Antibodies raised against whole sporangia of Sclerospora graminicola reacted specifically with sporangia and mycelium of this fungus. Agglutination studies localized the antigen on the cell-wall surfaces. Using this antibody, an indirect enzyme-linked immunosorbent assay (ELISA) with a biotin-avidin amplification system was developed for the quantification of S. graminicola in the host tissues of Pennisetum glaucum. This sensitive assay system, which could detect 3·1–5·5 μg dry weight of fungal biomass, was used to monitor the increase in fungal biomass during disease progression. 相似文献

The labile organic carbon(C) and C-related enzymes are sensitive indicators capturing alterations of soil organic matter(SOM),even in a short-time scale.Although the effects of crop husbandry and land use change on these attributes have been well studied,there is no consensus about how plant phenology may impact them.This study aimed to determine the short-term effect of six distinct phenological stages(PS-1:full bloom;PS-2:fruit set;PS-3:pit hardening;PS-4:physiological maturity;PS-5:60 d after physiological maturity;and PS-6:fall) of peach on the changes in soil organic carbon(SOC) fractions of different oxidizability,labile C pools,and C-cycle enzyme activities in soils,for two consecutive years(2015 and 2016) in the North-Western Himalayas(NWH).Peach rhizosphere soils were sampled at the topsoil(0–15 cm) and subsoil(16–30 cm) layers,along with rhizosphere soils from adjacent perennial grasses,which served as a control.Values for most of the assessed parameters,including very labile C,labile C,microbial biomass C,permanganate oxidizable C,dissolved organic C,mineralizable C,amylase activity,and carboxymethyl-cellulase activity,were significantly(P ≤ 0.05) higher at PS-3 than at other phenological stages of peach.Conversely,a sudden decline in these soil variables was recorded at PS-5,followed by a slight buildup at PS-6,particularly in the topsoil of the peach orchard.Short-term changes in organic C fractions of different oxidizability,influenced by peach phenological stage,significantly(P ≤ 0.05) affected C management index,C pool index,and lability index.Both the C management index and lability index showed their highest values at PS-3 and their lowest values at PS-5,clearly indicating short-term accretion and depletion of SOC,in tandem with the peach phenological events.Principal component analysis suggested that a composite of soil indicators,including microbial biomass C,dissolved organic C,amylase,and invertase,could help detect short-term changes in SOC content.It is concluded that peach phenological events had a major impact on the short-term variations of the studied soil variables,which could be attributed to changes in the above-and belowground plant residues,as well as the extent of nutrients and water acquisition. 相似文献

A multilocus sequence analysis (MLSA) was carried out to delineate Aeromonas hydrophila from fish in Uganda. Five housekeeping genes including recA, gyrB, metG, gltA and pps; and the 16S rRNA gene were amplified and sequenced from a total of nine A. hydrophila isolates. The obtained sequences were edited, and consensus sequences generated for each gene locus. The housekeeping gene sequences were concatenated and phylogenetic analysis performed in MEGA version 7.0.2. Pairwise distances ranged from 0.000 to 0.118, highest within the gltA gene locus and lowest within the 16S rRNA gene. The average evolutionary diversity within isolates from the same source ranged between 0.002 and 0.037, and it was 0.033 between the different sources. Similar tree topologies were obtained from the different gene loci with recA, metG and gyrB being more consistent in discriminating isolates according to sources while the 16S rRNA gene had the lowest resolution. The concatenated tree had the highest discriminatory power. This study revealed that A. hydrophila strains infecting fish in Uganda are of diverse genotypes suggesting different sources of infection in a given outbreak. Efforts to minimize spread of the bacteria across sources should be emphasized to control infections of mixed genotypes. 相似文献

During 2008 and 2009, field experiments were conducted at the research farm of CSIR-Institute of Himalayan Bioresource Technology, Palampur, India to assess the effect of shade levels (no shade, 25% shade, 50% shade, 75% shade) and plant geometry levels [30 × 15 cm (north–south, N–S), 45 × 10 cm (N–S), 30 × 15 cm (east–west, E–W) and 45 × 10 cm (E–W)] on the growth, yield and quality of stevia. Plant development was fastest in controls and slowest in stevia plants growing at minimum light (75% shade). These plants showed delayed bud formation and flowering. Significantly, higher dry leaf yield was recorded when the crop was grown under 25% shading than under 50 and 75% shade. Total steviol glycosides content decreased with increasing shade levels. All shade levels resulted in the same leaf-to-stem ratio, which was significantly higher than that obtained under ambient light. Plant geometry significantly affected the number of leaves and leaf dry weight plant–1. During both years, dry leaf weight was highest in the wider row spacing (45 × 10 cm) than in the narrow row spacing (30 × 15 cm). Stevia spaced at 30 × 15 cm in an E–W direction recorded significantly higher total steviol glycosides at 2 months after transplanting (MAT). 相似文献

Resistin, founding member of the resistin-like molecule (RELM) hormone family, is secreted selectively from adipocytes and induces liver-specific antagonism of insulin action, thus providing a potential molecular link between obesity and diabetes. Crystal structures of resistin and RELMbeta reveal an unusual multimeric structure. Each protomer comprises a carboxy-terminal disulfide-rich beta-sandwich "head" domain and an amino-terminal alpha-helical "tail" segment. The alpha-helical segments associate to form three-stranded coiled coils, and surface-exposed interchain disulfide linkages mediate the formation of tail-to-tail hexamers. Analysis of serum samples shows that resistin circulates in two distinct assembly states, likely corresponding to hexamers and trimers. Infusion of a resistin mutant, lacking the intertrimer disulfide bonds, in pancreatic-insulin clamp studies reveals substantially more potent effects on hepatic insulin sensitivity than those observed with wild-type resistin. This result suggests that processing of the intertrimer disulfide bonds may reflect an obligatory step toward activation. 相似文献

The crustacean ectoparasite, Argulus poses one of the major threats to carp culture due to absence of any suitable control measure. The study was undertaken to determine the expression of immune-related genes in three major immunocompetent organs viz., kidney, skin and liver of rohu (Labeo rohita) during experimental freshwater lice Argulus siamensis infection. Results showed that the expression of TLR 22-like, lysozyme G and β2-microglobulin genes in kidney was significantly (P ≤ 0.05) down-regulated in lice-infected fish. On the other hand, no significant difference (P>0.05) in CXCa, lysozyme C, TNFα and complement component 3 (C3) expression was found between uninfected control and different degrees of lice infected fish. In the skin, the expression of TLR 22-like and TNFα genes were significantly up-regulated whereas that of C3 was significantly (P ≤ 0.05) down-regulated in lice-infected fish with respect to control fish. The expression of CXCa, lysozyme C and transferrin was not detected in the skin samples of fish. In the liver, the expression of CXCa, lysozyme G, β2-microglobulin and transferrin was significantly (P≤0.05) up-regulated in lice-infected fish with respect to control fish whereas expression of C3 was significantly (P ≤ 0.05) down-regulated in lice-infected fish. The expression of TLR 22-like, lysozyme C, TNFα genes was not detected in the liver samples of fish. This study indicates that majority of the genes showed down-regulation in kidney tissue whereas up-regulation in liver and skin tissues except C3 in Argulus-infected fish. We show that infection with this parasite irrespective of intensity can also result in immune gene expression changes in tissues situated away from the site of parasite attachment and feeding. The information obtained here could be useful towards understanding the susceptibility of rohu to argulosis and mechanisms involved in protection of rohu to ectoparasitic infections, which is causing immense economic losses to freshwater aquaculture sector. 相似文献
