Pectin methylesterase (PME) from green bell peppers (Capsicum annuum) was extracted and purified by affinity chromatography on a CNBr-Sepharose-PMEI column. A single protein peak with pectin methylesterase activity was observed. For the pepper PME, a biochemical characterization in terms of molar mass (MM), isoelectric points (pI), and kinetic parameters for activity and thermostability was performed. The optimum pH for PME activity at 22 degrees C was 7.5, and its optimum temperature at neutral pH was between 52.5 and 55.0 degrees C. The purified pepper PME required the presence of 0.13 M NaCl for optimum activity. Isothermal inactivation of purified pepper PME in 20 mM Tris buffer (pH 7.5) could be described by a fractional conversion model for lower temperatures (55-57 degrees C) and a biphasic model for higher temperatures (58-70 degrees C). A study to evaluate the changes in the fertility of Fluvisols under continuous cultivation of sugarcane was conducted in the vicinity of the Santa Rosalía sugar refinery. About 4000 ha of Fluvisols have been under sugarcane cultivation for more than 30 years. Three representative plots under monoculture for 5, 10, 20 and 30 years, were selected under on-farm conditions. Several samples were obtained from crop row and inter-row zones, at 0–30 cm soil depth, and composited into four samples for each plot. Soil samples were analyzed for pH, organic matter, total organic C, total nitrogen, available phosphorus, exchangeable potassium, calcium and magnesium, cation exchange capacity, texture and bulk density. There were significant changes in the soil chemical properties due to 30 years of monoculture, except for OM, exchangeable K, Ca, Mg and CEC. Total soil organic C, total N and P declined by 17, 21 and 37%, respectively, by 30 years of monoculture of sugarcane. These losses mainly occurred during the first 20 years of cultivation. However, the adverse effect of decline in soil fertility was not reflected in the sugarcane yields. The 67% increase in yields observed between 5 and 30 years of monoculture may be attributed to the adoption of recommended cultural practices and to farmer's experience. Soil physical properties were not affected by the intensive sugarcane cultivation. A complete adoption of recommended practices by SR among farmers and a revision of the N and P fertilization rate are necessary to maintain the fertility of sugarcane soils. 相似文献

High-performance size-exclusion chromatography (HPSEC), static light scattering (SLS) and dynamic light scattering (DLS) techniques were used for the structural characterization of amaranth starch, solubilized in water by microwave heating in a high-pressure vessel. Apparent average molar mass (M w) gyration radius (R G), and hydrodynamic radius (R H) values were obtained from Berry and Zimm treatment of light-scattering data. When heating time increased from 35 to 90 sec, the M w, R G, and R H decreased, demonstrating a possible polymer degradation due to temperature. Apparent M r values from HPSEC at 35 sec (27 ± 2 × 107 g/mol) and 50 sec (20 ± 2 × 107 g/mol) were lower than those determined by SLS (35 sec = 69 × 107 g/mol, 50 sec = 56 × 107 g/mol). However, at 70 and 90 sec, the inverse pattern was obtained. The fractal dimensions (d′f) from HPSEC study for samples dissolved for 35 (3.26), 50 (3.24), and 70 sec (3.14) are characteristic of a particle that has the internal structure of hard sphere, and for samples dissolved for 90 sec (2.19), are characteristic of a fully swollen, randomly branched macromolecule. From SLS, d′f decreased with increasing treatment time (d′f = 2.44, 2.18, 1.50, and 1.03 for 35, 50, 70, and 90 sec, respectively). The particle-scattering factors and Kratky plots, well-suited for studying the internal structure of a macromolecule, showed a sample degradation when treatment time increased. Results from DLS showed bimodal distributions with differences in the peak locations when treatment time increased. The ratio of R G to R H (ρ) for samples analyzed were between 0.88 and 1.3; these values are characteristic of a sphere or globular structure. 相似文献

This study comparatively evaluates phosphorus (P) use efficiency of safflower and sunflower in view of identifying low input oil crops. Sunflower accumulated more P than safflower at all equivalent P supplies. Sunflower was more efficient at their best P supplies to utilize absorbed P than safflower in terms of efficiency ratio and utilization index. Yield response curves interpreting the functional relationship between nutrient supply and yield using Michaelis-Menten equation indicate that sunflower is less sensitive to inadequate P supply and thus more P-efficient than safflower with respect to yield, as the term ‘Km’ is lower for sunflower. The minimal P amount per pot required to trigger yield formation (term ‘Cmin’) was lower in sunflower than safflower. Harvest Index in sunflower out-yielded that of safflower at all P supplies. Critical deficiency contents of sunflower were lower than that for safflower. It can be concluded that safflower cannot be considered as low-input crop. 相似文献

The effect of increasing chloride content in nutrient solution on nutrient composition in root environment, interaction of nutrients in leaves and yield of greenhouse tomato cv. ‘Grace F1’ grown in rockwool were searched. In Experiment I (2004–2005) the levels of 15, 30, 60, and 90 mg Cl·L?1 but in Experiment II (2006) 30, 60, 90 and 120 mg Cl·L?1 of nutrient solution were tested. The sources of chloride were water (9.6–10.7 mg Cl·L?1) and calcium chloride (CaCl2·2H2O) but the rest of nutrients and sodium in all treatments were on the same levels. It was found that increasing content of chloride from 15 to 60 mg Cl·L?1 enhanced the total and marketable fruit yield. Within the range of 60 to 90 mg Cl·L?1 the yield was on the optimum level but the content of 120 mg Cl·L?1 declined it. Increasing chloride content in the nutrient solutions was reflected in rising of chlorine content in leaves. The concentration of chloride above 60 mg C·L?1 reduced the content of nitrogen but above 90 mg C·L?1 declined the content of calcium, sulfur and zinc in leaves. The antagonism between Cl:N, Cl:Ca: Cl:S and Cl:Zn was appeared. More variable interaction were between Cl:K and Cl:B. At the low levels of chloride, from 15 to 60 mg Cl·L?1, potassium and boron content were decreased but at the higher ones, from 90 to 120 mg·L?1, these nutrients had increasing course. It was not found out the effect of chloride contents on macro and microelement contents in nutrient solution emitted from drippers however their content upraising in root medium (rockwool). The highest increase was found out for Na 95.1 and 64.9 % (Exp. I and II - respectively), next for Ca (76.0, 70.1 %), Cu (62.5 and 71.0 %), Cl (43.6, 24.4), B (33.3, 21.0 %), N-NO3 (30.4, 49.6 %), Zn (29.5, 32.8 %), S-SO4 (25.9, 25.5 %), K (24.5, 24.1 %), Fe (19.8, 19.2 %), Mn (17.5, 21.3 %) and Mg (14.9, 11.7). Advantageous effect of chloride on tomato yield justified the need to introduce for the practice adequate chlorine nutrition, and recommend to maintain 60 to 90 mg Cl·L?1 in nutrient solution. The method to measure hardness and other viscoelastic properties of intact wheat kernels is presented. Wheat with 9.3% moisture showed high elastic behavior compared with wheat tempered at 22.5% moisture that showed a plastic behavior. Load‐deformation curves showed that bread wheat behaves as a more plastic material than durum wheat, which is a more elastic material. Yield point of all the wheat samples was ≈18.5 N, independent of wheat type and moisture content. The height of the wheat kernel increased linearly, and the compression area increased exponentially, with increasing moisture content. The modulus of elasticity of wheat ranged from 99.2 MPa for 22.5% moisture content to 394.8 MPa for 9.3% moisture content. Young's modulus range for soft wheat such as Salamanca, Saturno, and Cortazar cultivars was 232.2–308.5 MPa, as compared with Rayón bread wheat at 321.5 MPa and the Altar, Sofía, and Rafi cultivars of durum wheat that had elastic moduli of 438.7–485.8 MPa. The compression force and final stress decreased from 69.9 N and 40.1 MPa in soft wheat to 90 N and 78.9 MPa in durum, respectively. Total work range was 14.7 MPa/sec in soft wheat to 19.7 MPa/sec for durum wheat and, as expected, was higher in the durum and bread wheat than in soft wheat. Effect of tetracycline (TC) administration on the proteomic profile of pig muscle was evaluated by 2D electrophoresis and MALDI-TOF mass spectrometry. The TC content at slaughter was determined in L. dorsi samples by HPLC-DAD. Mean residual concentration of TC in the muscle of treated animals, calculated as the sum of TC and epi-TC was 126.3 microg/kg, indicating a rapid elimination of TC in this tissue. Several differential spots (n = 54, p < 0.05) were observed in protein profiles from control and treated animals. MALDI-TOF identification gave a positive match for 5 differential spots, that is, glycerol-3-phosphate dehydrogenase 1 (G3PD1), phosphoglycerate kinase 1, novelprotein (0610037L13Rik), leucine aminopeptidase 3 (LAP), and hypothetical protein isoform 2. Results show that proteomics could be a useful tool to reveal pharmacological treatments with TC, even if the possible uses of differential spots as biomarkers to detect illegal administration of TC require further studies. Different spot patterns as a consequence of TC treatments seem to be another interesting issue for the consequences on tissue metabolism and meat quality. 相似文献

The phenolic fraction of virgin olive oil influences both its quality and oxidative stability. One of the principal threats of the quality of olive fruit is the olive fly ( Bactrocera oleae) as it alters the chemical composition. The attack of this olive pest has been studied in order to evaluate its influence on the quality of virgin olive oil (free acidity, peroxide value, fatty acid composition, water content, oxidative stability, phenols, and antioxidant power of phenolic fraction). The study was performed using several virgin olive oils obtained from olives with different degrees of fly infestation. They were acquired in different Italian industrial mills from the Abruzzo region. Qualitative and quantitative analyses of phenolic profiles were performed by capillary electrophoresis-diode array detection, and electrochemical evaluation of the antioxidant power of the phenolic fraction was also carried out. These analyses demonstrated that the degree of fly attack was positively correlated with free acidity ( r = 0.77, p < 0.05) and oxidized products ( r = 0.58, p < 0.05), and negatively related to the oxidative stability index ( r = -0.54, p < 0.05) and phenolic content ( r = -0.50, p < 0.05), mainly with secoiridoid compounds. However, it has been confirmed that the phenolic fraction of olive oil depends on several parameters and that a clear correlation does not exist between the percentages of fly attack and phenolic content. 相似文献
