AIM: To study the expression and its kinetics of rice phenylalanine ammonia-lyase gene encoding into E. coli as the basis of treatment for phenylketouria. METHODS: The phenylalanine ammonia-lyase-1-cDNA(rPAL-1-cDNA) from rice was recombined into E. coli high expression vector pET-28c and transformed into E. coli host strain BL21DE3. Engineering bacteria was then inducted by isopropyl-β-D-thiogalactoside (IPTG) for 1, 3, 5, 7 hours, in order to obtain high level expression. RESULTS: After induction, the expression level of fusion protein was 21.40%, 30.60%, 35.40%, 35.43% respectively. The fusion protein exhibited a band of 78.6 kD on SDS-PAGE analysis, but was not found in controls.The target protein was mainly existed in the form of inclusion body. CONCLUSION:Rice PAL gene expressing E. coli was established by gentic engineering technique. 相似文献

AIM To investigate the role of curcumin (CUR) in lung ischemia/reperfusion (I/R) injury (LIRI) and its relationship with autophagy. METHODS 40 SD rats were randomly divided into sham operation (sham) group, I/R group, solvent (DMSO) group, CUR group and CUR+rapamycin (CUR-Rap) group. The rats were intraperitoneally injected with normal saline, DMSO, CUR or CUR+Rap before operation. After the rat LIRI model was established, the lung tissues were taken to measure W/D, TLW, IAR, and the contents of SOD and MDA were also measured to indicate the oxidative stress level. Light and electron microscopes were used to observed the morphology and ultrastrucure of lung tissues. The expression levels of autophagy-related proteins were determined by Western blot to evaluate autophagy levels. RESULTS Compared with sham group, wet weight/dry weight (W/D), total lung water (TLW), injured alveoli rate(IAR) and malondialdehyde (MDA) content in all other groups were increased, superoxide dismutase (SOD) activity was decreased, the levels of autophagy were increased (P <0.05), and lung tissue injury and cell ultrastructural damage were aggravated in CUR group. Compared with DMSO group, W/D, TLW and IAR and MDA content were decreased, SOD activity was decreased, autophagy levels were also decreased (P <0.05), and lung tissue and cell ultrastructural damage were attenuated. Compared with CUR group, W/D, TLW, IAR and MDA content were increased, SOD activity declined, the autophagy levels were increased (P <0.05), and damage of lung tissues and cells were more serious in CUR-Rap group. CONCLUSION Curcumin attenuates the lung I/R injury in rats, and its mechanism may be related to the reduction of oxidative stress and the inhibition of autophagy. 相似文献

稻麦两熟制条件下钵苗机插方式对不同类型水稻品种米质的影响 总被引:3,自引:0,他引:3
为探明钵苗机插方式在稻麦两熟地区对水稻米质性状的影响及其品种类型间差异,本文以常规粳稻、杂交籼稻和籼粳交水稻为材料,比较了钵苗机插、毯苗机插和机械直播3种种植方式在稻麦两熟制高产栽培条件下的稻米加工品质、外观品质、营养品质、蒸煮食味品质特性变化及其品种类型间差异。结果表明,钵苗机插的整精米率比毯苗机插和机械直播分别提高1.6%和4.9%(P0.05),整精米产量分别提高7.8%和25.9%(P0.05),改善了稻米加工品质;与毯苗机插和机械直播相比,钵苗机插的稻米垩白米率、垩白面积和垩白度显著增加,对稻米外观品质有不利影响,但钵苗机插精米蛋白质含量较高,有利于稻米营养品质的改良,其精米蛋白质产量分别比毯苗机插和机械直播提高8.7%和28.0%;3种种植方式相比,钵苗机插稻米蒸煮食味品质表现出一定程度的变优趋势,其中,稻米直链淀粉含量、消减值和糊化温度呈钵苗机插毯苗机插机械直播的趋势,胶稠度、峰值黏度、热浆黏度、最终黏度和崩解值呈钵苗机插毯苗机插机械直播的趋势;食味值于种植方式间无显著差异,呈籼粳交水稻和常规粳稻钵苗机插毯苗机插机械直播,杂交籼稻毯苗机插钵苗机插机械直播的趋势。由此得出,钵苗机插不仅能增加稻谷产量,还能改善部分稻米品质特征,是稻麦两熟地区实现高产高效优质协调水稻生产的潜力型机械化种植方式。 相似文献

针对Tripod并联机器人工作空间一般用数值搜索方法研究,难以给出边界曲面准确表达的问题,提出一种曲面扫略分析与机械结构形位分析结合的方法来解析Tripod并联机器人的工作空间。以正逆解的算法为基础,得到并联机器人各单开链扫略空间范围及其包络曲面表达式,用三维软件布尔运算得出各支链公共的工作空间,在考虑虎克铰约束和杆件干涉的情况下,再依据机械结构形位分析得到机器人实际工作空间及边界曲面表达式,并对工作空间的奇异性进行了分析。通过三坐标测量仪器测出实际的工作空间与理论分析解析出来的空间进行对比来验证解析方法的正确性,从而为机器人机械结构参数设计和工作空间计算奠定基础。 相似文献

AIM: To study the effect of fibroblast growth factor receptor 1 (FGFR1) expression knock-down on the viability, apoptosis, invasion and migration of infantile hemangioma endothelial cells (HemECs). METHODS: FGFR1 was down-regulated by FGFR1 small interfering RNA (si-FGFR1) transfection. The viability of the cells was measured by CCK-8 assay. The apoptotic rate was analyzed by flow cytometry and the invasion and migration abilities were determined by Transwell assay. The protein levels of phosphatidylinositol 3-kinase (PI3K), protein kinase B (AKT), and phosphorylated AKT (p-AKT) were examined by Western blot. RESULTS: Transfection of si-FGFR1 into HemECs had significant effects on inhibiting cell viability (P<0.05), promoting apoptosis (P<0.05), and decreasing cell invasion and migration abilities (P<0.05). The results of Western blot showed that knockdown of FGFR1 gene expression in the cells reduced the protein levels of PI3K and p-AKT (P<0.05), and had no significant effect on AKT protein level. CONCLUSION: Knock-down of FGFR1 expression changes the biological characteristics of endothelial cells in infantile hemangiomas by regulating PI3K/AKT signaling pathway. 相似文献
