With respect to the assessment of residue situation and as a part of preclinical trials to determine the biological activities of potential gonadotropin releasing hormone (GnRH) residues in porcine organisms the GnRH agonist Gonadorelin[6-D-Phe] (D-Phe(6)-LHRH) was administered either enterally or intramuscularly (i.m.) to female Goettinger miniature pigs in order to evaluate the GnRH-induced luteinizing hormone (LH) surge. Gilts received an (i) enteral application of 10 mg D-Phe(6)-LHRH via a probang (enteral group, n=7), (ii) i.m. injection of 0.1 mg D-Phe(6)-LHRH (parenteral group, n=5), or (iii) saline injection (control group, n=4). The GnRH and saline applications were repeated every second day with up to seven repetitions. Blood samples were collected via previously fitted jugular catheters immediately before injections, over an 8 h period in 1 h intervals beginning 2 h after injections, and at 24, 26, 28 and 30 h after applications. Enteral application of D-Phe(6)-LHRH induced an LH surge in 23 of 30 treatments. All gilts in the parenteral group exhibited LH release after each D-Phe(6)-LHRH application (P<0.05), whereas no LH surges were observed after saline injection in the control group. A significant (P<0.05) LH rise to mean maximum LH concentrations of 3.25 +/- 0.43 and 3.05 +/- 0.26 ng/ml occurred in both the enteral and parenteral groups, but there was no difference in the time interval after GnRH (2.6 +/- 0.3 vs. 2.3 +/- 0.3 h) and the mean duration of LH peak (6.5 +/- 0.4 and 6.8 +/- 0.3 h) between the treatment groups. In conclusion, (i) enteral application of 10 mg D-Phe(6)-LHRH induced LH release in a physiological range from the pituitary of female minipigs, and (ii) neither an accumulative effect nor a cumulative LH response were found after repeated GnRH application. Furthermore, (iii) in regard to consumer protection and gonadotropin secretion, D-Phe(6)-LHRH residues can be excluded from having long-term effects.  相似文献

Earthworm activity may have an effect on nitrous oxide (N2O) emissions from crop residue. However, the importance of this effect and its main controlling variables are largely unknown. The main objective of this study was to determine under which conditions and to what extent earthworm activity impacts N2O emissions from grass residue. For this purpose we initiated a 90-day (experiment I) and a 50-day (experiment II) laboratory mesocosm experiment using a Typic Fluvaquent pasture soil with silt loam texture. In all treatments, residue was applied, and emissions of N2O and carbon dioxide (CO2) were measured. In experiment I the residue was applied on top of the soil surface and we tested (a) the effects of the anecic earthworm species Aporrectodea longa (Ude) vs. the epigeic species Lumbricus rubellus (Hoffmeister) and (b) interactions between earthworm activity and bulk density (1.06 vs. 1.61 g cm−3). In experiment II we tested the effect of L. rubellus after residue was artificially incorporated in the soil. In experiment I, N2O emissions in the presence of earthworms significantly increased from 55.7 to 789.1 μg N2O-N kg−1 soil (L. rubellus; p<0.001) or to 227.2 μg N2O-N kg−1 soil (A. longa; p<0.05). This effect was not dependent on bulk density. However, if the residue was incorporated into the soil (experiment II) the earthworm effect disappeared and emissions were higher (1064.2 μg N2O-N kg−1 soil). At the end of the experiment and after removal of earthworms, a drying/wetting and freezing/thawing cycle resulted in significantly higher emissions of N2O and CO2 from soil with prior presence of L. rubellus. Soil with prior presence of L. rubellus also had higher potential denitrification. We conclude that the main effect of earthworm activity on N2O emissions is through mixing residue into the soil, switching residue decomposition from an aerobic and low denitrification pathway to one with significant denitrification and N2O production. Furthermore, A. longa activity resulted in more stable soil organic matter than L. rubellus.  相似文献

Laboratory studies were conducted to characterize the 1-octanol/water partition coefficients of pharmaceutically active substances carbamazepine, clofibric acid, diclofenac, ibuprofen, and propyphenazone. Partition coefficients determined by shake flask experiments (OECD guideline 107) varied between log K OW 1.51 for carbamazepine, 2.88 for clofibric acid, 1.90 for diclofenac, 2.48 for ibuprofen, and 2.02 for propyphenazone. Comparison of these values with the literature values revealed rather significant differences for most of the compounds. The partitioning coefficients of the acidic compounds diclofenac and ibuprofen agreed much better with sorption and mobility data from previously conducted experiments, whereas K OW values for carbamazepine were lower and for clofibric acid higher than expected from experiments. Only K OW values for propyphenazone were in the same range as reported in the literature and expected from column experiments.  相似文献

Arsenic (As) and cadmium (Cd) in soils can affect soil microbial function and community composition and, therefore, may have effects on soil ecosystem functioning. The aim of our study was to assess the effects of long-term As and Cd contamination on soil microbial community composition and soil enzyme activities. We analyzed soils that have been contaminated 25 years ago and at present still show enhanced levels of either As, 18 and 39 mg kg−1, or Cd, 34 and 134 mg kg−1. Soil without heavy metal addition served as control. Polymerase chain reaction (PCR) followed by denaturing gradient gel electrophoresis (DGGE) showed that bacterial community composition in As and Cd contaminated soils differed from that in the control soil. The same was true for the microbial community composition assessed by analysis of respiratory quinones. Soil fungi and Proteobacteria appeared to be tolerant towards As and Cd, while other groups of bacteria were reduced. The decline in alkaline phosphatase, arylsulphatase, protease and urease activities in the As- and Cd-contaminated soils was correlated with a decrease of respiratory quinones occuring in Actinobacteria and Firmicutes. Xylanase activity was unaffected or elevated in the contaminated soils which was correlated with a higher abundance of fungal quinones, and quinones found in Proteobacteria.  相似文献

Earthworm activity has been reported to lead to increased production of the greenhouse gas nitrous oxide (N2O). This is due to emissions from worms themselves, their casts and drilosphere, as well as to general changes in soil structure. However, it remains to be determined how important this effect is on N2O fluxes from agricultural systems under realistic conditions in terms of earthworm density, soil moisture, tillage activity and residue loads. We quantified the effect of earthworm presence on N2O emissions from a pasture after simulated ploughing of the sod (‘grassland renovation’) for different soil moisture contents during a 62-day mesocosm study. Sod (with associated soil) and topsoil were separately collected from a loamy Typic Fluvaquent. Treatments included low (L), medium (M) and high (H) moisture content, in combination with: only soil (S); soil+incorporated sod (SG); soil+incorporated sod+the anecic earthworm Aporrectodea longa (SGE). Nitrous oxide and carbon dioxide (CO2) fluxes were measured for 62 d. At the end of the incubation period, we determined N2O production under water-saturated conditions, potential denitrification and potential mineralization of the soil after removing the earthworms. Cumulative N2O and CO2 fluxes over 62 d from incorporated sod were highest for treatment HSGE (973 μg N2O-N and 302 mg CO2-C kg−1 soil) and lowest for LSG (64 μg N2O-N and 188 mg CO2-C kg−1 soil). Both cumulative fluxes were significantly different for soil moisture (p<0.001), but not for earthworm presence. However, we observed highly significant earthworm effects on N2O fluxes that reversed over time for the H treatments. During the first phase (day 3-day 12), earthworm presence increased N2O emissions with approximately 30%. After a transitional phase, earthworm presence resulted in consistently lower (approximately 50%) emissions from day 44 onwards. Emissions from earthworms themselves were negligible compared to overall soil fluxes. After 62 d, original soil moisture significantly affected potential denitrification, with highest fluxes from the L treatments, and no significant earthworm effect. We conclude that after grassland ploughing, anecic earthworm presence may ultimately lead to lower N2O emissions after an initial phase of elevated emissions. However, the earthworm effect was both determined and exceeded by soil moisture conditions. The observed effects of earthworm activity on N2O emissions were due to the effect of earthworms on soil structure rather than to emissions from the worms themselves.  相似文献

In a pot experiment, the P‐efficient wheat (Triticum aestivum L.) cultivar Goldmark was grown in ten soils from South Australia covering a wide range of pH (four acidic, two neutral, and four alkaline soils) with low to moderate P availability. Phosphorus (100 mg P kg–1) was supplied as FePO4 to acidic soils, CaHPO4 to alkaline, and 1:1 mixture of FePO4 and CaHPO4 to neutral soils. Phosphorus uptake was correlated with P availability measured by anion‐exchange resin and microbial biomass P in the rhizosphere. Growth and P uptake were best in the neutral soils, lower in the acidic, and poorest in the alkaline soils. The good growth in the neutral soils could be explained by a combination of extensive soil exploitation by the roots and high phosphatase activity in the rhizosphere, indicating microbial facilitation of organic‐P mineralization. The plant effect (soil exploitation by roots) appeared to dominate in the acidic soils. Alkaline phosphatase and diesterase activities in acidic soils were lower than in neutral soils, but strongly increased in the rhizosphere compared with the bulk soil, suggesting that microorganisms contribute to P uptake in these acidic soils. Shoot and root growth and P uptake per unit root length were lowest in the alkaline soils. Despite high alkaline phosphatase and diesterase activities in the alkaline soils, microbial biomass P was low, suggesting that the enzymes could not mineralize sufficient organic P to meet the demands of plants and microorganisms. Microbial‐community composition, assessed by fatty acid methylester (FAME) analysis, was strongly dependent on soil pH, whereas other soil properties (organic‐C or CaCO3 content) were less important or not important at all (soil texture).  相似文献

The capsaicinoid content in fruits of Capsicum annuum decreased within several days to a level of only 10% of the starting value when cells were disrupted by homogenization. This decrease was not observed in fruits that were carefully cut into halves. The analysis of one half made it possible to determine the reference content at time zero for the second half. A much lower decrease was observed when minced fruits were stored under nitrogen, whereas storage under oxygen resulted in considerable losses of capsaicinoids, indicating oxidative processes as a cause for the decrease of capsaicinoid content.  相似文献
