Bacterial leaf blight (BLB), caused by Xanthomonas axonopodis pv. vignicola (Xav), is widespread in major cowpea [Vigna unguiculata (L.) Walp.] growing regions of the world. Considering the resource poor nature of cowpea farmers, development and introduction of cultivars resistant to the disease is the best option. Identification of DNA markers and marker‐assisted selection will increase precision of breeding for resistance to diseases like bacterial leaf blight. Hence, an attempt was made to detect QTL for resistance to BLB using 194 F2 : 3 progeny derived from the cross ‘C‐152’ (susceptible parent) × ‘V‐16’ (resistant parent). These progeny were screened for resistance to bacterial blight by the leaf inoculation method. Platykurtic distribution of per cent disease index scores indicated quantitative inheritance of resistance to bacterial leaf blight. A genetic map with 96 markers (79 SSR and 17 CISP) constructed from the 194 F2 individuals was used to perform QTL analysis. Out of three major QTL identified, one was on LG 8 (qtlblb‐1) and two on LG 11 (qtlblb‐2 and qtlblb‐3). The PCR product generated by the primer VuMt337 encoded for RIN2‐like mRNA that positively regulate RPM1‐ and RPS2‐dependent hypersensitive response. The QTL qtlblb‐1 explained 30.58% phenotypic variation followed by qtlblb‐2 and qtlblb‐3 with 10.77% and 10.63%, respectively. The major QTL region on LG 8 was introgressed from cultivar V‐16 into the bacterial leaf blight susceptible variety C‐152 through marker‐assisted backcrossing (MABC).  相似文献

Field experiments were carried out during rainy (kharif) and winter (rabi) seasons (June–April) of 2008–2010 at Indian Agricultural Research Institute (IARI), New Delhi, to study the productivity, nutrients uptake, iron (Fe) use-efficiency and economics of aerobic rice-wheat cropping system as influenced by mulching and Fe nutrition. The highest yield attributes, grain and straw yields (5.41 tonnes ha?1 and 6.56 tonnes ha?1, respectively) and nutrient uptake in rice was recorded with transplanted and puddled rice (TPR) followed by aerobic rice with Sesbania aculeata mulch. However, residual effect of aerobic rice with wheat straw mulch was more pronounced on yield attributes, grain and straw yields (4.20 and 6.70 tonnes ha?1, respectively) and nutrient uptake in succeeding wheat and remained at par with aerobic rice with Sesbania mulch. Application of iron sulfate (FeSO4) at 50 kg ha?1 + 2 foliar sprays of 2% FeSO4 was found to be the best in terms of all the yield attributes, grain and straw yield (5.09 and 6.17 tonnes ha?1, respectively) and nutrient uptake and remained at par with 3 foliar sprays of 2% FeSO4. Although residual effect of iron application failed to increase the yield attributes, yield and nutrient uptake nitrogen, phosphorus and potassium (N, P, K) except Fe. The highest system productivity, nutrient uptake, gross returns, net returns, B: C ratio and lowest cost of cultivation were recorded with aerobic rice with wheat straw and Sesbania aculeata mulch. Application of FeSO4 at 50 kg ha?1 + two foliar sprays of 2% FeSO4 was found better in respect of system productivity, nutrient uptake, gross returns, net returns, B:C ratio and cost of cultivation in aerobic rice-wheat cropping system. The Fe use efficiency values viz. partial factor productivity (kg grain kg?1 Fe), agronomic efficiency (kg grain increased kg?1 Fe applied), agrophysiological efficiency (kg grain kg?1 Fe uptake), physiological efficiency (kg biomass kg?1 Fe uptake), apparent recovery (%) utilization efficiency and harvest index (%) of applied Fe were significantly affected due to methods of rice production and various Fe nutrition treatments in aerobic rice and aerobic rice-wheat cropping system.  相似文献

Influence of drought and salt stress on different morphological and physiological growth parameters in Capsicum annuum inoculated with our isolates was estimated during the present study. Bulkhorderia cepacia was reported to possess the maximal, whereas Citrobacter feurendii the least plant growth promoting efficacy under salt and drought stress. ACC Deaminase activity of purified B. cepacia, C. feurendii and Serratia marcescens was 12.8 ± 0.44, 12.3 ± 0.56 and 11.7 ± 0.53 μM αKB mg?1 min?1 respectively. Under drought stress, B. cepacia showed maximum tolerance as it produced 4.893 ± 0.06 mg/mg protein of exopolysaccharide followed by C. feurendii and S. marcescens that produced 4.23 ± 0.03 and 3.46 ± 0.05 mg/mg protein, respectively. Chlorophyll “a” concentration was recorded 5.7 gm L?1 in B. cepacia inoculated plant (without stress) and was sustained till 2.9 gm L?1 even under the highest tested drought period. Chlorophyll “a” concentration in the B. cepacia inoculated plant under the highest tested NaCl concentration was 3.2 gm L?1. Thus, bacterial inoculation mitigates the effects of salinity by the proliferation of root system, increasing plant biomass proving to be potential bioinoculum for alleviating abiotic stress.  相似文献

The aim of this study was to determine the genetics of polymorph systems of Transferrin in Garole sheep breed. The present study was conducted on 95 adult Garole sheep comprising 52 ewes and 43 rams, maintained at Sheep and Goat Breeding Farm of West Bengal University of Animal and Fishery Sciences, West Bengal, during the period from April–September, 2009. The polymorphism of transferrin was determined through SDS-Polyacrylamide gel electrophoresis technique. It was found that the transferrin type was controlled by five codominant alleles (TfA, TfB, TfC TfD and TfE) in Garole sheep. These five alleles, because of co-dominant nature of inheritance, determined the occurrence of nine transferrin genotypes in the analyzed flock. Four (TfAA, TfBB, TfCC and TfDD) of these were homozygous and the remaining five (TfAD, TfBC, TfBD, TfCD and TfDE) heterozygous. It was found that the TfDD genotype (0.263) was predominant while TfDE genotype (0.042) was least common in the analyzed flock. Frequencies of other genotypes were as: TfCD(0.242), TfBD(0.126), TfCC(0.084), TfBB(0.074), TfAA(0.063), TfAD and TfBC (0.053 for each genotype ) in whole population. From the result it was found that in whole population combined, the heterozygotic genotypic frequency (0.516) was more than that of homozygotic genotypic frequency (0.484). Considerable variations were recognized in the frequencies of transferrin alleles. In the whole population frequencies of transferrin alleles were found to be TfA = 0.089, TfB = 0.163, TfC = 0.232, TfD = 0.495 and TfE = 0.021. Transferrin system has shown an absence of genetic equilibrium among the analyzed herd (χ2 value = 51.31). In conclusion, there were polymorphism in Transferrin types and the presence of differences among the frequencies of the five alleles by categories could be a source of genetic variation in Garole sheep.  相似文献

Phenolic content and discoloration were determined for 13 commercial potato cultivars and 6 potato clones grown at 2 locations over a period of 5 successive years. Tubers from 6 potato cultivars and 5 potato clones resistant to the golden nematode were compared with tubers from 7 potato cultivars and 1 clone known to be susceptible to the golden nematode. Tubers from resistant plants were lower in phenols and discolored less than tubers from susceptible plants.  相似文献

The effect of freezing on the chemical composition of Katahdin and Atlantic potatoes was studied. Tubers were exposed to a temperature of ?20°C for 24 hrs., then thawed for 1 hr. at 20°C prior to analysis. Frozenthawed tubers were compared with unfrozen controls for total lipid, phospholipid, fatty acid and mineral content. Following freezing, a significant (p<.01) decrease was observed in crude lipid and phospholipid content of both cortex and pith sections. Generally, an increase in palmitic acid and a decrease in linolenic acid occurred in the phospholipid fractions of both varieties. A decrease in both macro and micro mineral content also occurred following freezing.  相似文献

1. An investigation was carried out into the effects of dietary α-tocopherol (α-T) concentration and source of supplemental oil on performance, activity of anti-oxidative enzymes and some immune responses in broilers from day-old to 41 d of age. 2. Three dietary concentrations of α-T (10, 50 and 100 mg/kg) with three sources of supplemental oil (sunflower - SFO, palm - PMO and safflower - SAO) were provided using a 3 × 3 experimental design. 3. Body weight gain and food conversion efficiency were not affected by either interaction or concentrations of α-T and sources of oil in diet. 4. Concentrations of total protein, globulin, triglycerides and cholesterol in sera increased significantly with dietary α-T concentration irrespective of the source of oil. Significantly higher concentration of serum albumin was evident in broilers fed on the SFO-based diet and the concentration of globulin was higher in groups fed on those diets containing PMO and SAO. 5. The lipid peroxidation (LP), measured as MDA release, decreased with the concentration of α-T in a dose-related manner with SFO- and SAO-based diets, although not with the PMO-based diet. With different oil sources, LP was significantly lower with the PMO-based diet compared to the others. Activities of glutathione peroxidase and RBC catalase increased and heterophil: lymphocyte ratio was reduced with concentration of α-T for each source of oil tested. 6. Assays for humoral and cell-mediated immune responses indicated no effect of the source of dietary supplemental oil or interaction, although an increasing concentration of dietary α-T improved cell-mediated immune responses. 7. It is concluded that sunflower oil, palm oil and safflower oil can be used as sources of oil for broiler diets without having any effect on performance, immune responses or the activity of anti-oxidizing enzymes. Higher concentrations of dietary α-tocopherol (50 or 100 mg/kg) reduced lipid peroxidation activity and enhanced activities of anti-oxidative enzymes, they also improved the cell-mediated immune responses in commercial broilers.  相似文献

The influence of phosphorus fertilization on the content of ascorbic acid, lipids, total nitrogen, protein, and non-protein nitrogen of Katahdin potatoes was investigated. Phosphorus in the form of mono ammonium phosphate was applied at rates of 0, 56, 112, and 168 Kg/ha (0, 50, 100, and 150 lb/acre). Ascorbic acid, total nitrogen, protein and non-protein nitrogen content were significantly increased (p<.01) and the phospholipid content was significantly reduced (p<.01) by phosphorus application.  相似文献

The present study was undertaken to estimate effect of various genetic and non-genetic factors on persistency of milk production and to identify the most appropriate persistency method that fits best in our environment. In the present study, effects of different non-genetic factors, viz. year, season, days to attain peak yield, and genetic group based on the level of exotic inheritance on persistency of milk yield in crossbred cattle were studied. Data comprised of 686 first lactation daily milk yield records of crossbred cattle that were maintained at GADVASU dairy farm over a period of 25 years from 1991 to 2015 were utilized to calculate persistency coefficients by four methods, viz., Ludwick and Peterson method (P1), Mahadevan method (P2), ratio method (P3), and Prasad et al. method (P4). Overall least squares means for persistency by Ludwick and Peterson method (P1), Mahadevan method (P2), ratio method (P3), and Prasad et al. method (P4) were 0.896?±?0.096, 1.385?±?0.224, 187.207?±?26.398, and 0.621?±?0.098, respectively. Effect of sires was significant (P?<?0.05) on P2 and P4 methods. Effect of genetic group on all four methods was non-significant. Period of calving had significant (P?<?0.01) effect on persistency of milk yield (P2, P3, and P4 methods). Effect of season of calving on persistency of milk yield was found to be significant in all estimates obtained by the four methods. Summer and autumn calvers were most persistent whereas spring and winter calvers were least persistent for (P2, P3, and P4 methods). Persistency of milk yield was significantly (P?<?0.05) affected by days to attain peak yield in P1 and P2 methods. Maximum persistency was obtained in animals attaining peak at 41–57 days of lactation and minimum in <?41 days for Mahadevan method and ratio method. The highest heritability of persistency and minimum value of standard error was estimated as 0.275?±?0.11 for the Mahadevan method followed by the Prasad method (0.197?±?0.10) by half sib correlation method. The maximum coefficient of variation which indicates available variability was estimated as 20.788% for persistency by the Mahadevan method followed by 18.969% for the Prasad method. The highest correlation was also observed between P1 and P3 methods by Spearman’s and Pearson’s correlation for least squares breeding value of the sires. On the basis of heritability, standard error of heritability, and coefficient of variation, it can be concluded that the Mahadevan method followed by the Prasad method suits best to our environment for animals in first lactation as well as they can be utilized for effective selection for higher persistency in crossbred animals of Punjab.  相似文献

An attempt was made for the first time in India to develop the performance indices for quantifying the relative emphasis to be given to production, reproduction, health and longevity traits for selection and genetic improvement of Sahiwal cattle in a sustainable manner. The study was conducted using information related to various production, reproduction, health and longevity traits spread over a period of 29 years in Sahiwal cattle. Using income and expenditure method, the relative economic values for 305 days milk yield (305DMY), average daily milk yield (ADMY), calving to first insemination interval (CFI), days open (DO) and longevity (LNG) were estimated as 1, 1, −6.62, −6.62 and 5.96 in Sahiwal cattle. A total of three performance indices were constructed using three-trait combination, that is production, reproduction and longevity before and after excluding the days suffered by the animals. Correlation between the aggregate genotype and index (RIH) was computed to determine the accuracy of each performance index, and based on highest RIH value, best performance index was identified for selection of Sahiwal cattle. Developed indices were further assessed for robustness by increasing the relative economic values of the traits by 25% and 50%. The performance index (305DMY, DO and LNG) was found to be the best index before as well as after excluding days suffered by the animals, but in later case emphasis to be given to different traits was found to be more balanced. The best index indicated that about 47, 42 and 11% relative emphasis to be assigned to 305 days milk yield, days open and longevity for selection of Sahiwal animals for sustainable genetic improvement.  相似文献
