Citrus postbloom fruit drop (PFD) is caused by Colletotrichum acutatum and C. gloeosporioides. These pathogens attack the flowers and cause premature fruit drop and the retention of fruit calyces. This study was designed to characterize the spatial and temporal dynamics of PFD in commercial citrus‐growing areas to better understand the disease spread. Experiments were carried out in three young orchards (500 trees each) in two municipalities in Sao Paulo State, Brazil. Symptoms of PFD on the flowers and presence of persistent calyces were assessed in each of three orchards for three years. Logistic, Gompertz and monomolecular models were fitted to the incidence data over time from the trees with symptoms. The spatial pattern of diseased trees was characterized by a dispersion index and by Taylor′s power law. An autologistic model was used for the spatiotemporal analysis. The logistic model provided the best fit to the disease incidence data, which had a fast progress rate of 0·53 per day. During the early epidemic of PFD, the spatial pattern of diseased trees was random, which suggested that inoculum spread was due to mechanisms other than rain splash. As the disease incidence increased (up to 12·6%), the spatial pattern of diseased trees became aggregated. The rapid rate of disease progress and the distribution of PFD suggest that dispersal of the pathogen is possibly related to a mechanism other than splash dispersal, which is more typical of other fruit diseases caused by Colletotrichum spp.  相似文献

The aim of this study was to investigate changes in defence compounds of common bean cultivars with different levels of resistance to the fungus Sclerotinia sclerotiorum and determine the relation of the compounds to pathogen tolerance. The lines were inoculated with the pathogen and assessed for enzymatic and non-enzymatic parameters related to plant defence: peroxidases (POX), polyphenol oxidases (PPO), catalase (CAT), superoxide dismutase (SOD) and ascorbate peroxidase (APX), total soluble phenol and lignin contents. Stem tissue samples were collected from two regions of the plant for biochemical analyses. Stem tissue samples were collected from two regions of the plant for biochemical analyses. In the position one, 5 cm of the stem was collected from the region with necrosis caused by the pathogen, and in the position two, 5 cm of the stem was collected from the end of the position one at the times of 12, 24, 48, 72, 96 and 120 h after inoculation (HAI). Greater lignin and total soluble phenol contents and greater induction of POX and SOD activity in inoculated plants in the region near the inoculation (position one) indicate local activation with later signalling for activation of defence mechanisms in other regions of the plant. The genotype with a greater level of resistance was superior to the susceptible one in regard to lignin production and the activities of POX, APX and SOD defence enzymes. These results suggest that a combination of these defence responses in common bean may contribute to greater plant resistance to the pathogen and that these enzymes have potential use in selection of common bean genotypes.  相似文献

In this study, colonization and disease development of axenically-grown tomato plants by Clavibacter michiganensis subsp. michiganensis (Cmm), the causative agent of bacterial wilt and canker, was investigated. For this, a spontaneous rifampicin resistant strain of Cmm was tagged with a marker that expressed a green fluorescent protein (GFP) in a stable way and which possessed a similar virulence to the parental strain. In vitro plants were drop-inoculated at the stem base and the population dynamics was determined by dilution pour-plating in a selective medium. At 3 h after inoculation, Cmm was already present in low densities in roots, stems and leaves. At 16 dpi, Cmm was found throughout the entire plant in high densities of ca. 1010 cfu g?1. Symptoms developed in the in vitro plants typical for Cmm, such as canker, wilting and growth reduction. The presence of Cmm in vascular and parenchymatic tissue of in vitro tomato plants was confirmed by epifluorescence stereo- and confocal laser scanning microscopy. This study showed that in vitro tomato plants can be effectively used for detailed studies on interactions between Cmm and its host, in particular if a GFP-tagged strain of the pathogen is used.  相似文献

The partitioning behavior of the glutenin proteins was evaluated in aqueous two-phase systems (ATPSs) formed by sulfate salts (lithium or sodium) and poly(ethylene glycol) (PEG) with average molar mass of 1500 g mol−1 or 4000 g mol−1. The partition coefficients for the glutenin proteins in each ATPS were investigated as a function of the temperature (278.2 K–318.2 K), tie line length (TLL) and electrolyte nature. In all ATPS, the majority of glutenin proteins spontaneously concentrate in the polymer-rich phase (Kp > 1). The partition coefficient is very dependent on the salt nature and the ATPS formed by PEG + lithium sulfate presents higher Kp values as compared with the ATPS formed PEG + sodium sulfate. An increase of molar mass of polymer promotes a decrease of Kp. Thermodynamic parameters of transfer (ΔtrG, ΔtrH and ΔtrS) were obtained by the application of the Van’t Hoff equation (VHE). The values obtained by VHE indicate that the transfer of glutenin proteins to the polymer-rich phase has an enthalpic origin.  相似文献

Best Linear Unbiased Prediction (BLUP) has become the most widely used method for genetic assessment of animal and perennial species, and it is potentially relevant for annual crops. The objective of this study was to assess this method for selection within non-inbred families in recurrent breeding programs. The ‘animal model’ was fitted. The data were expansion volume (EV) and grain yield of plants in recombination plots of two to three selection cycles in the popcorn population Viçosa, with half- and full-sib progenies. The ASReml program was used to perform the analyses. For both EV and yield the breeding values predicted from the additive and additive-dominant models were highly correlated. Multi-generation BLUP was, in general, more accurate than single-generation analysis. These two methods resulted in highly correlated predicted breeding values. The dominance genetic values predicted from the single- and multi-generation analysis were also highly correlated. The pedigree information reduced the percentage of coincidences among the selected individuals relative to phenotypic selection mainly in the population structured in half-sib families. Based on breeding values predicted by BLUP analysis, the most efficient selection procedure was mass selection.  相似文献

The present study investigated the growth performance of juvenile barramundi Lates calcarifer (Bloch) fed conspecific prey (CP), formulated diet (FD) and a mix of both (MIX) using a bioenergetics approach. Fish in the MIX treatment ingested significantly more CP than FD by dry mass. However, prey mass ingestion and cannibalism rate in the MIX treatment were significantly lower than in the CP treatment. This indicates that the provision of alternative food does not complete mitigate cannibalism, but it can significantly reduce cannibalism in barramundi. Fish in the FD treatment showed a significantly higher daily food intake than fish in the CP or MIX treatments. However, fish fed conspecifics showed significantly better feed conversion efficiencies, apparent digestibility rates and growth performances. Exclusive cannibalistic barramundi assimilated significantly more energy consumed, with 1.5% of energy lost in faeces comparing with 7.3% of siblings feeding exclusively on FD. Consequently, exclusive cannibals channelized more energy consumption into growth (57.1%) than those fed solely on FD (43.9%). Therefore, high energy allocation into growth promoted better feed conversion efficiency and growth performance of cannibalistic barramundi than siblings fed solely on FDs. This study implies that fast‐growing cannibals may continuously prey on slow‐growing conspecifics due to growth advantage through cannibalism. Therefore, size heterogeneity should be reduced at initial stocking and controlled as fish grow to avoid the emergence of new cannibals.  相似文献

The Amazon River prawn Macrobrachium amazonicum is endemic to tropical South America and is being intensively exploited by artisanal fisheries in Brazil. Limited information is available about the nutritional requirements of M. amazonicum, although the production of this species is now technically feasible. The digestive process in this species is still unclear and investigation into the digestive cells of its hepatopancreatic epithelium is required. Thus, the hepatopancreas from 15 specimens were fixed in Karnovsky Solution and processed for Transmission Electron Microscopy. Our results indicate that E cells are located at the distal portion of the hepatopancreatic tubule and are involved in mitotic activity. The cylindrical R cells are sparse and are only found in the proximal portion of the hepatopancreatic tubule. According to its ultrastructural characteristics, this cell is involved in pinocytosis. M cells are generally found near the R cells. The F cells are scattered throughout the length of the hepatopancreatic tubules, and B cells are observed mainly in the proximal and middle regions. F cells and B cells are likely related to, respectively, the synthesis of enzymes and the intracelular digestion. R‐ and M cells are probably related to material storage. Thus, these findings provide basic information on the cell types that perform protein digestion in M. amazonicum, and will be useful in further nutritional research. The identification and characterization of digestive cells is an important step towards understanding the digestive mechanisms.  相似文献

Hypothetically, humic substances (HSs) can improve the response of plants in sandy soils. The objective was to assess the effect of applying an organomineral fertilizer enriched with HSs on soybean plants grown with and without water stress. The experimental design was entirely randomized, in a greenhouse, in a 2 × 5 factorial setup (two moisture levels and five fertilizer doses: 0, 1, 2, 4, and 8 mL dm?3), for a total of 10 treatments, with eight repetitions, using as substrate a Psamment. The organomineral fertilizer was applied in the soil 21 days after plant emergence, and the water regimes were established 1 week thereafter. Plant height, shoot and root dry mass, mineral nutrition, and grain yield were evaluated. There was a positive response to the doses, with the most efficient one under water stress being 6.5 mL dm?3 and HSs favored the uptake of micronutrients.  相似文献

Virus-free supernatants of cultured swine monocytes infected by African swine fever virus (ASFV) suppressed in vitro proliferation of porcine and human blood mononuclear cells in response to phytohemagglutinin and the in vivo primary immune response of C57BL/6 mice against sheep RBC. The supernatants were fractionated by discontinuous ion-exchange chromatography and subfractionated by double-step preparative isoelectric focusing. The pool of the most purified active subfractions (F5'EP-ASFV) is made up of heat-unstable material, can be stained by silver nitrate, and has an isoelectric point of 3.88, a maximal optical density at 280 nm, and a mass of 36,000 daltons. In vivo kinetic studies in nonimmunized C57BL/6 mice were performed on days 1, 2, 3, 5, and 7 after injection with 50 micrograms of F5'EP-ASFV protein. Compared with the untreated mice, the treated mice had a noticeable increase in nonspecific immunoglobulin-secreting splenic plaque-forming cells (PFC) with the following isotype profile: IgG2a greater than IgG2b greater than IgG3 greater than IgG1 congruent to IgM. Three days after treatment with the active material, specific IgM PFC against sheep RBC increased up to 23-fold. In C57BL/6 mice immunized against sheep RBC 2 days after treatment with F5'EP-ASFV, the increase in nonspecific PFC was followed by a suppression of specific PFC response in the respective isotype. When C57BL/6 mice were treated after priming with sheep RBC, however, there was little or no suppression of specific PFC and the increase in nonspecific PFC was considerably lower than that in the other F5'EP-ASFV-treated mice. In this case, kinetic curves of specific vs nonspecific PFC of each isotype were mirror images. Mice treated with 200 micrograms of F5'EP-ASFV protein died with hemorrhagic diastasis.  相似文献
