Although methicillin-resistant Staphylococcus aureus (MRSA) were generally isolated from human beings; these agents were recently isolated from various animal species. It has been shown that MRSA isolates are not only resistant to beta-lactam antibiotics, but can also be resistant to the other commonly used antibiotics. In this study, 18 phenotypic methicillin resistant S. aureus isolates from bovine mastitis cases were analyzed by PCR for the presence of mecA gene encoding methicillin resistance and aac(6′)/aph(2″), aph(3′)-IIIa and ant(4′)-Ia genes encoding aminoglycoside resistance. Out of 18 S. aureus isolates (oxacillin MICs, ≥4 μg/ml), 3 were positive for mecA gene. Only one from 3 mecA positive isolates was positive for genes encoding aminoglycoside-modifying enzymes and this isolate carried aac(6′)/aph(2″) in combination with aph(3′)-IIIa gene. The aph(3′)-IIIa gene was detected in 3 isolates. These three isolates carrying the aminoglycoside-modifying enzyme genes were resistant to gentamicin, kanamycin and neomycin. The mecA gene of 3 MRSA isolates was sequenced. All three mecA genes of these isolates were identical to that found in human MRSA strains, except a one-base substitution at nucleotide position 757. From the data presented in this study, it can be concluded that MRSA isolated from bovine mastitis may be originated from human beings, but further studies are needed to investigate the possibility of zoonotic transfer of MRSA.  相似文献

High zinc (Zn) concentration of seeds has beneficial effects both on seed vigor and human nutrition. This study investigated the effect of Zn biofortification on growth of young durum wheat (Triticum durum cv. Yelken) seedlings under varied Zn and water supply. The seeds differing in Zn concentrations were obtained by spraying ZnSO4 to durum wheat plants at different rates under field conditions. Three groups of seeds were obtained with the following Zn concentrations: 9, 20, and 50 mg Zn kg?1. The seeds differing in Zn were tested for germination rate, seedling height, shoot dry matter production, and shoot Zn concentration under limited and well irrigated conditions in a Zn‐deficient soil with and without Zn application. In an additional experiment carried out in solution culture, root and shoot growth and superoxide dismutase activity (SOD) of seedlings were studied under low and adequate Zn supply. Low seed Zn concentration resulted in significant decreases in seedling height both in Zn‐deficient and sufficient soil, but more clearly under water‐limited soil condition. Decrease in seed germination due to low seed Zn was also more evident under limited water supply. Increasing seed Zn concentration significantly restored impairments in seedling development. Drought‐induced decrease in seedling growth at a given seed Zn concentration was much higher when soil was Zn‐deficient. Increasing seed Zn concentration also significantly improved SOD activity in seedlings grown under low Zn supply, but not under adequate Zn supply. The results suggest that using Zn‐biofortified seeds assures better seed vigor and seedling growth, particularly when Zn and water are limited in the growth medium. The role of a higher antioxidative potential (i.e., higher SOD activity) is discussed as a possible major factor in better germination and development of seedlings resulting from Zn‐biofortified seeds.  相似文献

The effects of organic versus conventional crop management practices (crop rotation, crop protection, and fertility management strategies) on wheat yields and grain metal (Al, Cd, Cu, Ni, Pb, and Zn) concentrations were investigated in a long-term field trial. The interactions between crop management practices and the season that the crop was grown were investigated using univariate and redundancy analysis approaches. Grain yields were highest where conventional fertility management and crop protection practices were used, but growing wheat after a previous crop of grass/clover was shown to partially compensate for yield reductions due to the use of organic fertility management. All metals except for Pb were significantly affected by crop management practices and the year that the wheat was grown. Grain Cd and Cu levels were higher on average when conventional fertility management practices were used. Al and Cu were higher on average when conventional crop protection practices were used. The results demonstrate that there is potential to manage metal concentrations in the diet by adopting specific crop management practices shown to affect crop uptake of metals.  相似文献

This paper introduces a new sampling design in a finite population setting, where potential sampling units have a wealth of auxiliary information that can be used to rank them into partially ordered sets. The proposed sampling design selects a set of sampling units. These units are judgment ranked without measurement by using available auxiliary information. The ranking process allows ties among ranks whenever units cannot be ranked accurately with high confidence. The ranking information from all sources is combined in a meaningful way to construct strength-of-agreement weights. These weights are then used to select a single sampling unit for full measurement in each set. Three different levels of sampling design, level-0, level-1, and level-2, are investigated. They differ in their replacement policies. Level-0 sampling designs construct the sample by sampling with replacement, level-1 sampling designs constructs the sample without replacement of the fully measured unit in each set, and level-2 sampling designs construct the sample without replacement on the entire set. For these three designs, we estimate the first and second order inclusion probabilities and construct estimators for the population total and mean. We develop a bootstrap resampling procedure to estimate the variances of the estimators and to construct percentile confidence intervals for the population mean and total. We show that the new sampling designs provide a substantial amount of efficiency gain over their competitor designs in the literature.  相似文献

In this study, effects of lyophilization on the functional properties of acid modified and autoclaved corn starch preparations were investigated. RS contents and pasting properties of these starch preparations were also determined. Significant increases in solubility were observed as the hydrolysis level of the lyophilized samples increased. All of the acid-modified gelatinized–autoclaved–lyophilized samples had higher water binding values than those of native starch and heat treated oven-dried native starch. Acid-modified gelatinized–autoclaved–lyophilized samples (with storage at 95°C: GASL or without storage: GAL, before lyophilization) improved emulsion properties of soy protein solution significantly. Acid modification seems to be a prerequisite to achieve improving effect of lyophilization. While native starch did not contain any RS, the level increased to 8.1% due to gelatinization, autoclaving and oven-drying (Control 2). The RS content of Control 2 was higher than that of gelatinized–autoclaved–lyophilized native starch (N-GAL, 2.9%). The samples stored prior to lyophilization had higher RS contents as compared to the corresponding unstored samples. The highest RS content (12.4%) was observed in 2h-GASL sample. Cold viscosity was observed in the RVA curves of N-GASL, 2h-GASL, N-GAL and 2h-GAL samples. A possible mechanism/model is suggested to explain the lack or existence of a cold viscosity.  相似文献

The objective of the present work was to evaluate the effects of three different rates of long chain n-3 polyunsaturated fatty acid (LC n-3 PUFA) on the sperm number of Astacus leptodactylus. The experiment was carried out with the four following treatments: control group (not supplemented LC n-3 PUFA), D1 (1 %), D2 (2 %) and D3 (3 %). The sperm number of A. leptodactylus was counted; the reproductive system was weighted; and the macroscopical analysis of the testes and vasa deferentia (DVD) were conducted. In addition, gonado-somatic index (GI) and testes index (TI) were also calculated. The results showed that there was a significant difference between the crayfish fed LC n-3 PUFA supplemented diets (D1, D2, D3) and control in mean sperm number (P < 0.05). The highest sperm number was found for the crayfish fed D3 (6.27 ± 0.14 × 108 sperm/distal vas deferens section). The results also showed that the reproductive system of the crayfish fed D3 and D2 was significantly heavier than that of the crayfish fed the control and D1 (P < 0.05). On the other hand, there was not a significant difference in the testes weight and vasa deferentia weight between the crayfish fed the control and D1, and between the crayfish fed D2 and D3 (P < 0.05). GI of the crayfish fed D3 and D2 was significantly higher than that of the crayfish fed control and D1 (P > 0.05), but there was not a significant difference in the TI of the crayfish fed control, D1, D2 and D3 (P > 0.05). In conclusion, diets containing different levels of LC n-3 PUFA significantly increase the sperm number of A. leptodactylus, and they are crucial for the production of higher number of sperm in this species.  相似文献

Buyukcekmece Reservoir, located in the western outskirts of Istanbul, is one of the major water resources of Istanbul, and supplies drinking water to about 4 million people. Erosion in the catchment of the reservoir is an important problem in terms of its longer-term sustainability for water supply. There is an urgent need to obtain reliable quantitative data regarding erosion and deposition rates within the catchment to assess the magnitude of the problem and to plan catchment management strategies. In the absence of existing data, attention has focussed on the potential for using 137Cs measurements to provide retrospective estimates of medium-term soil erosion rates within the catchment over the past ca. 40 years. To date, the 137Cs approach has not been used to document soil redistribution rates in Turkey and this contribution reports an attempt to confirm the viability of the approach and the results of a preliminary investigation of rates of soil loss from uncultivated areas within the catchment. The soil redistribution rates estimated using the profile distribution conversion model varied from − 16.11 (erosion) to 4.59 (deposition) t/ha/year.  相似文献

Nineteen goslings with pulmonary and systemic aspergillosis were the subject of the study. The lungs and air sacs were the main sites affected by the disease, and were generally characterised by diffuse yellowish-white granulomas. In 7 cases with pulmonary and air-sac involvement the granulomas were scattered to the serosal linings of the gastrointestinal and upper respiratory tracts, to the liver, spleen and kidneys, and in two cases also to the bursa of Fabricius, musculus (m.) longus colli and adventitia of aorta. The granulomas were often characterised by a necrotic centre surrounded by heterophils, macrophages, lymphocyte and plasma cells, and in late granulomas by multinucleated foreign-body giant cells, and again by an outer thin fibrous capsule. Numerous fungal hyphae were found within the necrotic debris of the granulomas by Gridley and PAS staining techniques. Immunohistochemistry reliably confirmed aspergillosis in all of the cases. Fungal elements in the lungs of goslings severely affected by the disease stained heavily within the centre of the granulomas, whereas few antigens reacted in the chronic cases. Fungal fragments, which were not discernible using routine fungal stains, reacted clearly in the cytoplasm of macrophages and giant cells. Thus, although fungal elements within the granulomas were histologically indicative of aspergillosis, immunohistochemistry also had to be applied to obtain a definitive diagnosis of the disease and to differentiate it from many of the filamentous fungi.  相似文献

Pectinolytic bacteria were isolated from 48 potato plants showing the symptoms of blackleg and collected in different fields of commercial potato production areas at Samsun, Amasya, Corum and Yozgat provinces in Turkey in 2015. The survey resulted in the isolation of 26 pectinolytic strains that belonged to P. atrosepticum, P. carotovorum subsp. brasiliense, P. carotovorum subsp. carotovorum and P. parmentieri species based on molecular identification with species-specific PCR and phenotypic characterization. The identified strains indicated typical biochemical characteristics of the assigned species. For 16 representative Pectobacterium isolates 10 unique rep-PCR band patterns were obtained. The 16S rRNA and recA and gapA gene fragment sequencing confirmed the species identity of the isolates. The phenotypic characterization of the strains revealed that for all assays but one (cellulase, protease activity, swimming but not swarming), the tested Pectobacterium species were significantly different from each other proving the diversity of the strains belonging to these genera. Recent outbreaks of blackleg and/or soft rot in potato production areas in Turkey may pose a threat on other crops, as tomato, pepper, cucumber, onion, cabbage, broccoli and sugar beet are cultivated in the same provinces.  相似文献

Evidence clearly shows that cationic micronutrients in spray solutions reduce the herbicidal effectiveness of glyphosate for weed control due to the formation of metal-glyphosate complexes. The formation of these glyphosate-metal complexes in plant tissue may also impair micronutrient nutrition of nontarget plants when exposed to glyphosate drift or glyphosate residues in soil. In the present study, the effects of simulated glyphosate drift on plant growth and uptake, translocation, and accumulation (tissue concentration) of iron (Fe), manganese (Mn), zinc (Zn), and copper (Cu) were investigated in sunflower (Helianthus annuus L.) plants grown in nutrient solution under controlled environmental conditions. Glyphosate was sprayed on plant shoots at different rates between 1.25 and 6.0% of the recommended dosage (i.e., 0.39 and 1.89 mM glyphosate isopropylamine salt). Glyphosate applications significantly decreased root and shoot dry matter production and chlorophyll concentrations of young leaves and shoot tips. The basal parts of the youngest leaves and shoot tips were severely chlorotic. These effects became apparent within 48 h after the glyphosate spray. Glyphosate also caused substantial decreases in leaf concentration of Fe and Mn while the concentration of Zn and Cu was less affected. In short-term uptake experiments with radiolabeled Fe (59Fe), Mn (54Mn), and Zn (65Zn), root uptake of 59Fe and 54Mn was significantly reduced in 12 and 24 h after application of 6% of the recommended dosage of glyphosate, respectively. Glyphosate resulted in almost complete inhibition of root-to-shoot translocation of 59Fe within 12 h and 54Mn within 24 h after application. These results suggest that glyphosate residues or drift may result in severe impairments in Fe and Mn nutrition of nontarget plants, possibly due to the formation of poorly soluble glyphosate-metal complexes in plant tissues and/or rhizosphere interactions.  相似文献
