Summary Plant viruses cause considerable losses to crops and none of the three current approaches to disease control are likely to give the long-term answer. The new concept of non-conventional protection, in which the expression of a viral or virus-related sequence in the plant genome interferes with the virus infection cycle, holds considerable promise for designing new resistance or protection genes. A series of targets in the viral genome is identified and a range of mechanisms for attacking those targets is discussed. There are several current systems which have been proved to give protection, at least to a certain extent, and many systems which are being researched upon for the future. Some of these are described to give a picture of the current situation and of the thinking for the future. The problems of field deployment of the transgenic plants are discussed, especially those associated with the risk to the environment. Various questions which molecular biologists and plant breeders will have to consider include what are desirable characters to have in protection genes and how these new genes should be deployed.  相似文献

High plasma vascular endothelial growth factor (VEGF) concentrations are associated with radiation resistance and poor prognosis. After an exposure to ionizing radiation in cell culture an early phase and a late phase of increased VEGF have been documented. The activation was dependent on the radiation dose. Therefore, the purpose of this study was to measure baseline plasma VEGF and changes in VEGF over the course of fractionated radiation therapy in dogs with spontaneous tumors. Dogs with tumors had a significantly higher pretreatment plasma VEGF than did dogs without tumors. Immediately after irradiation no increased plasma VEGF was observed. Over the course of radiation therapy there was an increased plasma VEGF in dogs treated with low doses per fraction/high total dose, whereas plasma VEGF remained stable in dogs irradiated with high doses per fraction/low total dose. The regulatory mechanisms are very complex, and therefore the value of plasma VEGF measurements as an indirect marker of angiogenesis induced by radiotherapy is limited.  相似文献

Botrytis cinerea causes gray mold disease and affects hundreds of plant species, including tomato (Lycopersicon esculentum). The wild nightshade, Solanum lycopersicoides, is cross compatible with tomato and is more resistant to B. cinerea, thus representing a potential source for crop improvement. Tests involving droplet inoculation of detached leaves and spray inoculation of entire seedlings demonstrated that resistance to B. cinerea varies among S. lycopersicoides accessions, with S. lycopersicoides LA2951 being the most resistant accession tested. Expression of resistance in the intergeneric hybrid (L. esculentum cv. 'VF36' × S. lycopersicoides LA2951) suggested that resistance is at least partially dominant in tomato. A green fluorescent protein-tagged B. cinerea strain was used for confocal microscopic comparison of infection in leaves of S. lycopersicoides and tomato. Even though S. lycopersicoides supported spore germination, there was evidence for hyphal lysis and death 3 days after inoculation, at a time when lesions were expanding on susceptible tomato plants. The reduced frequency of B. cinerea lesion spread on S. lycopersicoides explains why this fungus produced fewer spores in this wild nightshade than in tomato.  相似文献

Signs of severe otitis media in 20% of dairy calves on one farm were associated with Mycoplasma bovis infection, based on isolation from the external ear canal and nares. Affected calves seroconverted to M. bovis and no other significant bacteria were isolated. Infection was considered likely to have originated from cows in the milking herd based on evidence of seroconversion and detection of infection in a milk sample. M. bovis infection should be considered when investigating otitis problems in calves.  相似文献

The objective of this study was to determine whether mosquitoes, Aedes vexans (Meigen), could serve as biological vectors of porcine reproductive and respiratory syndrome virus (PRRSV). Specifically, the study assessed the duration of viability and the site of PRRSV within mosquitoes, and evaluated whether PRRSV could be transmitted to a susceptible pig by mosquitoes following a 7- to 14-day incubation period after feeding on an infected pig. For the first experiment, a total of 100 mosquitoes were allowed to feed on a pig, experimentally infected with PRRSV (day 7 post-inoculation) and were then maintained alive under laboratory conditions. A set of 10 mosquitoes were collected at 0 hour (h), 6 h, 12 h, 24 h, 48 h, 72 h, 5 days (d), 7 d, 10 d, and 14 d post-feeding (pf). Samples of exterior surface washes, salivary glands, thorax carcasses, and gut homogenates were collected from each set of mosquitoes, and tested for PRRSV. Infectious PRRSV was detected by polymerase chain reaction and swine bioassay only from the gut homogenates of mosquitoes collected at 0 h and 6 h pf. For the second experiment, a total of 30 mosquitoes were allowed to feed on a pig, experimentally infected with PRRSV and the mosquitoes were then maintained under laboratory conditions. On each of day 7, 10, and 14 pf, a set of 10 mosquitoes were allowed to feed on a susceptible pig. Transmission of PRRSV to susceptible pigs did not occur, and PRRSV was not detected from the mosquitoes. These findings indicate that mosquitoes are not likely to serve as biological vectors of PRRSV.  相似文献

The new growth retardants mepiquat chloride (a quaternary ammonium compound of the piperidinium type, coded BAS 083 00W) and a ternary sulphonium carbamate (coded BTS 44 584) were compared with ancymidol (A-Rest or Reducymol) and chlormequat chloride (Cycocel) for their ability to dwarf virus-free and virus-infected (ordinary) Mid-Century Hybrid lily cultivar ‘Enchantment’. Single compost drenches were given 3 weeks after transferring plants to the glasshouse.Compared with the virus-infected control plants, virus-free controls were 62% taller and 33% greater in leaf spread; floret number was approximately doubled and floret diameter was increased by 12%, but flowering was delayed by 6.5 days. The length of the lower stem bearing dead leaves was 15% of the total in virus-infected controls and 3% of the total in virus-free controls.Ancymidol, chlormequat chloride and mepiquat chloride were all effective growth retardants, and were without adverse side-effects. On a concentration basis, mepiquat chloride was 2–2.5 times as active as chlormequat chloride in reducing stem extension, but ancymidol was several orders of magnitude more active than both those compounds. With BTS 44 584, the maximum reduction in stem length obtained was 16% compared to controls. The decreases in stem extension were more pronounced, in both percentage and absolute terms, in the virus-free bulbs. Nevertheless, for a given retardant treatment, plants of the virus-free stock remained taller than those of the virus-infected stock. As a result, 2–3 times the amount of an active retardant was required by virus-free than by virus-infected plants in order to produce similar stem lengths at flowering. Effects of the retardants on the time of flowering, flower number, floret diameter and leaf spread were small, but increasing concentrations of all chemicals progressively increased the proportion of the stem bearing dead leaves.The effects of ancymidol, chlormequat chloride and mepiquat chloride persisted in the year after treatment.  相似文献

Sheep and rabbit antisera were produced against lysates of E. coli strain 711 (pVir). This K-12 strain carries the Vir plasmid which codes for Cytotoxic Necrotizing Factor type 2 (CNF2). Immunoglobulin G (IgG) fractions of both immune sera were subsequently purified by a two-step precipitation method. To increase the specificity for CNF2, the sheep IgG preparation was extensively adsorbed against both a sonicated extract of isogenic K-12 strain 711 and intact phenol-treated cells of vaccine strain 711 (pVir). An enzyme-linked immunosorbent assay (ELISA) was developed to detect clinical isolates of E. coli producing CNF2, using the final preparations of rabbit and sheep IgG in a double sandwich technique. The results obtained with this CNF2-ELISA were compared to those obtained with the conventional HeLa cell cytotoxicity assay. The testing of 133 E. coli strains (49 CNF2 positive strains and 84 negative strains) resulted in no false-negative and no false-positive. Therefore, the CNF2-ELISA offers a good alternative to the HeLa cell culture assay for the detection of CNF2-producing strains where facilities for and experience with cell cultures is lacking.  相似文献

A systems approach has been taken to a review of agricultural education programmes and as the essential theme of resultant curricula at Hawkesbury Agricultural College in Australia. The systems thinking and practices which have guided, and been shaped by, the innovations are outlined, and the rationale and framework of the major programme are described. The subsequent emphasis has been placed on effective learning for agricultural managers and their technologist advisors. It is argued that problem solving and learning are essentially the same psychological processes and that taking a systems approach to investigating problem situations provides a more useful paradigm for learning about agriculture than reductionist, discipline-based approaches. Experiential learning and autonomy in learning are seen as consistent with this and are basic features of the programmes. A conceptual framework for problem solving that incorporates soft and hard systems and scientific reductionist methodologies has been developed. A contingency approach to situation improving is emerging as a less restrictive and more realistic alternative to a normative approach to problem solving.  相似文献
