It is generally accepted that during the early stages of residue decomposition, easily available compounds are decomposed, leading to a relative increase in more recalcitrant compounds in the later stages of decomposition and that these changes in substrate availability are associated with changes in microbial community composition. However most studies on residue decomposition are conducted over several weeks or months; little is known about the changes in microbial community composition in the first weeks of decomposition. To address this knowledge gap, we incubated wheat residues inoculated with a microbial suspension in mesh bags buried in sand for 30 days, with sampling on days 0, 2, 4, 6, 8, 10, 15, 20, 25 and 30. Of the C added with the residues, 10, 18 and 25% had been respired on days 10, 20 and 30, respectively. The sum of PLFAs (phospholipid fatty acids), as an indicator of microbial biomass, increased strongly in the first 4 days and then decreased. The concentration of bacterial fatty acids was maximal on days 2 and 4, whereas the concentration of fungal fatty acids peaked on day 15. Microbial community composition (based on PLFA patterns) changed rapidly, with significant changes in the first 8 days and from day 8 to day 20. There were no significant changes in microbial community composition after day 20. The concentration of water-soluble C decreased strongly in the first 8 days, suggesting that the rapid changes in microbial community in this period are related to the changes in water-soluble C. Residue C chemistry, assessed by 13C NMR spectroscopy, changed little during the incubation period. This study showed that microbial community composition in decomposing residues changes rapidly in the first 1-2 weeks, which is, at least partly, the result of competition for the easily available compounds in the water-soluble fraction. After depletion of the water-soluble compounds, the microbial community composition changes more slowly.  相似文献

Uphill shifts of alpine species have been associated with climate warming; however, land-use change can act simultaneously. We surveyed two related alpine burnet moth species in the Pyrenees to determine whether there was an uphill shift in the range of these two species and, if so, whether this was driven by climate and/or land-use changes. In 2008/09 we revisited 28 sites at the lower altitudinal range limits at which the species had been recorded between 1958 and 1986 (data were obtained from museum collections, distribution atlas, researchers’ field notes). We also recorded the recent upper range limits. The arctic-alpine Zygaena exulans showed an uphill shift (148 m ± SD 87 m/decade) at all sites; at four sites the species has even become extinct. Changes in the lower altitudinal range limits were attributed to climate change. For the endemic species Zygaena anthyllidis we only ascertained an uphill shift at one third of the sites investigated (60 m ± SD 74 m/decade); these shifts were due to changes in grazing intensities. As both species colonize very high summits, further uphill shifts of the upper limits are no longer possible. The studied con-generic species are affected by different global change drivers. Our data indicate the considerable role of land-use in conservation and suggest that it can even exceed the impact of climate change. Ongoing climate change is likely to jeopardize the long-term survival of Z. exulans in the Pyrenees; Z. anthyllidis, however, may be preserved by appropriate management in the subalpine belt.  相似文献

Mulches can improve soil properties, but little is known about nutrient availability in mulched soil that contains plant residues and the effect of mulching with manures. The aim of this study was to determine the effects of mulching with high or low C/N organic materials, in which low C/N materials differed in decomposability, and the presence of wheat straw in the soil on plant growth and N uptake, soil N availability and microbial biomass N within about four months after mulching. Three organic materials were used: mature wheat straw (W, C/N 80), young faba bean shoots (FB, C/N 7), and sheep manure (SM, C/N 8). There were eight treatments differing in amendment methods (mulching or mixing with W or both) and mulching materials (W, FB or SM). Treatments that were only mulched with W, FB or SM are referred to as m‐treatments. In m/s‐treatments, after W was mixed into the soil, W, FB or SM were placed on the soil surface as mulch. Two other treatments included an unamended control and soil mixed with W. Wheat was planted 0, 35 or 70 days after mulching (referred to as 0, 35, and 70 DAM) and grown for 35 days. Faba bean mulch increased shoot dry weight, shoot N uptake and available N compared to wheat or sheep manure mulch, particularly in the m‐treatments. Shoot dry weight was higher in m‐treatments than corresponding m/s‐treatments with the same mulch type. Shoot N uptake was higher in 70 DAM than in 0 DAM in all treatments and 0.3 to three‐fold higher in m‐treatments than the corresponding m/s‐treatments. Microbial biomass N was higher in 0 DAM than in 35 and 70 DAM in most treatments and up to two‐fold higher in m/s‐treatments than the corresponding m‐treatments. Available N in m/s‐treatments was two to six‐fold higher than m‐treatments in 0 DAM, but differed little in older mulch ages of W and SM. It can be concluded that compared to soil with only mulch, mixing of wheat straw into soil reduced plant growth and N uptake, particularly in the early stages of mulching (0 and 35 DAM). However, the presence of wheat in mulched soil may provide a longer lasting source of N for plants and reduce the risk of N leaching from rapidly decomposing low C/N mulch due to greater microbial biomass N uptake than only soil with mulch.  相似文献

225 wheat varieties from Kazakhstan, Mongolia and Russia were analysed for their resistance to powdery mildew and leaf rust diseases. A set of 11 different Blumeria graminis f. sp. tritici isolates was used to test for powdery mildew resistance and a set of 10 different Puccinia triticina isolates for leaf rust. 115 cultivars/lines were susceptible to powdery mildew and 32 cultivars/lines showed an intermediate resistance response. 21 cultivars/lines revealed the response pattern of individual resistance genes Pm1, Pm2, Pm3, pm5, Pm6, Pm8, Pm9, Pm17 and Pm22, respectively, therefrom three line showed a combination of two resistance genes and two varieties a combination of three genes. 50 cultivars/lines showed resistance to some specific isolates but an assignment to known resistance genes or gene combinations was not possible, whereas seven lines were completely resistant to all used isolates. The leaf rust test showed that 83 cultivars/lines were susceptible and 11 lines revealed intermediate resistance response. 62 cultivars/lines could be assumed to possess major resistance genes Lr1, Lr3, Lr10, Lr23 and Lr26, respectively, therefrom seven cultivars possessed a combination of two resistance genes. Lr3 was the most widespread resistance gene, occurring in 42 cultivars/lines. 13 lines were completely resistant to all used isolates, however, the response patterns of 56 cultivars/lines did not match to any known gene or gene combination. In 13 varieties the T1BL·1RS wheat-rye translocation could be identified, in five cultivars resistance gene Pm8 was suppressed.  相似文献

The effect of drying and rewetting (DRW) on C mineralization has been studied extensively but mostly in absence of freshly added residues. But in agricultural soils large amounts of residues can be present after harvest; therefore, the impact of DRW in soil after residue addition is of interest. Further, sandy soils may be ameliorated by adding clay‐rich subsoil which could change the response of microbes to DRW. The aim of this study was to investigate the effect of DRW on microbial activity and growth in soils that were modified by mixing clay subsoil into sandy top soil and wheat residues were added. We conducted an incubation experiment by mixing finely ground wheat residue (20 g kg–1) into top loamy sand soil with clay‐rich subsoil at 0, 5, 10, 20, 30, and 40% (w/w). At each clay addition rate, two moisture treatments were imposed: constantly moist control (CM) at 75% WHC or dry and rewet. Soil respiration was measured continuously, and microbial biomass C (MBC) was determined on day 5 (before drying), when the soil was dried, after 5 d dry, and 5 d after rewetting. In the constantly moist treatment, increasing addition rate of clay subsoil decreased cumulative respiration per g soil, but had no effect on cumulative respiration per g total organic C (TOC), indicating that the lower respiration with clay subsoil was due to the low TOC content of the sand‐clay mixes. Clay subsoil addition did not affect the MBC concentration per g TOC but reduced the concentration of K2SO4 extractable C per g TOC. In the DRW treatment, cumulative respiration per g TOC during the dry phase increased with increasing clay subsoil addition rate. Rewetting of dry soil caused a flush of respiration in all soils but cumulative respiration at the end of the experiment remained lower than in the constantly moist soils. Respiration rates after rewetting were higher than at the corresponding days in constantly moist soils only at clay subsoil addition rates of 20 to 40%. We conclude that in presence of residues, addition of clay subsoil to a sandy top soil improves microbial activity during the dry phase and upon rewetting but has little effect on microbial biomass.  相似文献

Previous attempts have shown the potential of T cells in immunotherapy of cancer. Here, we report on the clinical activity of a bispecific antibody construct called blinatumomab, which has the potential to engage all cytotoxic T cells in patients for lysis of cancer cells. Doses as low as 0.005 milligrams per square meter per day in non-Hodgkin's lymphoma patients led to an elimination of target cells in blood. Partial and complete tumor regressions were first observed at a dose level of 0.015 milligrams, and all seven patients treated at a dose level of 0.06 milligrams experienced a tumor regression. Blinatumomab also led to clearance of tumor cells from bone marrow and liver. T cell-engaging antibodies appear to have therapeutic potential for the treatment of malignant diseases.  相似文献

A series of twenty-three linear and branched chain mono acetylene lipids were isolated from the Caribbean Sea sponge Cribrochalina vasculum. Seventeen of the compounds, 1–17, are new, while six, 18–23, were previously characterized from the same sponge. Some of the new acetylene-3-hydroxy alkanes 1, 6, 7, 8, 10 were tested for selective cytotoxicity in non-small cell lung carcinoma (NSCLC) cells over WI-38 normal diploid lung fibroblasts. Compound 7, presented clear tumor selective activity while, 1 and 8, showed selectivity at lower doses and 6 and 10, were not active towards NSCLC cells at all. The earlier reported selective cytotoxicity of some acetylene-3-hydroxy alkanes (scal-18 and 23), in NSCLC cells and/or other tumor cell types were also confirmed for 19, 20 and 22. To further study the structure activity relationships (SAR) of this group of compounds, we synthesized several derivatives of acetylene-3-hydroxy alkanes, rac-18, scal-S-18, R-18, rac-27, rac-32, R-32, S-32, rac-33, rac-41, rac-42, rac-43, rac-45, rac-48 and rac-49, along with other 3-substituted derivatives, rac-35, rac-36, rac-37, rac-38, rac-39 and rac-40, and assessed their cytotoxic activity against NSCLC cells and diploid fibroblasts. SAR studies revealed that the alcohol moiety at position 3 and its absolute R configuration both were essential for the tumor cell line selective activity while for its cytotoxic magnitude the alkyl chain length and branching were of less significance.  相似文献
