In aerobic rice cultivation systems, compost mulching and incorporation are important to rehabilitate the soil. Microbial-fortified compost is increasingly accepted as a safe approach in agro-waste management to recycling of crop residuals in agriculture soil and also to promote growth and suppress disease. This study aims to examine the stability and viability of the selected plant growth-promoting microorganisms (PGPM) in rice straw compost (RSC) over incubation period and its bio-efficacy in promoting rice (Oryza sativa) plant growth, productivity, soil health, and controlling of Pyricularia oryzae in aerobic cultivation conditions. Six selected PGPM: Pseudomonas aeruginosa (UPMP1), Corynebacterium agropyri (UPMP7), Enterobacter gergoviae (UPMP9), Bacillus amyloliquefaciens (UPMS3), Trichoderma harzianum (UPMT1), and Trichoderma virens (UPMT2) were used as a consortium of microbial inoculants to develop the microbial-fortified rice straw compost (MRSC). The MRSC was incorporated into mineral soil used for aerobic rice cultivation and its bio-efficacy was evaluated at harvest. The viability of Trichoderma spp. found stabilized at 6.78–6.00 log cfu/g and declined for all the bacterial isolates. At harvest, soil amended with MRSC significantly increased in plant height, leaf area index, 1000 grain weight, and productivity. The MRSC amended plots had significant low in rice blast disease severity with area under disease progress curve (AUDCP) of 748.22 unit/square, as compared to control (1782.67 unit/square). The physicochemical and microbiological properties of soil amended with MRSC were improved at harvest. The application of MRSC has potential to improve plant growth, productivity, rice blast disease management, and soil health of rice under aerobic cultivation systems.  相似文献

Triplicate groups of Mystus nemurus (Cuvier & Valenciennes) were fed isoenergetic semipurified diets containing seven dietary protein levels from 200 to 500 g kg–1 diet for 10 weeks. Dietary protein was supplied by graded amounts of a protein mixture (tuna muscle meal:casein:gelatine) at a fixed ratio of 50:37.5:12.5. Mystus nemurus fingerlings of initial weight 7.6 ± 0.2 g were fed close to apparent satiation at 2.5% of their body weight per day in two equal feedings. Growth performance and feed utilization efficiency increased linearly with dietary protein level from 202 to 410 g kg–1 diet and declined with protein levels of 471 g kg–1 diet or above. Protein efficiency ratio and apparent net protein utilization started to decline when the fish were fed with dietary protein levels exceeding 471 g kg–1 diet. Fish fed with lower protein diets (202–295 g kg–1 diet) had significantly ( P  < 0.05) higher carcass lipid content compared with fish fed with higher protein diets. Carcass lipid contents were inversely related to moisture content. Dietary protein did not significantly affect fish carcass protein and ash content. Using two-slope broken-line analysis, the dietary protein requirement for M. nemurus based on percentage weight gain was estimated to be 440 g kg–1 diet with a protein to energy ratio of 20 mg protein kJ–1 gross energy. This level of protein in the diet is recommended for maximum growth of M. nemurus fingerlings weighing between 7 and 18 g under the experimental conditions used in this study.  相似文献

This study dealt with a restoration project conducted at South‐East New Territories Landfill in Hong Kong, in order to screen suitable tree and shrub species (both native and pioneer species) for revegetation. For engineered landfills, landfill gas migration and leachate contamination to the topsoil are rarely problematic, but the lack of nutrients and moisture and poor physical soil conditions may jeopardize potential woodland establishment. The growth performance of 25 woody plant species subjected to 12 different soil amelioration and seedlings planting methods was compared. The results showed that the general performance (mortality rate, apical height, crown diameter and basal diameter) of pioneer species (notably Acacia species) was much better than that of native species in all blocks of woodland mix and scrub mix. The notch planting method was effective in helping seedling roots to have better contact with soil for water uptake, which subsequently increased the survival rates of seedlings, leading to a better tree coverage. Soil ameliorations (horticultural soil with fertilizer and horse and pig manure) generally increased the seedling survival rate and improved plant growth. Tree coverage on all the plant trial blocks was mainly established by the three Acacia species. A planting distance of 1 m was recommended for woodland establishment, and such dense seedlings planting would achieve a better tree coverage under a shorter period of time (than the distance of 3 m). Soybean isoflavones have multiple beneficial health effects especially on estrogen-deficient diseases such as menopausal symptoms. In this study, isoflavones were produced from soybean flour, and the extraction and purification parameters were optimized to give a high yield of total isoflavones, about 0.62 mg of aglycones/g of soybean flour, which is >2 times the initial yield. HPLC analysis and MTT cell proliferation assay using MCF-7 cells revealed that the product thus obtained not only contained a high content of isoflavone aglycones but also had estrogenic activity. MTT data also revealed that both genistein and daidzein exhibited estrogenic effects at lower concentrations and antiproliferative effects at higher concentrations, and 1 microM genistein and 10 microM daidzein exerted significant estrogenic activities, which were not more than that of the endogenous level of 17beta-estradiol (E2). The production method developed can be used as a guideline for manufacturing soy isoflavones, and the MTT assay was demonstrated to be suitable for quality control on isoflavone products. The results on the estrogenic properties of isoflavones can be used as reference data for their effective and safe usages in estrogenic therapy.  相似文献

We have completed the genetic characterization of all eight gene segments for four low pathogenic avian influenza (LPAI) viruses. The objective of this study was to detect the presence of novel signatures that may serve as early warning indicators of the conversion of LPAI viruses to high pathogenic avian influenza (HPAI) viruses. This study included three H5N2 and one H5N3 viruses that were isolated from live poultry imported into Singapore as part of the national avian influenza virus (AIV) surveillance program. Based on the molecular criterion of the World Organisation for Animal Health (OIE), sequence analysis with the translated amino acid (aa) sequence of the hemagglutinin (HA) gene revealed the absence of multibasic aa at the HA cleavage site, identifying all four virus isolates as LPAI. Detailed phylogenetic tree analyses using the HA and neuraminidase (NA) genes clustered these isolates in the Eurasian H5 lineage, but away from the HPAI H5 subtypes. This analysis further revealed that the internal genes clustered to different avian and swine subtypes, suggesting that the four isolates may possibly share their ancestry with these different influenza subtypes. Our results suggest that the four LPAI isolates in this study contained mainly avian signatures, and the phylogenetic tree for the internal genes further suggests the potential for reassortment with other different circulating avian subtypes. This is the first comprehensive report on the genetic characterization of LPAI H5N2/3 viruses isolated in South-East Asia.  相似文献

A hexameric rhamnose-specific lectin with a molecular mass of 205kDa and exhibiting some N-terminal sequence similarity to other fish lectins was isolated from roe of the grass carp (Ctenopharyngodon idellus) by affinity chromatography on rhamnose-Sepharose and ion exchange chromatography by fast protein liquid chromatography on a Mono S column. The lectin exhibited mitogenic activity toward murine splenocytes with a potency lower than that of the plant lectin ConA. It exerted a stimulatory effect at a concentration of 10 micro g/ml on the phagocytic activity of seabream (Sparus sarba) macrophages. It was unstable toward heat (temperature > or =40 degrees C), acid (0.1M HCl), alkali (0.1M NaOH), trypsin and succinylation.  相似文献

The responses of 24 white yam ( Dioscorea rotundata ) cultivars to mechanical and vector transmission with each of three viruses infecting yams were assessed through enzyme-linked immunosorbent assay (ELISA) and symptom development. The viruses were Dioscorea alata virus (DAV), genus Potyvirus ; Dioscorea alata bacilliform virus (DaBV), genus Badnavirus ; and Cucumber mosaic virus (CMV), genus Cucumovirus . Only TDr 95-128, a landrace cultivar from Nigeria, developed symptoms of infection with CMV and DaBV following mechanical and vector transmission, respectively. PAS-ELISA showed that nine genotypes remained uninfected by DAV and 11 were uninfected by CMV or DaBV. Genotypes TDr 747 and TDr 1640 both showed resistance to all three viruses.  相似文献

The objective of this study was to evaluate the effects of fishmeal (FM) replacement with corn protein concentrate (CPC) on growth performance, nutrient utilization, gut morphology and skin coloration of red hybrid tilapia, Oreochromis sp. Five isonitrogenous (350 g/kg crude protein) and isolipidic (10 g/kg lipid) diets were formulated to contain CPC that substituted 0%, 25%, 50%, 75% or 100% FM. Diets were fed to triplicate groups of tilapia (mean initial weight, 10.33 ± 0.02 g) twice daily for 63 days. The results showed that replacing up to 50% FM in red hybrid tilapia diet with CPC did not show any significant adverse effects on growth, feed utilization, haematocrit counts, condition factor and gut morphology of tilapia (p > 0.05). However, replacing 75% or 100% FM with CPC had deleterious effects (p < 0.05). Carotenoids in CPC contributed to skin yellowness, which was significantly higher in the diet where 100% FM was replaced with CPC. Using regression analysis, the optimal substitution level of FM by CPC was estimated at 25% for percentage weight gain, 33% for FCR and 29% for protein efficiency ratio. CPC could be used as a single plant protein source to substitute up to 50% FM in red hybrid tilapia diets.  相似文献

Genetically matched pluripotent embryonic stem (ES) cells generated via nuclear transfer or parthenogenesis (pES cells) are a potential source of histocompatible cells and tissues for transplantation. After parthenogenetic activation of murine oocytes and interruption of meiosis I or II, we isolated and genotyped pES cells and characterized those that carried the full complement of major histocompatibility complex (MHC) antigens of the oocyte donor. Differentiated tissues from these pES cells engrafted in immunocompetent MHC-matched mouse recipients, demonstrating that selected pES cells can serve as a source of histocompatible tissues for transplantation.  相似文献
