Mast cell tumour (MCT) is one of the most frequent skin tumours in dogs. Due to their unpredictable biological behaviour, MCTs often cause several therapeutic frustrations, leading to investigation regarding prognostic markers. Lysyl oxidase (LOX) is an enzyme that promotes extracellular matrix stability and contributes to cell migration, angiogenesis and epithelial-mesenchymal transition. Its expression positively correlates with poor prognoses in several human and canine mammary cancers. The aim of this study was to characterise the immunohistochemical expression of LOX in MCT samples and compare it with histological grading and post-surgical survival. Twenty-six tumours were submitted to immunohistochemistry for LOX expression evaluation. All samples were positive for LOX, with variable percentages of cytoplasmic and nuclear positivity. Cytoplasmic positivity was significantly higher in high-grade MCTs (P = .0297). Our results indicate that high expression of cytoplasmic LOX in neoplastic mast cells is an indicator of poor prognosis for canine cutaneous MCTs.  相似文献

Seedlings of four populations each of Pinus pinaster, P. halepensis, P. canariensis and P. pinea were grown in controlled conditions to evaluate both inter- and intra-specific differences in response to watering. We submitted half of the plants to a moderate water stress and after 22 weeks, we recorded height, stem diameter and root, stem and leaves dry weight. Patterns and amounts of phenotypic changes, including changes in biomass allocation, were analysed. We found a scant response in P. canariensis, P. pinaster and P. halepensis presented high population divergence for phenotypic changes, and P. pinea showed marked allocational shifts and no population divergence. The phenotypic changes observed within species are interpreted as a plastic response. The variation encountered within P. halepensis and P. pinaster may be indicative of specialisation to either resource-rich or resource-poor habitats, being populations from favourable sites more plastic. P. pinea exhibited a very uniform plastic response, indicating generalist behaviour.  相似文献

Two Rotstop® preparations, one containing a Phlebiopsis gigantea strain from Finland (Rotstop F) and the other one from Sweden (Rotstop S), were used in different concentrations to treat thinning stumps of Picea abies against spore infections by Heterobasidion spp. in southern Sweden. Trees were cut on three sites during the summer of 2004, and 285 stumps were treated manually ensuring 100% coverage. Spore concentrations in the treatment suspensions were ca 5 × 106 and 10 × 106 spores/l, and approximately 10 ml of suspension was applied per 100 cm2 of stump surface. An additional 31 stumps on one of the sites were treated mechanically with Rotstop S; in this treatment the spore concentration was high, about 20 × 106 spores/l, but the coverage was incomplete. Three months later there was a significant reduction in frequency and relative areas of Heterobasidion spp. infections on stumps with manual treatment compared with the untreated stumps. However, there was no significant difference between the preparations or the concentration of active ingredient in terms of their reduction of Heterobasidion infection. Mechanical treatment with incomplete coverage failed to control infection. Therefore, in conditions of moderate Heterobasidion spore load in the environment there seems to be no reason to use higher concentrations of P. gigantea in the treatment of spruce stumps.  相似文献

The evaluation uses the longest available time series for beech and oak defoliation in Germany. The data from Hesse, starting from 1984, show a typical pattern: for the first 12 years, a continuous increase in defoliation was observed ranging from an average value of 14% in 1984 to a peak value of 30%. This was followed by a subsequent decrease in the loss of foliage accompanied by a high variability, until the last monitoring in 2003, where an average value of 25% defoliation was observed. For both tree species, the years of trend reversal were identical. The same pattern was observed in the German federal states: Rhineland-Palatinate, North Rhine-Westphalia, and Bavaria. The year of trend reversal was identical in Hesse and Rhineland-Palatinate. In North Rhine-Westphalia, it occurred 1 year earlier and in Bavaria 3 years earlier. Whereas defoliation trends were clearly demonstrated, tree mortality did not appear on a large scale. The sample trees were grouped into four discrete clusters according to their annual defoliation values from 1984 to 2003. In 1996, the clusters represent 15, 25, 35, and 50% defoliation values. Regarding beech in Hesse, there was no overlap in the defoliation curves observed among the different clusters. These four clusters having different degrees of defoliation over the whole time span of 20 years were used for a further detailed statistical analysis. For discrete variables like crown spacing and—in the case of beech—fruit bearing, mosaic plots were applied in order to visualize relations of low dimensional contingency tables, with defoliation trends being used as the response variable. The data show for beech a very clear relation between defoliation and age, relative crown spacing, stand composition, and fruit bearing. Regarding oak, besides age and relative crown spacing, the years with significant appearance of biotic stress factors—leaf eating insects—show a clear relation to trends of defoliation. The statistical model used in this study—logistic regression—allows applying a multinomial response variable and a number of continuous or categorical explanatory variables. With this approach, an iterative optimized selection of effect variables was used to test the relevance of different variables on the defoliation pattern of the same four clusters mentioned above. For this, the variables were grouped in an iterative process with five steps, starting with a few basic variables of tree and site information, and ending with a total of more than 20 variables in the fifth step. The process selects first the variables which are of significance on the defoliation, and calculates the possible errors in the grouping of the different trees to the four clusters. In this analysis of beech, the basic tree and stand variables: age, relative crown spacing, stand composition and fruit bearing proved to be the most relevant group of parameters, with the other variables explaining the variation of defoliation only to a minor extent. More complex model levels do not change any basic selected variables; however, Cation Exchange Capacity (CEC), C/N-ratio, Al- and Ca-proportion of CEC are additionally selected and give a hint of the relevance of soil conditions. Regarding beech, the errors of the statistical model are lower compared to oak.  相似文献

ABSTRACT

During 2008–2011, model field experiments were carried out at the Joni?k?lis Experimental Station of Lithuanian Research Centre for Agriculture and Forestry on a clay loam Endocalcaric Endogleyic Cambisol. The study was aimed to establish the comparison of various postharvest practices (mineral nitrogen fertiliser alone or together with a bioactivator Penergetic k, livestock slurry, red clover biomass and straw incorporation in the soil by a stubble cultivator at a 10 cm depth) on the acceleration of the initial (nine-month period) decomposition of winter wheat straw. During this period, straw mass decomposition intensity was 20.7–29.1%, carbon (C) concentration decreased by 6.5–22.8%, while an increase in nitrogen (N) by 1.1–2.2 times was observed. The highest straw decomposition rate was recorded when after straw incorporation autumn was warm and humid. That year straw mass C to N ratio (C/N) was 38–46. Under less-favourable autumn conditions, the highest decomposition of straw was achieved, having applied mineral N (with and without Penergetic) and livestock slurry and having incorporated the straw in the soil (C/N = 40–55). A slower decomposition rate was observed for the straw spread on the soil surface with mineral N addition or on undersown red clover.  相似文献

BACKGROUND: Infection with Ehrlichia canis causes a highly variable, multisystemic disease in dogs. Nevertheless, many clinicians in Rio de Janeiro, Brazil, use the presence of only thrombocytopenia to make a presumptive diagnosis of E canis infection. OBJECTIVE: The objective of this study was to determine the prevalence of E canis in thrombocytopenic dogs from Rio de Janeiro, Brazil, using polymerase chain reaction (PCR). METHODS: Following DNA extraction of whole blood samples from 226 dogs, PCR assays were done using primers for rickettsial DNA (including Ehrlichia spp, Anaplasma platys and A phagocytophilum) and using E canis-specific primers (16S rRNA gene). Dogs were grouped as thrombocytopenic and nonthrombocytopenic based on platelet counts. The null hypothesis that there was no difference in the prevalence of E canis in these groups was rejected at P<.05. RESULTS: Thirty-six (32.1%) of the thrombocytopenic dogs and 4 (3.5%) of the nonthrombocytopenic dogs were positive for rickettsial gene sequences (P<.0001). Further, 30 (26.8%) of thrombocytopenic dogs and 4 (3.5%) nonthrombocytopenic dogs were positive for E canis-specific gene sequences (P<.0001). CONCLUSIONS: Although the prevalence of E canis infection was higher in thrombocytopenic dogs, less than one third of these dogs had demonstrable E canis infection. Thus, thrombocytopenia is not specific for the detection of E canis infection and should not be used solely to establish a diagnosis of canine ehrlichiosis, even in a geographic area with relatively high disease prevalence.  相似文献

Anaplasma marginale infection in Europe has been limited to the Mediterranean and eastern countries, to Austria and to very sporadic cases in Switzerland. There are no reports of its occurrence in the countries north of Switzerland. A severe outbreak of anaplasmosis in August 2002 in a cattle farm in the canton Grisons, Switzerland, north of the Alps, with more than 300 cattle that had to be culled, came unexpected and gave reason to hypothesize presence of an increased yet undetected prevalence of A. marginale in Switzerland. Randomly selected bovine serum samples collected in 1998 and 2003 were tested using a competitive inhibitory ELISA (cELISA) to test the hypothesis. Our validation of the diagnostic sensitivity and specificity of this test, done in the outbreak herd, yielded 99.2 and 83.3%, respectively, probably underestimating the true specificity. The true seroprevalence of anaplasmosis in Swiss cattle determined by cELISA was likely to be zero with upper 95% confidence limits of 2.49% in the canton Grisons and 1.17% in the rest of Switzerland, respectively, in 1998. For 2003, these estimates were even lower. There was no significant difference in apparent prevalences between 1998 and 2003. In search of a possible reservoir, three chamoises out of 46 free ranging wild ruminants from the Swiss National Park, Grisons, tested positive in the cELISA. This reaction is in accordance with A. marginale or a cross reacting agent such as Anaplasma ovis. From our results we conclude that the hypothesis of an increased prevalence of anaplasmosis in cattle in Switzerland must be rejected.  相似文献
