Generation, investigation, and manipulation of nanostructured materials are of fundamental and practical importance for several disciplines, including materials science and medicine. Recently, atmospheric new particle formation in the nanometer-size range has been found to be a global phenomenon. Still, its detailed mechanisms are mostly unknown, largely depending on the incapability to generate and measure nanoparticles in a controlled way. In our experiments, an organic vapor (n-propanol) condenses on molecular ions, as well as on charged and uncharged inorganic nanoparticles, via initial activation by heterogeneous nucleation. We found a smooth transition in activation behavior as a function of size and activation to occur well before the onset of homogeneous nucleation. Furthermore, nucleation enhancement for charged particles and a substantial negative sign preference were quantitatively detected.  相似文献

A continuous high-resolution Western Mediterranean sea surface temperature (SST) alkenone record spanning the past 250,000 years shows that abrupt changes were more common at warming than at cooling. During marine isotope stage (MIS) 6, SST oscillated following a stadial-interstadial pattern but at lower intensities and rates of change than in the Dansgaard/Oeschger events of MIS 3. Some of the most prominent events occurred over MISs 5 and 7, after prolonged warm periods of high stability. Climate during the whole period was predominantly maintained in interglacial-interstadial conditions, whereas the duration of stadials was much shorter.  相似文献

Phototaxis and photophobic responses of green algae are mediated by rhodopsins with microbial-type chromophores. We report a complementary DNA sequence in the green alga Chlamydomonas reinhardtii that encodes a microbial opsin-related protein, which we term Channelopsin-1. The hydrophobic core region of the protein shows homology to the light-activated proton pump bacteriorhodopsin. Expression of Channelopsin-1, or only the hydrophobic core, in Xenopus laevis oocytes in the presence of all-trans retinal produces a light-gated conductance that shows characteristics of a channel selectively permeable for protons. We suggest that Channelrhodopsins are involved in phototaxis of green algae.  相似文献

Cell-to-cell communication is a crucial prerequisite for the development and maintenance of multicellular organisms. To date, diverse mechanisms of intercellular exchange of information have been documented, including chemical synapses, gap junctions, and plasmodesmata. Here, we describe highly sensitive nanotubular structures formed de novo between cells that create complex networks. These structures facilitate the selective transfer of membrane vesicles and organelles but seem to impede the flow of small molecules. Accordingly, we propose a novel biological principle of cell-to-cell interaction based on membrane continuity and intercellular transfer of organelles.  相似文献

The seeds ofLupinus mutabilis show on average a protein content of 42.6% and a fat content of 18.7%. Unfortunately they contain up to 4.5% of bitter alkaloids which must be removed before consumption. Edible oil and protein concentrate were produced from the seeds at an industrial level. The oil is of good quality and is recommended for human nutrition. The main fatty acid is oleic while the main tocopherol is γ-tocopherol. During the refining process, which includes a washing process with aqueous acids for debittering, the alkaloid content of the oil is reduced to 5 ppm. During the extraction of the oil the alkaloids are concentrated in the oil cake, from which they can be removed by washing with an ethanol-water mixture. A protein concentrate containing 72.3% protein and 0.06% residual alkaloids is obtained. The analysis of alkaloid pattern by gas-chromatography showed that the alkaloid composition of the crude oil differs totally from that of the seed. The relative percentage of the alkaloid esters and that of the Tetrahydrorhombifoline are very much higher in the oil than in the seed. During the debittering process of the oil cake the relative percentage of the hydroxylupanines increases.  相似文献

The Mediterranean sponge Hamigera hamigera (family Anchinoideae) was studied since its total extract showed deterrent activity in a fish feeding assay. Eight compounds were isolated from the biologically active fractions and four of these proved to be new natural products, hamigeroxalamic acid (1), hamigeramine (2), hamigeramide (3) and hamiguanosinol (5). The structures of the new compounds were elucidated by 1D and 2D NMR spectroscopy and mass spectrometry.  相似文献

A survey of the production of polyunsaturated aldehydes (PUA) of manipulated plankton communities is presented here. PUA are phytoplankton-derived metabolites that are proposed to play an important role in chemically mediated plankton interactions. Blooms of different intensities of the diatom Skeletonema marinoi were generated in eight mesocosms filled with water from the surrounding fjord by adding different amounts of a starting culture and nutrients. This set-up allowed us to follow PUA production of the plankton community over the entire induced bloom development, and to compare it with the natural levels of PUA. We found that S. marinoi is a major source for the particulate PUA 2,4-heptadienal and 2,4-octadienal (defined as PUA released upon wounding of the diatom cells) during the entire bloom development. Just before, and during, the decline of the induced diatom blooms, these PUA were also detected in up to 1 nM concentrations dissolved in the water. In addition, we detected high levels of the PUA 2,4-decadienal that was not produced by the diatom S. marinoi. Particulate decadienal correlated well with the cell counts of the prymnesiophyte Phaeocystis sp. that also developed in the fertilized mesocosms. Particulate decadienal levels were often even higher than those of diatom-derived PUA, indicating that PUA sources other than diatoms should be considered when it comes to the evaluation of the impact of these metabolites.  相似文献

Climate change drives a northward shift of biomes in high-latitude regions. This might have consequences on the decomposition of plant litter entering the soil, including its lignin component, which is one of the most abundant components of vascular plants. In order to elucidate the combined effect of climate and soil characteristics on the decomposition pattern of lignin, we investigated lignin contents and its degree of oxidative decomposition within soil profiles along a climosequence in western Siberia. Soil samples were collected from organic topsoil to mineral subsoil at six sites along a 1500-km latitudinal transect, stretching from tundra, through taiga and forest steppe to typical steppe. The stage of lignin degradation, as mirrored by decreasing organic carbon-normalized lignin contents and increasing oxidative alteration of the remnant lignin (acid-to-aldehyde ratios of vanillyl- and syringyl-units [(Ac/Al)V and (Ac/Al)S]) within soil horizons, increased from tundra to forest steppe and then decreased to the steppe. Principal component analysis, involving also climatic conditions such as mean annual temperature and aridity index, showed that the different states of lignin degradation between horizons related well to the activity of phenoloxidases and peroxidases, enzymes involved in lignin depolymerization that are produced primarily by fungi and less importantly by bacteria. The low microbial lignin decomposition in the tundra was likely due to low temperature and high soil moisture, which do not favour the fungi. Increasing temperature and decreasing soil moisture, facilitating a higher abundance of fungi, led to increased fungal lignin decomposition towards the forest-steppe biome, while drought and high pH might be responsible for the reduced lignin decomposition in the steppe. We infer that a shift of biomes to the north, driven by climate change, might promote lignin decomposition in the northern parts, whereas in the south a further retardation might be likely.  相似文献

Background

Little is known about the effects of gypsum application to remediate saline–sodic soils in the tropics and the role of microbial indicators in soil reclamation.

Aims

Our study aimed at (1) remediating a highly weathered, irrigated sodic Lixisol under prolonged urban crop production by clean water and gypsum application and (2) to determine the remediation effects on soil microbial indices.

Methods

A three-factorial on-farm experiment with maize (Zea mays L.) was used to study effects on soil microbial biomass of (1) soil degradation at two levels of salinity, (2) irrigation with clean water and wastewater, and (3) the impact of added gypsum during a typical growing season.

Results

At the high-degradation site, the 0.5 M K2SO4 extractable carbon (C) content was 40% higher than at the low-degradation site. In addition, microbial biomass carbon (MBC) and microbial biomass nitrogen (MBN) were 20% lower than at the low-degradation site, while fungal ergosterol was even 40% lower, leading to a 33% lower ergosterol/MBC ratio. Wastewater irrigation increased MBN but decreased ergosterol content at the low-degradation site while having no effect at the high-degradation site. Gypsum amendment led to higher MBN at the low-degradation site but to lower MBN at the high-degradation site. Gypsum amendment always increased the ergosterol content whereby this increase was stronger at the low-degradation site, especially in combination with wastewater irrigation.

Conclusions

From a microbial perspective, high soil degradation levels should be avoided by treatment of a saline–sodic wastewater prior to its use for irrigation rather than relying on future remediation strategies of affected field sites.  相似文献

Mycoplasma conjunctivae is considered the major cause of infectious keratoconjunctivitis (IKC) in Alpine ibex (Capra i. ibex) and chamois (Rupicapra r. rupicapra). While it is known that domestic sheep can act as healthy carriers for M. conjunctivae, this question has not been addressed in wild ungulates so far. In this study, bacteriological investigations and field observations were performed to assess whether free-ranging Alpine ibex can be healthy carriers of M. conjunctivae. Among 136 ibex without clinical signs of IKC, M. conjunctivae was identified 26 times (19.1%) by TaqMan PCR. To assess the potential pathogenicity of M. conjunctivae strains isolated from asymptomatic eyes, strains from three healthy ibex and from 15 IKC-ibex and IKC-chamois were analysed genetically by DNA sequence analysis of the variable part of the lppS gene. No significant differences were observed between strains from asymptomatic and clinically affected animals, reflecting the assumption that healthy ibex may act as carriers for M. conjunctivae strains that may be pathogenic for other individuals. Our results further indicate that development of IKC is associated with M. conjunctivae load in the eyes. In addition, a questionnaire survey revealed that IKC is generally less common in ibex than chamois and that infection in wild ungulates is not necessarily linked to the presence of sheep. These data support the hypothesis that apparently healthy ibex may be important in the epizootiology of IKC and indicate that host predilection may play a role in IKC development.  相似文献
