The immunological biomarkers profiles were evaluated using Luminex as putative measures to monitor canine mammary carcinomas (MCs). Forty female dogs were categorized into benign mixed tumour (MC‐BMT = 28) and mammary carcinoma (MC=12). The ascendant biomarker signatures were used to compare the groups. For example, a higher frequency of MC‐BMT animals producing IL‐6, CXCL‐8 and CXCL‐10 was observed, whereas for the MC group IL‐2 and CXCL‐8 were detected. MC‐BMT animals without metastasis had an increase in the levels of IL‐2, CXCL‐8, CXCL‐10, IL‐6, TNF‐α, IL‐15 and a decrease in IL‐10 and CXCL‐8. MC‐BMT animals with metastasis showed only an increase in CXCL‐10 and a decrease in IL‐18. After comparing the ascendant signatures following the presence of metastasis in both groups, a higher frequency of dogs exhibiting IL‐10 production was observed. Pearson correlation (P = 0.0273) and receiver operating characteristic (ROC) curve analysis revealed that this pattern was associated with worse outcome and lower survival rates in MC animals.  相似文献

The diagnosis of acute babesiosis by direct examination of blood smears has some limitations and the indirect serological methods currently in use are designed for detection of IgG, which may not be detectable at an early stage of infection. There is a need, therefore, for rapid and reliable procedures to diagnose acute infections. An ELISA system using a crude antigenic preparation of Babesia bovis was standardized for the detection of IgM antibodies. Optimal dilutions of the antigen, using positive and negative reference sera, were determined by checkerboard titrations. Serum samples of cattle imported from tick-free areas collected before and during an immunization process were used to validate the tests. The specificity was 94% and sensitivity 100%. Specific IgM antibodies against B. bovis first appeared on the 11th day post-inoculation (p.i.) in animals infested with Boophilus microplus ticks and on the 19th day p.i. in animals which had been inoculated with infected blood. Antibody titers decreased after Day 33; however, all animals remained positive until the end of the experiment (124 days).  相似文献

This study aimed to determine whether using inoculants and re-ensiling in sorghum silages affect the intake and feeding behaviour, apparent digestibility, digestible energy, and nitrogen balance of sheep in maintenance. Half the sorghum was inoculated on the day of harvest, and the other half received the same inoculation volume but with water. Sorghum was ensiled in 100 experimental metal drum silos of 200 L covered with plastic: 50 silos with inoculants and 50 without them. After 56 days, 25 drums with inoculants and 25 without were exposed to air for 48 h. The other silos remained closed. The treatments were arranged in a 2 × 2 factorial scheme with inoculation and re-ensiling. The sheep received the silages after 211 days of ensiling and 155 days of re-ensiling to determine the intake, apparent digestibility, and feeding behaviour of animals. The experimental design was a simultaneous double 4 × 4 Latin square. The re-ensiling process increased silage pH, N-NH3.TN−1, butyric acid, and propionic acid. The inoculant increased propionic acid in ensiled and re-ensiled silages. Lactic acid concentration presented a statistical interaction with the inoculated and ensiled silage, 58% higher than the other treatment averages. The ADFap digestibility showed a complex interaction, in which the control ensiled and the inoculated and re-ensiled silages were about 35% lower than the inoculated ensiled silage. The Nbal:Nint ratio (g.g−1) in the inoculated and re-ensiled silage had lower nitrogen retention than intake compared with the other treatments. These results indicate that exposing inoculated silages to air does not compromise their use in sheep feeding.  相似文献

The physiological responses of sugarcane (Saccharum officinarum L.) to oxidative stress induced by methyl viologen (paraquat) were examined with respect to photochemical activity, chlorophyll content, lipid peroxidation and superoxide dismutase (SOD) and ascorbate peroxidase (APX) activities. Thirty-day-old sugarcane plants were sprayed with 0, 2, 4, 6 and 8 mM methyl viologen (MV). Chlorophyll fluorescence was measured after 18 h and biochemical analyses were performed after 24 and 48 h. Concentrations of MV above 2 mM caused significant damage to photosystem II (PSII) activity. Potential and effective quantum efficiency of PSII and apparent electron transport rate were greatly reduced or practically abolished. Both chlorophyll and soluble protein contents steadily decreased with MV concentrations above 2 mM after 24 h of exposure, which became more pronounced after 48 h, achieving a 3-fold decrease. Insoluble protein contents were little affected by MV. Oxidative stress induced by MV was evidenced by increases in lipid peroxidation. Specific activity of SOD increased, even after 48 h of exposure to the highest concentrations of MV, but total activity on a fresh weight basis did not change significantly. Nondenaturing PAGE assayed with H2O2 and KCN showed that treatment with MV did not change Cu/Zn-SOD and Mn-SOD isoform activities. In contrast, APX specific activity increased at 2 mM MV but then dropped at higher doses. Oxidative damage induced by MV was inversely related to APX activity. It is suggested that the major MV-induced oxidative damages in sugarcane leaves were related to excess H2O2, probably in chloroplasts, caused by an imbalance between SOD and APX activities, in which APX was a limiting step. Reduced photochemical activity allowed the early detection of the ensuing oxidative stress.  相似文献

The seasonal variation of the tick Boophilus microplus in the `Regiaõ Metalúrgica' region of Minas Gerais state was studied using 26 Bos taurus (Holstein)×Bos indicus (Gir) calves, of which 16 animals were 3/4 Holstein×1/4 Gir and 10 were 3/4 Gir×1/4 Holstein. Monthly tick counts were made between March 1996 and March 1998 and, although B. microplus infestations occurred throughout the year, they were greater during the rainy season. Rainfall was the climatic factor that most affected the seasonal variation. The animals with a preponderance of Holstein blood presented infestations that were significantly (p<0.05) greater than those of the other cross. Five of the animals (19.2%) maintained 45.5% of the parasite load of the entire group.  相似文献

Histidine plays an important role in haemoglobin synthesis, antioxidant defence system and stress resistance, and it is thought to be a limiting amino acid in most meagre diets. In this study, two different diets, a control and a diet supplemented with 3% histidine, were tested in 60‐day‐old meagre (initial weight—6.2 g) for 82 days. Survival varied between 92% and 94%, and neither SGR nor whole fish composition seemed to be affected by the diet. When fish were submitted to a stress test (outside water for 30 s), it was observed that the levels of glucose and lactate significantly increased in both treatments. The concentration of cortisol increased after the stress test only in the His 30 g/kg group. Muscle cellularity was affected by the diets, where the control group showed a higher mean fibre area and lower fibre density. These results, associated with a lower frequency of smaller fibres, suggest a decrease in fibre recruitment and a higher fibre hypertrophy. In conclusion, the supplementation of diets with 3% led to a more pronounced response to the stress test in terms of cortisol levels and increased muscle fibre recruitment, decreasing muscle fibre hypertrophy contribution to muscle growth.  相似文献

We have recently induced two powdery mildew (Erysiphe pisi Syd) resistant mutants in Pisum sativum L. via ethylnitrosourea (ENU) mutagenesis. Both mutations (er1mut1 and er1mut2) affected the same locus er1 that determines most of the identified natural sources of powdery mildew resistance (PMR) in this crop. The mutated gene er1mut2 was mapped to a linkage group of 16 DNA markers combining three main strategies: near isogenic lines (NILs) analysis, bulked segregant analysis and genetic mapping of randomly identified polymorphic markers, together with three DNA-markers techniques: ISSR, RAPDs and AFLPs. Markers located closer to the PMR locus, OPO061100y (0.5 cM), OPT06480 (3.3 cM) and AGG/CAA125 (5.5 cM), were cloned and converted into SCAR markers. Markers AH1R850 and AHR920y were found to be allelic and converted into the co-dominant marker ScAH1 (16.3 cM). Two previously known DNA markers, ScOPE161600 and A5420y, were mapped at 9.6 and 23.0 cM from the PMR locus, respectively. The novel markers identified in this study are currently being transferred to a new F2 mapping population derived from a cross between the induced PMR mutant line F(er1mut2) and a more genetically distant susceptible line of Pisum sativum var. arvense.  相似文献

Cacao (Theobroma cacao L) is mostly grown on soils with low natural fertility. On such soils nitrogen (N) is one of the most yield limiting nutrients for cacao. Information is lacking on N use efficiency in cacao. A greenhouse experiment was conducted to evaluate growth response and N use efficiency by two cacao genotypes. The genotypes used were TSH-565 and ICS-9 and N rates adapted were 0, 120, 240, 360, and 480 mg N /pot. In both genotypes, increasing levels of applied N improved growth (stem girth, dry weight of shoot and roots and shoot/root ratio), and concentration and uptake of N. Genotypes differed significantly for stem girth and ICS -9 produced greater stem girth compared with TSH-565. Nitrogen uptake had a linear relationship with root dry weight of the two genotypes. In both genotypes, increasing levels of applied N overall increased N-uptake efficiency (NEFF = N concentration in shoot x shoot/root), but decreased N-use efficiency by shoot and roots (NUE = g dry matter of shoot or root/mg N) and N-use efficiency of carbohydrate (NUEC = mg of total carbohydrates in shoot/mg of N in shoot). Both genotypes responded differently to applied N, despite the existence of close genetic relatedness between them. The method used here appears to be suitable method for identification of cacao genotypes that are efficient in uptake and utilization of N.  相似文献
