Nematotoxic effect of an aqueous extract of Artemisia annua and its components caffeic acid, chlorogenic acid (5-caffeoylquinic acid, 5-CQA), artemisinin and the related semi-synthetic artesunate, was investigated on the root-knot nematode Meloidogyne incognita and the potato cyst nematode Globodera rostochiensis and on the virus-vector dagger nematode Xiphinema index. Juveniles of M. incognita and G. rostochiensis and females of X. index were exposed to 500, 250 and 125 μl ml?1 solutions of the A. annua aqueous extract, caffeic acid, chlorogenic acid and artesunate and to 50 μl ml?1 solution of artemisinin for 2, 4, 8 and 24 h. Egg masses of M. incognita and cysts of G. rostochiensis were exposed for 24, 48, 96 h and 1 or 2 weeks only to the extract solutions. Aqueous extract was highly effective on G. rostochiensis juveniles, whereas M. incognita juveniles were affected only at long exposure times. Adversely, egg hatch inhibition was strong on M. incognita and poor or minimal on G. rostochiensis. Females of X. index were sensitive only to long exposures to the highest extract concentration. Both caffeic and chlorogenic acid did not affect juveniles of M. incognita but were highly active on G. rostochiensis juveniles and X. index females even at the lowest concentration. Artesunate toxicity was almost zero on M. incognita and low on X. index females, but high on G. rostochiensis juveniles. Artemisinin solution was lethal to more than 50 % of G. rostochiensis juveniles within 24 h, but did not affect M. incognita juveniles and X. index females. Results suggest different roles of the tested compounds in the biocidal activity on each target nematode species. The extract of A. annua and its main phytochemicals seem to have a potential to be developed into new nematicidal formulates, though their activity should be validated in the soil. 相似文献

New esterase substrates were synthesized using cis-, trans- and cis-trans-permethrinic acid chloride and then used to measure pyrethroid-cleaving enzymes in insects. The new substrates, namely cis-, trans- and cis-trans-7-coumaryl permethrates (7-CP), show a structure very similar to permethrin insecticide and yield fluorescent products on hydrolysis. These substrates were hydrolyzed by a commercial porcine preparation that provided esterase-specific activity, and were stable at different pH values (5.2-7.8). Studies made with house fly, Musca domestica (L.), homogenates showed that these compounds are appropriate for determining pyrethroid hydrolysis activity on individual insects. The measured activity of house fly esterase was 870 relative fluorescence units (RFU) min(-1) with cis-7-CP as substrate, 1117 RFU min(-1) with trans-7-CP as substrate and 1423 RFU min(-1) with cis-trans-7-CP as substrate. The fluorescent substrates for pyrethroid-cleaving esterases described in this paper have advantages over methods already given in the literature. They are substrates with structures very similar to pyrethroids, the cleavage of which can be followed by an increase in fluorescence emission at 440 nm; it takes only about 5 min to measure the reaction, and moreover the high sensitivity of the fluorescence technique allows the quantification of esterase activity on individual insects. 相似文献

The rapid immunofilter paper assay (RIPA) was developed to detect Tomato spotted wilt virus (TSWV), Groundnut ringspot virus (GRSV), and Tomato chlorotic spot virus (TCSV) using antisera against recombinant nucleocapsid (N) proteins of each tospovirus. The two-step RIPA was sensitive enough to detect each pecies specifically in only 30min. Canine cognitive dysfunction syndrome (CDS) is a neurodegenerative disorder of aged dogs characterized by a progressive decline in cognitive function. In humans and laboratory animals, a variety of neurotransmitter abnormalities have been described in patients affected by age-related dementia. Specifically, the regulatory role of the catecholaminergic, serotonergic, and cholinergic systems has been outlined. The aim of the present study was to measure blood monoamine levels, platelet α2-adrenergic receptors, and lymphocyte muscarinic receptors in healthy adult and aged dogs and in dogs affected by canine cognitive dysfunction. Based on clinical and behavioral examination, 40 dogs were divided into 3 groups: healthy adults (n = 14), aged dogs (n = 17), and aged dogs affected by canine cognitive dysfunction (n = 9). A significant reduction in plasma levels of norepinephrine and dopamine was observed both in aged dogs (0.16 ± 0.02 ng/mL, P < 0.01; 0.11 ± 02 ng/mL, P < 0.01, respectively) and in CDS dogs (0.14 ± 0.03 ng/mL, P < 0.05; 0.10 ± 00.005 ng/mL, P < 0.01, respectively) compared with adults (0.29 ± 0.04 ng/mL and 0.15 ± 0.02 ng/mL, respectively). No significant differences were observed among groups for α2-adrenergic receptor concentrations. Canine lymphocytes express 2 distinct classes of muscarinic receptors, characterized by high (HA) and low affinity (LA) for [3H]-N-methyl-scopolamine. A significant age-dependent decrease in HA muscarinic receptors was observed. However, no differences were found between aged dogs (87.65 ± 11.08 sites/cell × 102) and in CDS dogs (90.17 ± 6.75 sites/cell × 102 ) for HA muscarinic receptor concentrations. As far as LA muscarinic receptors are concerned, CDS dogs showed a significant increase (393.48 ± 63 sites/cell × 102; P < 0.05) with respect to healthy adult dogs (188.84 ± 16.50 sites/cell × 102). Our results suggest that the reduction in HA muscarinic receptor-binding sites could be representative of the physiological aging process, whereas the increase in lymphocyte LA muscarinic receptor levels could be related to the cognitive decline. 相似文献

A simple oxygraphic method, for which the theoretical and experimental bases have been recently revised, has been successfully applied to evaluate the peroxyl radical chain-breaking characteristics of some typical food antioxidants in micelle systems, among which is a system that reproduces conditions present in the upper part of the digestive tract, where the absorption and digestion of lipids occur. This method permits one to obtain from a single experimental run the peroxyl radical trapping capacity (PRTC, that is, the number of moles of peroxyl radicals trapped by a given amount of food), the peroxyl radical trapping efficiency (PRTE, that is, the reciprocal of the amount of food that reduces to half the steady-state concentration of peroxyl radicals), and the half-life of the antioxidant ( t(1/2)) when only a small fraction of peroxyl radicals reacts with the antioxidants present in foods. Examples of application of the method to various types of foodstuffs have been reported, assessing the general validity of the method in the simple and fast evaluation of the above-reported fundamental antioxidant characteristics of foods. 相似文献

Large-scale reforestation programmes are a major source of unwarranted gene flow and can have profound consequences on local genetic diversity. Recently, ash has been introduced to Ireland from continental Europe to stock plantations but has often exhibited poor stem form. It was not known whether these trees were Fraxinus excelsior, Fraxinus angustifolia or interspecific hybrids that are known to occur in continental Europe. F. excelsior is the only native ash in Ireland, so the introduced populations represent a potential threat for the genetic diversity of native populations. We characterized the introduced trees within two plantations (Clonee and Kildalkey) using morphological characters and six microsatellite markers. Samples from continental Europe were included for comparison. Plantations exhibited higher genetic diversity than control populations because our data suggest they contain a mixture of several provenances. There was a small but significant differentiation between plantations and control populations (ФCT = 0.0211). Bayesian analysis to infer population structure and to assign introduced individuals to reference populations clearly demonstrated the presence of hybrid individuals within the plantations. The percentage of trees detected with potential hybrid origin ranged from 28 to 58 % depending on the plantation and the threshold data analysis level chosen. Most hybrids could be considered cryptic because there was a lack of intermediate morphology for hybrid individuals that mainly clustered with F. excelsior. The results indicated that the source of material at the two plantation sites differed. Management options to minimize the impact of these introduced populations are discussed. 相似文献

This study was designed to investigate the role of hybrids in maize Fusarium section Liseola interaction and fumonisin production, with particular emphasis on the occurrence and accumulation of hidden fumonisins in maize (masking phenomenon). In this 2 year study, naturally infected field crops were chosen with 10 maize hybrids, six of them grown in both years. Maize samples collected in 2010 showed a higher incidence of fungal infection as well as higher fumonisin contamination than those obtained in 2009 but a very similar incidence of F. section Liseola. Fumonisin masking was confirmed in raw maize, with a lower amount of hidden forms as compared to free fumonisins detected in the year with higher contamination. The chemical composition of the different hybrids was determined and correlated with the contamination data: the results obtained highlight the main role of fatty acids, with a higher fumonisin contamination in hybrids showing a higher linoleic acid content and a higher masking action in hybrids with higher oleic to linoleic ratio. These results represent a good basis to explain maize hybrid susceptibility to F. section Liseola infection, fumonisin contamination, and masking not related to a specific commercial hybrid but extendable to all hybrids. 相似文献
