We screened the mycorrhizal species for an inoculum protocol would green pepper seedling production and compensate for nutrient deficiency. Three greenhouse studies (on three successive years) were conducted under nursery conditions using five arbuscular mycorrhizal (AM) fungi and their mixture. The AM fungi, Glomus mosseae, G. clarum, G. caledonium, G. intraradices and G. etunicatum propagated on maize roots were used. The AM fungi were inoculated to seed stages and half of the seed stages inoculated plants were re-inoculated at the seedling stages. In Experiment I, plants were harvested once and in Experiments II and III, plants were harvested twice at different developmental stages.  相似文献

Lichen phenolics that are known to leach out from lichen thalli during rainfall episodes are considered to be allelopathic or antimicrobial agents. On the other hand, small organic molecules originating from aboveground vegetation generally constitute an important C source for the soil microbial community. We studied the effects of leaching lichen acids on soil microbial processes by chemically removing usnic (UA) and perlatolic acid (PA) from the extracellular space of mat-forming lichen, Cladina stellaris, a dominant component of the ground vegetation in boreal oligotrophic forests. Removal of UA and PA did not affect the intracellular contents of the lichen. We hypothesized that if lichen acids have an adverse effect on soil biota, microbial respiration would increase underneath chemically manipulated lichen mats. In turn, if these substances provide energy source for the microorganisms, microbial respiration would decline under lichens from which PA and UA has been removed. Microbial metabolic activity, qCO2 was significantly lower under the lichens from which UA and PA had been removed, thus supporting the latter hypothesis. Removing UA and PA did not affect the amount of microbial C, N and P, or soil nutrient concentrations. Our results suggest that the microbial community in the soil under a lichen carpet is adapted to the presence of these lichen secondary metabolites and can utilize them as a C source. The approach of removing lichen substances from the system could be used as an effective research tool as it can overcome several problems that are often common in the studies on allelopathic interactions.  相似文献

Arbuscular mycorrhizal (AM) symbioses may alter the competitive abilities of plant species and facilitate positive interactions such as nutrient transfer between plants. They are therefore particularly interesting components in agroforestry systems. We studied spatial variation of AM colonisation on a cut-and-carry fodder production site (agroforestry plot) of the legume tree Gliricidia sepium and the fodder grass Dichanthium aristatum. Roots of the two plant species were sampled under the tree canopies and on the adjacent grass plot at 1 and 3.5 m from the first tree row where G. sepium roots also occur. Roots of D. aristatum were also sampled on a nearby grass monocrop. Colonisation of arbuscules, vesicles and hyphae in root samples was visually determined, and AM fungal species were identified by DNA sequencing. Colonisation and frequency of types of AM formations varied statistically significantly between the species and sampling points. Arbuscular colonisation in G. sepium roots was higher under the tree canopies than on the adjacent grass plot. Soil nutrient content, particularly P and N, and interspecies competition are the most probable explanations for the observed variation in AM colonisation. Both arbuscular colonisation and arbuscule:vesicle ratio in D. aristatum roots was lower on the D. aristatum monocrop than on the agroforestry plot under or near the tree canopies. Intercropping could stimulate AM symbiosis in D. aristatum. Both plant species formed symbiosis with Rhizophagus intraradices, indicating potential for interplant N transfer via common mycelial networks of AM-forming fungi.  相似文献

Calbindin-D28K (Ca-D28K) is a calcium-binding protein. In the kidney, Ca-D28K is present in the distal nephron, but not in the proximal nephron. This site-specific distribution in the kidney indicates that Ca-D28K is a potential marker for the differentiation of the distal nephron. In this study, we have examined the expression of Ca-D28K in 25 sporadic cases of chicken nephroblastomas. All cases of nephroblastomas were composed of atypical tubular structures, blastemal cells, and fibrous stroma in varying degrees of differentiation. Immunohistochemically in all nephroblastoma specimens, Ca-D28K was expressed in the epithelial cells of the subsets of tubular structures, but not in the blastema or the stroma. These results suggested that the tubuli in the nephroblastomas are able to differentiate into the phenotype of distal nephrons. Furthermore, Ca-D28K might develop as a novel diagnostic marker for nephroblastomas because this molecule is reported to be completely negative in other renal tumors, including renal cell carcinoma, chromophobe carcinomas, and oncocytoma.  相似文献

Despite the promising antioxidant action of Lamiaceae herbs in vitro, human studies on these potential sources of dietary antioxidants have remained scarce. In this work, the phenolic acids recovered in human urine after single ingestion of Origanum onites extract were analyzed. The excretion was increased 4- and 2-fold during 0-24 and 24-48 h of the follow-up, respectively. The mean increase in the excretion of phenolic compounds exceeded the ingested amount of identified phenolic acids. The result can be partly explained by rosmarinic acid, the main identified phenolic constituent in the extract, as well as flavonoids present in minor amounts, presumably being metabolized into a double amount of simple phenolic metabolites. Furthermore, unidentified phenolic constituents in the extract partly contribute to the excretory increase. The main metabolite, p-hydroxybenzoic acid, was excreted rapidly. The results show that constituents of oregano extract and, in particular, their metabolites may contribute to the dietary intake of phenolic antioxidants.  相似文献

It has previously been suggested that plasma membrane ATPase (PM H+-ATPase, EC 3.6.1.3.) is a site of incipient freezing injury because activity increases following cold acclimation and there are published data indicating that activity of PM H+-ATPase is modulated by changes in lipids associated with the enzyme. To test and extend these findings in a tree species, we analyzed PM H+-ATPase activity and the fatty acid (FA) composition of glycerolipids in purified plasma membranes (PMs) prepared by the two-phase partition method from current-year needles of adult red pine (Pinus resinosa Ait.) trees. Freezing tolerance of the needles decreased from -56 degrees C in March to -9 degrees C in May, and increased from -15 degrees C in September to -148 degrees C in January. Specific activity of vanadate-sensitive PM H+-ATPase increased more than two-fold following cold acclimation, despite a concurrent increase in protein concentration. During de-acclimation, decreases in PM H+-ATPase activity and freezing tolerance were accompanied by decreases in the proportions of oleic (18:1) and linoleic (18:2) acids and increases in the proportions of palmitic (16:0) and linolenic (18:3) acids in total glycerolipids extracted from the plasma membrane fraction. This pattern of changes in PM H+-ATPase activity and the 18:1, 18:2 and 18:3 fatty acids was reversed during cold acclimation. In the PM fractions, changes in FA unsaturation, expressed as the double bond index (1 x 18:1 + 2 x 18:2 + 3 x 18:3), were closely correlated with changes in H+-ATPase specific activity (r2 = 0.995). Changes in freezing tolerance were well correlated with DBI (r2 = 0.877) and ATPase specific activity (r2 = 0.833) in the PM fraction. Total ATPase activity in microsomal fractions also closely followed changes in freezing tolerance (r2 = 0.969). We conclude that, as in herbaceous plants, simultaneous seasonal changes in PM H+-ATPase activity and fatty acid composition occur during cold acclimation and de-acclimation in an extremely winter hardy tree species under natural conditions, lending support to the hypothesis that FA-regulated PM H+-ATPase activity is involved in the cellular response underlying cold acclimation and de-acclimation.  相似文献

The potential of grafted watermelon for resistance toFusarium oxysporum f.sp.niveum on some Curcurbitaceae,Lagenaria, Luffa, Benincasa and commercial rootstocks was evaluated. Effects of grafting on yield and quality of diseased plants were evaluated. All grafted plants and rootstocks were resistant to the three known races (0, 1, and 2) ofF. oxysporum f.sp.niveum except watermelon cv. ‘Crimson Tide’, which was susceptible to race 2. Fruit yield was positively (21–112%) affected byLagenaria rootstocks but negatively affected (200–267%) byCucurbita rootstocks when compared with the control. While only minor differences in fruit quality were determined in control and grafted plants onLagenaria rootstocks, the quality parameters for watermelon grafted ontoCucurbita rootstocks were lower than in the control. The reasons for low yield and quality might be due to an incompatibility betweenCucurbita rootstocks and watermelon. Volatile emission profile of strawberry (Fragaria x ananassa Duch.) plants (cvs. Polka and Honeoye) damaged by cyclamen mite (Phytonemus pallidus Banks) or leaf beetle Galerucella tenella (L.) (cv. Polka) was analyzed to determine the potential of these strawberry plants to emit herbivore-induced volatiles. The total volatile emissions as well as emissions of many green leaf volatiles (e.g., (Z)-3-hexen-1-ol and (Z)-3-hexenyl acetate) and methyl salicylate were greater from cyclamen mite-damaged strawberry plants than from intact plants. Leaf beetle feeding increased emissions of monoterpenes (Z)-ocimene and (E)-beta-ocimene, sesquiterpenes (E)-beta-caryophyllene, (E,E)-alpha-farnesene, and germacrene-D, and a homoterpene (3E)-4,8-dimethyl-1,3,7-nonatriene (DMNT) significantly. Nevertheless, the na?ve generalist predatory mites, Neoseiulus cucumeris, Neoseiulus californicus, and Euseius finlandicus did not prefer P. pallidus- or G. tenella-damaged plants over intact plants in a Y-tube olfactometer, suggesting that these predatory mite species are not attracted by the herbivore-induced volatiles being released from young strawberry plants.  相似文献
