The role of nitric oxide and reactive oxygen species, molecules indispensable for plant-pathogen signalling, was studied in the Lactuca spp.-Bremia lactucae pathosystem. Using a leaf disc model the translaminar effect of various compounds affecting their metabolism was studied by light microscopy. Time course studies revealed a slowdown in the development of B. lactucae (race BL16) infection structures by rutin (scavenger of reactive nitrogen and oxygen species) and SNP (NO donor) within 48 h post inoculation, followed by a retardation of sporulation. Application of the specific NO scavenger, PTIO, accelerated penetration of B. lactucae but had no further effects on the plant-pathogen interaction. Inhibitors of NO synthase (L-NAME) and nitrate reductase (sodium tungstate) did not influence pathogen development. Our results suggest that drastic change in the NO: hydrogen peroxide ratio seems to determine the pathogen’s fate. NO synthase-like activity significantly increased early after B. lactucae challenge in resistant L. virosa. Confocal laser scanning microscopy revealed the accumulation of nitric oxide in the penetrated cells, pointing to a role in the initiation of the hypersensitive reaction. The tips of germ tubes and appressoria of B. lactucae also accumulated NO, suggesting an essential role for this molecule in penetration of the biotrophic pathogen. Additionally, temporal changes in endogenous levels of rutin and quercetin in extracts from Lactuca spp. leaves will be discussed in connection to their role as part of the antioxidative machinery that influences the plants’ susceptibility/resistance to lettuce downy mildew.  相似文献

Two experiments were conducted on sheep to determine the effect of dietary supplementation with zinc and a medicinal plant mixture on haematological parameters and microbial activity in the rumen and large intestine. In Experiment 1, 24 male lambs were randomly divided into four groups: One group was fed an unsupplemented basal diet (control), and three groups were fed a diet supplemented with 70 mg Zn/kg diet in the form of Zn sulphate (ZnSO4), a Zn‐chelate of glycine hydrate (Zn‐Gly) or a Zn‐proteinate (Zn‐Pro), for five months. The ruminal content was collected separately from each lamb, and batch cultures of ruminal fluid were incubated in vitro with mixture of medicinal plants (Mix) with different roughage:concentrate ratios (800:200 and 400:600, w/w). Bioactive compounds in Mix were quantified by UPLC/MS/MS. In Experiment 2, four sheep were fed a diet consisting of meadow hay and barley grain (400:600, w/w), with Zn‐Gly (70 mg Zn/kg diet), Mix (10% replacement of meadow hay) or Zn‐Gly and Mix (Zn‐Gly‐Mix) as supplements in a Latin square design. Mix decreased total gas (p < 0.001) and methane (p < 0.01) production in vitro. In Experiment 1, caecal isobutyrate and isovalerate concentrations varied among the dietary treatments (p < 0.01). The isovalerate concentration of the zinc‐supplemented groups in the distal colon was higher (p < 0.001) compared with the control. In Experiment 2, the molar proportion of isobutyrate was the highest in the faeces of the sheep fed the diet with Zn‐Gly‐Mix (p < 0.01). The plasma zinc concentration was higher in the groups fed a diet supplemented with zinc (< 0.001). The haematological profile and antioxidant status did not differ between the dietary groups (p > 0.05). The diets containing medicinal plants and organic zinc thus helped to modulate the characteristics of fermentation in ruminants.  相似文献

Among the bacterial pathogens of Eucalyptus in Brazil, Ralstonia solanacearum is considered one of the most important because of the characteristics of the pathogen, like the high diversity among the strains related to host range, high virulence, broad geographical distribution and its damage to the crop in recent years. Given its importance and the lack of research on this pathosystem, the present study aimed to perform a molecular characterization of different strains of infected Eucalyptus plants in Brazil. A total of 19 bacterial cultures isolated from Eucalyptus in different regions of Brazil were analysed. A 372‐bp product generated by multiplex‐PCR amplification using Nmult primers identified all the strains analysed as belonging to phylotype II. Eighteen strains were grouped into subclade IIA and one into subclade IIB. The phylogenetic tree generated from the gene sequences of endoglucanase (egl) confirmed the classification of the strains into phylotype II and separated the strains into sequevars. Strains AMC22, IBSBF2568 and IBSBF2576 were grouped into a single clade, as were strains UFV18 and UFV20, with 89% and 78% a posteriori probability, respectively, forming two new potential sequevars not yet defined. We also identified strains belonging to sequevars 41 (100% probability) and 37 (88% probability). However, most of the strains did not fit into any previously described sequevar and did not form distinct clades. The results of the analysis of fragments amplified using the ERIC‐PCR technique indicated the existence of genetic diversity among the strains studied, with a generally high correlation between similarity and the geographical origin of the strains.  相似文献

Olive trees (Olea europaea L.) are considered moderately tolerant to salinity, with clear differences found among cultivars. One‐year‐old self‐rooted olive plants of the Croatian cv. Oblica and Italian cv. Leccino were grown for 90 d in nutrient solutions containing 0, 66, or 166 mM NaCl, respectively. The shoot length and the number of nodes and leaves for both cultivars were not affected by salinity up to 66 mM NaCl. However, at 166 mM NaCl, growth of Leccino was reduced earlier and to a higher extent than growth of Oblica. After 10 d of exposure to 66 and 166 mM NaCl, increased activity of superoxide dismutase (SOD) was observed in Leccino, whereas there was almost no response in Oblica. Reduced SOD activity in Leccino at 166 mM NaCl was observed after prolonged stress (90 d), whereas in Oblica SOD was increased at 66 mM compared to control or 166 mM NaCl. Electrolyte and K+ leakage were increased and relative water content decreased as NaCl concentration increased with similar intensity of response measured in both cultivars. Oblica exhibited an ability to keep a higher K+ : Na+ ratio at all salinity levels compared to Leccino, but since no difference was found in leaf K+ concentration, this was mainly achieved by less Na+ ions reaching the younger leaves. The antioxidative system represents a component of the complex olive salt‐tolerance mechanism, and it seems that the role of SOD in protection from oxidative stress depends on sodium accumulation in leaves.  相似文献

ABSTRACT

A meta-analysis of 297 treatment data from the Vezaiciai Branch of the Lithuanian Research Centre for Agriculture and Forestry long-term field experiment published from 2006 to 2015 was used to characterize the changes in SOC under different fertilization treatments and residue management practices in Lithuania’s acid soil. A meta-analysis was performed to quantify the relative annual change (RAC) of SOC content and the average RAC rate of SOC under four fertilization modes (farmyard manure (FYM) (40?t?ha?1)); alternative organic fertilizers (in the manure background (40?t?ha?1)); FYM (60?t?ha?1); alternative organic fertilizers (in the manure background (60?t?ha?1)) in two soil backgrounds (naturally acid and limed soil). The average RAC under four fertilization modes was 1.46 g?kg?1?yr?1, indicating that long-term fertilization had considerable SOC sequestration potential. Incorporation of alternative organic fertilizers in unlimed soil showed negative effects (?0.39 and ?0.66 g?kg?1?yr?1) in the observed long-term experiment. The RAC in the limed soil with incorporated organic fertilizers (FYM and alternative organic fertilizers), compared to the control, and varied from 0.25 g?kg?1?yr?1 in the treatment with incorporated alternative organic fertilizers (in the manure background (40?t?ha?1)) to 0.71 g?kg?1?yr?1 in the soil with FYM (60?t?ha?1). In this study, the average RAC rate of SOC under organic fertilization treatments in limed soil (5.07–6.54%) was longer than organic fertilization in unlimed soil (2.11–3.49%), which might be attributed to the application of organic manure that would result in a slow release of fertilizer efficiency. Our results indicate that the application of manure (40 or 60?t?ha?1) showed the greatest potential for C sequestration in agricultural soil and produced the longest SOC sequestration duration.  相似文献

Long-term soil cultivation at the same depth affects soil characteristics and crop productivity. The aim of the study was to investigate the impact of a long-term different intensity soil tillage methods and deep loosening on weed number, weed agrobiological group and soil seed bank changes in till Bathygleyic Dystric Glossic Retisol soil under the climatic conditions of the Western Lithuania (geographical coordinates 55°43′38″N, 21°27′43″E). The study included different soil tillage methods (conventional ploughing, shallow ploughing and shallow ploughless tillage) and deep loosening. During investigational years, the greatest weed number in crops and the greatest weed seed number in the seed bank were determined in the soil reduced tillage (shallow ploughing and shallow ploughless tillage). The weed number in crops of conventional ploughing soil was 35.8% lover compared to reduced tillage soil. The weed seed number in the seed bank of conventional ploughing was 49.6% lover compared to reduced tillage Decreasing soil tillage intensity resulted in weed seeds concentration in the upper topsoil. A one-time deep loosening had a significant effect during the crop rotation: the weed number in crops and weed seed number in the seed bank were determined to have increased by 26.6% and 51.6% in conventional ploughing soil and by 11.9% and 23.2% shallow ploughless soil respectively. However, after deep loosening, the number of Poa annua in crops decreased 2.9 times in plots of conventional ploughing and 1.7 times – in plots of shallow ploughing soil.  相似文献

After harvest, sugarcane residues left on the soil surface can alter nitrogen (N) dynamics in the plant-soil system. In Oxisols, the nitrogen fertilizer applied had its effects on the levels of ammonium and nitrate in the soil, N concentration in the plant leaves, and on the growth and productivity of second ratoon plants. The N rates tested were of 0, 60, 120, 180, and 240 kg ha?1. Each treatment was replicated four times. Four months after the experiment was started, ammonium and nitrate concentration in the soil, N levels in plant leaves, and plant growth were evaluated. Productivity was evaluated 11 months after the experiment was set. By increasing the content of mineral N in soil, plant growth variables reflected differences in the production of stems; however, it did not affect foliar N. The use of leaf analysis was not important to assess the nutritional status of nitrogen in the ratoon sugarcane. Nitrogen concentration in soil was affected by nitrogen fertilization, but not the N content in leaves. The rate of 138 kg N ha?1enabled greater production of sugarcane stalks (140 t ha?1).  相似文献

Recovery of phenols from olive mill wastewater (OMWW) was studied, comparing five sample preparation methods: filtration, solid-phase (SPE), liquid-liquid (LLE) and ultrasonic (US)-assisted extraction of liquid and solid (freeze-dried) OMWW. Results showed that ultrasonication is a good alternative to conventional solvent extractions, providing higher recoveries at both levels of individual and total phenol yields. Sonication of liquid OMWW in organic solvent was more efficient vs its nonassisted counterpart (agitation), but did not provide a representative phenol chromatogram due to ethyl acetate use. By contrast, the US-assisted extraction of freeze-dried OMWW (3 × 20 min) in 100% methanol (1.5 g/25 mL, w/v) offered the highest qualitative-quantitative phenol yields without any US-induced alterations. Moreover, freeze-drying is an excellent preservation of initial liquid OMWW, holding a great potential for delayed analysis. This study is also the first report that Slovenian OMWW may be utilized as a valuable source of phenols, especially hydroxytyrosol and tyrosol.  相似文献

Earlier studies of postmining heaps near Sokolov, Czech Republic (0–46 years old) showed that massive changes in plant community composition occur around 23 year of succession when the heaps are colonized by the earthworms Lumbricus rubellus (Hoffm.) and Aporrectodea caliginosa (Savigny). The aim of the current study was to test the hypothesis that the introduction of earthworms into a postmining soil enhances growth of late succession plant species. In a laboratory experiment, earthworms significantly increased biomass of Festuca rubra and Trifolium hybridum grown in soil from a 17-year-old site. The biomass increase corresponded to a significant decrease in pH and an increase in oxidable C, total N, and exchangeable P, K, and Ca content. A second laboratory experiment showed higher biomass production of late successional plant community (Arrhenatherum elatius, Agrostis capillaris, Centaurea jacea, Plantago lanceolata, Lotus corniculatus, and Trifolium medium) in soil from late successional stage (46 years old); the introduction of earthworms into soil from an early successional stage (17 years old) increased biomass production. In a field experiment, introduction of L. rubellus to enclosures containing a 17-year-old soil not colonized by earthworms significantly increased the biomass of grasses after 1 year. The results support the hypothesis that colonization of postmining areas by earthworms can substantially modify soil properties and plant growth.  相似文献
