methylation of NPC1 is associated with risk of cardiovascular disease (CVD). Methods: Fifty CVD patients and 50 healthy subjects as the control group were recruited in this study. Promoter methylation of NPC1 gene was defined using a nested-methylation specific polymerase chain reaction method. Statistical analyses were done using the chi-square, t-test or ANOVA tests. Results: Our study showed that the frequency of semi-methylated promoter (methylated/unmethylated status) was significantly higher in CVD patients than that in controls (OR = 6.521, 95% CI = 2.211-19.215, P = 0.008). However, a completely methylated promoter (methylated/methylated status) was not detected in any subjects in either of the two groups tested. Additionally, the analysis of clinical data according to the methylation status of NPC1 gene demonstrated that serum levels of total cholesterol, total triglycerides, high low-density lipoprotein cholesterol (LDL-C) and low high-density lipoprotein cholesterol (HDL-C) are influenced by NPC1 methylation, so that subjects with a completely unmethylated promoter (Unmethylated/unmethylated status) held lower levels of total triglycerides, total cholesterol, LDL-C and higher levels of HDL-C. Conclusion: Our findings propose that the NPC1 promoter methylation is a probable mechanism that can result in reduced/impaired NPC1 expression/activity and may thus contribute to progression of CVD. In this study, cotton/nylon blended fabrics were treated with atmospheric air plasma at various times (30–60 s) and were subsequently printed with pastes containing carbon black nanoparticles. Properties of plasma treated fabrics such as visible-near infrared (Vis-NIR) reflectance, water contact angle, air permeability, and color fastness were measured. It was shown that increasing plasma treatment time decreases reflection level of treated fabrics in Vis-NIR region. Plasma treatment also enhanced the hydrophobicity of cotton/nylon fabrics observed by an increase in water contact angle. Plasma treated samples for 60 s demonstrated lower air permeability than those treated for 30 s. Furthermore, printed samples possessed acceptable levels of fastness against washing, light and crocking.  相似文献

Plants are capable of reducing environmental pollutions through uptaking contaminants in their tissues. In the study site, twenty one-year-old leaves and shoots as well as twenty soil samples were sampled and analyzed by Inductively Coupled Plasma (ICP). We investigated the uptaking ability of lead (Pb) and cadmium (Cd) by the leaves and shoots of Platanus orientalis, Robinia pseudoacacia and Fraxinus rotundifolia in Karaj city, the western Tehran, Iran. We also evaluated the total metal accumulation capacity by using metal accumulation index (MAI). Results indicated that there was no significant difference in uptaking contents of Cd and Pb in the leaves among the trees. However accumulation of Cd in shoots of F. rotundifolia and R. pseudoacacia was significantly higher than that of P. orientalis. The accumulation of Pb in shoots of R. pseudoacacia was significantly higher than the other species. The amount of Pb in the soil of the study area was significantly higher than Cd. Concentrations of Cd and Pb in leaves of the three species are in the ranges of 2.4 2.7 mg Kg-1 and 7.1 14.4 mg Kg-1 , respectively. R. pseudoacacia had the highest MAI value for leaves (2.21) and F. rotundifolia had the highest MAI value for shoots (2.4).  相似文献

We selected 15 beech trees (Fagus orientalis) from the study area in a mountainous region from 520 to 1310 m above sea level. Ground observations of the beech tree growth process from January to December 2004 in 7- to 15-day intervals were performed both visually and by measuring leaf chlorophyll concentration (chlorophyll meter SPAD-502). Results for the regression analysis showed that the leaved period positively correlated with the air temperature (r?=?.894). Anthesis and SPAD measurements have a negative relationship with air temperature and as temperature increases they appear earlier in the year. Anthesis and SPAD measurements had correlation coefficients of r?=??.883 and r?=??.855, respectively. The anthesis and bud bursting and leaf tip appearance have a negative relationship with increasing altitude from sea level and as altitude increases they appear later in the year. To make phenological events in deciduous broadleaf forests recognizable, we used a seasonal 8-day composite Moderate Resolution Imaging Spectroradiometer/Normalized Difference Vegetation Index (MODIS/NDVI). Prior to leaf expansion, NDVI increased from the 109th day of the year or earlier. Existence of the evergreen plants (Ruscus hyrcanus) on the ground and snow-melting exert an influence on inconsistency of phenology monitoring of MODIS/NDVI images. Regression analysis showed that there is a positive relationship between NDVI and SPAD measurements (chlorophyll content) in beech trees.  相似文献

Microbial indices and their spatial patterns are strongly affected by environmental factors. Spatial variability of soil properties is one of the most important causes of variability in soil microbial indices. This research was conducted in the Caspian forest to assess spatial variabilities and frequency distributions of microbial properties.Ninety soil samples were taken using a grid sampling design 40 9 40 m. Microbial indices, organic carbon,nitrogen and pH were determined. Soil variable distributions showed that microbial indices had abnormal distributions. Logarithmic transformation produced normal distribution. Spatial continuity using geostatistical(variogram) was studied and maps obtained by point kriging.The variograms revealed the presence of spatial autocorrelation. The results indicate that spatial dependence of soil microbial indices was affected by non-intrinsic factors and forest management procedures. The maps show that soil microbial indices and soil properties have spatial variability. The spatial pattern of microbial indices was correlated to organic carbon and nitrogen.  相似文献

Oil from seeds of Diospyros lotus was extracted using a conventional method with two different solvents:hexane and petroleum ether. A central composite design with response surface methodology were used to optimize the process. A second-order polynomial equation was employed, and ANOVA was applied to evaluate the impact of various operating parameters including extraction temperature(x_1; 44.9–70.1 °C), extraction time(x_2;5.0–10.0 h) and solvent to solid ratio(x_3;11.6–28.4 mL g-1), on oil yield. Experiments to validate the model showed decent conformity between predicted and actual values. Extraction conditions for optimal oil yield were 61 °C, 8.75 h extraction duration and 19.25 mL g-1 solvent to solid ratio. Under these conditions, the oil yield was predicted to be 5.1340%. Oil samples obtained were then analyzed using gas chromatography. The fatty acid composition revealed the major fatty acids to be oleic acid(C18:1) and linoleic acid(C18:2). The analysis of oil also demonstrated a decent ratio between omega-3 and omega-6 fatty acids. The structure of seeds was imaged using scanning electron microscopy. Oil quality was analyzed thermogravimetrically and by Fourier transform infrared spectroscopy. The assigned nutritional features of the D. lotus oil suggested that it can be used as an edible oil in pharmaceutical and food industry in the future.  相似文献

The introgression of wild chromosomal segments into popular rice varieties is one of the potential approaches for developing varieties for drought stress condition. Sixteen genotypes, including nine indica, two tropical japonica and five chromosome segments substitution lines(CSSLs) with different levels of tolerance/susceptibility to drought stress, were selected for diversity study. Sixty-three microsatellite markers were utilized for assessing genetic diversity. A total of 95 alleles were amplified, and out of them, 60 were polymorphic. Six unique alleles, amplified by the microsatellite loci RM276, RM472,RM488, RM537, RM541 and RM28089, were identified in six genotypes, namely FR13A, Brahamanakhi,RUF44, Swarna-sub1, Brahamanakhi and Satyabhama. The highest genetic similarity was found among CSSLs. Polymorphism information content(PIC) value varied from 0 to 1.00 with an average of 0.66 per locus. Twenty-eight microsatellites were found to be polymorphic, which could be used in marker-assisted selection programme. All the sixteen genotypes were grouped into two major clusters at genetic similarity of 0.64. In the cluster I, five CSSLs identified as diverse genotypes had wild ancestor segments responsible for drought tolerance, and hence they could be utilized as potential donors. The popular Indian varieties, Swarna-sub1 and IR64-sub1, could be used as recurrent parents in the future breeding program for developing varieties for abiotic stresses such as submergence and drought.  相似文献
