The sulfur-induced embrittlement of nickel has long been wrapped in mystery as to why and how sulfur weakens the grain boundaries of nickel and why a critical intergranular sulfur concentration is required. From first-principles calculations, we found that a large grain-boundary expansion is caused by a short-range overlap repulsion among densely segregated and neighboring sulfur atoms. This expansion results in a drastic grain-boundary decohesion that reduces the grain-boundary tensile strength by one order of magnitude. This decohesion may directly cause the embrittlement, because the critical sulfur concentration of this decohesion agrees well with experimental data on the embrittlement. 相似文献

Centrioles are cylindrical, ninefold symmetrical structures with peripheral triplet microtubules strictly required to template cilia and flagella. The highly conserved protein SAS-6 constitutes the center of the cartwheel assembly that scaffolds centrioles early in their biogenesis. We determined the x-ray structure of the amino-terminal domain of SAS-6 from zebrafish, and we show that recombinant SAS-6 self-associates in vitro into assemblies that resemble cartwheel centers. Point mutations are consistent with the notion that centriole formation in vivo depends on the interactions that define the self-assemblies observed here. Thus, these interactions are probably essential to the structural organization of cartwheel centers. 相似文献

Stem growth of ‘White Rose’ cut seed potatoes, during exposure to ethylene concentrations up to 10 ppm for 2 weeks at 20 C, was markedly retarded and thickened. Development of the vascular system was abnormal, resulting in small xylem vessels. Root promotion was inhibited, but growth of lateral buds and roots became normal after removal from ethylene. Sound, hand harvested, whole potatoes were exposed to continuous flowing atmosphere containing air, air with 3 or 5% CO2, and air with concentrations of 0.5, 1.0, and 10ppm C2H4 for periods of 1 to 11 days at 20°C. At 1-day intervals, potatoes were bruised and black spot development assessed. Development of black spot in potatoes exposed to C2H4 was equal to or less than in potatoes at harvest. The severity remained low during 3-day exposure to 1 ppm C2H4 and after transfer to 4 days in air, then increased. Severity of black spot increased in potatoes exposed to air, with intermediate response from exposure to air with CO2. With an increase in sprouting, differences in black spot among treatments diminished. Less black spot developed in immature than in mature potatoes. 相似文献

The impact of winter cover crops, specifically wheat (Triticum aestivum L.), red clover (Trifolium pratense L.), and rapeseed (Brassica napus L.) or winter fallow, on community composition of arbuscular mycorrhizal fungi (AMF) in subsequent soybean roots was investigated in a 5-year field trial on andosolic soils in Japan. Soybean roots were sampled at full-flowering and analyzed for AMF communities using a partial LSU rDNA region. Phylogenetic analysis detected 22 AMF phylotypes, including eight Glomus, three Gigaspora, two Scutellospora, three Acaulospora, two Rhizophagus, and one of Funneliformis, Diversispora, Paraglomus, and an unknown glomeromycete in the roots. The 5-year rotation of different winter cover crops or winter fallow did not impact the molecular diversity of AMF communities colonizing the roots of subsequent soybean. In all of the rotations, Glomus and Gigaspora phylotypes were common to soybean roots over the 5-year period. Redundancy analysis (RDA) demonstrated that AMF communities in the roots of subsequent soybean were not significantly different among winter cover crop rotations or fallow. However, AMF communities in soybean roots were clearly influenced by rotation year suggesting that climate or other environmental factors were more important than winter cover cropping system management. 相似文献

The properties of garlic (Allium sativum L.) are attributed to organosulfur compounds. Although these compounds change during cultivation and storage, there is no report of their simultaneous analysis. Here, a newly developed analytical method with a rapid and simple sample preparation to determine four sulfoxides and three gamma-glutamyl peptides in garlic is reported. All garlic samples were simply extracted with 90% methanol solution containing 0.01 N hydrochloric acid and prepared for analysis. Alliin, isoalliin, methiin, cycloalliin, and gamma-l-glutamyl-S-methyl-l-cysteine were determined by normal-phase HPLC using an aminopropyl-bonded column. gamma-l-Glutamyl-S-(2-propenyl)-l-cysteine and gamma-l-glutamyl-S-(trans-1-propenyl)-l-cysteine were separated on an octadecylsilane column. The overall recoveries were 97.1-102.3%, and the relative standard deviation values of intra- and interday precision were lower than 2.6 and 4.6%, respectively. This newly developed method offers some advantages over the currently accepted techniques including specificity, speed, and ease of use and would be useful for chemical and biological studies of garlic and its preparations. 相似文献

ABSTRACT Reducing the arsenic (As) concentration in rice grains is of great interest from a human health perspective. Iron (Fe) materials immobilize As in soils, thereby effectively reducing the As concentration in rice grains. We investigated the effect of by-product Fe materials obtained from the casting industry on the As mobility in two soils (soil A and soil B) by a long-term (approximately 100 days) flooded soil incubation experiment. The examined Fe materials were spent steel shot (SSS), fine spent casting sand (SCS) containing steel shot, and two kinds of residual Fe materials (RIMs) from steel shot production. Commercial Fe materials used to immobilize As (zero-valent Fe and ferrihydrite) were tested for comparison. The dissolved As in soil solution of controls for soil A and soil B reached approximately 100 and 800 μg L?1, respectively. The effect on As immobilization of all the by-product Fe materials increased with time and was comparable to or greater than that of commercial ferrihydrite, except for SCS. The additions of SSS and RIMs decreased by more than 90% of the dissolved As in soil A and decreased by more than 50% in soil B after 100 days incubation. Overall, the effect of the by-product Fe materials on the solubility of silicon and phosphorus was much less than that of the commercial Fe materials. Considering the cost advantage over commercial Fe materials, the Fe materials obtained from the casting industry as by-products are promising amendments for the immobilization of As in paddy soils. 相似文献

In our wheat breeding program to introduce the low amylose character of Tanikei A6099 to elite lines, five waxy lines were unexpectedly obtained from 249 doubled haploid lines of the F1 hybrid of Saikai 168 × Tanikei A6099. The amylose content of all the waxy lines was <1% and the blue value was <0.1. Starch granule-bound proteins were extracted and subjected to modified sodium dodecyl sulfate polyacrylamide gel electrophoresis. The waxy lines lacked the Wx protein. Starch paste viscosity measurements gave pasting profiles of waxy wheat starch that were quite different from those of nonwaxy wheats but similar to those of waxy maize. However, the peak viscosity of waxy wheats was much higher than that of the waxy maize. 相似文献
