In 1998, the Organic Arable Farming Experiment Gladbacherhof (OAFEG) was started in order to explore the impact of different organic arable production systems (mixed farming, stockless farming with rotational ley, stockless cash crop farming) and of different tillage intensities (conventional plough as a full inversion tillage, two-layer plough, inversion tillage at reduced depth, non-inversion tillage) on sustainability parameters. In this article, we present results on the development of soil organic matter (SOM) levels. Starting with organic mixed farming with approximately 0.7 livestock units (LU) per ha cattle before set-up of the experiment, only the mixed farming system in the experiment was able to maintain SOM levels. The stockless system with ley maintained soil organic carbon (SOC), but lost soil total nitrogen (STN), and the stockless cash crop system had a significant SOM loss in the magnitude of 7.7 t SOM ha?1, or roughly 8.4% of the initial SOM mass. Reducing tillage intensity had no impact on SOM masses, but only on organic matter stratification in soils. We conclude that specialization of organic farms towards stockless arable crop production requires special attention on SOM reproduction to avoid detrimental effects. Further, reduced tillage intensity does not necessarily have a positive effect on SOM.  相似文献

Chromatographic, chemical, and spectroscopic techniques were used to characterize the physicochemical properties of napin purified by preparative chromatography. The molar extinction coefficient was determined (epsilon = 0.56), and static and dynamic light scattering measurements enabled the average molecular weight (M(w) = 13919), the second virial coefficient (A(2) = 23.95 x 10(-)(5) mol cm(3) g(-)(2)), and the hydrodynamic radius (R(H) = 1.98 nm) to be determined. No conformational changes were observed by fluorescence and circular dichroism measurements in different buffers at pH 3, 4.6, 7, and 12, confirming the high pH stability of this protein. From MALDI-TOF analysis and after enzymatic digestion, it was found that this purified sample, extracted from the rapeseed variety Express, contained mainly isoform 2SS3_BRANA.  相似文献

Summary To evaluate the role of plasmids in soil communities antibiotic-resistant bacteria have been isolated from soil. Among them, 419 l-aminopeptidase positive strains (Gram-negative) and 28 l-aminopeptidase negative strains (Gram-positive) were screened for the presence of plasmids. None of the Gram-negative organisms contained plasmids. Among the Gram-positive bacteria plasmid-harboring strains were detected.  相似文献

This study was conducted to assess the abundance of iron-oxidizing bacteria and biological sulfur oxidation potential from soil impacted by coal and coal refuse from two coal-burning electric power facilities located at the U.S. Department of Energy's Savannah River Site (Aiken, S.C.) and the South Carolina Electric and Gas Site at Beech Island, S. C. Significantly higher MPN counts of iron-oxidizing bacteria were obtained from samples collected at the confluence of a coal storage runoff containment basin, a coal reject (refuse) pile, and an adjacent wetland at the Savannah River Site. Significant differences in pH, sulfate-S, ferrous- and ferric-iron were also obtained between sampling locations. No significant differences in ferric/ferrous ratios were determined. These ratios however, exceeded a value of 2.0 when sample pH values were less than 4.5. Under optimal conditions, biological thiosulfate-S oxidation potentials (in vitro) showed a 4- to 7-day lag in the appearance of sulfate-S, and a final pH (after twenty-four days of perfusion) of 1.97 to 3.90. These results indicate that contamination of subsurface water by acidic leachate derived from thionic bacterial activity will occur if coal and coal refuse piles are not confined by an impermeable surface or containment facility.  相似文献

Available evidence shows that sweetpotato originated from either Central or South American lowlands with subsequent dispersal to North America, Europe, Africa, Asia and the pacific islands. A total of 71 polymorphic RAPD molecular markers were used to assess the genetic relationships amongst 74 sweetpotato varieties originating from a total of 23 sweetpotato producing countries within six geographical regions, namely, South America, Central America/Caribbean, United States of America (USA), East Africa, Asia and Oceania. An Analysis of Molecular Variance (AMOVA) indicated that 93.4% of the total variance was due to the differences between genotypes within regions. The difference between regions was significant (P < 0.001) but only contributed 6.6% to the variance. Genetic distance (PhiST) calculated with AMOVA and multidimensional scaling (MDS) revealed that the South American and the Central American/Caribbean genotypes formed two separate clusters. East African varieties, which have unique characteristics from other traditional varieties, were distinct from other traditional varieties from South America and Oceania. These results support the reported hypothesis of the origin and dispersal of the sweetpotato and indicate that the primary centre of diversity probably has two distinct genepools. It is proposed that the dispersal of the sweetpotato from its origin may have mainly involved varieties from Central America/Caribbean as opposed to varieties from South America. There is an indication that new genepools may be evolving in Africa and Asia due to hybridisation and adaptation to the local environments.  相似文献

We studied the influence of an immobilized laccase from Trametes versicolor on non-extractable residue (NER) formation of the systemic fungicide 14C-metalaxyl in soil. We added the enzyme (130 mU/g DW) to soil sterilized by gamma irradiation and observed that the amount of NER (6.3 % of applied radioactivity) after 10 days of incubation was enhanced about twofold compared to the sterile soil without laccase addition. Residues formed within samples without enhanced enzyme activity were mainly bound via ester linkages to all fractions of humic matter, i.e., fulvic acids, humic acids, non-humines, and humines, respectively. In contrast, residues formed in presence of immobilized laccase were more strongly bound by covalent linkages such as ether and C-C bonds, especially to humic acids. After chemical degradation of the humic matter, it could be observed that all NER contained the first major transformation product, i.e., metalaxyl acid. The findings underline that the residue formation of metalaxyl in soil may be partly catalyzed by immobilized extracellular oxidative enzymes through oxidative coupling reactions within the humic matter.  相似文献

Purpose

Flood events are expected to increase both in intensity and frequency due to climate change in the near future. From an environmental toxicology perspective, there is concern that such flood events could lead to the remobilization of contaminated sediment layers in rivers. The aim of this pilot study was to establish a novel and interdisciplinary framework combining methods of hydrodynamic engineering and ecotoxicological assessment to enable investigation of the potential risks associated with such remobilization events.

Materials and methods

Formulated sediment was prepared according to OECD guideline 218 and spiked with a mixture of four polycyclic aromatic hydrocarbons (phenanthrene, chrysene, pyrene, benzo[a]pyrene) at concentrations of 3.3?C8.3?mg?kg?1 dry weight. Rainbow trout (Oncorhynchus mykiss) were exposed as test animals to re-suspended sediments in three out of five experiments. The experiments were carried out in an annular flume designed to investigate transport behaviour of fine-grained sediments. Several physicochemical (e.g. pH) and sedimentological (e.g. turbidity) parameters were measured to characterise environmental conditions and erosion behaviour of sediments. Furthermore, exposure concentrations were measured by means of an in vitro assay (7-ethoxyresorufin-o-deethylase (EROD), RTL-W1 cell assay) and chemical analysis.

Results and discussion

Preparation and spiking of large amounts of formulated sediments were feasible but not practical. Successful spiking could be confirmed by the bioanalytical methods with the spiked sediments showing significantly elevated EROD induction compared to control sediments. Conditions within the annular flume remained stable throughout all experiments and were adequate to support rainbow trout. Flood events were successfully simulated, resulting in re-suspension of formulated sediment. Different erosion behaviours of sediments during the simulated flood events were observed and could be associated with changes in microbial composition of sediments due to differences in storage conditions. Therefore, maintaining constant storage conditions of formulated sediments is crucial to enable consistency and comparability among erosion experiments.

Conclusions

This study clearly demonstrated the feasibility of a combined hydro-toxicological approach in support of the investigation of the potential ecotoxicological relevance of sediment re-suspension events. However, based on the results presented here, it is recommended to include additional physicochemical parameters, such as redox potential and conductivity, and to extend the experimental setup to natural sediments and different aquatic organisms. Future studies will use natural sediments containing representative microbial communities and extracellular polymeric substances to enable extrapolation from the annular flume to conditions in natural flowing waters.  相似文献

Present research in Germany on water, wind, and tillage erosion is examined. Existing soil‐protection strategies, laws, and guidelines are outlined. These are the result of extensive scientific investigations and the adoption of existing international knowledge. The most common methodologies currently used in Germany are reviewed. They allow an estimation of the water‐ and wind‐erosion risks as well as methods for their reduction. The results enable an aim‐orientated, site‐specific, risk‐adequate approach to implementing control measures in an easily understood framework. An examination of the methods for nuclear magnetic cross-relaxation spectroscopy (CRS) data collection and analysis was conducted using water and an aqueous waxy corn starch suspension to better perform and interpret the results obtained using CRS. The CRS data collection properties evaluated were the time to achieve steady state saturation, the direct saturation of liquid protons, generation of transverse magnetization, and dependence of the offset frequency and radio frequency (RF) field strength of longitudinal relaxation in the presence of RF saturation. Effects were evaluated for variations of input values of RF saturation field strength, apparent cross-relaxation rate, and solid longitudinal relaxation rate on the results for solid content and solid internal mobility from fitting NMR data to modified theoretical expressions. Discrepancies between fitted and stoichiometric values for the solid to liquid proton ratio were investigated. The fitting procedure used a Gaussian line shape for RF saturation of the solid-like spin system and a Lorentzian line shape for RF saturation of the liquid-spin system. Conditions under which acceptable results can be obtained with limited data sets are discussed.  相似文献
