The purpose of this study was to develop and investigate selection strategies that aim at maximizing long-term genetic response while conserving gene diversity and controlling inbreeding in populations of limited effective size, assuming complete knowledge of all genes affecting a quantitative trait. Three selection strategies were proposed to select on 100 quantitative trait loci (QTL) and compared with truncation selection on breeding value. Alternative selection strategies aimed at maximizing the average breeding value of parents with a penalty on (1) the number of unfavourable QTL genotypes among parents (OS-I), (2) the negative of the logarithm of the frequency of the favourable allele at each QTL among parents (OS-II), and (3) the average pedigree relationship among parents (OS-III). When all QTL and their effects were known, the strategies examined were able to obtain extra long-term responses, conserve QTL diversity and reduce inbreeding, compared with truncation selection. Strategy OS-II was the most effective in conserving QTL diversity and OS-III in reducing inbreeding. By changing the magnitude of the penalties applied, the impact on long-term response, inbreeding and diversity can be controlled. Extra long-term responses over truncation selection of OS-I and OS-II were even greater when effects of QTL were estimated rather than assumed known, indicating the applicability of results to practical strategies for marker-assisted selection. This investigation aims to classify, describe and evaluate the sustainability of dairy goat production systems (GPS) in South Spain Sierra de Cadiz. The research took place throughout 25 goat farms during the 2001–2002 campaigns, with the method posed by Masera et al. (1999) [Masera, O., Astier, M., S., López-Ridaura. 1999. Sustentabilidad y manejo de recursos naturales. El marco de evaluación MESMIS (Sustainability and natural resource management. The MESMIS evaluation framework). Mundi-Prensa, S.A., Gira, IE-UNAM, México. 109 pp.] and adapted to animal production systems, as the guideline and framework to evaluate sustainability.
The principal component, namely energy input from grazing (eigenvalue 1.329) which comprises the indicators total area per goat (factorial value 0.664) together with net energy obtained from grazing (factorial value 0.903) allowed to differentiate significantly between semi extensive (SES), semi intensive (SIS) and intensive (IS) goat production systems.
Intensification of the GPS tends to be inefficient, especially in terms of net margin per litre of milk produced (p < 0.05). A higher degree of adaptability of IS (64.8%) derives from a higher investment on new production strategies. Likewise, higher self-management capacity of SES (60.9%) fosters standards of productivity (76.0%) and stability (42.9%). The SIS presented the highest equity values (67.8%).
On the whole, sustainability of GPS tends to decrease as the degree of intensification increases: SES = 57.3%; SIS = 55.7% and IS = 53.1%. The reduction of the dependency on external input alongside with the optimization of natural resources management would surely improve the standard of sustainability. 相似文献
Background: Progesterone receptor (PR) antagonist aglepristone (RU534) has been used successfully for pregnancy termination and therapy of pyometra, vaginal tumors, and mammary hyperplasia in bitches and queens. All of these conditions share with canine mammary carcinomas the expression of PR. Objectives: To study the effect of RU534 on proliferation and apoptosis in canine mammary carcinomas in relation to PR expression. Animals: Twenty‐seven nonspayed bitches with mammary carcinomas were treated with either 2 doses of 20 mg/kg RU534 (n = 22, RU534‐treated group) or oil placebo (n = 5, control group) on days 1 and 8. Methods: Tumor samples were collected before (day 1) and after (day 15) treatment for immunohistochemistry. PR expression, proliferation index (PI), and apoptotic index (AI) were determined using antibodies against PR, Ki67, and cleaved lamin A/C antigens, respectively. The effect of treatment on these parameters was analyzed. Results: Differential expression of PR between day 1 (59.1% PR‐positive tumors) and day 15 (36.4% PR‐positive tumors) was observed in RU534‐treated tumors exclusively. After RU534 treatment, mean PI was significantly decreased in PR‐positive but unchanged in PR‐negative RU534‐treated tumors. A reduction of ≥20% in PI was found in 61.5% of RU534‐treated tumors with PR expression. Conversely, no effect on AI was observed after RU534 treatment. Conclusions and Clinical Importance: Neoadjuvant RU534 treatment had PR expression‐related inhibiting effects on proliferation of canine mammary carcinoma cells. 相似文献

The purpose of this study was to investigate the hemolymph kinetics and depuration time of oxytetracycline following intramuscular administration at doses of 2, 8 and 40 mg/kg body weight, respectively. The concentration of OTC in hemolymph was assayed using solid phase extraction and high performance liquid chromatography. The elimination half‐life of the terminal part of the elimination phase (t1/2β) ranged from 87.9 to 114.3 h. The total body clearance (CLb) was 0.0430 L/kg/h at the lower dose, 0.0123 L/kg/h at the medium dose and 0.0013 L/kg/h at the higher dose. The apparent volume of the central compartment (Vc) was found to be 1.383, 0.699 and 0.143 L/kg respectively. The depuration time for each dose was 13.6, 29.6 and 57.6 days, respectively. Results from the present study suggest that the 40 mg/kg dose might have the best therapeutic efficacy following intramuscular administration. 相似文献
The proposed technique consists of placing a catheter in the olfactory recess of the lateral ventricle of the brain of a sheep. Twenty adult ewes were used in the study. The olfactory recess is in the frontal region 25 +/- 3 mm below the skin, just caudal to the line joining the supraorbital foramen and approximately 8-10 mm from the median plane. The approach is relatively easy under normal experimental conditions. The wall of the olfactory bulb, however, is fragile, and the animals must not have been injured by Oestrus ovis. 相似文献
A sandwich enzymeimmunoassay (EIA) for pregnant mare serum gonadotropin (PMSG) using a microtiter plate was developed. Sensitivity of the assay to PMSG was 15.6 mIU/ml (0.2 ng/well). The PMSG levels in serum were measured with the EIA in superovulated and anti-PMSG rabbit antiserum treated mice and heifers. In mice, the PMSG blood level was measurable in the serum 4-6 days after intraperitoneal injection of 5-30 IU of PMSG. The administration of anti-PMSG antiserum at the same dose level as PMSG caused a rapid decrease in the PMSG blood level, declining to undetectable levels within 17 hours. In heifers, the PMSG level was measurable at 10-11 days after the injection of 2500 or 3000 IU of PMSG. When antiserum was injected 48 hours after the PMSG injection, the clearance rate of PMSG was affected by the route of the administration. The administration of 3000 units of anti-PMSG antiserum intravenously caused a rapid decline and the disappearance of circulating PMSG within 17 hours. When 3000 units of anti-PMSG antiserum was injected intra-muscularly, the PMSG blood level also decreased and became unmeasurable 24 hours after administration; however, it was still detectable for up to 17 hours. These results indicate that the administration of anti-PMSG antiserum at the proper timing and dosage could lead to successful superovulation through the improvement of hormonal conditions. 相似文献
Lactose in normal saline was administered intravenously to a group of Zebu cattle infected with Trypanosoma vivax to determine the blood plasma kinetics at onset of an experimental infection and its ability to protect tissues against damage as part of preliminary studies to determine its suitability for use in the treatment of trypanosomosis. Significantly (P < 0.01) higher lactose concentrations were observed in the T. vivax-infected bulls at 30 min and 1 h (P < 0.05) post-infection (p.i.) and by 4 h p.i. the plasma lactose remained above the level prior to infusion, after which it fell slightly below the pre-infusion level in the uninfected group. Calculated pharmacokinetic parameters revealed delayed excretion of lactose in the T. vivax-infected group soon after infection. The total body clearance (Cl(B)) was significantly (P < 0.05) reduced. The biological half-life (t1/2), elimination rate constant (k(el)) and apparent volume of distribution (V(d)) were relatively decreased (P > 0.05) as a result of the T. vivax infection. Retention of lactose in the plasma was attributed to decreased plasma clearance. It is suggested that the presence of trypanosomes in circulation rather than organic lesions could have been responsible for the delay observed in the excretion of lactose. At 12 weeks p.i., when the experiment was terminated, the group infected and given lactose infusion (despite higher parasitaemia) had no gross or histopathological lesions in the brain, spleen, lymph nodes, heart, kidneys, liver and testes. However, the group infected but not infused with lactose were emaciated, had pale mucosae, watery blood, general muscular atrophy, serous atrophy of coronary fat and other adipose tissue, hepatomegaly, splenomegaly, swollen and oedematous lymph nodes, all of which are suggestive of trypanosomosis. Histopathological lesions included narrowing of Bowman's space and hypercellularity of glomerular tufts in the kidneys with the mean glomerular tuft nuclear indices (GTNs) in the group significantly higher (P < 0.01) than the mean GTNs of the lactose-infused and control bulls. Degenerative changes occurred in the myocardium, spleen, testes and epididymides. The tesicular and epididymal lesions are indicative of male reproductive dysfunction. 相似文献
