Abstract. Nitrate leaching was measured for four years at the Royal Agricultural College 's Coates Farm in the Cotswolds, England. Coates is a typical Cotswold mixed farm with thin, well-drained calcareous soils especially prone to leaching. Over the duration of this study there were dairy, sheep and arable enterprises on the farm. A 'Farm Gate' nitrogen (N) budget was constructed. Small 120 m × 20 m 'farmlets' were sited in ten fields across the farm, covering all parts of the rotation, as the sites for detailed measurements. Each farmlet received the same management as the rest of the field in which they were situated. Using ceramic probes inserted to 60 cm, soil water was sampled every two weeks throughout the winter drainage season. The annual drainage varied from 135 mm under grassland in 1996/7 to 600 mm under cereals in 1998/9. Average N losses by leaching were determined mostly by rainfall and were 65 kg N ha–1 yr–1, accounting for 25% of the N inputs. Especially leaky parts of the rotation were the ploughing out of a lucerne ley and the grazing of stubble turnips with sheep, both typical Cotswold farm practices. The research highlights some of the difficulties in developing practicable, profitable management practices to decrease nitrate losses.  相似文献

Although a principal source of energy and protein for millions of the world's poorest people, the nutritional value of sorghum (Sorghum bicolor L. Moench) is diminished because of low digestibility of grain protein and starch. To address this problem, we analyzed the properties of two sorghum lines that have a common pedigree but differ in digestibility. Consistent with results based on a ruminal fluid assay, the protein and starch of one line (KS48) was more thoroughly digested than that of the other (KS51) using in vitro assays based on pepsin and α-amylase. The indigestibility of KS51 relative to KS48 was shown to be due to (i) a greater abundance of disulfide-bonded proteins; (ii) presence in KS51 of non-waxy starch and the accompanying granule-bound starch synthase; and (iii) the differing nature of the protein matrix and its interaction with starch. The current findings suggest that each of these factors should be considered in efforts to enhance the nutritional value of sorghum grain.  相似文献

Copy number variants affect both disease and normal phenotypic variation, but those lying within heavily duplicated, highly identical sequence have been difficult to assay. By analyzing short-read mapping depth for 159 human genomes, we demonstrated accurate estimation of absolute copy number for duplications as small as 1.9 kilobase pairs, ranging from 0 to 48 copies. We identified 4.1 million "singly unique nucleotide" positions informative in distinguishing specific copies and used them to genotype the copy and content of specific paralogs within highly duplicated gene families. These data identify human-specific expansions in genes associated with brain development, reveal extensive population genetic diversity, and detect signatures consistent with gene conversion in the human species. Our approach makes ~1000 genes accessible to genetic studies of disease association.  相似文献

Abstract. As part of a study of recession farming on the 'fadama' lands on a segment of the Komadugu-Yobe floodplain, a survey was carried out to investigate the local farmers' perception of soil types and management practices. The farmers are clearly aware of the differences in soil type on the fadama and they possess unique skills in managing their farm lands. The farmers classify fadama soils for recession farming by assessing soil texture and soil drainage conditions by feel and observation. Integrating such local knowledge into soil surveys will lead to better practical definition of mapping units and give soil names that have more meaning for the farmers.  相似文献

Calorie restriction (CR) increases life span in yeast independently of Sir2. Lamming et al. (Reports, 16 September 2005, p. 1861) recently proposed that Sir2-independent life-span extension by CR is mediated by the Sir2 paralogs Hst1 and Hst2. Contradictory to this, we find that CR greatly increases life span in cells lacking Sir2, Hst1, and Hst2, which suggests that CR is not mediated by Sir2, Hst2, or Hst1.  相似文献

A retrospective study of laminitis was carried out to identify risk factors associated with this disease on an East Anglian farm with approximately 1000 animals living in an area of 1000 acres. Medical records between January 1997 and May 2000 and between April 2005 and March 2008 were reviewed, and the age, sex, weight (kg), height (inches [in] and hands [H]) and weight-to-height ratio (kg/in) was recorded. The prevalence, incidence and seasonality of laminitis were determined and their relationship to the monthly temperature, rainfall and hours of sunshine was evaluated. Averaged over the six years, the highest prevalence (2.6 per cent) and incidence (16 cases/1000 animals) of laminitis occurred in May. The findings of a multivariate analysis revealed that females (P=0.007, odds ratio [OR] 1.46, 95 per cent confidence interval [CI] 1.1053 to 1.9646) and light animals (P ≤ 0.001, OR=0.995, 95 per cent CI =0.9932 to 0.9963) had the greatest risk of developing laminitis. A positive association was found between hours of sunshine and incidence (P=0.007, relative risk [RR] 1.009, 95 per cent CI 1.001 to 1.012) and prevalence (P=0.002, RR 1.008, 95 per cent CI 1.003 to 1.012) of laminitis. The data suggest that there is a relationship between season, sex of the animal and the development of laminitis.  相似文献

Abstract. The main inputs, outputs and transfers of potassium (K) in soils and swards under typical south west England conditions were determined during 1999/00 and 2000/01 to establish soil and field gate K budgets under different fertilizer nitrogen (N) (0 and 280 kg ha−1 yr−1) and drainage (undrained and drained) treatments. Plots receiving fertilizer N also received farmyard manure (FYM). Potassium soil budgets ranged, on average for the two years, from −5 (+N, drained) to +9 (no N and undrained) kg K ha−1 yr−1 and field gate budgets from +23 (+N, drained) to +89 (+N, undrained). The main inputs and outputs to the soil K budgets were fertilizer application (65%) and plant uptake (93%). Animals had a minor effect on K export but a major impact on K recycling. Nitrogen fertilizer application and drainage increased K uptake by the grass and, with it, the efficiency of K used. It also depleted easily available soil K, which could be associated with smaller K losses by leaching.  相似文献

Slot blot hybridisation studies with total genomic DNA probes were used to compare Leptospira interrogans serovar hardjo strain Hardjoprajitno, strain Hardjobovis and a number of other Leptospira interrogans serovars. Strains Hardjoprajitno and Hardjobovis were found to have little genetic relationship with each other when compared to some of the other serovars tested. Hardjoprajitno is closely related to serovar icterohaemorrhagiae and not to Hardjobovis whereas Hardjobovis is closely related to serovars vietnam, balcanica and javanica but not to serovar icterohaemorrhagiae; this places strain Hardjoprajitno in the species L interrogans and strain Hardjobovis in the species L borgpetersoni. Because of this lack of genetic relatedness between strains Hardjoprajitno and Hardjobovis, it is proposed to remove the prefix Hardjo from the strain name Hardjobovis and call it L borgpetersoni serovar hardjo strain Bovis.  相似文献

Titers of two systemic neonicotinoid insecticides in citrus trees were measured in conjunction with conventional evaluations of their impact on glassy-winged sharpshooter (Homalodisca coagulata (Say); GWSS) populations. Xylem fluid samples were collected at regular intervals and from multiple locations within field-grown citrus trees to determine imidacloprid and thiamethoxam concentrations using commercial ELISA kits. Uptake profiles varied considerably with peak mean titers of imidacloprid occurring 6-8 weeks after application compared with 2 weeks for thiamethoxam. The persistence of each compound also varied as near-peak levels of imidacloprid were sustained for another 6-10 weeks before gradually declining. In contrast, thiamethoxam titers declined more rapidly after the initial peak, possibly reflecting an application rate only one-quarter of that used for imidacloprid. Within-tree distributions were more similar for the two compounds, with no significant effect due to height of the sample (upper or lower half) or to the quadrant location within the tree, with the exception of one quadrant in the thiamethoxam-treated trees. Substantial reductions in GWSS nymphs and adults were observed in imidacloprid-treated trees during the 2001 trial and were sustained for 4-5 months after treatment. Treatment effects on nymphs were not as well pronounced in the 2002 trial, when overall GWSS infestations were much reduced from the previous year. However, consistently lower adult infestations were still observed in 2002 for both treatments compared with untreated trees. Information on the spatial and temporal profiles in citrus trees was obtained for both compounds to complement field impact data and improve understanding of their pest management potential.  相似文献
