Dipeptidyl peptidase I (DPP I; EC 3.4.14.1) was purified from porcine skeletal muscle after several steps such as heat treatment, ammonium sulfate fractionation, gel filtration chromatography, and HPLC anion exchange chromatography. The purified enzyme showed a native molecular mass of approximately 200 kDa on Sephacryl S-200 column chromatography. Two protein bands of 65 and 42 kDa were obtained by SDS-PAGE, indicating its oligomeric nature. Maximum activity was reached at pH 5.5 and 55 degrees C. DPP I shared some common substrate specificities, both on synthetic derivatives and on real peptides, with porcine muscle DPP III. The enzyme required reducing agents for full activation, although the halide requirement was not proved. DPP I was inhibited by the assayed cysteine peptidase inhibitors except p-CMB. The serine peptidase inhibitor 3, 4-DCI also inhibited the enzyme as did the divalent cations Co(2+), Mn(2+), and Zn(2+). On the basis of its properties, DPP I may contribute to the generation of dipeptides during the processing of meat and/or meat products, including cooked ham.  相似文献

Abstract

The material balance of all the chemical species associated with the phosphate sorption reaction by two amorphous clays, silica-alumina gel and synthetic goethite was investigated adjusting the initial pH to 4.0, 5.0, 6.0, and 7.0. Phosphate sorption was found to decrease with increasing initial pH. During phosphate sorption, the equilibrium pH rose, adsorption of Cl- decreased to a state of negative adsorption, adsorption of Na+ increased markedly, silicate was released, and Al in the solution was precipitated. With increasing initial pH, the amount of apparent released OH- increased, the amount of Na+ adsorption increased, and the amount of released silicate decreased. The overall relationship of the material balance is explained in terms of the charge balance associated with phosphate sorption.  相似文献

Unlike mercury, which has an extensive +1 oxidation state chemistry, zinc usually adopts the +2 oxidation state. Decamethyldizincocene, Zn2(eta5-C5Me5)2, an organometallic compound of Zn(I) formally derived from the dimetallic [Zn-Zn]2+ unit, has been isolated from the low-temperature (-10 degrees C) reaction of Zn(C5Me5)2 and Zn(C2H5)2 in diethyl ether. X-ray studies show that it contains two eclipsed Zn(eta5-C5Me5) fragments with a Zn-Zn distance (+/- standard deviation) of 2.305(+/-3) angstroms, indicative of a metal-metal bonding interaction.  相似文献

A sample of 58 natural ryegrass populations (Lolium perenne L.) from the French and Spanish oceanic coast was evaluated for three seasonal growth traits (i.e. spring, autumn and winter vigour traits) in 5 evaluation sites (three in France and two in Spain). This sample showed a high level of genotype (population) × environment (evaluation sites) interactions for the three agronomic traits. A factor regression analysis, using both isozyme frequencies of populations and climatic factors of evaluation sites as covariates, was carried out on a sub-sample of 30 populations in order to explain these interactions. This method succeeded in explaining most population × location interactions by the product of two covariates. For instance, for spring vigour trait, 72.8% of the interaction term could be explained by the use of two covariates: the PGI2-20 allelic frequency of populations and the minimum temperature of the coldest month of evaluation sites. This study shows the advantage of such a method for plant breeders who need to identify promising ryegrass populations for their breeding objectives. Internal migration has been recognized as the major influence in terms of population redistribution across urban systems, but it is not a homogeneous phenomenon. Within the context of internal rural‐urban migration decline and the negative changes in migratory balances in the metropolitan area of Mexico City, the core of enquiry in this paper is the approach to growth and consolidation of an internal urban–urban migration system in the early twenty‐first century (2000–2015). This process has taken place through two main networks, among metropolitan areas not corresponding to the principal city and among intermediate cities. Internal migration is a complex process that involves both individual and spatial characteristics and which leads to spatially uneven development in the long term. Data from three censuses of the population of Mexico (2000, 2010, and 2015) show a transition to a more urban–urban migration pattern, with skilled migrants tending to have metropolitan and urban destinations, whereas less‐skilled migrants prefer rural and small urban destinations.  相似文献

Efforts to manage National Forests in the USA for wood production, while protecting water quality, are currently constrained by models that do not address the temporal dynamics of variable non-point source (NPS) areas. NPS areas are diffuse sources of contaminants contributed mostly by runoff as a result of different land use activities. Riparian vegetative buffers are often used to control contaminants from NPS areas but defining suitable widths require different policy considerations. In this study, the approach for defining suitable buffer widths is to apply a distributed process-based model that predicts potential NPS areas prone to generating runoff in relation to overland flow distances. A case study of the concept was applied to the 72 km2 Pete King watershed located in the Clearwater National Forest (CNF) in central Idaho, USA. This grid modeling approach is based on a Geographic Information System (GIS) and it integrates the soil moisture routing (SMR) model with probabilistic analysis. The SMR model is a daily water balance model that simulates the hydrology of forested watersheds using real or stochastically generated climate data, a digital elevation model, soil, and land use data. The probabilistic analysis incorporates the variability of soil depth and accounts for uncertainties associated with the prediction of NPS areas using Monte Carlo simulation. A 1-year simulation for the case study location was performed to examine the spatial and temporal changes in NPS areas prone to generating runoff. The results of the simulation indicate that the seasonal variability of saturated areas determines the spatial dynamics of the potential NPS pollution. Use of this model for the design of riparian buffer widths would increase the effectiveness of decision-making in forest management and planning by mapping or delineating NPS areas likely to transport contaminants to perennial surface water bodies.  相似文献

The current characteristics in the global economy have defined “knowledge-intensive services” (KIS) as part of the so-called “industries of the future” due to the attributes of their employment and central function in the development processes in science, technology, art, and culture. This study outlines as a central hypothesis that the localization of KIS depends on different factors according to the type of knowledge being developed (analytical, synthetic, and symbolic). The main objective in this work is to explore the spatial economic factors associated with the localization and changes (positive and negative) when using KIS in the “national urban system” in México between 2004 and 2014. To do so, the services sector is defined according to the “intensity” and the “types” of knowledge that they develop. An analysis of spatial econometrics is carried out that distinguishes between the geographic (local) effect and the spatial (non-local) effect of the independent variables using data and microdata from the period 2004 to 2014. The results partly correspond with the central hypothesis of the study and offer possibilities of public policies to take advantage of and to promote the benefits of KIS, especially regarding cities in developing countries looking to become part of the 21st century economy.  相似文献

Pepper huasteco yellow vein virus (PHYVV) is the main virus of pepper crop in Mexico. No resistant cultivars are available and resistance breeding is hampered by the lack of knowledge of heritability (h 2) of PHYVV resistance. This is a continuation of previous studies and the objectives were to analyze the h 2 and the behavior of the resistant trait to PHYVV. Four resistant assays were done with three resistant wild lines (UAS12, UAS13 and UAS10) of Capsicum annuum in the S4, S5, S6 and S7 generation under greenhouse conditions. Plants from all tests were inoculated with PHYVV through Bemisia tabaci. Line UAS12 was the most resistant showing a significantly proportion of resistant plants, less disease symptoms and longer incubation time, followed by the lines UAS13 and UAS10 in all assays. Distribution of symptoms showed a bimodal tendency in all the trials, suggesting that two groups of genes are involved in this resistance trait. The lines UAS12, UAS13 and UAS10 showed the same pattern of response to selection with an average of h 2 of 0.17, 0.06, 0.02 and 0.00 in the S4, S5, S6 and S7, respectively. These results indicate that all lines responded positively to the selection in the S4, S5 and S6, whereas in the S7 there was no response by the possible exhaustion of variation. Line UAS12 is the most promising genotype and the lines UAS13 and UAS10 are genetic resources that can be supplemented to breed the resistance of PHYVV. These results provides basic information for resistance breeding.  相似文献

The whitefly-transmitted Tomato chlorosis virus (ToCV) (genus Crinivirus) is associated with yield and quality losses in field and greenhouse-grown tomatoes (Solanum lycopersicum) in South America. Therefore, the search for sources of ToCV resistance/tolerance is a major breeding priority for this region. A germplasm of 33 Solanum (Lycopersicon) accessions (comprising cultivated and wild species) was evaluated for ToCV reaction in multi-year assays conducted under natural and experimental whitefly vector exposure in Uruguay and Brazil. Reaction to ToCV was assessed employing a symptom severity scale and systemic virus infection was evaluated via RT-PCR and/or molecular hybridization assays. A subgroup of accessions was also evaluated for whitefly reaction in two free-choice bioassays carried out in Uruguay (with Trialeurodes vaporariorum) and Brazil (with Bemisia tabaci Middle-East-Asia-Minor1—MEAM1?=?biotype B). The most stable sources of ToCV tolerance were identified in Solanum habrochaites PI 127827 (mild symptoms and low viral titers) and S. lycopersicum ‘LT05’ (mild symptoms but with high viral titers). These two accessions were efficiently colonized by both whitefly species, thus excluding the potential involvement of vector-resistance mechanisms. Other promising breeding sources were Solanum peruvianum (sensu lato) ‘CGO 6711’ (mild symptoms and low virus titers), Solanum chilense LA1967 (mild symptoms, but with high levels of B. tabaci MEAM1 oviposition) and Solanum pennellii LA0716 (intermediate symptoms and low level of B. tabaci MEAM1 oviposition). Additional studies are necessary to elucidate the genetic basis of the tolerance/resistance identified in this set of Solanum (Lycopersicon) accessions.  相似文献

With a view to the selection of plants for the re-vegetation of contaminated, semi-arid land, two populations of the perennial species Bituminaria bituminosa (Fabaceae) from the south of Spain were studied: one (“LA”) from a non-contaminated soil and the other (“C2”) from a similar soil having elevated total levels of Pb and Zn (1,112 and 4,249 μg g?1, respectively). For sand-cultured plants receiving nutrient solution, flow cytometry showed that heavy metals, at the concentrations measured in aqueous extracts from contaminated soils, had only slight genotoxic effects on root tip cell nuclei. Both populations were also grown in both soils, in two pot experiments. In the first, shoot biomass of LA and C2 in the contaminated soil was decreased to similar extents, with respect to the “clean” soil. Tissue heavy metal concentrations were unlikely to have been phytotoxic, except in the case of shoot Zn for population LA, but there were tissue deficiencies of P and K for populations LA and C2, respectively. In the second pot assay, the stimulation of growth by NPK fertiliser confirmed that even though this soil had high total heavy metal levels, nutrient availability was the principal factor limiting growth. The lesser transport of heavy metals (Cd, Mn and Zn) to the shoot by the population from the contaminated site is a factor that should be considered when selecting B. bituminosa lines for the phytostabilisation of such sites.  相似文献
