Since the ban of malachite green in the fish farming industry, finding alternative ways of controlling Saprolegnia infections has become of utmost importance. Much effort has been made to elucidate the mechanisms by which Saprolegnia invades fish eggs. Little is known about the defence mechanisms of the hosts, making some eggs more prone to infection than others. One clue might lie in the composition of the eggs. As the immune system in the embryos is not developed yet, the difference in infection levels could be explained by factors influenced by the mother herself, by either transferring passive immunity, influencing the physical aspects of the eggs or both. One of the physical aspects that could be influenced by the female is the chorion, the extracellular coat surrounding the fish egg, which is in fact the first major barrier to be overcome by Saprolegnia spp. Our results suggest that a thicker chorion in eggs from Atlantic salmon gives a better protection against Saprolegnia spp. In addition to the identification of differences in sensitivity of eggs in a fish farm set‐up, we were able to confirm these results in a laboratory‐controlled challenge experiment.  相似文献

Soil organic matter (SOM) is considered an important indicator of soil quality, which can be impacted by crop production practices such as tillage. In this study, two long‐term tillage regimes (conventional tillage [CT] and no tillage [NT], conducted for 36 years) were compared in continuous sorghum production in a sub‐tropical environment in southeast Texas. The positive effects of long‐term NT practice were more conspicuous at the soil surface compared with the deeper soil profiles. The SOC was greater (1.5 t C ha?1 greater) in the NT system compared with the CT system. Results from an incubation study indicate that the rate of C‐min at 0–5 cm soil depth was significantly greater (164 μg of CO2–C g?1 of soil greater) in NT than that of CT, but this trend was reversed at 10–20 cm depth wherein the C‐min rates were 106 μg of CO2–C g?1 of soil greater in CT compared with NT, which is likely because of soil disturbance during the study. Soil cumulative CO2‐C emissions were greater in the CT system (7.28 g m?2) than in the NT system (5.19 g m?2), which is primarily attributed to high soil temperature conditions in the CT system. Sorghum grain yield however was not influenced by the differences in SOC content in this long‐term experiment. Overall, the present study found that long‐term conservation tillage improved SOC stock and reduced carbon loss, thus had a positive impact on soil health and sustainability.  相似文献

We describe a simple technique for the germination of arbuscular mycorrhizal (AM)–fungal spores and their multiplication in pots. Glomus fasciculatum, G. mosseae, and Gigaspora margarita were used. A single wheat seedling was tied to a glass slide, previously covered with filter paper with the help of thread. One single surface‐sterilized AM‐fungal spore was placed on the middle portion of the root of the wheat seedling using a sterilized syringe. The slide was placed vertically in a 100 mL glass beaker filled with 25 mL of root exudates–water (1:4, v/v) solution, which was collected by growing twenty wheat seedlings in a 150 mL beaker filled with 100 mL sterilized distilled water for 7 d. The slide was observed daily using a compound microscope to follow the time course of germination. In this technique, the spore is directly in contact with the host root, and a visualization of spore germination, hyphal development, and appressorium formation is possible without disrupting fungal growth or the establishment of the symbiosis. The method allows to document the germination events and to assess hyphal‐elongation rates by photographing the same spore on consecutive days. The inoculated seedling was used to initiate single‐spore multiplication in a sterilized (autoclave on 3 alternate days at 120°C for 120 min at 1.05 kg cm–2 pressure) potted sandy soil (150 mL volume) into which the slide with the inoculated seedling was inserted carefully through a previously made slit. The wheat seedlings in all pots (4 treatments and 15 replications) became colonized by mycorrhiza, confirming that the establishment of the AM‐fungal symbiosis is highly reproducible. Our technique permits the relatively undisturbed growth of the symbiotic partners, the visualization of germinating AM‐fungal spores, and their multiplication in pots. This simple and low‐cost method facilitates the production of pure lines of AM fungi from single spores, allowing for the study of intraspecific variation and potentiality for cytological, biochemical, physiological, and taxonomical studies.  相似文献

Wild meat harvests in African moist forests are presumed to exceed production, even in the case of traditional societies still using rudimentary hunting methods. Though some approximations do exist of the volume of bushmeat harvested in some Central African moist forest areas, estimates based on extensive and simultaneous sampling, within a large geographical region, are not available. Here, we present the results of the first reported study of this kind. During a period of 5 month, we counted bushmeat carcasses deposited in 89 urban and rural markets in a 35,000 km2 area between the Cross River in Nigeria and the Sanaga River in Cameroon. We used these data to calculate annual bushmeat volume traded by site, species and overall in the study area.Mammals represented >90% of the bushmeat carcasses sold in all sites. Reptiles were also abundant, but birds and amphibians were relatively scarce. Estimates of carcasses extracted and crude biomass per site varied significantly between countries. In Nigeria, biomass (kg) extracted for sale per km2 per year, was three times greater (600 kg/km2) than in Cameroon. Conservative estimates for the entire study area indicate that >900,000 reptiles, birds and mammals are sold each year by the rural and urban population, corresponding to around 12,000 tonnes of terrestrial vertebrates. We also assessed the relationship between bushmeat harvested for sale and distance of the study settlements from the main protected areas (Cross River and Korup National Parks). The number of carcasses and biomass sold was negatively related to the proximity to the national parks in >50% of species in Nigeria, and in 40% of species in Cameroon.Our cross-site comparison documents the staggering volume of wild species affected by hunting in the region. We also conclude that species within the main protected areas in both countries are likely to be negatively affected by the current and future demand for bushmeat in the surrounding areas.  相似文献

A simple, reproducible and new method (modified dehydrogenase activity measurement after 72 h; DHA 72) was developed to assess plant residue quality for decomposition. The method resulted in similar assessments of residue quality than measurements of the C:N ratio, lignin:N ratio, and lignin and polyphenol concentration (PRQI). Among 25 plant residues tested, there was a large variation of DHA 72 as found in other well‐established methods. Based on cluster analysis, plant residues were grouped in four different classes; highly decomposable, moderately decomposable, slowly decomposable, and least decomposable. It is concluded that DHA 72 can very well be used to screen plant residues for residue quality judgment. The proposed method is very simple and easy to handle.  相似文献

Fertigation has the potential to reduce extra chemical load by improving nutrient and water use efficiency of coconut (Cocos nucifera L.), but studies demonstrating the fertilizer reduction through drip irrigation in comparison to conventional ring basin method are rare in the Eastern Indo-Gangetic Plains (IGP) of South Asia. A long-term field experiment was conducted during 2007–2013 in West Bengal, India, to study the effect of fertigation on coconut var. DXT. The experiment was laid out in a randomised complete block design (RCBD) with six treatments (control – no fertilizers and water applied with drip irrigation; 25%, 50%, 75% and 100% of the recommended dose of fertilizer (RDF), each applied with drip irrigation; and 100% of the RDF and water applied with ring basin method of irrigation (i.e., conventional method)). Nuts yield was significantly higher for 75% of RDF (24.44 t ha?1 year?1) followed by 100% of RDF, each drip irrigation (23.79 t ha?1 year?1) compared to control (21.89 t ha?1 year?1). Copra yield was significantly higher for 75% of RDF (3.19 t ha?1) compared to 100% of RDF (3.12 t ha?1) and no fertilizer (1.87 t ha?1). Nitrogen (N), phosphorus (P) and potassium (K) contents of soil increased by 4.9%, 10.4% and 9.4%, respectively, with 75% of RDF applied through drip irrigation. Microbial population showed inverse relationship with amount of fertilizer application. The most water-use efficient fertigation treatment was 75% RDF (13.48 kg copra m?3) followed by 100% RDF (13.18 kg copra m?3) with drip irrigation as compared to conventional way soil application of fertilizers through ring basin method of irrigation (4.23 kg copra m?3). Role of N on yield variability was most prominent by both available soil N status (R2 = 0.49**) and leaf N concentration (R2 = 0.51**). The study indicated that there is a great scope for reducing the N, P and K fertilizers by up to 25% of the present RDFs for coconut when applied through drip irrigation compared to ring basin method of irrigation for its higher productivity and profitability through efficient use of nutrients and water in the Eastern IGP of South Asia.  相似文献
