Agricultural soils are important sources of greenhouse gases (GHGs). Soil properties and environmental factors have complex interactions which influence the dynamics of these GHG fluxes. Four arable and five grassland soils which represent the range of soil textures and climatic conditions of the main agricultural areas in the UK were incubated at two different moisture contents (50 or 80% water holding capacity) and with or without inorganic fertiliser application (70 kg N ha−1 ammonium nitrate) over 22 days. Emissions of N2O, CO2 and CH4 were measured twice per week by headspace gas sampling, and cumulative fluxes were calculated. Multiple regression modelling was carried out to determine which factors (soil mineral N, organic carbon and total nitrogen contents, C:N ratios, clay contents and pH) that best explained the variation in GHG fluxes. Clay, mineral N and soil C contents were found to be the most important explanatory variables controlling GHG fluxes in this study. However, none of the measured variables explained a significant amount of variation in CO2 fluxes from the arable soils. The results were generally consistent with previously published work. However, N2O emissions from the two Scottish soils were substantially more sensitive to inorganic N fertiliser application at 80% water holding capacity than the other soils, with the N2O emissions being up to 107 times higher than the other studied soils.  相似文献

OBJECTIVE: To describe the microanatomic features of pancreatic islets and the immunohistochemical distribution of glucose transporter (GLUT) molecules in the pancreas and other tissues of New World camelids. ANIMALS: 7 healthy adult New World camelids, 2 neonatal camelids with developmental skeletal abnormalities, and 2 BALB/c mice. PROCEDURE: Samples of pancreas, liver, skeletal muscle, mammary gland, brain, and adipose tissue were collected postmortem from camelids and mice. Pancreatic tissue sections from camelids were assessed microscopically. Sections of all tissues from camelids and mice (positive control specimens) were examined after staining with antibodies against GLUT-1, -2, -3, and -4 molecules. RESULTS: In camelids, pancreatic islets were prominent and lacked connective tissue capsules. Numerous individual endocrine-type cells were visible distant from the islets. Findings in neonatal and adult tissues were similar; however, the former appeared to have more non-islet-associated endocrine cells. Via immunostaining, GLUT-2 molecules were detected on pancreatic endocrine cells and hepatocytes in camelids, GLUT-1 molecules were detected on the capillary endothelium of the CNS, GLUT-3 molecules were detected throughout the gray matter, and GLUT-4 molecules were not detected in any camelid tissues. Staining characteristics of neonatal and adult tissues were similar. CONCLUSIONS AND CLINICAL RELEVANCE: In New World camelids, microanatomic features of pancreatic islets are similar to those of other mammals. Data suggest that the poor glucose clearance and poor insulin response to hyperglycemia in adult camelids cannot be attributed to a lack of islet cells or lack of GLUT molecules on the outer membrane of those cells.  相似文献

OBJECTIVE: To assess serum 17-alpha-hydroxyprogesterone (17OHP) and corticosterone concentrations in dogs with nonadrenal neoplasia and dogs being screened for hyperadrenocorticism. DESIGN: Prospective study. ANIMALS: 16 clinically normal dogs, 35 dogs with nonadrenal neoplasia, and 127 dogs with suspected hyperadrenocorticism. PROCEDURE: ACTH stimulation tests were performed in all dogs. Baseline serum cortisol and corticosterone concentrations were measured in the healthy dogs; baseline serum cortisol concentration and ACTH-stimulated cortisol, corticosterone, and 17OHP concentrations were measured in all dogs. Endogenous plasma ACTH concentration was also measured before administration of ACTH in dogs with neoplasia. RESULTS: In 35 dogs with neoplasia, 31.4% had high serum 17OHP concentration and 22.9% had high serum corticosterone concentration. Of the 127 dogs with suspected hyperadrenocorticism, 59 (46.5%) had high ACTH-stimulated cortisol concentrations; of those, 42 of 59 (71.2%) and 32 of 53 (60.4%) had high serum 17OHP and corticosterone concentrations, respectively. Of dogs with serum cortisol concentration within reference range after ACTH administration, 9 of 68 (13.2%) and 7 of 67 (10.4%) had high serum 17OHP and corticosterone concentrations, respectively. In the dogs with neoplasia and dogs suspected of having hyperadrenocorticism, post-ACTH serum hormone concentrations were significantly correlated. CONCLUSIONS AND CLINICAL RELEVANCE: Serum concentrations of 17OHP or corticosterone after administration of ACTH may be high in dogs with nonadrenal neoplasia and no evidence of hyperadrenocorticism. Changes in serum 17OHP or corticosterone concentrations after administration of ACTH are proportionate with changes in cortisol concentration.  相似文献

Pathogenicity data from surveys of Puccinia triticina (formerly P. recondita f. sp. tritici) conducted in western Europe in 1995 were analysed to compare the structure of regional populations of the pathogen. Many of the populations differed in phenotypic diversity and pathotypic composition, even though they occurred within a single epidemiological unit, suggesting that local factors may influence the establishment and propagation of individual pathotypes in the regional populations. Neighbouring regions were more similar than distant regions, and all regions shared at least one pathotype, except populations in northern Italy and Scotland. A high degree of similarity was found between populations in northern France and Great Britain, providing strong evidence of free movement of inoculum between these regions. Resistance genes were postulated for a selection of 91 wheat cultivars, representing those most commonly grown in western Europe in 1995. Thirteen cultivars lacked detectable seedling resistance genes and the remaining 78 possessed from one to three resistance genes; those detected were Lr1, Lr3a, Lr10, Lr13, Lr14a, Lr17b, Lr20, Lr26 and Lr37. The most commonly detected resistance gene was Lr13, which was present singly or in combination with other resistance genes in 48 cultivars (53%). The gene Lr14a was detected in 18 cultivars, Lr26 was present in 16 cultivars. The role of host selection in the composition of the regional populations of P. triticina in western Europe in 1995 was difficult to assess on the basis of the results obtained, since virulence data were not available for Lr13 and Lr14a. Genetics aims to understand the relation between genotype and phenotype. However, because complete deletion of most yeast genes ( approximately 80%) has no obvious phenotypic consequence in rich medium, it is difficult to study their functions. To uncover phenotypes for this nonessential fraction of the genome, we performed 1144 chemical genomic assays on the yeast whole-genome heterozygous and homozygous deletion collections and quantified the growth fitness of each deletion strain in the presence of chemical or environmental stress conditions. We found that 97% of gene deletions exhibited a measurable growth phenotype, suggesting that nearly all genes are essential for optimal growth in at least one condition.  相似文献

Forest systems cover more than 4.1×109 ha of the Earth's land area. The future response and feedbacks of forest systems to atmospheric pollutants and projected climate change may be significant. Boreal, temperate and tropical forest systems play a prominent role in carbon (C), nitrogen (N) and sulfur (S) biogeochemical cycles at regional and global scales. The timing and magnitude of future changes in forest systems will depend on environmental factors such as a changing global climate, an accumulation of CO2 in the atmosphere, and increase global mineralization of nutrients such as N and S. The interactive effects of all these factors on the world's forest regions are complex and not intuitively obvious and are likely to differ among geographic regions. Although the potential effects of some atmospheric pollutants on forest systems have been observed or simulated, large uncertainty exists in our ability to project future forest distribution, composition and productivity under transient or nontransient global climate change scenarios. The potential to manage and adapt forests to future global environmental conditions varies widely among nations. Mitigation practices, such as liming or fertilization to ameliorate excess NOx or SOx or forest management to sequester CO2 are now being applied in selected nations worldwide.The U.S. Government's right to a non-exclusive, royalty free licence in and to any copyright is acknowledged.  相似文献

Grain compositional components impacting barley (Hordeum vulgare L.) use in food, feed and fuel products, must be combined with improved agronomic traits to produce a commercially viable barley cultivar. Little current information is available on grain composition and variability among winter barley genotypes. This study was conducted to determine the variability among modern hulled and hulless winter barley genotypes in grain composition. Barley types varied significantly in grain and kernel weight, starch, beta-glucan, oil and ash content, but not in protein concentration. Hulless barley had significantly higher grain test weight and starch concentration than hulled and malting types, and significantly higher beta-glucan than malting barley. Hulless barley had significantly lower kernel weights, oil, and ash concentrations than hulled and malting types. Higher starch and lower fiber and ash in grain of hulless barley versus hulled feed or malting type barley are characteristics that increase hulless barley desirability. Selection for high starch concentration among all barley types is feasible and will facilitate development of barley cultivars better suited for use in feed, malt, and ethanol production.  相似文献

Tephritid fruit flies are serious economic pests worldwide. As larvae, they feed and develop within the pulp of host fruits, making infestation difficult to detect by visual inspection. At U.S. ports of entry, incoming produce shipments are checked for infestation by manually cutting open a small sample of fruit and searching for tephritid larvae. Consequently, there is a need for more sensitive, high-throughput screening methods. This study evaluated gas chromatography (GC) as a potential technology for improved detection of hidden infestation. Grapefruits (Citrus × paradisi Macfad.) infested with immature stages of the Caribbean fruit fly Anastrepha suspensa (Loew) (Diptera: Tephritidae) were examined to determine if infested fruit emitted a chemical profile distinct from that of non-infested fruit. Peaks identified by GC analysis were grouped into three classes. Chemicals detected in similar quantities in all samples, or slightly elevated in infested samples, were regarded as non-diagnostic background volatiles. Chemicals highly elevated after oviposition, during the last instar exit stage, and in experimentally-pierced fruit were interpreted to be indicators of citrus peel injury, and included d-limonene and β-ocimene. Chemicals elevated exclusively in the larval infestation stages were considered indicators of feeding damage and potentially diagnostic of infestation, and included hexyl butanoate and an unidentified compound. The peaks associated with injury and feeding were also detectable with a portable ultra-fast GC analyzer that required less than 80 s per sample. Further studies will investigate the potential application of these results for development of a rapid, non-destructive screening method for detection of tephritid infestation.  相似文献

Hexaploid triticale (X Triticosecale Wittmack) (2n= 6x= 42, AABBRR) and wheat (Triticum aestivum L.) (2n= 6x= 42, AABBDD) differ in their R and D-genomes. This produces differences in both agronomic and end-use quality characteristics. Our objective was to determine how introgressions of individual chromosomes from the D-genome of wheat affect these characteristics of a winter triticale 'Presto'. We studied the effects of 18 D-genome chromosome substitution lines, 15 sib-lines as controls, and five check cultivars at Lincoln, NE in 1996, using a randomized complete block design with two replications. The experiment was repeated at Lincoln and Mead, NE in 1997 and 1998 with 15 substitution lines that survived the first winter in Lincoln, along with their 12 control sibs and five check cultivars. Few D-genome chromosomes had positive effects. Chromosomes 2D, 4D, and 6D significantly reduced plant height when substituted for 2R, 4B, and 6R, respectively. No grain yield increases were associated with any of the D-genome chromosomes tested, but three substitutions decreased the grain yield. Depending on the allele of the hardness gene present, chromosome 5D increased or decreased kernel hardness when substituted for 5R or 5A, respectively. Introgressions of chromosomes 1D and 6D improved end-use quality characteristics of Presto. These results suggest that apart from beneficial effects of individual loci located on the D-genome chromosomes, no major benefit can be expected from D-genome chromosome substitutions.  相似文献
