ABSTRACT Phytophthora infestans isolates collected from potato and tomato crops from various parts of Nepal during the 1999 and 2000 crop seasons were characterized for nuclear and mitochondrial DNA polymorphisms using restriction fragment length polymorphism markers. The nuclear DNA probe RG57 detected 11 multilocus genotypes among 280 isolates. Three genotypes were detected 21 times or more, constituting 94% of the total population, whereas frequencies of other genotypes ranged from 0.004 to 0.014. The overall genotypic diversity as estimated by the Gleason index was 1.78. Most of the overall diversity was present at the highest level (i.e., interregional, 46%), indicating limited gene flow among regions. Cluster analysis of multilocus genotypes derived from RG57 and mating type data for Nepalese isolates and representative isolates worldwide showed Nepalese isolates grouping into four clusters. Characterization of 67 isolates for mitochondrial DNA polymorphisms revealed the presence of two mt-haplotypes, Ia and Ib with the proportions of 0.88 and 0.12, respectively. Polymorphisms in nuclear and mitochondrial DNA revealed a moderate level of diversity in this population. Genotype NP3 had an identical RG57 fingerprint to US1 and had mt-haplotype Ib, confirming the presence of an old population in Nepal. Most of the genotypes had a different RG57 fingerprint than that of US1 and mt-haplotype Ia, the common characteristics of new populations. The presence of a new population at high proportions in Nepal was consistent with the global trend of mt-haplotype distribution, and suggests the displacement of old populations. This study indicates at least three possible introductions of P. infestans to Nepal.  相似文献

Documentation of ethnomedicinal use of plants has been considered a high priority to support the discovery of new drugs to benefit mankind. This paper deals with the field observations recorded on the use of medicinal plants in traditional health care systems of the Tharus of three Villages of Nawalparasi district of central Nepal. The Tharus are isolated forest dwellers who have lived harmoniously with nature over the centuries and accumulated a vast knowledge of plant use to cure various ailments. First-hand information on the ethnomedicinal uses of 110 plants species belonging to 52 families were documented from the study. These plant species are used by the Tharus to treat ailments ranging from fever and headache to cough and cold, rheumatic pain and fracture to urinary tract infection and menstrual disorders. The scientific names of these plants are presented in tabular format according to alphabetic order, family, parts used, medicine preparation process and use and method of application. Increasing accessibility of modern health care facilities, low recognition of traditional healers and decreasing interest amongst the young generation has resulted in declining trends in the use of traditional medicine among the Tharus. Thus, the documentation of such knowledge is an important step for preservation of traditional knowledge, conservation and management of valuable plant resources and commercialization of ethnomedicines.  相似文献

Summary Potato late blight has appeared in epidemic proportions in Nepal since the mid 1990s and fungicides have been reported to be decreasingly effective in managing the disease.Phytophthora infestans isolates were collected from potato crops during 1999–2000 and analysis of 371 isolates for mating types and 270 isolates for metalaxyl sensitivity revealed the presence of both new and old populations. This is the first report on the presence of metalaxyl-resistant isolates in Nepal. The frequencies of A1 and A2 were 83 and 17%, respectively. Metalaxyl-resistant, intermediate and sensitive isolates were recorded as 10%, 12% and 78% respectively. Metalaxyl resistance was distributed in both mating types. Sites with a history of metalaxyl use had a significantly higher number of resistant and intermediate isolates ({ie337-1}, P<0.01) than sites where metalaxyl had not been used. This study confirms the changes in population structure ofP. infestans in Nepal.  相似文献

Summary A total of 251Phytophthora infestans isolates collected during 1999–2000 potato growing seasons from nine potato growing areas of Nepal were tested for virulence using a set of eleven potato differentials. The virulence test revealed the presence of 30 races with the most frequent being 4.11, 4 and 11. Twenty-nine percent of isolates belonged to simple races having none to one virulence factor, whereas 71% had complex virulence factors governed by 2 to 7 genes. Gleason and Shannon indices of 5.24 and 2.59, respectively, showed a moderate level of diversity in NepaleseP. infestans populations. Shannon indices comparison among regional populations revealed significant differences (t-test, P<0.001) in the diversity. The population from hills were the most diverse followed byterai and the high hill populations. The virulence complexities were minimal in Nepalese populations. The implications of these findings in association with the potato cultivation system in Nepal are discussed.  相似文献

BACKGROUND: In the mid‐southern region of the United States, sugarcane borer, Diatraea saccharalis (F.), is a major target pest of transgenic maize expressing Bacillus thuringiensis (Bt) proteins. Novel transgenic maize technologies containing two or more pyramided Bt genes for controlling lepidopteran pests have recently become commercially available. Insect resistance management (IRM) is an important issue in the sustainable use of Bt crop technologies. The objective of this study was to determine the frequency of resistance alleles in field populations of D. saccharalis to the new pyramided Bt maize technologies. RESULTS: A total of 382 F2 family lines derived from 735 feral larvae/pupae of D. saccharalis collected from six locations in Louisiana and Mississippi during 2008 and 2009 were screened for resistance to three new Bt maize technologies: MON 89034, Genuity? VT Triple Pro? and SmartStax?. None of the 382 F2 isoline families survived on the Bt maize leaf tissue for ≥ 12 days in the F2 screen. The joint frequency for two‐ or three‐gene resistance models with 95% probability in these populations was estimated to be < 0.0063 to MON 89034 and < 0.003 to VT Triple Pro? and SmartStax?. CONCLUSION: These results suggest that the resistance allele frequency in D. saccharalis to the three pyramided Bt maize technologies is low in the mid‐southern region of the United States, which should meet the rare resistance assumption of the currently used IRM strategy for Bt maize. White spot syndrome virus (WSSV) infection was induced in tiger shrimp, Penaeus monodon, under laboratory conditions, and histopathological changes in subcuticular epithelial cells of the eye stalk and pleopod were studied sequentially at different time post-challenge. Routine histological techniques using paraffin embedded tissues, as well as frozen tissues, were used to document WSSV infection. Histological manifestations such as cellular hypertrophy in the subcuticular epithelial cells of the eyestalk and pleopod could be detected as early as 18 h post-infection (p.i.) before the manifestation of clinical signs of the disease. However, no histopathological changes could be detected before 18 h p.i.. Hypertrophy of the nuclei in the epithelial cells was pronounced after 24 h p.i. Marked necrosis, and eosinophilic intranuclear inclusions, characteristic of early stages of WSSV infection were observed between 24–36 h p.i. Clinical signs of the disease appeared at 48 h p.i. The presence of WSSV at early asymptomatic stages of p.i. has been tested in parallel samples using polymerase chain reaction, for further confirmation of WSSV. This paper discusses the potential of a non-lethal and rapid histopathological diagnostic method to document WSSV infection, using the eyestalk or pleopod, when expensive DNA based diagnostics are not available or affordable.  相似文献

Wild meat harvests in African moist forests are presumed to exceed production, even in the case of traditional societies still using rudimentary hunting methods. Though some approximations do exist of the volume of bushmeat harvested in some Central African moist forest areas, estimates based on extensive and simultaneous sampling, within a large geographical region, are not available. Here, we present the results of the first reported study of this kind. During a period of 5 month, we counted bushmeat carcasses deposited in 89 urban and rural markets in a 35,000 km2 area between the Cross River in Nigeria and the Sanaga River in Cameroon. We used these data to calculate annual bushmeat volume traded by site, species and overall in the study area.Mammals represented >90% of the bushmeat carcasses sold in all sites. Reptiles were also abundant, but birds and amphibians were relatively scarce. Estimates of carcasses extracted and crude biomass per site varied significantly between countries. In Nigeria, biomass (kg) extracted for sale per km2 per year, was three times greater (600 kg/km2) than in Cameroon. Conservative estimates for the entire study area indicate that >900,000 reptiles, birds and mammals are sold each year by the rural and urban population, corresponding to around 12,000 tonnes of terrestrial vertebrates. We also assessed the relationship between bushmeat harvested for sale and distance of the study settlements from the main protected areas (Cross River and Korup National Parks). The number of carcasses and biomass sold was negatively related to the proximity to the national parks in >50% of species in Nigeria, and in 40% of species in Cameroon.Our cross-site comparison documents the staggering volume of wild species affected by hunting in the region. We also conclude that species within the main protected areas in both countries are likely to be negatively affected by the current and future demand for bushmeat in the surrounding areas.  相似文献
