Commercial areas containing Eucalyptus plantations have expanded in recent years due to increased demands for pulp, paper and bioenergy. One of the threats that can reduce Eucalyptus production is the eucalyptus rust disease caused by Austropuccinia psidii, a biotrophic fungus that affects a broad range of Myrtaceae. An accurate diagnosis tool for the early detection of rust disease could be useful in breeding programs for selection of resistant plants against rust, in phytosanitary purposes or in rust epidemics studies. The aim of the present work was to develop a SYBR Green-based quantitative real-time PCR (qPCR) assay for the early detection and quantification of A. psidii in Eucalyptus grandis leaves. Three sets of primers based on the A. psidii ribosomal DNA intergenic space region (IGS), beta-tubulin and elongation factor genes were designed and evaluated. The assays using the IGS primer set resulted in the highest detection efficiency, detecting a lower limit of 0.5 pg of A. psidii DNA. Under artificial inoculation in plants, A. psidii was detected immediately after pathogen inoculation until 240 h post-inoculation using qPCR. In field validation of the method, A. psidii was detected using qPCR in naturally infected leaves with or without rust symptoms. This easy and fast method can be used for an efficient detection of A. psidii in E. grandis leaves. The implications of this tool for rust studies are discussed below.  相似文献

Atmospheric pollution is an important concern in urban environments.The impact of urban pollution on the physiological,biochemical and anatomical properties of leaves of oriental plane(Platanus orientalis L.)was studied.The leaves were collected from an urban site(Tehran,Iran)and a non-urban forest park(Chitgar Forest Park).Anatomical(stomata,parenchyma,and cuticle)and physiological(chlorophyll content,enzyme activities)properties were analyzed.The concentrations of Cd(cadmium),Pb(lead),Ni(nickel),and Cr(chromium)in leaves were significantly higher and Zn(zinc)lower at the urban site relative to the forest park.Chlorophyll a,total chlorophyll,and carotenoid content in leaves of the urban site were significantly less than those of the forest park,but there was no significant difference in chlorophyll b.In addition,the activities of superoxide dismutase,catalase,ascorbate peroxidase,and guaiacol peroxidase in urban leaves were significantly higher than in the leaves of the forest park.In the urban area,leaves experienced a significant decrease in leaf surface area,stomata density,stomata pore area,epidermis,and spongy mesophyll thickness,but a significant increase in cuticle and palisade thicknesses relative to forest park leaves.The increase in enzyme activities may indicate that the trees are attempting to cope with increased reactive oxygen species(ROS)because of pollution-induced stress.Our study suggests that oriental plane trees alter their physiological and anatomical properties when living in a polluted urban environment.  相似文献

This paper reports an experimental study on compressive, flexural and fatigue behavior of polyurethane cast elastomers (PCE) reinforced with warp-knitted spacer fabrics (WKSF). It aims to investigate new applications for these fabrics as the reinforcements for elastomeric parts such as shoe soles, rubber floor coverings, vibration dampening and shock absorbing pads, etc. A series of polyester WKSF with different thickness, structure of outer layer fabric and spacer yarns density was prepared and converted to PCE reinforced WKSF using the hand molding method. All the samples, including the neat PCE, were subjected to static compression, flat and spherical compression, three-point bending and flexural fatigue tests. The results showed that reinforcing PCE with WKSF, considerably enhances its spherical compressive strength (concentrated loading), flexural strength and fatigue resistance. However, it deteriorates flat compressive strength (distributed loading) and recovery behavior after static compression loading. The effect of fiber weight fraction, thickness, structure of outer layer fabric and spacer yarns density on the mentioned properties of the composites was discussed in the paper.  相似文献

Improved understanding of the influence of climate on agricultural production is needed to cope with expected changes in temperature and precipitation, and an increasing number of undernourished people in food insecure regions. Many studies have shown the importance of seasonal climatic means in explaining crop yields. However, climate variability is expected to increase in some regions and have significant consequences on food production beyond the impacts of changes in climatic means. Here, we examined the relationship between seasonal climate and crop yields in Tanzania, focusing on maize, sorghum and rice. The impacts of both seasonal means and variability on yields were measured at the subnational scale using various statistical methods and climate data. The results indicate that both intra- and interseasonal changes in temperature and precipitation influence cereal yields in Tanzania. Seasonal temperature increases have the most important impact on yields. This study shows that in Tanzania, by 2050, projected seasonal temperature increases by 2 °C reduce average maize, sorghum, and rice yields by 13%, 8.8%, and 7.6% respectively. Potential changes in seasonal total precipitation as well as intra-seasonal temperature and precipitation variability may also impact crop yields by 2050, albeit to a lesser extent. A 20% increase in intra-seasonal precipitation variability reduces agricultural yields by 4.2%, 7.2%, and 7.6% respectively for maize, sorghum, and rice. Using our preferred model, we show that we underestimate the climatic impacts by 2050 on crop yields in Tanzania by 3.6%, 8.9%, and 28.6% for maize, sorghum and rice respectively if we focus only on climatic means and ignore climate variability. This study highlights that, in addition to shifts in growing season means, changes in intra-seasonal variability of weather may be important for future yields in Tanzania. Additionally, we argue for a need to invest in improving the climate records in these regions to enhance our understanding of these relationships.  相似文献

Anguina obesa n. sp., a new species of the genus, causing small seed galls inside the ovaries of foxtail weed plants (Alopecurus mysuroides Huds.) is described and illustrated based on its morphological and molecular characters. The new species is characterized by its 1516–2564 μm long obese females irregularly ventrally curved after fixation, having six lines in lateral fields, 6–9 μm long stylet with well-developed rounded knobs, constriction at junction of isthmus with the pharyngeal bulb, monodelphic-prodelphic female reproductive system, and conical, 60–80 μm long tail. Males of the new species are characterized with their slender 936–1420 μm long body, 25–30 μm long tylenchoid spicules, and bursa not reaching tail tip. Second stage juveniles of the new species were also common inside the galls and also recovered from soil in type locality. The new species is morphologically close to Anguina agropyronifloris, A. amsinckiae, A. paludicola and A. tumefaciens, but is more closely related to A. paludicola, from which it can be separated based on differences in morphological characters and internal transcribed spacer sequence. In Bayesian inference using sequences of the aforementioned genomic fragment, the new species formed a clade with A. agrostis, A. funesta, A. graminis, A. phalaridis and some unidentified isolates, with robust Bayesian posterior probability (BPP). The morphologically closest species, A. paludicola, occupied a separate position, outside of the clade containing the new species. The sequences of two other genomic fragments, 18S and 28S rDNA (D2/D3 region) were also made available for the new species. Morphological comparisons of the new species with the related species are discussed.  相似文献

The study aims at performing a comparative assessment of two types of burn wound treatment. The present study was designed to prepare crosslinked and blended two natural polymers nanofiber scaffolds using gelatin (GE) and hyaluronic acid (HA). The GE/HA composite nanofibrous membranes with varied GE/HA weight ratio have also been successfully fabricated by an electrospinning method. The average diameter of GE/HA fibers was in the range of 20 to 150 nm. In vivo efficacy was also investigated based on a deep second degree burns model for Wistar rats. At 14 days post-operation, the dermal defect basically recovered its normal condition. A percentage of wound closure of GE/HA composite nanofibrous membranes and ChitoHeal gel reached up to 81.9 % and 77.8 % respectively, compared with 65 % of the untreated control (p<0.05). Also, histological parameters were assessed on postoperative day 7 and 14. The results of in vivo experiments showed that more epidermis was formed in the gel and scaffold groups compared to the control group. The numbers of inflammatory cells in these two groups were also smaller as compared with the control group, which could well be the reason for the delayed healing in the control group.  相似文献
