For the purpose of investigation of working mechanisms in endocrine disruptors, we evaluated the dose-related effects of fetal and/or neonatal exposure to an estrogenic compound on the male reproductive organs in adult mice, particularly with respect to gene expression of steroidogenic acute regulatory protein (StAR). The pregnant ICR mice were given subcutaneous injections of 10 micro g/day/animal of diethylstilbestrol (DES) to subject the fetal mice to in utero exposure (IUE). Subsequently, the newborn male mice were subjected to neonatal exposure (NE) by treatment with vehicle or 0.1-10 micro g/day/animal of DES. Fertility rates of each group were as follows: control, 100%; IUE only, 60%; IUE+NE 0.1 micro g, 25%; IUE+NE 1 micro g, 0%; IUE+NE 10 micro g, 0%. In general histology, germ cell layers in the seminiferous tubules were thinned in the group of IUE+NE 10 micro g. Hypoplasia of the Leydig cells, in which the staining intensity of eosin was diminished, was also observed in the groups of IUE+NE 0.1-10 micro g. The androgen receptor (AR) and estrogen receptor alpha (ERalpha) immunoexpression in the Leydig cells of IUE+NE 1-10 micro g was slightly lower than that in the controls. Long-term dysfunction of the hypothalamo-pituitary-testicular axis, including sustained hypoproduction of gonadotropin and testosterone, and altered expressions of steroid hormone receptors and StAR genes were observed. The hypothalamo-pituitary control of gonadotropin secretion may be affected by the smaller doses of estrogenic agents than the reproductive organs. Furthermore, the fertility rate in the male mice exposed to this estrogenic agent was closely correlated with the testosterone levels, and even more so with the rate-limiting factor of steroidogenesis, StAR. This finding suggests that endocrine disruptors have an important pronounced effect on StAR gene expression.  相似文献

Leaf spots were found on Christmas rose (Helleborus niger) in Yamagata Prefecture, Japan, in October 2006. The morphology of the causal fungus was very close to that of Colletotrichum truncatum. Classifying the species from the sequences of the internal transcribed spacer regions of ribosomal DNA was inconclusive, and the isolates were identified only as Colletotrichum sp. Artificial inoculation confirmed the pathogenicity of isolates to the host plant and some legumes. We propose the name anthracnose of Christmas rose for this disease by Colletotrichum sp.  相似文献

Immobilizing materials such as lherzolite could reduce metal bioavailability but the effectiveness of lherzolite on the extractability and bioavailability of cadmium (Cd) and zinc (Zn) is rarely investigated. We conducted a greenhouse experiment to investigate the effect of 5% application of lherzolite to a contaminated soil on the chemical fractionation of Cd and Zn and their uptake by radish (Raphanus sativus L.) and Japanese mustard spinach (Brassica rapa L. var. perviridis). Both plants were grown in a highly contaminated (with Cd and Zn) sandy loam soil. Plants were cultivated consecutively three times in the same pots. After the third cultivation, soil samples were collected and analyzed by sequential extraction procedure into five operationally defined fractions (F1—exchangeable, F2—carbonate-bound, F3—oxides-bound, F4—bound with organic matter, and F5—residual). Addition of lherzolite to soil decreased 50% of exchangeable (F1) Cd but it increased the carbonate (F2), oxide (F3), and organic (F4) fraction Cd. For Zn, application of lherzolite resulted into the reduction of both F1 (87%) and F2 (33%) fractions but it increased the F3, F4, and F5 fractions. The reduction in exchangeable fraction of Cd and Zn in the soil resulted in higher plant growth and lower concentrations of both Cd (64% to 92%) and Zn (78% to 99%) in plant tissues of both plant species grown. We may thus conclude that application of lherzolite resulted into lower availability of these metals in the soil leading to lower uptake of Cd and Zn by plant roots, lower toxicity, and ultimately higher plant growth.  相似文献

The distribution pattern of collagen fibril diameter in the equine superficial digital flexor tendon (SDFT) is known to differ in central and peripheral areas of some regions. This study reports the essence of collagen fibril differences among different regions of the equine SDFT by transmission electron microscopic (TEM) and high-voltage electron microscopic observations and biochemical analysis. The distribution of large collagen fibrils increased but the density of collagen fibrils decreased from the proximal metacarpal region to the distal metacarpal region. Large collagen fibrils with an irregular cross-sectional profile were found more frequently in the middle metacarpal region than in other regions. Three-dimensional reconstruction of images of irregularly shaped collagen fibrils revealed that these fibrils are formed through fusion of small collagen fibrils with large ones. The amount of decorin, which reportedly inhibits the lateral fusion of collagen fibrils, decreased in the direction of the distal metacarpal region. On the other hand, the size of decorin gradually increased in the direction of the distal metacarpal region. These results suggest that regional differences in collagen fibril distribution and density of collagen fibrils in the SDFT are due, at least in part, to fusion of collagen fibrils and the concomitant regional differences in the amount and size of decorin.  相似文献

Activities of certain cytoplasmic enzymes were measured in bovine T lymphoma (BTL-PC3 cells). The activities of hexokinase, pyruvate kinase and glucose-6-phosphate dehydrogenase in PC3 cells were elevated as much as 2 or 3-fold of those in bovine thymocyte. The high activities of these enzymes derived from activation of glucose metabolism may reflect the high growth potential of PC3 cells.  相似文献

Differences in tree species may lead to contrasting soil environments via differences in litter chemical quality and physical environmental factors, such as soil type and soil moisture. However, separating the effects of litter quality and physical environment is difficult under field conditions. Both litter quality and soil environment affect the species composition of the soil animal community. A diversity gradient of canopy tree species (11–25 species) located on homogeneous soil substrate at Tomakomai Experimental Forest of Hokkaido University was used to analyse the relationship between tree species diversity and oribatid mite community structure. Soil samples were collected from three levels of tree species richness (high, intermediate and low) with three replicates each, in July 2000. Leaf area index (LAI) was positively correlated with tree species diversity suggesting higher litter input into the soils with increasing tree diversity. However, the tree species diversity gradient affected neither accumulation of litter on the forest floor nor abundance and species richness of oribatid mites. Canopy and understory plant species richness, LAI, total soil carbon and biomass of epigeic and endogeic earthworms did not significantly affect mite community structure as indicated by redundancy analysis (RDA). The results suggest that oribatid mite community structure is minimally affected by tree species diversity and associated changes in litter diversity.  相似文献

In clinical trials, patients usually take many kinds of drugs at the same time. Thus, drug-drug interactions can often directly affect the therapeutic safety and efficacy of many drugs. Oral delivery is the most desirable means of drug administration. Changes in the activity of drug transporters may substantially influence the absorption of administered drugs from the intestine. However, there have been a few studies on food-drug interactions involving transporters. It is important to be aware of the potential of food-drug interactions and to act in order to prevent undesirable and harmful clinical consequences. Coenzyme Q10 (CoQ10) is very widely consumed by humans as a food supplement because of its recognition by the public as an important nutrient in supporting human health. Since intestinal efflux transporter P-glycoprotein (P-gp) is one of the major factors in drug-drug interactions, we focused on this transporter. We report here for the first time that CoQ10, which is widely used as a food supplement, affects the transport activity of P-gp.  相似文献

Various physiological functions of dietary glucosylceramides (GlcCer) have been reported, such as preventing colon cancer and improving skin barrier function. One potential GlcCer source used as a foodstuff is sea cucumber. In this study, our objective was to determine the effect of dietary GlcCer prepared from sea cucumber on plasma and liver lipids in cholesterol-fed mice. ICR mice were fed four different diets (control diet, sea cucumber GlcCer supplemented diet, high cholesterol supplemented diet, and high cholesterol + sea cucumber GlcCer supplemented diet). Dietary GlcCer decreased total cholesterol significantly in ICR mice. The mRNA expression of LDL receptor was increased significantly, while the expression of the gene CYP7A1, which is involved in bile acid formation, was decreased significantly compared with the control (diet without cholesterol). These results suggest that the expression of the cholesterol homeostasis gene in liver is modulated due to the cholesterol lowering effect of dietary GlcCer.  相似文献
