Lupinus albus seeds contain conglutin gamma (Cγ) protein, which exerts a hypoglycemic effect and positively modifies proteins involved in glucose homeostasis. Cγ could potentially be used to manage patients with impaired glucose metabolism, but there remains a need to evaluate its effects on hepatic glucose production. The present study aimed to analyze G6pc, Fbp1, and Pck1 gene expressions in two experimental animal models of impaired glucose metabolism. We also evaluated hepatic and renal tissue integrity following Cγ treatment. To generate an insulin resistance model, male Wistar rats were provided 30% sucrose solution ad libitum for 20 weeks. To generate a type 2 diabetes model (STZ), five-day-old rats were intraperitoneally injected with streptozotocin (150 mg/kg). Each animal model was randomized into three subgroups that received the following oral treatments daily for one week: 0.9% w/v NaCl (vehicle; IR-Ctrl and STZ-Ctrl); metformin 300 mg/kg (IR-Met and STZ-Met); and Cγ 150 mg/kg (IR-Cγ and STZ-Cγ). Biochemical parameters were assessed pre- and post-treatment using colorimetric or enzymatic methods. We also performed histological analysis of hepatic and renal tissue. G6pc, Fbp1, and Pck1 gene expressions were quantified using real-time PCR. No histological changes were observed in any group. Post-treatment G6pc gene expression was decreased in the IR-Cγ and STZ-Cγ groups. Post-treatment Fbp1 and Pck1 gene expressions were reduced in the IR-Cγ group but increased in STZ-Cγ animals. Overall, these findings suggest that Cγ is involved in reducing hepatic glucose production, mainly through G6pc inhibition in impaired glucose metabolism disorders.  相似文献

Quinoa protein concentrate (QPC) was extracted and digested under in vitro gastrointestinal conditions. The protein content of QPC was in the range between 52.40 and 65.01% depending on the assay used. Quinoa proteins were almost completely hydrolyzed by pepsin at pH of 1.2, 2.0, and 3.2. At high pH, only partial hydrolysis was observed. During the duodenal phase, no intact proteins were visible, indicating their susceptibility to the in vitro simulated digestive conditions. Zebrafish larvae model was used to evaluate the in vivo ability of gastrointestinal digests to inhibit lipid peroxidation. Gastric digestion at pH 1.2 showed the highest lipid peroxidation inhibition percentage (75.15%). The lipid peroxidation activity increased after the duodenal phase. The digest obtained at the end of the digestive process showed an inhibition percentage of 82.10%, comparable to that showed when using BHT as positive control (87.13%).  相似文献

 The native soils of Galicia (NW Spain) exhibit a biochemical equilibrium such that total soil N is a function of five biochemical and microbiological parameters: microbial biomass C, mineralized N, phosphomonoesterase, β-glucosidase and urease activities. To investigate whether the ratio of the total N calculated from biochemical soil properties (Nc) and the total N as measured by the Kjeldahl method (Nk; Nc/Nk) can be used as an index of soil quality, we determined these variables and consequently the ratio in three kinds of disturbed soils: an artificially Cu-contaminated soil, two lignite mine soils, and a number of arable soils. In none of the studied soils did the individual biochemical parameters respond consistently to the factors influencing soil quality, but in all cases soil degradation was reflected by the Nc/Nk value, which differed more or less markedly from 100%. Nc/Nk can therefore be used for the rapid evaluation of soil degradation, since it distinguishes among biochemically balanced soils, soils in a transient state of high microbiological and biochemical activity and degraded soils. It can also serve as a reliable basis for the rapid calculation of the "ecological dose" (ED50) of soil pollutants. The effect of three annually consecutive additions of pig slurry at two rates (90 and 150 m3 x ha(-1) x year(-1) on soils and soil humic acids (HAs) was investigated in a field experiment under semiarid conditions. Soils and pig slurries were analyzed by standard methods. The HAs were isolated from soils and pig slurry by a conventional procedure based on alkaline extraction, acidic precipitation to pH 1, purification by repeated alkaline dissolutions and acidic precipitations, water washing, dialysis, and final freeze-drying. The HAs obtained were analyzed for elemental (C, H, N, S, and O) and acidic functional group (carboxylic and phenolic) composition, and by UV-vis, FT-IR, fluorescence, and ESR spectroscopies. With respect to the control soil, the pig slurry amended soils had greater pH and electrical conductivity, slightly larger total N content, and smaller values of C/N ratio. A decrease of total organic C was observed only in soils amended for 2 and 3 years at the higher slurry rate. With respect to control soil HA, pig slurry HA was characterized by larger contents of S- and N-containing groups, smaller acidic functional group and organic free radical contents, a prevalent aliphatic character, extended molecular heterogeneity, and smaller aromatic polycondensation and humification degrees. Amendment with pig slurry HA determines a number of modifications in soil HAs, including increase of C, S, and COOH contents, C/N ratios, and aliphaticity and decrease of extraction yields and N, O, phenolic OH, and organic free radical contents. These effects are generally more evident after the first year of slurry application and tend to disappear with increasing number of treatments. Most probably, over the years the slightly humified slurry HA is mineralized through extended microbial oxidation, whereas only the most recalcitrant components, such as S-containing, phenolic, and aliphatic structures, are partially accumulated by incorporation into soil HA.  相似文献

A novel electronic nose based on solid-phase microextraction (SPME) coupled with a surface acoustic wave (SAW) sensor array has been used to analyze different quality virgin olive oils. A mathematical model was designed with 37 samples to distinguish lampante from the other virgin olive oils categories (extra-virgin and virgin), because lampante-virgin olive oils cannot be consumed without a previous refining process. The model, successfully validated with a test set of 16 samples, was able to classify 90% of the samples correctly. Misclassifications were explained by SPME-HRGC analyses and a second sensory evaluation.  相似文献

The effect of fermentation on the antioxidant compounds [vitamins C and E, total phenolic compounds (TPC), and reduced glutathione (GSH)], and antioxidant capacity [superoxide anion scavenging activity (SOD-like activity), peroxyl radical-trapping capacity (PRTC), inhibition of phosphatidylcholine (PC) peroxidation, and Trolox equivalent antioxidant capacity (TEAC)] of soybean (Glycine max cv. Merit) was studied. Fermentation was carried out in solid state in cracked seeds inoculated with Aspergillus oryzae, Rhizopus oryzae, Bacillus subtilis, and Lactobacillus plantarum and in liquid state either in cracked seeds or milled soybean flours fermented naturally by only the microorganisms present in the seeds or by inoculation with L. plantarum. Vitamin C was not detected in the studied samples. Fermentation caused a decrease in vitamin E activity, except when cracked seed was fermented with A. oryzae, R. oryzae, or B. subtilis that increased 31, 30, and 89%, respectively. Fermentation produced an increase in TPC content and did not affect or reduce the GSH content. Fermentation decreased SOD-like activity drastically, while PRTC increased except when it was carried out naturally in cracked seed. TEAC values rose sharply when soybeans were fermented with B. subtilis. Processed soybean extracts inhibited PC peroxidation in comparison with the control assay. On the basis of the results obtained, the relative contributions of vitamin E, TPC, and GSH to antioxidant capacity were calculated and results showed a very high TPC contribution and a low contribution of GSH and vitamin E activity. Optimum results for functional soybean flours were achieved when fermentation was carried out with B. subtilis inoculum.  相似文献

There are no specifically formulated dry foods for tench (Tinca tinca L.), which forces farmers to use diets formulated for other fish species. This has major drawbacks, such as high mortality, slow growth, and body deformities. A 120 day experiment was performed with five-month-old juvenile tench (initial mean weight: 0.388 g; total length: 31.78 mm) to evaluate decapsulated Artemia cysts as a supplement to a dry diet for other fish species. Three treatments, differing in the daily supplement, were tested: 1,800 freshly hatched nauplii, 1,800 cysts, and 300 cysts per g of tench biomass. Final survival ranged between 95.3% and 97.9%. Juvenile tench that received the supplement of 1,800 decapsulated Artemia cysts had a specific growth rate (1.28), weight (1.83 g), and total length (52.30 mm) significantly higher than those with the same amount of nauplii. The lowest supplement (300 cysts/g of fish biomass) allowed significantly lower growth and higher condition coefficient (1.40) than the rest. Animals with body deformities (1.06%) were only recorded in the groups that received the lowest cyst supplement. Results showed Artemia cysts are a suitable dietary supplement for juvenile tench, being an advantageous alternative to live nauplii.  相似文献

Recent advances in crayfish feeding have enabled the development of size grading studies from the start of first‐feeding. A 180‐day experiment aimed at intensive rearing of Pacifastacus leniusculus was carried out under controlled conditions, evaluating the effects of size grading at two different periods from the onset of exogenous feeding. Stage 2 juveniles were stocked in fibreglass tanks at a density of 100 m2, and fed a dry diet for salmonids combined with restricted amounts of Artemia cysts. Five groups were tested: no grading, grading at 60 days (large and small size) and grading at 100 days (large and small size). After 6 months, no significant differences were found in the survival among groups (mean: 73.06%). The highest final growth (pooled results from upper and lower classes: 17.39 mm carapace length, 1.43 g weight) was achieved by the crayfish sorted at 60 days, showing significant differences from the ungraded group. Smaller crayfish graded at 60 days grew significantly faster than smaller crayfish graded at 100 days. The food conversion ratio was lower in the graded groups (mean: 2.64), showing significant differences from the ungraded group (3.23). This study shows that size grading allows a better performance and an improved feeding efficiency.  相似文献
