A reduction of arbuscules in roots of grapevines (Vitis vinifera) observed when ring nematodes were added to field microplots led to the hypothesis that nematode feeding suppresses arbuscules by competing for root carbohydrates. Support for this hypothesis was tested by growing ‘Pinot noir’ grapevines in a factorial experiment with three levels of initial nematode densities (0, 0.1, 1.0 nematodes g?1 soil), two levels of light (full sun, 50% sun), and two levels of AMF (nonAMF, +AMF). Effects on plant growth were primarily driven by a light and AMF treatment interaction, such that low light increased stem dry matter accumulation at the expense of roots in +AMF vines only. Nematodes had only a minor influence on plant growth (leaf mass was reduced at the highest nematode density), but nematodes did not affect overall plant dry matter accumulation. Since nonAMF vines were severely limited by P and their growth was so poor, the impact of nematode and light treatments was further analyzed in +AMF plants only. Nematode populations, AMF colonization, and root carbohydrates were differentially affected by initial nematode density or light levels. Root biomass, and reducing sugar and starch concentrations in fine roots were reduced by low light, but the final nematode populations and arbuscule frequencies in roots were unaffected by light. Nematodes reduced arbuscules and starch concentrations in fine roots, but did not affect total colonization by AMF (hyphae, vesicles or arbuscules). Nematodes reduced plant P and K uptake at the highest density, and low light reduced Mg uptake. These findings are consistent with the hypothesis that ring nematodes suppress arbuscules in roots via competition for root carbohydrates. However, the lack of a treatment interaction between light and nematodes in our study suggests that ring nematode–AMF interactions in grape roots are controlled by more than competition for photosynthate.  相似文献

The depth dependence of physical properties along the Japan subduction zone interface was explored using teleseismic recordings of earthquake signals. Broadband body waves were inverted to determine the duration of rupture and source depth for 40 interplate thrust earthquakes located offshore of Honshu between 1989 and 1995. After scaling for differences in seismic moment, there is a systematic decrease in rupture duration with increasing depth along the subducting plate interface. This indicates increases in rupture velocity or stress drop with depth, likely related to variation in rigidity of sediments on the megathrust.  相似文献

The efficient representation of all species in conservation planning is problematic. Often, species distribution is assessed by dividing the land into a grid; complementary sets of grids, in which each taxon is represented at least once, are then sought. To determine if this approach provides useful surrogate information, species and higher taxon data for South African plants and animals were analyzed. Complementary species sets did not coincide and overlapped little with higher taxon sets. Survey extent and taxonomic knowledge did not affect this overlap. Thus, the assumptions of surrogacy, on which so much conservation planning is based, are not supported.  相似文献

A 5 yr old female spayed mastiff was evaluated for a 3-4 mo history of paraparesis and 3 days of acutely worse paraparesis and incontinence. On magnetic resonance imaging, a spinal cord lesion was present at the ninth thoracic vertebra. The lesion was hyperintense on T2-weighted images (T2-W), and a hyperintense rim was present on T1-weighted postcontrast images. Histologic examination showed a cystic mass lined by squamous epithelial cells. Histopathologic diagnosis was an intramedullary epidermoid spinal cyst. Epidermoid cyst should be a differential diagnosis in young dogs with a myelopathy and an intramedullary spinal cord lesion on magnetic resonance imaging examination.  相似文献

The chronic impact of ring nematode (Mesocriconema xenoplax) feeding on grapevine (Vitis vinifera) was studied under controlled conditions. ‘Pinot noir’ grapevines were exposed to ring nematode or kept nematode-free for three growing seasons and vines were either grown in full sunlight, 15% of full sun, or partially defoliated to manipulate vine carbohydrate status. Whole plants were destructively sampled to assess the impact of ring nematode on whole plant biomass, carbohydrate, and mineral nutrient accumulation. Vine shoot growth and total biomass was unaffected by ring nematode in the first growing season, although reserves of nonstructural carbohydrates (NSC), P, K, and Ca in the roots and wood were reduced in all canopy management treatments. Vine shoot growth and total biomass were reduced by ring nematode in Year 2, and greater declines in reserve NSC and most mineral nutrients had occurred. Reserves of NSC were affected more than biomass or nutrients during the second year. During the third year of exposure to ring nematode, vines in the 15% sun treatment were dying (prompting an earlier destructive harvest), even though these vines had similar biomass and NSC reserves as the partially defoliated vines at the end of the second year. The demise of the 15% sun vines was associated with higher ring nematodes per unit of root mass, as compared to either full sun or defoliated vines. Therefore, predicting plant response to this nematode requires an understanding of nematode density per quantity of roots, not nematodes per unit of soil which is how plant parasitic nematodes are currently enumerated.  相似文献

Abstract

AIM: To determine whether viable Mycobacterium avium subsp. paratuberculosis (Map) is present in skeletal muscle and blood in ewes with and without Johne's disease confirmed histologically.

METHODS: A total of 51 mixed-aged ewes in poor body condition from a farm with a history of clinical Johne's disease were culled and examined at necropsy. BACTEC radiometric culture was performed on samples of skeletal muscle from the biceps femoris, mononuclear cells in peripheral blood (hereafter referred to as blood), and ileum. Histological sections and Ziehl-Neelsen (ZN)-stained impression smears of terminal ileum and mesenteric lymph nodes were examined. Ewes were defined as having confirmed Johne's disease if there was histopathological evidence typical of the disease within the ileum and adjacent lymph nodes.

RESULTS: Eighteen of 21 (86%) ewes with confirmed clinical Johne's disease were culture-positive for Map from sites peripheral to the alimentary tract, comprising 15 from skeletal muscle and 13 from blood. Five of 30 (17%) ewes that did not have Johne's disease were culture-positive, with four from skeletal muscle and one from blood. The likelihood that ewes with confirmed Johne's disease had systemic Map infection compared with ewes without was determined as OR=30 (95% CI=6.3–142.0; p<0.001).

CONCLUSION: The prevalence of Map infection of skeletal muscle and blood in ewes with confirmed Johne's disease was 71% and 62% respectively, and in unaffected ewes was 13% for muscle and 3% for blood.

CLINICAL RELEVANCE: Skeletal muscle and blood are potential sources of exposure of humans to Map, and the risk appears higher from sheep with Johne's disease.  相似文献

Most perennial ryegrass (Lolium perenne) plants in pastures in New Zealand contain a naturally occurring fungal endophyte, Neotyphodium lolii. Endophyte-infected perennial ryegrass produces alkaloids that vary in concentration during the year. Some alkaloids enhance the persistence and productivity of ryegrass pastures by protecting them against insect attack. However, when other alkaloids are consumed they can reduce animal performance and lead to health problems. The alkaloid lolitrem B, for example, causes the neuromuscular disorder ryegrass staggers (Fletcher et al 1999 Fletcher, LR, Sutherland, BL and Fletcher, CG. 1999. “The impact of endophyte on the health and productivity of sheep grazing ryegrass-based pastures”. Clinical symptoms of ryegrass staggers in animals range from slight muscular tremors through to staggering and complete collapse. Severely affected animals create management problems and are prone to accidental death. Outbreaks occur sporadically, particularly in summer and autumn and affect sheep, cattle, deer, horses, llamas (Lama glama) and alpaca (Lama pacos). Lolitrem B appears to be a stable compound, which tends to be concentrated in the leaf sheath at the base of the ryegrass plant and in the seed-heads (di Menna et al 1992 di Menna, ME, Mortimer, PH, Prestidge, RA, Hawkes, AD and Sprosen, JM. 1992.
