An important determinative of malt quality is the malt beta-glucan content, which in turn depends on the initial barley beta-glucan content as well as the beta-glucan depolymerization by beta-glucanase (EC 3.2.1.73) during malting. Another enzyme, named beta-glucan solubilase, has been suggested to act prior to beta-glucanase; its existence, however, has not been unequivocally proven. We monitored changes in beta-glucan levels and in the development of beta-glucan-degrading enzymes during malting of five lots of contrasting barley genotypes. Two models of in vivo kinetics for beta-glucan degradation were then compared as follows: (i) a biphasic model based on the sequential action of beta-glucan solubilase and beta-glucanase and (ii) a monophasic model assuming that all beta-glucans are depolymerized by beta-glucanase without the previous intervention of another enzyme. Confirmatory regression analysis was used to test the fit of the models to the observed data. Our results show that beta-glucan degradation is mostly monophasic, although some enzyme other than beta-glucanase seems to be required for the early solubilization of a small fraction of insoluble beta-glucans (on average, 7% of total beta-glucans). Furthermore, the genotype-dependent kinetic rate constant (indicating beta-glucan degradability), in addition to beta-glucanase activity, is suggested to play a major role in malting quality.  相似文献

The storage response of cactus pears [Opuntia ficus-indica Miller (L.) cv. Gialla] was investigated over 6 weeks at 6 degrees C, plus an additional week of simulated marketing period (SMP) at 20 degrees C, after a 3-min dip treatment with thiabendazole (TBZ) at 1000 mg/L at 20 degrees C or 150 mg/L TBZ at 52 degrees C. Untreated fruits were used as control. Following TBZ treatments at 20 and 52 degrees C, total residues were recovered from the peel of cactus pear, as the concentration of residues in the pulp was negligible. Treatments with 1000 mg/L TBZ at 20 degrees C resulted in a 2.82 mg/kg residue uptake (active ingredient, whole-fruit basis), whereas treatment at 150 mg/L TBZ left 1.09 mg/kg. TBZ showed great persistence over both storage and SMP: on average, in the fruits treated at 20 and 52 degrees C, over 72 and 68%, respectively, of TBZ was still present after SMP. Postharvest treatments with 1000 mg/L TBZ at room temperature did not affect the expression of slight-to-moderate chilling injury (CI), but reduced severe CI by approximately 50% and decay development by 63.4% in comparison to those of untreated fruit after SMP. The effectiveness of TBZ was much higher with the treatment at 150 mg/L TBZ at 52 degrees C, providing 91% control of severe CI and approximately 89% suppression of decay; no treatment damage occurred during storage and SMP. External appearance was better in fruit treated with 150 mg/L TBZ at 52 degrees C. Respiration rate, titratable acidity, soluble solids concentration, and acetaldehyde in the flesh were not significantly influenced by treatments. Ethylene production rate and ethanol levels in the flesh were significantly higher in the TBZ-treated fruit as opposed to those in the untreated control fruit.  相似文献

The exploitation of South American river turtles as a food source has long been considered the main factor contributing to the decline of populations. Along a stretch of the Aguarico River (Ecuador), we investigated the spatial and temporal distribution of terecay (Podocnemis unifilis) nests, factors affecting nest outcome, and the effect of offering a reward for each hatchling captured on the pattern of egg consumption by the local human community. Flooding influence on egg mortality appears to be particularly important in this Amazonian region, destroying 63.1% of all nests. This amount of nests resulted more than sufficient to satisfy the local community’s consumption needs (28.2%). The proposed reward for each hatchling ensured the voluntary participation of the Cofan people in the terecay conservation project, leading to: (i) nests being harvested only from sites where there were likely no hatching possibilities, (ii) efficient management and protection of nesting beaches with abolition of poaching of nests and adult females, and (iii) transplantation of nests from sites that would be flooded (and whose yield exceeds human consumption). Therefore, we argue that in this area of Aguarico River there are both biologically and socially favourable conditions for the establishment of a sustainable harvest of terecay eggs. Possible factors determining high nest mortality due to flooding in this area, as well as opportunities to make the project evolve toward economic self-sustainability, will also be discussed.  相似文献

The essential oil obtained from different parts of Ferula glauca L. (formerly considered as a subspecies of F. communis) growing in Marche (central Italy), was analyzed for the first time by GC-FID and GC-MS. The major volatiles were (E)-caryophyllene and caryophyllene oxide in leaves, alpha-pinene, myrcene and germacrene D in flowers, alpha- and beta-pinene in fruits, (E)-beta-farnesene, myristicin and elemicin in roots, respectively. The differences in composition detected with respect to F. communis, made the volatile fraction a reliable marker to distinguish between them, and confirm the botanical data at the base of their discrimination. Furthermore, the oil was assayed for its antimicrobial activity by the broth microdilution method. B. subtilis was found to be the most sensitive microorganism, with the lowest MIC values.  相似文献

ObjectiveTo evaluate the incidence of myoclonus (involuntary movements during anaesthesia, unrelated to inadequate hypnosis or analgesia, and of sufficient severity to require treatment) in dogs anaesthetized with a TIVA of propofol with or without the use of fentanyl.Study designRetrospective clinical study.AnimalsDogs, undergoing general anaesthesia for clinical procedures between January 2012 and January 2013 and subject to TIVA with propofol.MethodsA retrospective analysis reviewed the medical and anaesthetic records. Animals with existing or potential neurological or neuromuscular pathology in the anamnesis or upon clinical examination and cases with incomplete clinical records were excluded. Myoclonus was considered as involuntary muscle contractions which did not cease following a bolus administration of propofol or fentanyl and, due to their intensity and duration, made continuation of the procedure impracticable without other drug administration. Tremors, paddling or muscle spasms, explicable as insufficient hypnosis or analgesia, and transient excitatory phenomena only present during the awakening phase, were not considered as myoclonus.ResultsOut of a total of 492 dogs undergoing anaesthesia, six mixed breed dogs (1.2%), one male and five females, American Society of Anaesthesiologists (ASA) physical status I, median (range) weight 20.5 (7–37) kg and age 1.5 (1–5) years had myoclonus according to the aforementioned definition. In all subjects, myoclonus appeared within 20 minutes after induction of anaesthesia, and mainly involved the limb muscles. All subjects appeared to be in an adequate plane of anaesthesia before and during myoclonus.Conclusions and clinical relevanceThis study shows that 1.2% of dogs, undergoing TIVA with propofol with or without fentanyl administration, developed myoclonus, which required to be, and were treated successfully pharmacologically. Relationships between “available” soil phosphorus and foliar phosphorus content of Tectona grandis (teak) in West-Africa Significant relationships were analysed between the foliar phosphorus content of Tectona grandis L. and the topsoil phosphorus contents on a broad range of soils in West African plantations of Tectona grandis L. (teak), except for soils very poor in clay or organic matter (< 15% T or < 1% C). Of all methods used (P-H2O, P-Mehlich, P-CAL, P-Bray-1, P-Soltanpour, P-Dabin, P-Resin, P-H2SO4, P-550, P-org) the fraction of soil P (P-550) dissolved in 1 n H2SO4 after dry ashing at 550 °C (r = 0.548***, n = 63) and the method of Bray-1 (r = 0.505***, n = 63) show the best results. The first reflects 80-90% of the organic P content and is recommended for the evaluation of the phosphorus supply. Using multiple correlation analysis we found a highly significant relationship (R = 0.794***, R2 = 0.63, n = 58) between foliar P as depending variable and P-Bray (positive), the soil sand content (negative) as well as soil pH (positive). A sufficient phosphorus supply of teak foliage (0.2% P) occured at soil levels of 300-320 kg P per ha and 15 cm (or more than 150 mg P/kg in topsoils free of stones (P-550)) in the studied area between Benin and Liberia.  相似文献

In vitro grown kiwifruit (Actinidia deliciosa, Liang and Ferguson) plantlets were treated with two humic fractions distinguished by two different relative molecular mass and characterized through their elemental composition and 13Carbon (C) nuclear magnetic resonance. The effects exerted on plant growth, root morphology, and nutrition were evidenced by means of leaves and roots mineral content, root respiration, and nitrate reductase activity. The two humic substances differentially influenced the studied parameters. The lower molecular fraction humic substance, endowed with a higher content of phenolic and carboxylic groups, caused an enhanced content of micro- and macro-elements, together with a higher root respiration at lower concentrations (0.5–1 mg C L? 1). The high molecular fraction humic substance, needed higher concentrations (> 5 mg C L? 1) in order to achieve similar effects. This fraction also caused major changes on root morphology. The ability to improve micronutrient assimilation, in particular iron, confirmed the agronomic importance of humic substances on soil fertility.  相似文献

Two monoclonal antibodies were obtained against the apple proliferation phytoplasma that provide easy, rapid, specific and sensitive serological detection. They reacted specifically by using ELISA and immunofluorescence techniques with apple proliferation-infected periwinkles and apple trees from different regions in northern Italy and Slovenia, but not with several other phytoplasma isolates. We did not observe any monoclonal antibody reaction even using phytoplasmas belonging to the same phylogenetic group such as European stone fruit yellows and pear decline. Two serological techniques, immunofluorescence and ELISA, were compared with DAPI staining and PCR. From July until leaf fall ELISA was as sensitive as PCR but was more rapid and convenient than PCR; immunofluorescence was useful for specific detection of apple proliferation phytoplasma on roots throughout the year. Serological techniques could be conveniently applied in the roots, stems and leaves of apple trees depending on specific phenological stages of the plants.  相似文献

The effects of four light intensities (1000 lx, 500 lx, 50 lx, 3 lx) on growth, survival and feeding activity in common sole (Solea solea L.) larvae were studied from 4 to 51 days post hatching (dph). During the pelagic larval stage (4–12 dph), larvae reared at 3 lx showed a lower growth. From 19 onwards, the larvae reared under 3 lx displayed a significant ( 0.05) higher SGR than the other treatments and a higher final weight compared to 1000 lx and 500 lx. Survival rate was higher under intermediate light intensities (500 and 50 lx). Larvae reared at 3 lx displayed a significant delay in the degree of metamorphosis compared to the other treatments, while at 33 dph metamorphosis was completed under all treatments. Histological examination revealed the importance of vision and light in the first feeding of this species, while after metamorphosis, the full development of other sensory organs indicated that feeding activity is also mediated by chemosensory perception. Results indicate that high light intensity seems to be more suitable during the pelagic larvae, while the opposite would ensure better growth from the onset of metamorphosis to the benthic phase.  相似文献

The essential oils and supercritical CO(2) extracts of wild Daucus carota L. subsp. carota growing spontaneously in Sardinia and in Portugal were investigated. The main components in the Sardinian essential oil of flowering and mature umbels with seeds are beta-bisabolene (17.6-51.0%) and 11-alpha-(H)-himachal-4-en-1-beta-ol (9.0-21.6%); instead, the oils from Portuguese samples are predominantly composed of geranyl acetate (5.2-65.0%) and alpha-pinene (3.5-37.9%). Supercritical extracts contain lower amounts of monoterpenes and higher amounts of sesquiterpene hydrocarbons. Antifungal activities of the Sardinian oils were the highest, particularly for dermatophytes and Cryptococcus neoformans, with MIC values of 0.16-0.64 microL mL(-1).  相似文献
