Plants are generally assumed to influence the surrounding soil microflora through rhizodeposition. However, the role of rhizodeposits, and especially organic acids, in structuring the bacterial communities is still poorly understood. In this study, we asked the question whether plants differing in organic acid secretion have a different impact on the soil bacterial communities, and if this is the case, to which extent this impact is due to different organic acid concentrations in the rhizosphere. To investigate this question, we compared white lupin and wheat. The former is a high organic acid-secreting species, while the latter secretes only low amounts of carboxylates. We grew the plants in large microcosms including root-free control compartments for one year (replanted every second month) and analyzed the spatio-temporal changes in soil ATP concentrations, as well as in diversity and structure of bacterial communities (using DNA- and RNA-based DGGE) along a root-soil gradient after two, six and twelve month's cultivation. Our results showed: i) that white lupin and wheat differed in their impact on soil ATP concentrations and on the structure of root bacterial communities; ii) that cultivation time was a key factor in explaining the observed differences in all the parameters studied; and iii) that the amounts of organic acids accounted for a significant proportion (15%) of the variability within root active communities. These results indicate that plants influence their associated bacterial communities in a species-specific way and that for communities living in the direct vicinity of roots (rhizoplane-endorhizosphere), a significant part of this influence can be attributed to root-secreted organic acids. 相似文献

We used authentication tests developed for ancient DNA to evaluate claims by Asara et al. (Reports, 13 April 2007, p. 280) of collagen peptide sequences recovered from mastodon and Tyrannosaurus rex fossils. Although the mastodon samples pass these tests, absence of amino acid composition data, lack of evidence for peptide deamidation, and association of alpha1(I) collagen sequences with amphibians rather than birds suggest that T. rex does not. 相似文献

Bambara groundnut is a leguminous crop that originated in West Africa. It is a drought-tolerant crop and has the potential to provide improved food and nutritional security in the dry areas of Africa. However, the plant contains anti-nutritional factors that affect its food value chain. The objective of this study was to assess the content of four major anti-nutritional factors in 30 bambara groundnut accessions in South Africa. The seeds of the bambara groundnut landraces were ground to make a fine powder and assessed for condensed tannins (CTs), phytic acid phosphate (PAP), polyphenol, and trypsin inhibitor content. Each assay was done in triplicate. The content of the four anti-nutritional compounds varied widely among the bambara groundnut accessions. The mean values for CTs ranged between 0.2 and 6.2 mg/g of prodephinidin equivalent, whereas that of the PAP was between 1.4 and 4.9 mg/g. A low mean polyphenol content of 0.4 mg/g of GA (gallic acid) equivalent was observed among the accessions. The level of trypsin inhibitor ranged from 5.3 to 73.4 TI/mg. With the exception of CTs and trypsin inhibitor, no significant correlation was observed among the four anti-nutritional factors assessed in this study. An automated, rapid, sensitive, and label-free biosensor-based assay for folate-binding protein (FBP) in bovine milk utilizing surface plasmon resonance optical detection is described. The active concentration of FBP is estimated from its specific interaction with a pteroyl-l-glutamic (folic) acid derivative immobilized on the sensor surface in a direct binding assay format. Milk, colostrum, and milk powders are prepared for analysis by dilution into buffer. Analysis conditions, including ligand immobilization, flow rate, contact time, and regeneration, have been defined, and nonspecific binding considerations were evaluated. Performance parameters include a working range for FBP in buffer of 0-200 ng/mL, a method detection limit of 0.13 microg/mL in fluid milk, overall instrument response RSD(R) of 0.64%, a mean interassay RSD(R) of 7.3% for skim milk powder, and surface stability over ca. 200 samples. The technique was applied to the measurement of active FBP content of consumer milks, the heat classification of skim milk powders manufactured under a wide range of thermal processing protocols, and change during early bovine lactation. 相似文献

To understand evolutionary paths connecting diverse biological forms, we defined a three-dimensional genotypic space separating two flower color morphs of Antirrhinum. A hybrid zone between morphs showed a steep cline specifically at genes controlling flower color differences, indicating that these loci are under selection. Antirrhinum species with diverse floral phenotypes formed a U-shaped cloud within the genotypic space. We propose that this cloud defines an evolutionary path that allows flower color to evolve while circumventing less-adaptive regions. Hybridization between morphs located in different arms of the U-shaped path yields low-fitness genotypes, accounting for the observed steep clines at hybrid zones. 相似文献

New 2,2-dimethylcyclopropanecarboxylic acids bearing halogenated 1,3-alkadienyl substituents at position 3 of the cyclopropane ring were synthesised and the insecticidial activities of their 3-phenoxybenzyl and α-cyano-3-phenoxybenzyl esters against housefly (Musca domestica), Colorado potato beetle (Leptinotarsa decemlineata) and Egyptian cotton leafworm (Spodoptera littoralis) were determined. The activity of these compounds varied according to the relative position of the halogen substituents on the diene chain. The genetic modification in fruit and vegetables could lead to changes in metabolic pathways and, therefore, to the variation of the molecular pattern, with particular attention to antioxidant compounds not well-described in the literature. The aim of the present study was to compare the quality composition of transgenic wheat ( Triticum durum L.), corn ( Zea mays L.), and tomato ( Lycopersicum esculentum Mill.) to the nontransgenic control with a similar genetic background. In the first experiment, Ofanto wheat cultivar containing the tobacco rab1 gene and nontransgenic Ofanto were used. The second experiment compared two transgenic lines of corn containing Bacillus thuringiensis "Cry toxin" gene (PR33P67 and Pegaso Bt) to their nontransgenic forms. The third experiment was conducted on transgenic tomato ( Lycopersicum esculentum Mill.) containing the Agrobacterium rhizogenes rolD gene and its nontransgenic control (cv. Tondino). Conventional and genetically modified crops were compared in terms of fatty acids content, unsaponifiable fraction of antioxidants, total phenols, polyphenols, carotenoids, vitamin C, total antioxidant activity, and mineral composition. No significant differences were observed for qualitative traits analyzed in wheat and corn samples. In tomato samples, the total antioxidant activity (TAA), measured by FRAP assay, and the naringenin content showed a lower value in genetically modified organism (GMO) samples (0.35 mmol of Fe (2+) 100 g (-1) and 2.82 mg 100 g (-1), respectively), in comparison to its nontransgenic control (0.41 mmol of Fe (2+) 100 g (-1) and 4.17 mg 100 g (-1), respectively). On the basis of the principle of substantial equivalence, as articulated by the World Health Organization, the Organization for Economic Cooperation and Development, and the United Nations Food and Agriculture Organization, these data support the conclusion that GM events are nutritionally similar to conventional varieties of wheat, corn, and tomato on the market today. 相似文献
