To evaluate the importance of arbuscular mycorrhizal fungi (AMF) to crop production, it is imperative to move beyond the plow layer to include the full soil profile impacted by plant roots. To illustrate this, we investigated the vertical distribution of AMF biomass and community structure within the top 100 cm of soil in soybean (Glycine max (L.) Merr., cv: Enrei) rotational systems cropped to wheat (Triticuma estivum L. cv: Bandowase) or left fallow using fatty acid methyl ester (FAME) biomarkers and molecular analysis, respectively. AMF biomass, as measured by concentration of C16:1cis11, declined during fallow and with increasing soil depth. Greater than 50 % of the stored AMF biomass was found at depths below 35 cm. Phylogenetic analysis revealed 16 AMF phylotypes, including nine Glomus, two Gigaspora, two Scutellospora, and one each of Diversispora, Paraglomus, and an unknown glomeromycete, at different sampling depths in this study. Cluster analysis based on the number and abundance of each AMF phylotype formed two distinct clusters separating wheat from fallow rotations. There was no distinct relationship with soil depth beyond clustering AMF communities above and below 20 cm under wheat. Redundancy analysis (RDA) and hierarchical cluster analysis demonstrated that AMF communities by soil depth within each rotation were not significantly different. However, AMF communities were clearly influenced by crop rotation, where the distribution of specific AMF phylotypes responded to the presence of the wheat crop.  相似文献

Equine paratyphoid is caused by Salmonella enterica serovar Abortusequi, and manifests mainly as abortion in the mare. We compared S. Abortusequi strains isolated in Japan and other countries using pulsed-field gel electrophoresis (PFGE) and fluorescent amplified-fragment length polymorphism (FAFLP) analysis. PFGE analysis of S. Abortusequi strains gave 21-27 fragments ranging in size from 33 to 602kb. Although two PFGE profiles were observed among the 20 S. Abortusequi isolates in Japan, the restriction fragments originating from the chromosome were common between the two profiles. The similarity index of the two profiles was 90.9%, while those between Japanese and five other S. Abortusequi strains were 29.8-37.5%. On the other hand, FAFLP analysis of S. Abortusequi strains generated 64-67 amplified fragments ranging in size from 100 to 400bp. One polymorphic fragment was observed among the 20 S. Abortusequi isolates in Japan. These data indicate the close relation of this agent in Japan. S. Abortusequi strains sharing a common ancestry might have been conserved in Japan.  相似文献

We assessed fumonisin production during the ensiling of rice grain. Rice grain was harvested at the full‐ripe stage and prepared as rough rice, crushed rough rice, brown rice or crushed brown rice. Each material was ensiled under six conditions: (1) no fungus, anaerobic; (2) no fungus, aerobic; (3) water added, anaerobic; (4) water and fumonisin‐producing fungus added, anaerobic; (5) water and fumonisin‐producing fungus added, aerobic; or (6) fumonisin‐producing fungus added to autoclaved material, aerobic. After 40 days of ensilage, we analyzed the silage fermentative quality and fumonisin concentration. The fermentative quality of all materials was good in treatments (3) and (4) (pH < 4), reasonable in treatment (5) (pH = 5~6) and unacceptable in treatments (1) and (2) (pH > 6.5). The fumonisin concentration was low in all materials in treatments (1) to (4), slightly increased in the three materials other than rough rice in treatment (5), and enormously increased in all materials in treatment (6). The results indicate that the fumonisin‐producing fungus does not produce fumonisin in anaerobic conditions. It is important that an anaerobic condition be maintained during ensiling in order to reduce the fumonisin content in rice grain silage.  相似文献

Waxy wheat (Triticum aestivum L.), a type of wheat that was first developed in Japan, produces grain amylopectin. I carried out three researches. Firstly, using Japanese adults, we carried out sensory evaluation of three types of mochi (a dumpling-like cake made of steamed and pounded grains) prepared from waxy wheat, normal wheat and waxy rice, and demonstrated that waxy wheat mochi had low adhesiveness and had the greatest ease of swallowing among the samples tested. Secondly, using the members of the JSDR (the Japanese Society of Dysphagia Rehabilitation) who attended the annual meeting, we performed another texture evaluations between waxy wheat and waxy rice mochi. The respondents reported waxy wheat mochi was less sticky and thus easier to chew and swallow than waxy rice mochi.  相似文献

The visual symptoms of damage by acid sulfate soil look quite similar to those due to soil moisture deficit. The present paper is to confirm if acid soil-tolerance is associated with drought tolerance in terms of leaf water potential, transpiration, and dry matter production. Seedlings of seven acid soil-tolerant (acid-tolerant), and three acid soil-intolerant varieties (acid-intolerant) were subjected to low pH conditions (pH 3.8) for 48 hours. The rate of water uptake was determined continuously by measuring radioactivity in the collar part (target area) of rice leaves exposed to 18F-labeled water (18F-water) using a Positron Emitting Tracer Imaging System (PETIS). The PETIS measurement showed that the radioactivity in the target position of leaves of acid-tolerant varieties increased faster than that of the intolerant varieties after the 18F-water was applied into the vial. In the plants subjected to low pH conditions, the transpiration rate (Tr) and leaf water potential (Ψ) decreased in both acid-tolerant and intolerant varieties. However, the extent of reduction was less in the acid-tolerant varieties than in the intolerant ones. Moreover, the dry matter production rate of the acid-tolerant varieties was significantly higher than that of the acid-intolerant ones in the low pH conditions. This study clearly shows that 18F-water uptake in a leaf was higher, and water relations were also better maintained in the acid-tolerant varieties compared with intolerant ones, resulting in higher growth rate in the acid-tolerant varieties, when plants were exposed to the acid solution conditions. We conclude that acid soil-tolerance is closely associated with leaf water relations in rice plants.  相似文献

Our previous studies demonstrated that an 8‐week intake of 5% (w/w) apple polyphenol (APP) in the diet improves muscle endurance of young‐adult rats. In order to identify a lower limit of the dietary contribution of APP to the effect, the experiments were designed for lower‐dose supplementation (8‐week feeding of 0.5% APP in AIN‐93G diet) to 12‐week‐old male Sprague‐Dawley rats. Results clearly showed that the 0.5% APP diet significantly up‐regulates slower myosin‐heavy‐chain (MyHC) isoform ratios (IIx and IIa relative to total MyHC) and myoglobin expression in lower hind‐limb muscles examined (P < 0.05). There was a trend to increased fatigue resistance detected from measurements of relative isometric plantar‐flexion force torque generated by a stimulus train delivered to the tibial nerve (F(98, 1372) = 1.246, P = 0.0574). Importantly, there was no significant difference in the animal body‐phenotypes or locomotor activity shown as total moving distance in light and dark periods. Therefore, the present study encourages the notion that even low APP‐intake may increase the proportions of fatigue‐resistant myofibers, and has promise as a strategy for modifying performance in human sports and improving function in age‐related muscle atrophy.  相似文献

Color variation within populations of the pea aphid influences relative susceptibility to predators and parasites. We have discovered that infection with a facultative endosymbiont of the genus Rickettsiella changes the insects' body color from red to green in natural populations. Approximately 8% of pea aphids collected in Western Europe carried the Rickettsiella infection. The infection increased amounts of blue-green polycyclic quinones, whereas it had less of an effect on yellow-red carotenoid pigments. The effect of the endosymbiont on body color is expected to influence prey-predator interactions, as well as interactions with other endosymbionts.  相似文献

Global climate change will result in extreme environments, such as droughts and floods. We investigated the individual and combined effects of droughts and floods of varying duration on sugarcane (Saccharum spp.) growth using a pot experiment under glasshouse conditions with the following six treatments: drought for 15 d, prolonged drought for 30 d, flood for 15 d, prolonged flood for 30 d, short flood followed by prolonged drought, and prolonged flood followed by prolonged drought. Plants that were subjected to drought conditions, including drought after a flood, had reduced CO2 assimilation (through stomatal closure) and leaf areas, whereas flood conditions showed no effect. During flooding, some roots died, and adventitious roots with well-developed aerenchyma appeared from the submerged nodes. At the time of harvest, there were no significant differences in stem fresh weight, sucrose content, or sugar yield between the treatments. However, ion content analysis revealed that flood conditions caused an accumulation of sodium in the bottom of stems and adventitious roots. Therefore, under flood conditions, plants may develop adventitious roots, which may offset the negative effects of root death, helping them to maintain their growth and yield.  相似文献

This study was conducted to investigate the effects of probiotics administration on fatty acid metabolism in Japanese Black cattle as per changes in blood fatty acid concentrations and blood biochemical tests. Eighteen clinically healthy Japanese Black female fattening cattle bred on the same fattening farm were randomly classified into the probiotics administration group (n=9) or the control group (n=9). In the probiotics administration group, 50 g of probiotics were started per animal per day at the age of 18 months, and the administration period was 2 months from the start date of the study. Blood was collected twice before starting the probiotics administration and at 2 months after starting the probiotics administration. In the probiotics administration group, palmitic, linoleic, arachidonic and α-linolenic acid tended to be higher at the end of the administration compared with those before probiotics administration. Additionally, as a result of multiple comparison test, monounsaturated fatty acids at Post was significantly higher, and the ω6 / ω3 ratio was significantly lower than in the control group. Vitamin A, E and albumin were significantly higher at the end of the administration than in the control group. In this study that administering probiotics to Japanese Black cattle in the late middle stage of fattening period did not have a significant effect on fatty acid metabolism during feed digestion and absorption, but suggested that may alter some blood fatty acids concentrations.  相似文献

To establish vaccine strains of H5 and H7 influenza viruses, A/duck/Hokkaido/Vac-1/04 (H5N1) [Vac-1/04 (H5N1)], A/duck/Hokkaido/Vac-3/07 (H5N1) [Vac-3/07 (H5N1)], and A/duck/Hokkaido/ Vac-2/04 (H7N7) [Vac-2/04 (H7N7)] were generated from non-pathogenic avian influenza viruses isolated from migratory ducks. Vac-1/04 (H5N1) and Vac-3/07 (H5N1) were generated by genetic reassortment between H5N2 or H5N3 virus as an HA gene provider and H7N1 or H6N1 viruses as an NA gene provider. Vac-2/04 (H7N7) was a genetic reassortant obtained using H7N7 and H9 N2 viruses to give high growth character of the H9N2 virus in chicken embryonated eggs. The results of sequence analyses and experimental infections revealed that these H5N1 and H7N7 reassortant viruses were non-pathogenic in chickens and embryos, and had good growth potential in embryonated eggs. These viruses should be useful to develop vaccines against H5 and H7 highly pathogenic avian influenza viruses.  相似文献
