As clinical toxoplasmosis is not considered a problem in pigs, the main reason to implement a control strategy against Toxoplasma gondii (T. gondii) in this species is to reduce the establishment of T. gondii tissue cysts in pork, consequently reducing the risk of the parasite entering the human food chain. Consumption of T. gondii tissue cysts from raw or undercooked meat is one of the main sources of human infection, with infected pork being considered a high risk. This study incorporates a mouse bioassay with molecular detection of T. gondii DNA to study the effectiveness of vaccination (incomplete S48 strain) in its ability to reduce tissue cyst burden in pigs, following oocyst (M4 strain) challenge. Results from the mouse bioassay show that 100% of mice which had received porcine tissues from vaccinated and challenged pigs survived compared with 51.1% of mice which received tissues from non-vaccinated and challenged pigs. The presence (or absence) of T. gondii DNA from individual mouse brains also confirmed these results. This indicates a reduction in viable T. gondii tissue cysts within tissues from pigs which have been previously vaccinated with the S48 strain. In addition, the study demonstrated that the main predilection sites for the parasite were found to be brain and highly vascular muscles (such as tongue, diaphragm, heart and masseter) of pigs, while meat cuts used as human food such as chop, loin, left tricep and left semitendinosus, had a lower burden of T. gondii tissue cysts. These promising results highlight the potential of S48 strain tachyzoites for reducing the number of T. gondii tissues cysts in pork and thus improving food safety.  相似文献

There has been interest within the pesticide regulatory community in developing a tool that can provide estimates of potential pesticide exposure in shallow groundwater across an intended use area. Therefore, industry initiated an investigative project based on the PRZM 3.12 model, which uses regional soils and weather in an easy to use interface. The goal of this proof-of-concept is to facilitate the refinement of groundwater exposure estimates. The focus of this paper is to report the effectiveness of the tool as a regional estimator of potential groundwater contamination.  相似文献

Eicosapentaenoic acid (EPA) derived from chemically hydrolyzed sardine oil was concentrated by urea fractionation using methanol at different temperatures (2, 4, and 6 degrees C) and urea/fatty acid ratios (2:1, 3:1, and 4:1 w/w) and purified by argentation neutral alumina column chromatography. The individual fatty acids were determined as fatty acid methyl esters (FAME) by gas-liquid chromatography and gas chromatography-mass spectroscopy as FAME and N-acyl pyrrolidides. In the mass fragmentation pattern of FAME, the base peak was assigned to be the 1-methoxyethenol moiety (m/z = 74) obtained by McLafferty rearrangement. Formation of the cyclic tropylium ion (m/z = 91) in fatty acids with four or more double bonds was apparent in FAME-PUFAs. The base peak of N-acyl pyrrolidides was the McLafferty rearrangement ion, 1-(pyrrolidin-1-yl)ethenol (m/z = 113). The highest concentration of EPA (47.78%) was obtained at the crystallization temperature of 4 degrees C with a urea/fatty acid ratio of 4:1 (w/w) with 93.74% yield. After complexation of saturated and less unsaturated fatty acids by urea complexation, argentation chromatography resulted in an EPA of high purity (99.6%) with an overall recovery of 54.09% using 50% diethyl ether/n-hexane as eluting solvent. The peroxide (POV) and thiobarbituric acid (TBS) values were found to be highest (4.0 mequiv of O2/kg and 5.2 mg of malondialdehyde/kg, respectively) during urea fractionation at the higher crystallization temperature (6 degrees C) and higher urea/fatty acid ratio (4:1). OBJECTIVE: The risk factors for fractures are incompletely understood. An outstanding question concerns the optimal amount of dietary calcium needed to minimise the risk of fracture. DESIGN: We examined the associations of dietary calcium and other nutrients with self-reported fracture risk in a prospective cohort study. Nutrient intakes were estimated using a semi-quantitative food-frequency questionnaire administered at recruitment. SETTING: The UK. Participants: A total of 26 749 women and 7947 men aged 20-89 years. RESULTS: Over an average of 5.2 years of follow-up, 1555 women and 343 men reported one or more fractures, 72% of these resulting from a fall. Among women, fracture risk was higher at lower calcium intakes, with a relative risk of 1.75 (95% confidence interval (CI) 1.33-2.29) among women with a calcium intake of < 525 mg day- 1 compared with women with a calcium intake of at least 1200 mg day- 1 (test for linear trend, P < 0.001). The association of dietary calcium with fracture risk was stronger among women aged under 50 years at recruitment than among women aged 50 and above. Dietary calcium intake was not associated with fracture risk in men. Fracture risk was not related to the dietary intake of any other nutrient examined. CONCLUSION: In this population, women with a low dietary calcium intake had an increased risk of bone fracture, and this association was more marked among younger women than among older women.  相似文献

ABSTRACT A meta-analysis of the effect of tebuconazole (e.g., Folicur 3.6F) on Fusarium head blight and deoxynivalenol (DON) content of wheat grain was performed using data collected from uniform fungicide trials (UFTs) conducted at multiple locations across U.S. wheat-growing regions. Response ratios (mean disease and DON levels from tebuconazole-treated plots, divided by mean disease and DON levels from untreated check plots) were calculated for each of 139 studies for tebuconazole effect on Fusarium head blight index (IND; field or plot-level disease severity, i.e., mean proportion of diseased spikelets per spike) and 101 studies for tebuconazole effect on DON contamination of harvested grain. A random-effects meta-analysis was performed on the log-transformed ratios, and the estimated mean log ratios were transformed to estimate the mean (expected) percent control for IND ( C(IND) ) and DON ( C(DON)). A mixed effects meta-analysis was then done to determine the effects of wheat type (spring versus winter wheat) and disease and DON levels in the controls on the log ratios. Tebuconazole was more effective at limiting IND than DON, with C(IND) and C(DON) values of 40.3 and 21.6%, respectively. The efficacy of tebuconazole as determined by the impact on both IND and DON was greater in spring wheat than in winter wheat (P < 0.01), with a 13.2% higher C(IND) and a 12.4% higher C(DON) in spring wheat than in winter wheat. In general, C(IND) and C(DON) were both at their lowest values (and not significantly different from 0) when mean IND and DON in the controls, respectively, were low (相似文献

We examined the distribution of sardine larvae relative to environmental conditions with the purpose of identifying and characterizing habitat that encourages high larval growth and survival, based on the 1983–1998 surveys of the California Cooperative Oceanic Fisheries Investigations (CalCOFI). Long-term averages show that sardine 'survivors' (spatially aggregated larvae ≥ 18 days old) were most abundant offshore, whereas sardine egg density, chlorophyll biomass and zooplankton volume were greatest inshore. In contrast, mesoscale eddies, observed in remotely sensed sea surface temperature imagery, were found only in offshore regions. To further examine the link between eddies – which often result in locally elevated chlorophyll and zooplankton – and sardine survival, we compared the distribution of larvae and eddies survey by survey. Sardine survivors were most abundant offshore in only one-quarter of the research surveys, and when they were most abundant offshore they were associated with eddies. This indicates that the offshore eddy habitat produced exceptionally large numbers of survivors, as evidenced by the disproportionate effect on the long-term average.  相似文献

Following independence from Ethiopia in 1993, Eritrea resumed exploiting Red Sea and Arabian fish species for the ornamental trade in 1995 as a means to earn foreign exchange from sparsely exploited marine resources. This paper describes the findings of research conducted in 1997, in collaboration with the Eritrean Ministry of Fisheries. The capture, transport and export of aquarium fish were reviewed and potential impacts and the status of management were investigated through liaison with stakeholders and researchers. From 1995 to 1997 two companies exported approximately 60,000 fish per year, mainly to the USA, worth US$65,000 (export value). Seventy-five species (from 22 families) were exported. Damselfishes made up two-thirds of total exports but more valuable families (angelfishes and butterflyfishes) were more economically significant. To earn revenue for Eritrea, a 20% export tax was imposed, although this was calculated from declarations by the operators. The emerging nature of the trade allowed detailed monitoring by the Ministry of Fisheries. However, management efforts were constrained by a lack of capacity for enforcement and baseline research. Several potential effects of the trade exist but other, land-based impacts may be more pressing concerns for Eritrea's reefs. Research priorities for management are discussed as well as the implications of mariculture of Eritrean species by other nations.  相似文献
