A method using Raman spectroscopy was recently developed for the determination of the degree of substitution of succinate in waxy maize starch. In this paper it is demonstrated that the method can be generalized to a wide range of starches of different amylose contents and botanical origins. Raman calibration sets were used to form regression equations for five types of succinylated starches, that is, waxy, regular, and two high-amylose maize samples (47 and 66% amylose, respectively) and wheat. The derived calibration curves can be used to find the degree of substitution in samples with unknown levels of succinylation. The Raman calibration lines had linear correlation coefficients of 0.995 or better and enable the fast and nondestructive determination of the degree of substitution of succinate for different types of starches with minimal sample preparation. Also discussed is the potential utility of Raman spectroscopy to simultaneously determine the degree of substitution of succinate and amylose content, using previously determined calibration curves developed for the amylose content of maize starches.  相似文献

Spinosad is an insect control agent that is derived from a naturally occurring soil bacterium and is effective on several classes of insects, especially Lepidopteran larvae. Spinosad is registered in many countries for use on a variety of crops, including cotton, corn, soybeans, fruits, and vegetables. Residue methods utilizing a magnetic particle-based immunoassay (IA) test kit have been developed and validated for determining spinosad in environmental and food matrices. These methods involve an extraction of the residues from the matrices with appropriate solvents. For some matrices, the sample extracts can be diluted and measured directly by IA without any cleanup. For other matrices, sample extracts are purified using liquid-liquid partitioning and/or solid phase extraction prior to measurement by IA. The methods determine the total residue of spinosad, which includes the active ingredients (spinosyns A and D) and several minor metabolites, including spinosyn B, spinosyn K, and N-demethylspinosyn D. The methods have validated limits of quantitation of 0.0001 microgram/mL in water, 0.05 microgram/g in sediment, and 0.010 microgram/g in crops, crop processed commodities, and animal tissues. This paper briefly summarizes the residue methodology and method validation data for spinosad in 34 food, feed, and environmental matrices.  相似文献

Airborne pollutants in the form of SO2 and heavy metals have been dispersed over the boreal forest in the Thompson area of northern Manitoba since 1961. Metal deposition has been found in soils and plant material to a distance of 35 km from the Inco nickel smelter. Studies were initiated in 1979 to determine effects of Ni and Cu on coniferous regeneration of jack pine (Pinus banksiana Lamb.) and black spruce (Picea mariana (Mill) BSP.).Studies included metal accumulation in cones, effects on seed quality, seed germination and seedling response in metal contaminated soils. Nickel and Cu accumulation in cones of both species declined rapidly with distance from the smelter. Seed quality and germination potential was not related to metals in cones or air pollutant influence in general. Nickel and Cu accumulation in surface organic soils was inversely related to distance from the smelter. A significant inverse correlation was determined between seedling growth and Cu/Ni concentrations in surface organic soils. Metal accumulation in soils within 5 km of the nickel smelter inhibited root growth of conifer seedlings.  相似文献

A dry column method for isolating N-nitrosopyrrolidine (NPYR) from fried, cure-pumped bacon and detection by gas chromatography-thermal energy analyzer (TEA) was studied collaboratively. Testing the results obtained from 11 collaborators for homogeneous variances among samples resulted in splitting the nonzero samples into 2 groups of sample levels, each with similar variances. Outlying results were identified by AOAC-recommended procedures, and laboratories having outliers within a group were excluded. Results from the 9 collaborators remaining in the low group yielded coefficients of variation (CV) of 6.00% and 7.47% for repeatability and reproducibility, respectively, and the 8 collaborators remaining in the high group yielded CV values of 5.64% and 13.72%, respectively. An 85.2% overall average recovery of the N-nitrosoazetidine internal standard was obtained with an average laboratory CV of 10.5%. The method has been adopted official first action as an alternative to the mineral oil distillation-TEA screening procedure.  相似文献

ABSTRACT

This study sought to identify whether piggery effluent-derived nitrogen sources can be formulated with urea and nitrification inhibitors to better synchronize nitrogen (N) supply with crop demand than conventional urea fertilizer alone. A 288 pot pasture growth and leaching growth accelerator trial (5 pasture cuts) was completed with a factorial treatment structure of three N sources (2.63 g N [kg soil]?1 applied as 100% urea-N, 8% struvite-N + 92% urea-N, and 8% piggery pond sludge-N + 92% urea-N), five rates of three nitrification inhibitors (including 3,4-Dimethylpyrazole phosphate, DMPP; limonene+ethanol; and dicyandiamide, DCD), and matrix encapsulated forms of these inhibitors. Applying a combination of piggery sludge with urea increased N uptake during the first 4 weeks of plant growth (by 65%), though total N uptake throughout the trial (22 weeks) did not differ across the N-sources. The microbial community of the soil to which the sludge was added was significantly different from the un-amended soil at the conclusion of the trial. All inhibitor formulations significantly decreased leaching losses of mineral-N relative to the control (by 14 to 61%). The use of DMPP decreased initial nutrient uptake, deferring uptake until later in the experiment. Inhibitor addition resulted in microbial community effects that persisted throughout the trial. The study demonstrated that a piggery-derived N-source and a nitrification inhibitor can be used to manipulate plant N uptake to occur later or earlier in a growing period with equal cumulative uptake, achieving an 11% increase in residual N store, and decreased N leaching losses.  相似文献

Acid rain results in losses of exchangeable base cations from soils, but the mechanism of base cation displacement from the forest floor is not clear, and has been hypothesized to involve mobilization of aluminum from the mineral soil. We attempted to test the hypothesis that losses of calcium from the forest floor were balanced by increases in Al in NewHampshire northern hardwoods. We measured exchangeable (six stands) and acid extractable (13 stands) Ca and Al in horizons of the forest floor over an interval of 15 years. Our sampling scheme was quite intensive, involving 50 or 60 blocks per stand, composited in groups of 10 for chemical analysis. Even at this level of effort, few stands exhibited changes large enough to be significant. Because of high spatial variability, differences would have had to be greater than about 50% to be statistically detectable. Differences in Ca and Al concentrations between Oi, Oe, Oa, and A horizons, however, were readily detected. Acid-extractable Al increased with depth, while Ca concentrations decreased; Ca-to-Al ratios decreased from 8.3 (charge basis) in the Oi to 0.2 in the A horizon. Therefore, a small change in sampling depth, or the inclusion of more or less A horizon material in the forest floor, could cause large differences in measured Ca and Al concentrations. To detect small changes in exchangeable cations over time would require sampling very intensively with careful control for comparability of horizons.  相似文献

Canopy and hydraulic conductance in young, mature and old Douglas-fir trees   总被引:1,自引:0,他引:1  
We tested for reductions in water transport with increasing tree size, a key component in determining whether gas exchange and growth are hydraulically limited in tall trees. During the summers of 1998 and 1999, we measured water transport with Granier-type, constant-heat sap flow probes, vapor pressure deficit, and leaf and soil water potentials in overstory Pseudotsuga menziesii (Mirb.) Franco trees in three stands differing in size and age (15, 32 and 60 m in height and about 20, 40 and 450 years in age, respectively) in a P. menziesii-dominated forest in the Pacific Northwest, USA. A total of 24 trees were equipped with sap flow sensors--six 60-m trees, nine 32-m trees and nine 15-m trees. Based on the sap flow measurements and leaf area information estimated from leaf area-sapwood area relationships, we estimated crown-averaged stomatal conductance (GS) and leaf-specific hydraulic conductance (KL). We tested the hypothesis that GS and KL vary inversely with tree height (15 > 32 > 60 m). Analysis of variance of GS ranked as 15 = 60 > 32 m during the early summer and 15 > 60 > 32 m during late season drought. Over the growing season, mean daily GS (+/- SE) was 29.2 +/- 4.4, 24.0 +/- 6.8 and 17.7 +/- 7.2 mmol m-2 s-1 for the 15-, 60- and 32-m trees, respectively. The value of K(L) differed among tree heights only during late season drought and ranked 15 > 32 = 60 m. A hydraulic mass balance suggests that greater sapwood conductivity in 60-m trees compared with 32- and 15-m trees is a likely cause for the departure of the above rankings from those predicted by height and leaf-to-sapwood area ratio.  相似文献

Magellan radar images of 15 percent of the planet show 135 craters of probable impact origin. Craters more than 15 km across tend to contain central peaks, multiple central peaks, and peak rings. Many craters smaller than 15 km exhibit multiple floors or appear in clusters; these phenomena are attributed to atmospheric breakup of incoming meteoroids. Additionally, the atmosphere appears to have prevented the formation of primary impact craters smaller than about 3 km and produced a deficiency in the number of craters smaller than about 25 km across. Ejecta is found at greater distances than that predicted by simple ballistic emplacement, and the distal ends of some ejecta deposits are lobate. These characteristics may represent surface flows of material initially entrained in the atmosphere. Many craters are surrounded by zones of low radar albedo whose origin may have been deformation of the surface by the shock or pressure wave associated with the incoming meteoroid. Craters are absent from several large areas such as a 5 million square kilometer region around Sappho Patera, where the most likely explanation for the dearth of craters is volcanic resurfacing. There is apparently a spectrum of surface ages on Venus ranging approximately from 0 to 800 million years, and therefore Venus must be a geologically active planet.  相似文献

X-ray crystal structures of three species related to the oxidative half of the reaction of the copper-containing quinoprotein amine oxidase from Escherichia coli have been determined. Crystals were freeze-trapped either anaerobically or aerobically after exposure to substrate, and structures were determined to resolutions between 2.1 and 2.4 angstroms. The oxidation state of the quinone cofactor was investigated by single-crystal spectrophotometry. The structures reveal the site of bound dioxygen and the proton transfer pathways involved in oxygen reduction. The quinone cofactor is regenerated from the iminoquinone intermediate by hydrolysis involving Asp383, the catalytic base in the reductive half-reaction. Product aldehyde inhibits the hydrolysis, making release of product the rate-determining step of the reaction in the crystal.  相似文献

Three-dimensional structure of an antigen-antibody complex at 2.8 A resolution   总被引:78,自引:0,他引:78  
The 2.8 A resolution three-dimensional structure of a complex between an antigen (lysozyme) and the Fab fragment from a monoclonal antibody against lysozyme has been determined and refined by x-ray crystallographic techniques. No conformational changes can be observed in the tertiary structure of lysozyme compared with that determined in native crystalline forms. The quaternary structure of Fab is that of an extended conformation. The antibody combining site is a rather flat surface with protuberances and depressions formed by its amino acid side chains. The antigen-antibody interface is tightly packed, with 16 lysozyme and 17 antibody residues making close contacts. The antigen contacting residues belong to two stretches of the lysozyme polypeptide chain: residues 18 to 27 and 116 to 129. All the complementarity-determining regions and two residues outside hypervariable positions of the antibody make contact with the antigen. Most of these contacts (10 residues out of 17) are made by the heavy chain, and in particular by its third complementarity-determining region. Antigen variability and antibody specificity and affinity are discussed on the basis of the determined structure.  相似文献
