Vertical water and nitrate movement into deeper soil layers on fields located in the south of Germany In Southern Bavaria, selected fields from deep loess and sandy sediments were sampled to a depth of maximum 10 m every 4 months, in order to determine the basic processes of water and nitrate movement in the course of the year. The downward movement of water and nitrate followed the principle of piston flow only in the intermediate vadose zone of loess soils with a leaching distance amounting 0.8 m per year. On the other hand, an accelerated as well as a delayed transport was observed in the zone of evapotranspiration of loess soils and in the whole profile of sandy soils, not being in accordance with the simple simulation model FLOTRA based on the classic theories of transport (Darcy flow, convection-dispersion-equation). Preferential flow led to the leaching of nitrate from the top soil to the lower boundary of the evapotranspiration zone of loess soils in 2 m depth, in winter and spring. In sandy soils nitrate was leached to the groundwater table in 4–6 m depth after extensive rainfall.  相似文献

(1) Flocculation of cephalin-cholesterol emulsions by blood serum is markedly influenced by the amount of light to which the serum-saline-antigen suspensions are exposed. Protection from bright light, natural and artificial, has eliminated many falsely positive reactions. (2) Other factors that appear to influence the cephalin flocculation procedure have been briefly mentioned. It is well established that high doses of monomeric immunoglobulin G (IgG) purified from pooled human plasma [intravenous immunoglobulin (IVIG)] confer anti-inflammatory activity in a variety of autoimmune settings. However, exactly how those effects are mediated is not clear because of the heterogeneity of IVIG. Recent studies have demonstrated that the anti-inflammatory activity of IgG is completely dependent on sialylation of the N-linked glycan of the IgG Fc fragment. Here we determine the precise glycan requirements for this anti-inflammatory activity, allowing us to engineer an appropriate IgG1 Fc fragment, and thus generate a fully recombinant, sialylated IgG1 Fc with greatly enhanced potency. This therapeutic molecule precisely defines the biologically active component of IVIG and helps guide development of an IVIG replacement with improved activity and availability.  相似文献

Pulmonary dysfunction was evaluated in pigs infected with porcine reproductive and respiratory syndrome virus (PRRSV, isolate VR-2332) and compared to clinical and pathological findings. Infected pigs developed fever, reduced appetite, respiratory distress and dullness at 9 days post-inoculation (dpi). Non-invasive pulmonary function tests using impulse oscillometry and rebreathing of test gases (He, CO) revealed peripheral airway obstruction, reduced lung compliance and reduced lung CO-transfer factor. PRRSV-induced pulmonary dysfunction was most marked at 9–18 dpi and was accompanied by a significantly increased respiratory rate and decreased tidal volume. Expiration was affected more than inspiration. On histopathological examination, multifocal areas of interstitial pneumonia (more severe and extensive at 10 dpi than 21 dpi) were identified as a possible structural basis for reduced lung compliance and gas exchange disturbances.  相似文献

Soil systematics and classification systems Part I: Fundamentals Soil‐ordering systems are primarily based and developed on one of two underlying principles: They are either categorized according to soil‐forming processes, or the formation of categories develops by chosen parameters. This perspective has already been established in the literature, though it is often confusing as many terms are defined and applied differently. In this contribution, the various definitions of systematics, classification, taxonomy, and identification will be clearly differentiated and summarized. The core of our work is to clearly define and contrast three terms: systematics, classification, and identification. Systematics is the fundamental scientific and deductive ordering of objects into systematic units. The purpose of this approach is to organize the entire spectrum of knowledge within a discipline into a transparent and manageable form. Classification, in direct contrast to systematics, is goal‐oriented and an inductive ordering of objects. Thus, the ordering scheme consists of classes which are clearly parameterized. Identification is the ordering of new objects into an already existing systematics or classification system. Close attention is paid to both the differences and the similarities between a systematics and a classification system, especially pertaining to their practical applications. The identification requires that the category‐forming characteristics can be measured (e.g., for soil systematics, these are the soil‐forming processes and factors). Currently, it is unfortunately not feasible to objectively quantify most soil‐forming processes. Thus, most attempts at categorizing soils by systematics are hypothetical and highly subjective in nature. The resulting identification derived from the soil systematics approach is open to questions and contestable, since a graded measuring system does not yet exist to verify these determinations. In contrast, a soil‐classification system does allow an objective soil‐profile identification, although such systems are conceived pragmatically and designed for a practical purpose (e.g., not scientifically based on process intensities). Unfortunately, such a classification system cannot be applied as a universal scientific categorization system due to this method of conception. Both categorization approaches are required in soil science in order to satisfy both the practical and the scientific aspects of the field. However, substantial research must be done to complete and verify systematics. The only viable short‐term solution is through the development of a graded classification system where the categories of the system are directly derived from the current systematics approach. In the long run both the exact investigation and the detailed modeling of the soil‐forming processes are inevitable.  相似文献

A method for the determination of total N-nitroso compounds (NOC) by chemical denitrosation and subsequent chemiluminescence detection of evolved NO is described. Denitrosation was accomplished with CuCl in HCl at 70 degrees C. The detection limit for N-nitrosoproline (NPRO) was 1 pmol. NO formation from NPRO was linear (R(2) = 0.999) from 4 pmol to 2 nmol. Among the possible interfering compounds tested, only S-nitroso compounds contribute any significant interference. This method had several advantages over other similar methods: (1) A commercially available one-piece reaction vessel and a NO analyzer with software were used. (2) NO release occurred rapidly and was easily measured and quantified. (3) Compared to HBr or HI, CuCl was more convenient to work with and safe. (4) CuCl was suitable for samples in aqueous and most organic solvents. The application of this method to food, personal care products, and human body fluids demonstrates its utility.  相似文献

With respect to their browning potential and in consideration of a combined recovery of pectin and phenolic compounds, peels of 14 cultivars and the flesh of nine cultivars of mango (Mangifera indica L.) fruits were analyzed for their contents of flavonol O- and xanthone C-glycosides by high-performance liquid chromatography (HPLC)-diode array detection-electrospray ionization mass spectrometry (ESI-MS). While total amounts of up to 4860 mg/kg dry matter demonstrated the peels to be a rich source of phenolic compounds, only traces could be detected in the flesh. The profile of flavonol glycosides of the peels proved to be highly characteristic and may therefore serve as a tool for authenticity control of mango puree concentrate, which is often produced from unpeeled fruits and represents an important intermediate for the production of mango nectars. Two compounds were isolated by preparative HPLC, and their structures were elucidated on the basis of ESI-MS as well as NMR spectroscopy, establishing the two compounds as rhamnetin 3-O-beta-galactopyranoside and rhamnetin 3-O-beta-glucopyranoside, respectively. In the peels of red-colored cultivars, cyanidin 3-O-galactoside and an anthocyanidin hexoside so far not reported in mango could tentatively be identified. The contents and degrees of esterification of pectins extracted from the lyophilized peels ranged from 12.2 to 21.2% and from 56.3 to 65.6%, respectively, suggesting mango peels also as a promising source of high-quality pectin.  相似文献

Flavonol O- and xanthone C-glycosides were extracted from mango (Mangifera indica L. cv. "Tommy Atkins") peels and characterized by high-performance liquid chromatography-electrospray ionization mass spectrometry. Among the fourteen compounds analyzed, seven quercetin O-glycosides, one kaempferol O-glycoside, and four xanthone C-glycosides were found. On the basis of their fragmentation pattern, the latter were identified as mangiferin and isomangiferin and their respective galloyl derivatives. A flavonol hexoside with m/z 477 was tentatively identified as a rhamnetin glycoside, which to the best of our knowledge, has not yet been reported in mango peels. The results obtained in the present study confirm that peels originating from mango fruit processing are a promising source of phenolic compounds that might be recovered and used as natural antioxidants or functional food ingredients.  相似文献
