To improve understanding of the mechanism of early ovarian development in eels, the effects of water temperature decrease on oocyte development, plasma levels of sex steroids [estradiol 17β (E2), testosterone (T), 11-ketotestosterone (11-KT)], and gonadotropin β-subunit [follicle-stimulating hormone (FSHβ), luteinizing hormone (LHβ)] messenger RNA (mRNA) expression levels were investigated. A total of 27 female Japanese eels Anguilla japonica were divided into initial, control, and test (water temperature decrease) groups. Starting on 22 September 2009, eels in the test group were reared in a tank with gradual temperature decrease from 25°C to 15°C over 39 days, while the control group was maintained at 25°C. The test group accumulated more oil droplets in their oocytes than did the other groups. Levels of sex steroids, especially 11-KT, were higher in the test group. In contrast, FSHβ and LHβ mRNA expression levels were lower in the test group. These results suggest that water temperature decrease only induced an early stage of ovarian development that was partly affected by an 11-KT increase. For further maturation, other environmental factors related to induction of gonadotropin increase appear to be needed.  相似文献

ABSTRACT

Sandy soils are usually dominant in tropical monsoon regions, due to the high weathering potential associated with high temperatures and precipitation. The organic matter content of sandy soils is low due to low clay content and high microbial activity. Therefore, soil management practices that alter the soil organic carbon (SOC) content may be important for the sustainable management of crop yields. Thus, the present study investigates the distribution of rice yield and SOC content under different land management practices and analyzes the relationship between rice yield and SOC with pertinent management practices (manure and fertilizer applications). The soil horizons from 0- to 40-cm depths were collected in each layer to measure SOC and soil properties at 64 sites. At each sampling site, farmers were given questionnaires and the record book for the standards for good agricultural practices of farm owners were gathered to assimilate information on rice yield and their practices during 2010–2014. The mean rice yield of the whole crop year and SOC were 2.93 Mg ha?1 and 47.09 Mg C ha?1, respectively, in the irrigated areas, and were 2.38 Mg ha?1 and 32.08 Mg C ha?1 in the rain-fed areas. Significantly higher values were obtained in the irrigated areas (p < 0.05). There was a significant positive correlation between rice yield and SOC in both the irrigated areas (R2 = 0.72, p < 0.01) and the rain-fed areas (R2 = 0.85, p < 0.01); however, the slopes of these regression equations were significantly different. In both irrigated and rain-fed areas, manure should be applied every year, with an optimal application rate of N, P, and K fertilizers being selected. The combination of manure, fertilizer, and increasing irrigation facilities the maintenance of SOC levels and substantially increases rice yields.  相似文献

The effects of a short chilled storage period before freezing, frozen storage temperature, and freezing rate on lipid oxidation of bluefin tuna (Thunnus thynnus) meat during frozen storage were investigated. After 12-months storage, all samples had increased in peroxide value though they were less at the lower temperatures (?45 and ?60°C). Peroxide values in all samples stored at ?20°C increased after 3 months storage, particularly those processed and stored 51 h after harvest. The lowest increase in peroxide value occurred in the samples frozen rapidly 3 h after harvest. Vitamin E levels decreased faster during frozen storage at ?20°C. There were no apparent differences in levels of triacylglycerides nor in n-3 fatty acid levels between treatments, storage periods, and storage temperatures. After 12-months storage, headspace oxidative volatiles were highest in samples stored at ?20°C and lowest in those stored at ?60°C. Lipid oxidation in tuna meat stored at ?45°C is similar to that at ?60°C, and rapid freezing rather than slow freezing should be used.  相似文献

Insect growth regulators (IGRs) can be developed by elucidating the molecular mechanisms of insect-specific biological events. Because insect molting, and metamorphosis are controlled by ecdysteroids, their biosynthetic pathways can serve as targets for IGR development. The glutathione S-transferase Noppera-bo (Nobo), which is conserved in dipteran and lepidopteran species, plays an essential role in ecdysteroid biosynthesis. Our previous study using 17β-estradiol as a molecular probe revealed that Asp113 of Drosophila melanogaster Nobo (DmNobo) is essential for its biological function. However, to develop IGRs with a greater Nobo inhibitory activity than 17β-estradiol, further structural information is warranted. Here, we report five novel non-steroidal DmNobo inhibitors. Analysis of crystal structures of complexes revealed that DmNobo binds these inhibitors in an Asp113-independent manner. Among amino acid residues at the substrate-recognition site, conformation of conserved Phe39 was dynamically altered upon inhibitor binding. Therefore, these inhibitors can serve as seed compounds for IGR development.  相似文献

Hierarchical Bayesian (HB) methods are useful tools for modeling multifaceted, nonlinear phenomena such as those encountered in ecology, and have been increasingly applied in environmental sciences, e.g., to estimate soil gas flux from different soil textures or sites. We have developed a model of soil carbon dioxide (CO2) flux based on soil temperature (T, 5 cm depth) and water-filled pore space (WFPS, 5 cm depth) using HB theory. The HB model was calibrated using a dataset of CO2 flux measured from bare soils belonging to four texture classes in 14 upland field sites in a watershed in central Hokkaido, Japan, in the nonsnow-cover season from 2003 to 2011. The numerical software HYDRUS-1D was used to simulate daily WFPS, and the estimated values were significantly correlated with the measured WFPS (R2 = 0.68, P < 0.001). Compared to a nonhierarchical Bayesian model (Bayesian pooled model), the CO2 predictions with the HB model more accurately represented texture-specific observations. The simulation–observation fit of the CO2 flux model was R2 = 0.64 (P < 0.001). More than 90% of the observed daily data were within the 95% confidence interval. The HB model exhibited high uncertainty for high CO2 flux values. The HB model calibration revealed differing sensitivity of CO2 flux to T and WFPS in different soil texture classes. CO2 flux increased with an increase in T, and it increased to a lesser degree with a finer texture, possibly because the clay and silt facilitated soil aggregation, thus reducing temperature fluctuations. WFPS values between 0.48 and 0.64 resulted in optimal conditions for CO2 flux. The minimum WFPS value increased with an increase in clay content (P < 0.05). Although only a small number of soil types were studied in only one season in this study, the HB model may provide a method for predicting how the effects of soil temperature and moisture on CO2 flux change with texture, and soil texture could be regarded as an upscaling factor in future research on regional extrapolation.  相似文献

Eggs of the Japanese eel Anguilla japonica collected in the western North Pacific were identified by onboard species-specific polymerase chain reaction (PCR) and DNA nucleotide sequencing after the cruise. Fish eggs of various species were collected by large plankton net tows at 12 stations along the southern part of the West Mariana Ridge on 19–25 May 2009. A total of 43 fish eggs were distinguished morphologically as possibly being of A. japonica. Thirty-one of those were analyzed by PCR, which included 15 eggs collected at 12°50–55′N, 141°15–20′E (in 5 tows) that showed positive results. The 16S ribosomal RNA (rRNA) gene sequences of eggs determined after the cruise indicated that 31 A. japonica eggs had been collected. The remaining eggs were of mesopelagic eel species (Serrivomeridae and Derichthyidae), or unidentified species. The morphological characteristics of the A. japonica eggs were consistent with those of artificially spawned eggs, except they had a slightly larger diameter. The egg diameter range did not overlap with those of mesopelagic eels of the Serrivomeridae, which often spawn in the same area as A. japonica. These results suggest that egg diameter and embryo shape can be used to morphologically identify naturally spawned A. japonica eggs.  相似文献
