Foliar diseases are the main biotic restriction reducing yield in wheat crops affecting both, grain number and/or grain weight, depending on developmental stage at which infection occurs (pre- or post-anthesis, respectively). Grain weight reductions due to foliar diseases were widely reported in the literature mostly associated with decreases on radiation interception during the grain filling period. However, different evidences in wheat showed variations on grain weight responses when fungicide was applied during the grain filling period, probably associated with the timing of fungicide application or with the amount of available resources per grain set when fungicides are applied. The present study was designed to determine the causes of grain weight reduction due to foliar diseases complex (including leaf rust, Septoria leaf blotch and tan spot) in wheat crops growing under contrasting agronomic and environmental conditions (i.e. different years, locations, cultivars and N supply). The experiments were carried out during 4 years under field conditions in different locations of Argentine and France. Five different commercial wheat cultivars were sown on early and late sowing dates; and two contrasting N availability and two fungicide treatments (protected and unprotected) were applied. Grain number was not affected by foliar diseases as their appeared after anthesis. Grain weight was strongly, poorly or not affected by foliar diseases and was not associated individually with both, the sink size and the source size. However, when the grain weight response due to fungicide application was plotted against the healthy area absorption per grain (HAAG), a significant negative association (r2 = 0.81; p < 0.0001) was found for the Argentine experiments. When the HAAG was corrected by the grain weight potential (HAAGW) all experiments conduced in Argentine and in France fit well to a common negative linear regression (r2 = 0.74, p < 0.0001) for the relationship between grain weight variation and HAAGW demonstrating that grain weight potential is an important feature to consider in diseases control programs. Foliar diseases forced the crop to use the accumulated reserved increasing the utilization rate of the water soluble carbohydrates (WSCUR), depleting as a consequence the water soluble content at physiological maturity (WSCPM) in all experiments. The association between WSCUR and the healthy area absorption per grain corrected by grain weight of healthy crops (HAAGW) suggest that foliar diseases in wheat cause source limitation, forcing to the crop to use the WSC reserve which could be insufficient to fill the grains previously formed.  相似文献

Grass cell wall degradability is conventionally related to the lignin content and to the ferulic-mediated cross-linking of lignins to polysaccharides. To better understand the variations in degradability, 22 maize inbred lines were subjected to image analyses of Fasga- and M?ule-stained stem sections and to chemical analyses of lignins and p-hydroxycinnamic acids. For the first time, the nearness of biochemical and histological estimates of lignin levels was established. Combination of histological and biochemical traits could explain 89% of the variations for cell wall degradability and define a maize ideotype for cell wall degradability. In addition to a reduced lignin level, such an ideotype would contain lignins richer in syringyl than in guaiacyl units and preferentially localized in the cortical region rather than in the pith. Such enrichment in syringyl units would favor wall degradability in grasses, contrary to dicots, and could be related to the fact that grass syringyl units are noticeably p-coumaroylated. This might affect the interaction capabilities of lignins and polysaccharides.  相似文献

Background

Root-knot nematodes (RKN) are major pest of olive tree (Olea europaea ssp. europaea), especially in nurseries and high-density orchards. Soil samples were collected from main olive growing areas of Morocco, to characterize Meloidogyne species and to discuss the contribution of biotic and abiotic factors in their spatial distribution.

Results

RKN were found in 159 soil samples out of 305 from nurseries (52.1% occurrence) and in 11 out of 49 soil samples from orchards (23.2% occurrence). Biochemical and molecular characterisation (PAGE esterase and SCAR) revealed the dominance of M. javanica both in nurseries and orchards with minor presence of M. incognita only in nurseries, and M. arenaria in only one nursery. RKN were distributed on aggregated basis. Frequent presence of M. javanica in orchards might have come from nurseries. In contrast, the detection of M. incognita in nurseries alone suggests that this species could not reproduce in orchards because of either the competition with other plant-parasitic nematodes or unfit local habitats. The impact of environmental variables (climate, habitat origin and physicochemical characteristics of the substrates) on the distribution of Meloidogyne species is also discussed.

Conclusion

Olive nurseries in Morocco are not able to guarantee the safety of rooted plants. As a result, olive production systems are exposed to strong RKN invasion risks. Consequently, the use of healthy substrates in nurseries may prevent plant-parasitic nematode induction in orchards.

Several theoretical models of the casein micelle structure have been proposed in the past, but the exact organization of the four individual caseins (α(s1), α(s2), β, and κ) within this supramolecular structure remains unknown. The present study aims at determining the topography of the casein micelle surface by following the interaction between 44 monoclonal antibodies specific for different epitopes of α(s1)-, α(s2)-, β-, and κ-casein and the casein micelle in real time and no labeling using a surface plasmon resonance (SPR)-based biosensor. Although the four individual caseins were found to be accessible for antibody binding, data confirmed that the C-terminal extremity of κ-casein was highly accessible and located at the periphery of the structure. When casein micelles were submitted to proteolysis, the C-terminal extremity of κ-casein was rapidly hydrolyzed. Disintegration of the micellar structure resulted in an increased access for antibodies to hydrophobic areas of α(s1)- and α(s2)-casein.  相似文献

Dairy industries are interested in knowing the heat treatment undergone by milk so as to control the quality of drinking milks or to control their heating systems. Among the different techniques available to characterize the heat treatment of milk, estimation of the denaturation of proteins has been widely used. However, because the concentration of the proteins in raw milk can fluctuate significantly, determining only the concentration of a native protein without knowing its concentration in the raw milk before undergoing heat treatment can lead to significant imprecision. The objective of this study was to develop, on Biacore 3000, a biosensor assay for determining the denaturation index of alpha-lactalbumin by quantifying separately the native and "heat-denatured" forms of alpha-lactalbumin with specific monoclonal antibodies. alpha-Lactalbumin denaturation index is independent of the concentration of alpha-lactalbumin in the original raw milk. The technique developed is discriminating, fast, repeatable, fully automated, and requires no pretreatment of the milk sample.  相似文献

Bulk and rhizosphere soil of rape and barley grown in a calcareous soil were pre-incubated for 7 days at 20 °C with Na235SO4 to partially label soil organic S. The soils were then incubated for 7 days more with increasing levels of two C sources as organic acids (succinic and malic acids) and as glucose (from 0 to 640 mg C kg−1 soil) with or without increasing levels of N (from 0 to 15 mg N kg−1 soil) in the form of ammonium nitrate, in order to mimic rhizodeposition inputs into soil. A second incubation experiment with a single highest dose of the used substrates was undertaken and two destructive soil samplings on days 17 and 35 were carried out. Both incubation experiments showed the intensities of S immobilization in the order: barley rhizosphere>rape rhizosphere>bulk soil. Glucose addition generated positive S priming effects in all studied soils after one week of incubation. Significant correlation coefficients were observed between immobilized-S and microbial biomass-S (r=0.95,p<0.001), arylsulfatase activity (ARS) and microbial biomass-S (r=0.65,p<0.05) on day 17 but not on day 35, whereas significant correlation coefficients were found between arylsulfatase activity and immobilized-S at both days 17 (r=0.79,p<0.01) and 35 (r=0.75,p<0.01). A marked decline of biomass-S noted in substrate-amended treatments at day 35 suggests a quick turnover of this compartment followed by its incorporation into the organic S. Finally, with organic acids high values of ARS per unit of biomass-S were recorded over the two studied dates in the rhizosphere soil of rape. It is concluded that the rhizosphere microbial biomass under rape exhibited more efficient arylsulfatase activity and hence greater turnover of organic S than that under the barley rhizosphere soil.  相似文献

One of the possible initiating factors in canine cranial cruciate ligament (CCL) rupture could be an abnormal pattern of ligament cell death. This study compared apoptotic cell death in sections of ruptured CCLs and normal controls, and examined nitric oxide (NO) production in joint tissues and correlated this to apoptosis. CCLs and cartilage from the lateral femoral condyle were harvested from 10 healthy dogs and 15 dogs with CCL rupture and ligaments were further processed to detect cleaved caspase-3 and to determine supernatant NO production in explant cultures. Apoptotic activity was greater in ruptured ligaments compared to controls. NO in ligaments showed a moderate but significant positive correlation with caspase-positive cells. The results suggest that increased apoptosis has a role in CCL rupture and that apoptosis may be influenced by local NO production.  相似文献

The aim of the present study was to evaluate some enzymes inhibitory effects of 11 plant species belonging to 9 families from Burkina Faso. Methanolic extracts were used for their Glutathione-s-transferase (GST), Acetylcholinesterase (AChE), Carboxylesterase (CES) and Xanthine Oxidase (XO) inhibitory activities at final concentration of 100 microg mL(-1). The total phenolics, flavonoids and tannins were also determined spectrophotometrically using Folin-Ciocalteu, AlCl3 and ammonium citrate iron reagents, respectively. Among the 11 species tested, the best inhibitory percentages were found with Euphorbia hirta, Sclerocarya birrea and Scoparia dulcis (inhibition > 40%) followed by Annona senegalensis, Annona squamosa, Polygala arenaria and Ceratotheca sesamoides (inhibition > 25%). The best total phenolic and tannin contents were found with S. birrea with 56.10 mg GAE/100 mg extract and 47.75 mg TAE/100 mg extract, respectively. E hirta presented the higher total flavonoids (9.96 mg QE/100 mg extract). It's was found that Sclerocarya birrea has inhibited all enzymes at more than 30% and this activity is correlated to total tannins contents. Contrary to S. birrea, the enzymatic activities of E. hirta and S. dulcis are correlated to total flavonoids contents. Present findings suggest that the methanolic extracts of those plant species are potential inhibitors of GST, AChE, CES and XO and confirm their traditional uses in the treatment of mental disorders, gout, painful inflammations and cardiovascular diseases.  相似文献

Foliar diseases are the main biotic cause of yield loss in wheat crops (Triticum aestivum L.) in Argentina and other regions around the world. Most of the studies on foliar diseases take a phytopathological perspective, but few studies have analyzed the problem with an eco-physiological approach aimed at the understanding of which crop traits are affected by foliar diseases. The present study was designed to determine the effects of a foliar disease complex (including leaf rust, Septoria leaf blotch and tan spot), on (i) grain yield and (ii) the physiological components of biomass production; intercepted radiation (RI) and radiation use efficiency (RUE), in bread wheat crops growing under contrasting agronomic and environmental conditions (i.e. different cultivars, years, location and nitrogen supply). The experiments were carried out during 4 years in different locations (three in the rolling pampas of Argentina and one in northern of France). Five different commercial wheat cultivars were sown on early (E) and late (L) sowing dates (SD); and two contrasting nitrogen availability and two fungicide treatments (protected and unprotected) were applied. Foliar diseases appeared during the grain filling period and affected both, leaf area duration (LAD) and healthy area duration (HAD) during that period. Foliar diseases reduced both, above-ground biomass at harvest (1533 and 1703 g m−2 for unprotected and protected treatments, respectively) and grain yield (646 and 748 g m−2 for unprotected and protected treatments, respectively) without important effects on harvest index. Biomass reductions after anthesis, due to the effects of foliar diseases, were associated with a reduced capacity of the canopy to absorb solar radiation more than any effect on RUE. However, RUE was consistently lower—when leaf rust was the predominant disease in the crop, suggesting that this biotrophic pathogen could affect the photosynthetic activity at the leaf or canopy level.  相似文献
