In the mammalian testis, the ubiquitin-proteasome system plays important roles in the process that promotes the formation of
mature sperm. We recently identified zygote-specific proteasome
assembly phaperone (ZPAC), which is specifically expressed in the mouse
gonads and zygote. ZPAC mediates a unique proteasome assembly pathway in the zygote, but the expression profile and function
of ZPAC in the testis is not fully understood. In this study, we investigated the possible role of ZPAC during mouse
spermatogenesis. First, we analyzed the expression of ZPAC and 20S proteasome subunit α4/PSMA7 in the adult mouse testis.
ZPAC and α4 were expressed in spermatogonia, spermatocytes, and round spermatids. In elongating spermatids, ZPAC was
expressed until step 10, whereas expression of α4 persisted until step 12. We then examined the expression profile of ZPAC
and α4 in a mouse model of experimental unilateral cryptorchidism. Consistent with appearance of morphologically impaired
germ cells following cryptorchidism, the ZPAC protein level was significantly decreased at 4 days post induction of
experimental cryptorchidism (D4) compared with the intact testis, although the amount of α4 protein persisted at least until
D10. Moreover, intense ZPAC staining was co-localized with staining of annexin V, an early indicator of apoptosis in
mammalian cells, in germ cells of cryptorchid testis, but ZPAC was also expressed in germ cells showing no detectable
expression of annexin V. These results suggest that ZPAC plays a role during spermatogenesis and raises the possibility that
20S proteasome mediated by ZPAC may be involved in the regulation of germ cell survival during spermatogenesis. 相似文献

Structures and dynamics of old-growth coniferous stands are affected by several types of disturbances including typhoons. We report the forest dynamics of four old-growth Chamaecyparis stands in central Japan that differ in the disturbance history of typhoons over a period of 23 years. The stem number, basal area and mortality were examined. In a predominant stand of C. obtusa (Sieb. et Zucc.) Endl., 24 % of the C. obtusa canopy trees died, mainly as a result of the severe damage of a strong typhoon that caused a single tree-fall gap and the following gap enlargements. In this stand, the total basal area decreased to 76.5 % of the initial value, although the mortality declined in recent years. In contrast, the other three stands decreased only slightly in the stem numbers (0.0–5.6 %) and increased in the basal areas of C. obtusa canopy trees. It is confirmed that the stand-level ingrowths of 300-year-old C. obtusa canopy trees could contribute to the increase in the stock of each stand. Our results support an idea that the dynamics of old-growth Chamaecyparis forests were greatly affected by typhoons. The stand structures will be gradually changed (with the processes of gap dynamics) and C. obtusa will continue to be dominant, potentially over hundreds of years. 相似文献

The hyphal tip was isolated from 13 weakly or moderately virulent strains of Helicobasidium mompa to remove double-stranded (ds) RNAs and demonstrate their role as the hypovirulence factor. All of 829 hyphal tip subcultures retained dsRNAs. However, strain v670 containing two large fragments (10kb) and one small fragment (ca. 2.3kb) of dsRNA lost the largest fragment in 3 of 63 subcultures analyzed. One of the three subcultures (v670hti) was used to inoculate carrots to regain virulence compared to the parental strain v670. When isolate v670hti was paired with v670, the largest fragment was reintroduced to v670hti, and its virulence was diminished. Northern blot analysis with two probes hybridizing dsRNA fragments in most H. mompa strains revealed that the largest fragment involved in hypovirulence was different from two other fragments that are common in Japan. These results indicate that the largest dsRNA fragment in strain v670 is associated with hypovirulence in H. mompa. 相似文献

An inoculation method using Lupinus luteus was developed for estimating virulence of isolates of the white root rot fungus, Rosellinia necatrix Prillieux. Fungal cultures grown on pieces of mulberry twigs were placed in contact with the hypocotyl of 3-week-old seedlings
growing in pots of soil. Disease development was uniform and reproducible in repeated experiments. Of 24 isolates with double-stranded
RNA, eight were weakly virulent. This method is useful throughout the year for estimating virulence of many isolates of the
fungus and for screening for hypovirulent isolates.

Isolates of the violet root rot fungus Helicobasidium mompa were collected from herbaceous and tree plants. Their host preference was studied by inoculation experiments using carrots,
sweet potatoes, and apple stocks. It was found that sweet potato isolates from Kyushu produced infection cushions on carrots
and sweet potatoes but not on apple stocks. Other isolates did not show host preference. Sweet potato isolates were also characterized
by ready hyphal mass (sclerotium) production. They were thought to have adapted to the habitat with high disturbance by annual
tillage.

New regulatory gene for pectate lyase (Pel) production of Erwinia chrysanthemi strain EC16 was searched by observing the gene dosage effect of each cosmid library in Pel production. From this survey, a
cosmid clone, p5A, had reduced in Pel production under both inducing and non-inducing conditions and caused less tissue maceration
of potato tubers. The 2.64-kb HindIII fragment from p5A was found to be responsible for this phenotype and to contain one major ORF consisting of 1107 nucleotides,
which had homology with ddlA (49.6%) and ddlB (49.6%) of Escherichia coli and with ddlA (44.6%) of Salmonella typhimurium. These genes had been shown to encode D-alanine-D-alanine ligase (Ddl), an enzyme which is involved in the synthesis of peptidoglycan. This ORF encodes 368 amino acid residues
and its molecular weight is estimated to be 43 kDa. This ORF of EC16 could complement ddl deficient mutant of Escherichia coli.

Paratropomyosin (PTM) composes myofibril functions to weaken the rigor linkages formed between actin and myosin during postmortem aging of muscles. PTM has the similar physico‐chemical properties as tropomyosin (TM) that is a regulatory protein of myofibrils. So far, it is unclear whether PTM is definitely different from TM, because the primary structure of PTM has not been determined yet. The aim of this study was to clarify structural difference of PTM from TM. PTM was prepared by column chromatography immediately after slaughter from broiler breast muscle, and purified by high‐performance liquid chromatography (HPLC). Purified PTM was successfully separated from TM, and the recovered PTM molecule was reduced with dithiothreitol to separate again by HPLC. Two subunits were obtained and peptides from each digested subunit by V8 protease were recovered by HPLC, and then amino acid sequences of the peptides were analyzed by protein sequencing. As a result, some amino acid residues were replaced from that of TMα1 isoform which is the major isoform of TM, and also was different between the two subunits. Therefore, it is concluded that PTM clearly differs from TM and it is suggested that functional difference in PTM from TM is attributed to amino acid replacements in subunits composing PTM. 相似文献
