The present study aimed to assess the effect of polymorphisms in the tumor necrosis factor α (TNF-α) promoter (A/A, A/G and
G/G) and exons (T/T, T/C and C/C) on immune function and reproductive performance in dairy cows. The occurrence of the first
postpartum ovulation within 3 weeks in the cows with the TNF-α promoter A/G and G/G genotypes was higher than in the A/A
group. Among the different TNF-α exon genotypes, the occurrence of early first postpartum ovulation was higher in the T/C and
C/C genotype groups than in the T/T group. Single nucleotide polymorphisms (SNPs) in the TNF-α gene did not affect the rate
of artificial insemination (AI) or duration from parturition to next conception (days open). The apoptosis rate of
polymorphonuclear leukocytes (PMNs) did not differ among the TNF-α promoter genotypes, but the PMN transmigration rate was
significantly higher for the A/A and A/G genotypes than for the G/G genotype. Interleukin 8 (IL-8) mRNA expression in PMNs
and peripheral blood mononuclear cells (PBMCs) before culture was significantly higher for the A/A genotype compared with the
G/G genotype. There were no significant differences between the genotypes in the mRNA expression of TNF-α, IL-6, IL-1β, and
toll-like receptor 4 (TLR4) in PMNs and PBMCs before and 4 h after culture. IL-8 and IL-1β production by PBMCs cultured for 4
h was significantly higher for the animals with the A/A genotype than for those with the G/G genotype. On the other hand, no
significant difference was observed in IL-8 and IL-1β production by PMNs among different TNF-α genotypes. Taken together,

Six adult healthy Beagles were used to investigate the hepatic perfusion dynamics of Levovist, a contrast agent used in contrast harmonic imaging (CHI). In addition, 8 dogs with hepatocellular carcinoma (HCC) and 2 dogs with metastatic hepatic hemangiosarcoma (HSA) were used to characterize both the CHI findings with Levovist. In the Beagles, the start of intravenously injected Levovist into the aorta between the cranial mesenteric and renal arteries and the portal vein at the hepatic hilum were 5.47 +/- 1.52 sec and 16.03 +/- 3.39 sec, respectively. As a characteristic CHI finding in the 8 dogs with HCC, the early arterial phase showed a fine network of blood flow enhanced at the surrounding region and within the tumor in all the 8 dogs (100%), and the post vascular phase demonstrated a defect in the whole tumor and an enhancement of the surrounding hepatic tissues in 7 dogs (87.5%). In the 2 dogs with HSA, characteristic finding in which the early arterial and late vascular phases showed a rim contrast enhancement pattern, and the post vascular phase revealed that the whole tumor lacked contrast enhancement and the surrounding hepatic tissues was clearly enhanced. In dogs, the start of the early arterial and late vascular phases, and the characterizations of the CHI findings in HCC and HSA were suggested to be similar to those in humans. Therefore, CHI is thought to be useful for the diagnosis of HCC and metastatic hepatic HSA in dogs as well as in humans. 相似文献

The effects of addition of alpha-casein (alpha-CN) to dried egg white (DEW) were investigated by measuring transparency, hardness, and water-holding capacity (WHC) of the heat-induced gels. A DEW concentration of 8% (w/w) was required for formation of a self-supporting gel following heating at 80 degrees C for 20 min at pH 7. Solutions of alpha-CN, even up to a protein concentration of 12% (w/w), did not gel under the same conditions. The addition of alpha-CN (0.5-4%) to 8% DEW caused the increase in gel hardness gels, as compared with DEW gels alone at a total amount of protein concentrations, and the mixed gels became transparent with the increase of added alpha-CN concentrations. The 10% mixed protein solutions of alpha-CN (3-6%) and DEW (4-7%) formed transparent gels, although each protein did not gel individually at their protein concentrations. Mixture with 2:8 mixing ratio of alpha-CN to DEW at a total protein concentration of 10% showed synergistic effects in improving DEW gel properties above pH 7 and below 25 mM NaCl. The improvements (hardness, transparency, and WHC) of DEW gel by alpha-CN seem to be caused mainly by the inhibition of alpha-CN against heat coagulation of DEW protein. 相似文献

Regiospecific profiles of fatty acids (FA) of triacylglycerols (TAG) and phospholipids (PL) isolated from red and black rices were investigated. The lipids extracted from red and black rices were separated by thin‐layer chromatography (TLC) into eight subfractions, respectively. With a few exceptions, the major lipid components were TAG (76.4–80.5%), free FA (7.2–9.8%), and phospholipids (3.5–3.6%), while hydrocarbons, steryl esters, diacylglycerols (1,3‐DAG and 1,2‐DAG), and monoacylglycerols were present in minor proportions (0.1–4.1%). The PL components isolated from the two cultivars were phosphatidyl choline (52.3–53.7%), phosphatidyl ethanolamine (22.3–23.1%), phosphatidyl inositol (20.6–21.3%), and others (<3.4%). No significant differences (P < 0.05) in FA distribution were found when these cultivars were compared. The principal characteristics of the FA distribution in the TAG and PL were evident in the rices between the two cultivars: unsaturated FA were predominantly located at the sn‐2 position (77.3–96.8%), while saturated FA primary occupied the sn‐1 or the sn‐3 position (35.0–78.7%) in these lipids. The results of this study could provide useful information to both consumers and producers during manufacture of traditional rice foods in Japan. 相似文献

The infection dynamics of avian haematozoa, which includes the genera Plasmodium, Haemoproteus, and Leucocytozoon, are complicated by a variety of environmental factors and host-parasite interactions. In Japan, the prevalence of haematozoa in wild birds has recently been determined in several local areas. However, no information on the annual prevalence of avian haematozoa in a single study site has been reported. Here, we investigated the long-term infection dynamics of haematozoa in wild birds inhabiting a mountain forest of Japan. Blood samples were collected from 415 wild birds captured in the Chichibu mountains in Saitama Prefecture at an altitude of 1650 m between 2007 and 2010. All obtained samples were examined for haematozoan infection using nested polymerase chain reaction (PCR) of the cytochrome b (cytb) genes of haematozoa. A total of 62 out of 415 (14.9%) forest birds were PCR positive for haematozoa. Relatively high infection rates of Leucocytozoon were found among several bird species (Parus ater, 64.3%; Parus montanus, 81.8%) and may be due to the host preference of vector black flies and host nestling pattern in this forest. Phylogenetic analysis of amplified cytb sequences revealed for the first time that a variety of lineages of avian haematozoa are distributed among wild bird hosts in a high-altitude forest stand in Japan. Notably, significant seasonal changes of the prevalence of avian haematozoa were not observed; however, continuous investigation will likely provide detailed information on host-parasite interactions, including local environmental factors, that influence the dynamics of avian haematozoan infections. 相似文献

A 15-day-old Holstein calf with lethargy and tachypnea presented to the Veterinary Teaching Hospital at Obihiro University of Agriculture and Veterinary Medicine for evaluation of suspected congenital heart defect. A Levine grade 6 systolic murmur was noted at right apical site auscultation and phonocardiogram also recorded systolic a murmur. Electrocardiography findings include increased R and S waves, R wave split, and negative T waves without arrhythmia. Echocardiography revealed a single ventricle with a trace of the right ventricular wall, atrioventricular valve regurgitation, and turbulent in a single ventricle. Arterial blood analysis showed a marked decrease in oxygen saturation of 78% and oxygen partial pressure of 44 mmHg. Post-mortem examination confirmed the diagnosis of a single left ventricle. 相似文献

The purpose of this study was to evaluate the effect of protein malnutrition on the skin epidermis of mice. A low protein diet induced thinning of the skin epidermis, a decrease of cell proliferative activity in epidermal cells and a decrease of stratum corneum hydration. Dityrosine was immunostained in the cytoplasm of epidermal cells in the low protein diet group. Plasma advanced oxidation protein product (AOPP) levels were significantly more increased in the low protein diet group than in the control diet group. These results suggest that protein malnutrition adversely affects the structure and water barrier and reservoir functions of the skin epidermis, and these pathological changes are associated with the expressions of protein oxidation markers, dityrosine and AOPP. 相似文献

Because of the increasing use of maize hybrids with genetically modified (GM) stacked events, the established and commonly used bulk sample methods for PCR quantification of GM maize in non-GM maize are prone to overestimate the GM organism (GMO) content, compared to the actual weight/weight percentage of GM maize in the grain sample. As an alternative method, we designed and assessed a group testing strategy in which the GMO content is statistically evaluated based on qualitative analyses of multiple small pools, consisting of 20 maize kernels each. This approach enables the GMO content evaluation on a weight/weight basis, irrespective of the presence of stacked-event kernels. To enhance the method's user-friendliness in routine application, we devised an easy-to-use PCR-based qualitative analytical method comprising a sample preparation step in which 20 maize kernels are ground in a lysis buffer and a subsequent PCR assay in which the lysate is directly used as a DNA template. AbstractSlag silicate fertilizer (SSF) is applied to paddy fields with different soil chemical properties to increase silicon (Si) concentration in rice (Oryza sativa L.) plants. However, the effects of soil chemical properties on Si availability of SSF to rice Si uptake is poorly understood. To investigate the relationships between chemical properties of soils and the effects of SSF application on the Si concentration in rice plants, a field experiment was conducted in 2007 and 2008 at 18 paddy fields on the Shounai Plain, Yamagata, Japan. Two treatments were implemented: SSF applied at 1.5 t ha?1 and a no-SSF control. The Si concentrations of rice tissues were measured at the tillering and ripening stages. The difference in the Si concentrations of rice tissues between treatments (ΔSi concentration) was used to evaluate the effect of SSF. The Si concentrations in the shoots and aboveground parts of the rice plants were significantly increased by the SSF application in six or more of the fields at the two growth stages, whereas the Si concentrations in the panicles of the rice plants at the ripening stage were not increased significantly in most fields. Results of two-way analysis of variance evidenced a significant effect of field on the ΔSi concentrations in the shoots and aboveground parts at both growth stages. Furthermore, the ΔSi concentrations in the same rice plant tissues and at the same growth stages in the first year and in the second year were found to be positively correlated. These results indicate that the effect of SSF on the Si concentration in shoots and aboveground parts of rice plants varies from field to field. The ΔSi concentrations in shoots and aboveground plant parts at both growth stages were also found to be negatively correlated with soil chemical properties, i.e., available Si, Si adsorption capacity, contents of Si adsorbents (acid oxalate-extractable iron and manganese) and the pH under flooded soil conditions. These findings imply that those soil chemical properties of paddy fields should be taken into account for better prediction of ΔSi concentration of rice plants. 相似文献
