Nitric oxide is detected by a new technique in which tunable infrared radiation from a spin-flip Raman laser is used to measure the absorption spectrumn of a gas sample by optoacoustic spectroscopy. Strawberry (Fragaria × ananassa Duch.) is rich source of dietary antioxidants, minerals and nutrients. Dietary antioxidants have been known as beneficial for enhancing the fitness, preventing certain diseases and even mitigating the effects of ageing. The objectives of the present study were to determine variability and inheritance of antioxidants, to identify antioxidant rich and productive genotypes, and to suggest suitable breeding approaches. The genotypes, namely Ofra, Chandler, Festival and Camarosa showed higher concentrations of dietary antioxidants and therefore could be useful in future breeding. Results indicate that the effect of the genotypes on antioxidant contents is stronger than that of the environment. The high heritability (>80%) and low genetic advance as percentage of mean (<40%) for ascorbic acid and β-carotene contents could be improved by heterosis breeding. However, selection and hybridization would be effective tools to enhance the phenols and anthocyanin content, and yield potential as these traits showed high heritability (>80%) and high genetic advance as percentage of mean (>40%). Positive direct effect on fruit yield was highest for phenol content (0.609) which is also fairly close to its correlation coefficient (0.765) indicating that a direct selection based on phenol content would be most effective and that the phenol content could be used as a reliable biochemical marker to identify the productive genotypes having higher amounts of dietary antioxidants. The in vitro performance of mango shoot culture is delimited by several factors, of which explant necrosis and media browning are considered as major constraints for successful exploitation of such an important commercial crop. Our results showed that source of explants had a significant effect on the performance of in vitro cultures of mango. The variations in survival of explants collected from glasshouse grown seedlings and field-grown stock plants indicated towards differences between their physiological/ontogenic age. Though, chronologically, both are young, the glasshouse grown shoots are ontogenetically younger and; therefore, responded better over field-grown shoots. Improved performance of explants harvested from mycorrhizal plants suggests integration of mycorrhizal biotechnology with tissue culture biotechnology in order to achieve better results as mycorrhiza being a well-known stress alleviator may help explants mitigate in vitro stresses. Furthermore, shoot tip explants of field-grown ‘Amrapali’ mango was assayed for their browning propensity at pre- and post-culture stages. Status of different bio-chemicals such as in vivo phenol, in vitro phenol exudation, phenylalanine ammonia lyase, peroxidase and polyphenol oxidase as affected by various pre-treatments were estimated to establish the relationship of these bio-chemicals with necrosis of mango shoot cultures. The different pre-treatments comprised etiolation of newly emerged shoots (5–7 days old) of stock plants using black polythene for 7 days, etiolation treatment + agitation of explants in antioxidant solution (antioxidants: ascorbic acid at 100 mg l−1 + citric acid at 50 mg l−1), etiolation treatment + agitation of explants in polyvinylpyrrolidone solution (PVP) at 0.2% and non-etiolated control. Of these, explants treated with PVP proved to be superior to other treatments with regards to explant necrosis percentage as such explants exhibited least activities of phenols and oxidative enzymes. Neural circuits regulate cytokine production to prevent potentially damaging inflammation. A prototypical vagus nerve circuit, the inflammatory reflex, inhibits tumor necrosis factor-α production in spleen by a mechanism requiring acetylcholine signaling through the α7 nicotinic acetylcholine receptor expressed on cytokine-producing macrophages. Nerve fibers in spleen lack the enzymatic machinery necessary for acetylcholine production; therefore, how does this neural circuit terminate in cholinergic signaling? We identified an acetylcholine-producing, memory phenotype T cell population in mice that is integral to the inflammatory reflex. These acetylcholine-producing T cells are required for inhibition of cytokine production by vagus nerve stimulation. Thus, action potentials originating in the vagus nerve regulate T cells, which in turn produce the neurotransmitter, acetylcholine, required to control innate immune responses. 相似文献

Bovine pregnancy-associated glycoprotein-1 (bPAG-1) is predicted to play an essential role during pregnancy and is labelled as a potential biochemical marker of pregnancy in ungulates. We have compared the generation of the glycosylated form of recombinant bPAG-1 (rbPAG-1) by human embryonic kidney 293 (HEK 293) and Chinese hamster ovary (CHO) cells in attached cultures and evaluated the adaptation of the rbPAG-1 transfected cell line to suspension culture. The PAG cDNA was cloned from placental RNA obtained from a slaughtered cow on day 55 of pregnancy. The PAG-pRcRSV expression vector was transfected into HEK 293 and CHO cells. Western blot analysis showed that clonal HEK 293 cells expressed rbPAG-1 better than CHO cells in attached cultures. Transfected HEK 293 cells were adapted to suspension culture in spinner flasks and the rbPAG-1 purified to homogeneity using ion-exchange, pepstatin-sepharose affinity chromatographies and preparative SDS-PAGE. The expression of rbPAG-1 was immunocharacterised using a polyclonal antibody. Our findings indicated that 293 cells are suitable for production of glycosylated form of rbPAG-1 and that the availability of the recombinant glycoprotein will aid in further studies to elucidate the function and structure of the protein. 相似文献

Homo and copolymers of monomers 2-(N-phthalimido) ethylmethacrylate (NPEMA) and 4-Chloro-3-methyl phenyl methacrylate (CMPMA) were prepared in N,N-dimethyl formamide (DMF) solution at 70 °C using 2,2-azobisisobutyronitrile (AIBN) as initiator. The solution of poly(NPEMA-co.-CMPMA) in 20 % DMF was used to fabrication electrospun nanofiber by electrospinning technique. IR data were primarily employed to characterize polymers. The formation of nanofibers was identified by SEM study. The metal ion uptake capacity of copolymers and nanofibers were obtain by batch equilibrium method using different metal ion solution. The antimicrobial activity of the copolymers, Polymer nanocomposites and their nanofibers were tested against different microbial organisms by using quantitative method. The main objective of this investigation was to find whether nanofiber are better remover of metal ions compared to copolymers. A series of monoazo disperse dyes based on 2-amino-4-phenylthiazole was prepared using variousN,N-dialkylaniline derivatives as the coupling component. The dyes were characterized by IR spectral studies, visible absorption
spectroscopy and elemental analysis. The dyeing performance of these dyes was assessed on cellulose triacetate and nylon fibers.
These dyes were found to give a wide range of colour shades varying from bright red to royal blue with very good depth, brightness
and levelness on fibers. The dyed fibers showed good to very good light fastness and very good to excellent fastness to washing,

A study was conducted to investigate the effect of dough formulation and hydration level on the rheological properties of pasta dough during pasta extrusion. Semolina 100%, whole wheat 100%, and the following mixtures semolina-whole wheat (49:51), semolina-flaxseed flour (90:10), whole wheat-flaxseed flour (90:10), and semolina-whole wheat-flaxseed flour (39:51:10) were the formulations used for the experiments. Dough was hydrated at 30, 32, and 34% moisture content. Pasta was extruded with a capillary and a semi-commercial pasta extruder to determine the apparent viscosity of the dough during extrusion conditions and its relationship to the behavior of the dough during pasta processing. Results showed that non-traditional pasta dough behaved like a shear thinning fluid that can be described by the Power Law model. Increased hydration levels and/or presence of flaxseed flour on the dough formulation decreased the apparent viscosity of the dough, which correlated with extrusion pressure, mechanical energy, and specific mechanical energy that were required to extrude the dough in the pasta extruder. The strong correlations found between the apparent viscosity of the dough and the pasta extrusion parameters indicates the possibility of using a capillary rheometer to determine the appropriate hydration level of ingredient formulations before extruding with a pasta press. 相似文献
