A reconsideration of natural dispersal distances of the pine pathogen Dothistroma septosporum 下载免费PDF全文
The natural spread of Dothistroma septosporum, the causal agent of a foliar disease of pines, was investigated at three sites in the south of England using trap plants. The pathogen is considered to be primarily rain‐splash dispersed, but this study shows that it can be spread many hundreds of metres from an inoculum source, demonstrating that dispersal is not solely via rain splash. The maximum distance the pathogen was recorded from any infection source was in excess of 1400 m, over five times the distance defined in the only previous work of this kind. Consequently, a reassessment of forest and production nursery management practices is called for, as these assume that the pathogen only spreads naturally over limited distances. Detection of the pathogen on trap plants over 100 m from the inoculum source was, in most cases, only possible using quantitative real‐time PCR diagnosis. The entire diagnostic procedure, from DNA extraction to amplification, was able to detect a minimum of approximately 17 D. septosporum cells in a pine needle sample, assuming only a moderate DNA extraction efficiency of 30%. 相似文献

Puccinia psidii has long been considered a significant threat to Australian plant industries and ecosystems. In April 2010, P. psidii was detected for the first time in Australia on the central coast of New South Wales (NSW). The fungus spread rapidly along the east coast and in December 2010 was found in Queensland (Qld) followed by Victoria a year later. Puccinia psidii was initially restricted to the southeastern part of Qld but spread as far north as Mossman. In Qld, 48 species of Myrtaceae are considered highly or extremely susceptible to the disease. The impact of P. psidii on individual trees and shrubs has ranged from minor leaf spots, foliage, stem and branch dieback to reduced fecundity. Tree death, as a result of repeated infection, has been recorded for Rhodomyrtus psidioides. Rust infection has also been recorded on flower buds, flowers and fruits of 28 host species. Morphological and molecular characteristics were used to confirm the identification of P. psidii from a range of Myrtaceae in Qld and compared with isolates from NSW and overseas. A reconstructed phylogeny based on the LSU and SSU regions of rDNA did not resolve the familial placement of P. psidii, but indicated that it does not belong to the Pucciniaceae. Bacterial wilt caused by Ralstonia solanacearum is a destructive disease for many crops. The aim of this study was to investigate the phylogenetic relationships and genetic structure of an R. solanacearum population from diverse origins in Taiwan. All 58 tested isolates belonged to phylotype I, except the two biovar 2 isolates from potato. These belonged to phylotype IIB sequevar 1 and were identical to known potato brown rot strains, which were probably introduced. Phylotype I isolates were grouped into 10 sequevars. Sequevar 15 was predominant (34 out of 56 isolates). Its distribution covered the whole island and it was largely associated with solanaceous crops such as tomato, and with tomato field soil. A total of 14 haplotypes were identified based on a partial endoglucanase gene sequence. Parsimony network analysis revealed that haplotype A was the oldest haplotype in the local population. It encompassed all but one of the sequevar 15 isolates. Large variation in virulence on tomato was observed among the 58 isolates, and seven pathotypes were identified. Significant genetic differentiation was detected among pathotypes. Moreover, genetic differentiation was detected between biovar 3 and biovar 4 subgroups and between the strains associated with solanaceous and non‐solanaceous species, but none was detected between strains from different geographic origins. The results suggest that the phylotype I population in Taiwan is homogeneous, while mutation and local adaptation to specific ecological niches keep shaping the population. 相似文献

Weed seeds in and on the soil are the primary cause of weed infestations in arable fields. Previous studies have documented reductions in weed seedbanks due to cropping system diversification through extended rotation sequences, but the impacts of different rotation systems on additions to and losses from weed seedbanks remain poorly understood. We conducted an experiment in Iowa, USA, to determine the fates of Setaria faberi and Abutilon theophrasti seeds in 2‐, 3‐ and 4‐year crop rotation systems when seed additions to the soil seedbank were restricted to a single pulse at the initiation of the study. Over the course of the experiment, seedlings were removed as they emerged and prevented from producing new seeds. After 41 months, seed population densities dropped >85% for S. faberi and >65% for A. theophrasti, but differences between rotation systems in the magnitude of seedbank reductions were not detected. Most of the reductions in seedbank densities took place from autumn through early spring in the first 5 months following seed deposition, before seedling emergence occurred, suggesting that seed predation and/or seed decay was important. For S. faberi, total cumulative seedling emergence and total seed mortality did not differ between rotation systems. In contrast, for A. theophrasti, seedling emergence was 71% lower and seed mortality was 83% greater in the 3‐ and 4‐year rotation systems than in the 2‐year system. Results of this study indicate that for certain weed species, such as A. theophrasti, crop rotation systems can strongly affect life‐history processes associated with soil seedbanks. 相似文献

The introduction of imidazolinone‐tolerant rice varieties has made selective Oryza sativa (weedy rice) control possible. We hypothesised that Italian weedy rice populations have variable degrees of susceptibility to imazamox prior to imidazolinone‐tolerant variety introduction. To this end, 149 Italian weedy rice populations collected from fields never before cultivated with imidazolinone‐tolerant varieties were tested in a glasshouse‐based, whole‐plant response screening study. Imazamox was applied to all populations post‐emergence at a rate of 70 g a.i. ha?1, resulting in 70–90% shoot biomass reduction in the majority of cases. The results prompted a second study of the seedling dose response of four weedy rice populations from the initial study group. Three imidazolinone‐tolerant and one conventional rice variety were also included. The seedling roots were cut six days after germination and exposed to different concentrations of imazamox. The root regrowth associated with each concentration‐exposure was then measured. Imazamox concentrations to inhibit weedy rice root growth by 50% varied by about two orders of magnitude, or between 0.0018 and 0.12 mm . Even with this result, imidazolinone‐tolerant varieties were at least 31.8 times less susceptible than weedy rice populations, suggesting that Italian weedy rice populations were not tolerant to imazamox before introduction of these varieties. 相似文献

Management of banana xanthomonas wilt (XW) (caused by Xanthomonas campestris pv. musacearum, Xcm) has been impeded by poor adoption of control options that are complex, cumbersome and costly. To improve XW management, this study investigated Xcm survival and latent infections in subsequent generations, survival of latently infected planting materials (suckers), incidence of latent infections in symptomless plants in mats having diseased plants, and XW status across farms and markets in districts previously devastated but currently endemic. On‐station experiments were protected from new infections. Latent bacteria at low levels were detected in up to 20% of the third generation suckers, with a significant (P < 0·05) reduction (43–20%) in subsequent generations. Only 3–6% of latently infected suckers succumbed to XW. Incidence of Xcm in symptomless suckers from farmers' fields (with up to 70% incidence) was low (3%) while it increased (8–25%) with disease severity in mats in controlled experiments. In the surveyed districts, incidence had significantly declined with yields observed to have recovered relative to earlier reports, although latent infections remained high. This study provides evidence that if new infections are prevented, fields with high XW incidence can be rejuvenated. It showed incomplete systemic movement of Xcm in mats coupled to a gradual decline of bacterial load in subsequent generations to levels that cannot initiate disease. These studies explain the current successes in farms practising single diseased plant removal instead of whole mat rouging, and gives hope to farmers lacking access to clean planting material. 相似文献

Assessing host specialization of Botrytis cinerea on lettuce and tomato by genotypic and phenotypic characterization 下载免费PDF全文
This study tested the hypothesis that Botyrtis cinerea shows host specialization on tomato and lettuce, using phenotypic and genotypic tools. Strains were isolated from tomato and lettuce grown together in the same greenhouse. Forty‐four lettuce strains and 42 tomato strains were investigated for their genetic diversity and their aggressiveness. Both gene diversity and allelic richness were significantly higher in lettuce strains than in tomato strains (P = 0·01). Cluster analysis revealed a clear division of the strains under study into two clusters. However, this structure did not separate the strains according to their host of origin. Tomato strains were significantly more aggressive than lettuce strains when inoculated on tomatoes (P = 0·001), but no significant differences in aggressiveness were observed when the strains were inoculated on lettuce (P = 0·17) or on apple (P = 0·87). The results suggest an absence of clear host specialization of B. cinerea on tomato and lettuce. 相似文献

Panama disease, caused by Fusarium oxysporum f. sp. cubense (Foc), is ranked among the most destructive diseases of banana. The use of resistant varieties is the most desirable and effective control measure. Information on the pathogen population structure is essential, as durability of the resistance and effective cultivar deployment are strongly linked to this structure. In this study, 214 Foc isolates from different banana producing states in three regions of Brazil (northeastern, southeastern and southern) were analysed. Initially, nine microsatellite markers (SSR) were tested, which revealed 52 distinct haplotypes distributed in the different geographical regions and cultivars. While amova analysis showed that 68·01% of the total variation occurred within states, correlation between genetic and geographical distances was only found in the southern region. Results indicated that isolates from different states comprise a single population, which is predominantly clonal. When isolates representing different haplotypes were inoculated in four banana cultivars, differences in severity were found, with the high severity values being caused by isolates from haplotypes H7, H31 and H41. The diversity found here points to the need for additional studies, as this characteristic may be related to Foc's evolutionary potential and possibly to its ability to overcome the resistance from breeding programme‐generated cultivars. This is the most comprehensive study on population biology of Foc in Brazil. 相似文献

Although fungicide resistance in crop pathogens is a global threat to food production, surprisingly little is known about the evolutionary processes associated with the emergence and spread of fungicide resistance. Early stages in the evolution of fungicide resistance were evaluated using the wheat pathogen Zymoseptoria tritici, taking advantage of an isolate collection spanning 20 years in Oregon, USA, and including two sites with differing intensity of fungicide use. Sequences of the mitochondrial cytb protein conferring single‐mutation resistance to QoI fungicides and the nuclear CYP51 gene implicated in multiple‐mutation resistance to azole fungicides were analysed. Mutations associated with resistance to both fungicides were absent in the 1992 isolates, but frequent in the 2012 collection, with higher frequencies of resistance alleles found at the field site with more intensive fungicide use. Results suggest that the QoI resistance evolved independently in several lineages, and resulted in significant mitochondrial genome bottlenecks. In contrast, the CYP51 gene showed signatures of diversifying selection and intragenic recombination among three phylogenetic clades. The findings support a recent emergence of resistance to the two fungicide classes in Oregon, facilitated by selection for mutations in the associated resistance genes. 相似文献
