Rheological properties of raw oat flour slurries were determined in experimental high β‐glucan (≤7.8%) and traditional oat lines (4–5% β‐glucan) grown in two consecutive years. Three different media were used to disperse oat flours: deionized water, silver nitrate solution (to inactivate endogenous enzymes), and alkali solution (to solubilize both water‐soluble and water‐insoluble β‐glucans). Significant correlations (P < 0.05) between viscosity of slurries and β‐glucan concentration obtained in either deionized water (r = 0.833), silver nitrate (r = 0.940), or alkali (r = 0.896) solutions showed that β‐glucans were the main contributor to oat extract viscosity. The highest correlation was obtained in silver nitrate solution, suggesting that inactivating endogenous enzymes is important to obtain high correlations. Predictive models of oat β‐glucan concentration based on the viscosity profile were developed using partial least squares (PLS) regression. Prediction of β‐glucan concentration based on viscosity was most effective in the silver nitrate solution (r = 0.949, correlation coefficient of predicted vs. analyzed β‐glucans) and least effective in the alkali solution (r = 0.870). These findings demonstrate that the β‐glucan in oat could be predicted by measuring the viscosity of raw flours in silver nitrate solution, and this method could be used as a screening tool for selective breeding.  相似文献

Experimental oat lines high in β‐glucan (6–7.8%) and traditional lines (3.9–5.7% β‐glucan) were used to evaluate the effect of β‐glucan on pasting (by rapid viscoanalysis) and thermal properties (by differential scanning calorimetry) of oat flours. Significant correlations established between β‐glucan concentration and the pasting parameters after amylolysis demonstrated the role of β‐glucans in oat pasting. The relative decrease of peak viscosity (PV) observed after enzymatic removal of β‐glucans was correlated with β‐glucan concentration (r = 0.880, P < 0.010) and reconfirmed their contribution to pasting. A significant increase of PV with β‐glucan concentration obtained under conditions of either autolysis (deionized water used for dispersion) (r = 0.89, P < 0.010) or inhibition (silver nitrate solution used for dispersion) (r = 0.91, P < 0.001) might be explained by an increase in water retention capacity caused by the β‐glucans. Predictive models of β‐glucan concentration based on the whole pasting profile or selected profile regions were developed using partial least squares (PLS) regression.Prediction of β‐glucan based on the whole profile obtained in the silver nitrate solution was the most effective (r = 0.93, correlation coefficient of predicted vs. analyzed β‐glucans, P < 0.050). No correlations were observed between the thermal properties of oat flours and the β‐glucan concentration.  相似文献

The objectives of this study were to determine the effects of the dietary docosahexaenoic acid (DHA) to arachidonic acid (ARA) ratio on the survival, growth, hypersaline stress resistance and tissue composition of black sea bass larvae raised from first feeding to metamorphic stages. Larvae were fed enriched rotifers Brachionus rotundiformis and Artemia nauplii containing two levels of DHA (0% and 10% total fatty acids=TFA) in conjunction with three levels of ARA (0%, 3% and 6% TFA). On d24ph, larvae fed the 10:6 (DHA:ARA) treatment showed significantly (P<0.05) higher survival (62.3%) than larvae fed 0:0 (DHA:ARA) (27.4%). Notochord length and dry weight were also significantly (P<0.05) greater in the 10:6 (DHA:ARA) treatment (8.65 mm, 2.14 mg) than in the 0:0 (DHA:ARA) (7.7 mm, 1.65 mg) treatment. During hypersaline (65 g L−1) challenge, no significant differences (P>0.05) were observed in the median survival time (ST50) between larvae fed 10% DHA (ST50=25.6 min) and larvae fed 0% DHA (ST50=18.2 min). The results suggested that black sea bass larvae fed prey containing 10% DHA with increasing ARA within the range of 0–6% showed improved growth and survival from first feeding through metamorphic stages.  相似文献

More knowledge is needed about variability of starch functional traits in adapted and exotic germplasm and possible genetic effects of these traits before conducting rigorous inheritance studies and breeding programs for starch quality. We studied and compared the range of variability for starch functional traits in a set of Corn Belt inbred lines with a set of exotic inbred lines from Argentina, Uruguay, and South Africa. Reciprocal hybrids of some of the lines within each set were compared with their parents. Functional traits were examined by using differential scanning calorimetry on starch extracted from single kernels of genotypes. The set of Corn Belt lines had a wider range of values for most traits than did the set of exotic lines. For both sets of lines, the maximum value for peak height index was as high as that previously reported for the waxy endosperm mutant. Although the Corn Belt lines exhibited a wider range of values for range of retrogradation than the exotic lines, the exotic lines showed a wider range of values for percentage retrogradation. Hybrid values were not consistently higher, lower, midpoint, or similar with respect to the values of their parents. This was true regardless of germplasm type or functional trait. Reciprocal cross values showed trends suggesting reciprocal differences, although there was no trend suggesting greater effect of the female parent. These traits seem to be controlled by many modifying effects in addition to major effects. Results indicate that sufficient variability exists in Corn Belt germplasm to conduct breeding and inheritance studies effectively and that there should be potential for breeding for functional traits.  相似文献

Decomposition of organic matter in inundated wetland soils requires a number of interdependent microbial processes that ultimately generate CO2 and CH4. Largely as the result of anaerobic decomposition, wetland soils store globally significant amounts of organic carbon and are currently net sources of CH4 to the atmosphere. Given the importance of wetlands in the global carbon cycle, it is important to understand controls on anaerobic decomposition in order to predict feedbacks between wetland soils and global climate change. One perplexing pattern observed in many wetland soils is the high proportion of CO2 resulting from anaerobic decomposition that cannot be explained by any measured pathway of microbial respiration. Recent studies have hypothesized that humic substances, and in particular solid-phase humic substances in wetland soils, can support anaerobic microbial respiration by acting as organic electron acceptors. Humic substances may thus account for much of the currently unexplained CO2 measured during decomposition in wetland soils. Here we demonstrate that humic acids extracted from a variety of wetland soils act as either electron donors or electron acceptors and alter the ratio of CO2:CH4 produced during anaerobic laboratory incubations. Our results suggest that soil-derived humic substances may play an important, and currently unexplored, role in anaerobic decomposition in wetland soils.  相似文献

Abstract: Two young adult dogs with gastrointestinal signs were each found to have an intra‐abdominal mass based on physical examination and diagnostic imaging. On exploratory laparotomy, small intestinal masses and mesenteric lymphadenopathy were found in both dogs; a liver mass was also found in dog 1. Cytologic and histologic examination of intestinal and liver masses and mesenteric lymph nodes revealed 2 distinct lymphoid cell populations: lymphoblasts and atypical Mott cells. With Romanowsky stains, the atypical Mott cells contained many discrete, clear to pale blue cytoplasmic inclusions consistent with Russell bodies that were positive by immunohistochemistry for IgM and CD79a in both dogs and for IgG in dog 2. The Mott cells and occasional lymphoblasts stained strongly positive with periodic acid‐Schiff. Using flow cytometric immunophenotyping in dog 1, 60% of peripheral blood mononuclear cells and 85% of cells in an affected lymph node were positive for CD21, CD79a, IgM, and MCH II, indicative of B‐cells. With electron microscopy, disorganized and dilated endoplasmic reticulum was seen in Mott cells in tumors from both dogs. Antigen receptor gene rearrangement analysis of lymph node and intestinal masses indicated a clonal B‐cell population. Based on cell morphology, tissue involvement, and evidence for clonal B‐cell proliferation, we diagnosed neoplasms involving Mott cells. To the authors' knowledge, this is the second report of Mott cell tumors or, more appropriately, B‐cell lymphoma with Mott cell differentiation, in dogs. More complete characterization of this neoplasm requires further investigation of additional cases. This lymphoproliferative disease should be considered as a differential diagnosis for canine gastrointestinal tumors.  相似文献

Flavonoids and hydroxycinnamic derivatives of turnip tops (Brassica rapa L. subsp. sylvestris L.) were characterized for the first time in four samples from different origins. Turnip tops exhibit a high polyphenols content (ranging from 107 to 191 mg/100 g, fresh weight) and a good antiradical activity, determined with the DPPH* test. After a liquid-liquid extraction and fractionation procedures, most flavonoids (isorhamnetin, kaempferol, and quercetin glycosides) and hydroxycinnamic derivatives were identified by means of HPLC-DAD/MS techniques. Isorhamnetin glycosides were the main flavonoid derivatives, differing from that found in the vegetables belonging to the Brassica oleracea group.  相似文献

Microbial‐based inoculants have been reported to stimulate plant growth and nutrient uptake. However, their effect may vary depending on the growth stage when evaluated or fertilizer applied. Thus, the objective of this study was to test the hypothesis that microbial‐based inoculants known to promote root growth and nutrient uptake will promote plant growth, enhance early root development, and increase nutrient concentrations of corn (Zea mays L.). Plants were evaluated at four different growth stages and in the presence of three different nitrogen (N) fertilizers. The microbial‐based treatments evaluated were: SoilBuilder™ (SB), a filtered metabolite extract of SoilBuilder™ (SBF), a mixture of four strains of plant growth‐promoting Bacillus spp (BM), and a water‐inoculated control. The experiment also included four fertilizer treatments: urea (U), urea‐ammonium nitrate (UAN), calcium‐ammonium nitrate (CAN), and an unfertilized control. Corn plants were evaluated at growth stages V2, V4, V6, and VT. Plant growth parameters for biomass, height, and SPAD readings were enhanced by the three microbial‐based treatments. A greater effect of microbial‐based treatments was observed when plants were evaluated at V6 and VT stages. Parameters of early root development such as total root length (TRL), root surface area (RSA), and length of fine roots were enhanced when microbial‐based treatments were applied. Concentrations of N, P, and K were also increased by microbial‐based treatments compared to the non‐inoculated control. Increases in plant N concentration due to microbial‐based treatments were on average 72% for CAN, 61% for UAN, 72% for urea, and 54% for the unfertilized control. Phosphorus concentration was increased most (138%) when BM was applied with CAN. In the same way, when CAN was present, K concentration was increased by 95% with BM and 65% when SB and SBF were applied. Overall, the results demonstrate that microbial‐based inoculants evaluated in this study can positively impact corn growth and nutrient concentration, especially during the late vegetative stages. Furthermore, the results indicate that the enhancement of nutrient concentrations (N, P, and K) in this case was related to the capacity of microbial‐based treatments to impact root morphology at early stages of corn growth.  相似文献
