The performance of flow cytometry (FC) was compared with immunofluorescence microscopy (IM) for detection of Giardia duodenalis in bovine feces. Samples from 36 adult dairy cows and 208 dairy calves were collected. Flow cytometry test characteristics were calculated using continuous, ordinal, and dichotomized results. Spearman correlation coefficients comparing the results of the 2 tests were 0.47 and 0.68 for cows and calves, respectively. Using IM as indicative of presence or absence of G. duodenalis cysts in each sample, likelihood ratios of FC results with 0, 1, and > or = 2 gated events indicated that samples with 1 gated event were likely to be positive in the cows but not in the calves. Immunofluorescence microscopy detected G. duodenalis in 69.7% and 48.1% of cows and calves, respectively. When dichotomizing the FC results at a cut-off point of 1 or 2 gated events, 46.3% and 19.9% of the cow and 51.9% and 35.1% of the calf samples, respectively, were classified as G. duodenalis-positive. Relative to IM, the sensitivity in the cows was 0.59 and 0.28, respectively, and 0.76 and 0.64, respectively, in the calves. At a cut-off point of 1, 65.7% and 73.1% of the cow and calf samples, respectively, were correctly classified in FC, and at a cut-off point of 2, 49.3% and 78.4% were correctly classified in the cows and calves, respectively. Flow cytometry was less sensitive than IM. Possible reasons and research needed to improve FC for G. duodenalis detection are discussed. 相似文献
The use of personal protective equipment by veterinary workers during radiographic imaging is inconsistent. While the self‐reported use of leaded aprons and thyroid shields approaches 100% in some studies, the use of leaded gloves and eyeglasses is much lower. Previous studies describing personal protective equipment use are based on self‐reporting. Objectives of this prospective, observational study were to describe use of leaded personal protective equipment during radiographic imaging by veterinary workers, and to compare observed use with self‐reported use. Use of leaded personal protective equipment during radiographic imaging by veterinary workers was observed over a 10 week period using two motion‐triggered video cameras, and a questionnaire was then completed by workers on their use of personal protective equipment. Workers restrained the animal during 91.8% (753/820) of exposures. An apron and a securely closed thyroid shield were worn for >99% of studies. Gloves were used correctly for 43.6% (156/358) of radiographic studies. Leaded eyeglasses were worn for 1.7% (6/358) of studies. Correct glove use was more frequent during regular working hours than after‐hours for both veterinarians (odds ratio 32.7, P = 0.001) and veterinary students (odds ratio 75.1, P < 0.001). The number of workers in the room was lower when animals were sedated (P = 0.002) or anesthetized (P = 0.017). Workers overestimated their frequency of glove use (P <0.001). In conclusion, workers use personal protective equipment less frequently in an unsupervised environment, and overestimate their use of personal protective equipment. Use of sedation or anesthesia decreases worker exposure to ionizing radiation. 相似文献

A method is presented for the quantification and structure confirmation of trichothecenes in wheat by high-performance liquid chromatography combined with multiple mass spectrometry (MS(n)()). Nine type A- and B-trichothecenes were determined (nivalenol, deoxynivalenol, fusarenon-X, 3-acetyldeoxynivalenol, 15-acetyldeoxynivalenol, neosolaniol, diacetoxyscirpenol, HT-2 toxin, and T-2 toxin). Extraction was carried out with acetonitrile/water. The extract was purified on a MycoSep column. Quantification was based on an internal standard and atmospheric pressure chemical ionization in the positive ion mode. Recoveries from spiked wheat were in the range of 80-106% at levels of 500 ppb. The limits of quantification for the whole method were between 10 and 100 ppb. Ion adduct formation with ammonium and acetate ions and MS(n) experiments provided information about substitution and fragmentation behavior of the mycotoxins. A scheme has been established for the partial structure elucidation of type A- and B-trichothecenes in fungal cultures. 相似文献
This study investigated the acute toxicity (LC50-24 hr) effects of the essential oil of Cymbopogon citratus for adult Argulus sp. and Dolops discoidalis, before and during oviposition. In vitro acute toxicity (LC50-24 hr) was tested using 20, 40, 60, 80, 100, 120, 140 and 160 μg/L of C. citratus essential oil, and two control groups (one with cultivation tank water and one with cultivation tank water + alcohol) were used. Specimens of Argulus sp. and D. discoidalis submitted to acute toxicity were evaluated using histological procedures. The major chemical compounds of C. citratus essential oil were geranial (47.5%), neral (35.6%) and myrcene (6.7%). The LC50-24 hr for Argulus sp. adults was 67.97 μg/L, while for D. discoidalis it was 59.55 µg/L. In the oviposition of both species of argulids, maximum mortality began with treatments of 140 μg/L, while the LC50-24 hr for Argulus sp. and D. discoidalis was 83.98 μg/L and 82.48 μg/L, respectively. In both argulid species exposed to C. citratus essential oil, morphological alterations were observed only in the eyes, and they occurred in the ommatidium and rhabdomeres and were dependent on the concentration of C. citratus essential oil and the parasite species. 相似文献
Lippia alba hydrolate (LAH) is produced as a secondary product in essential oil extraction. The objective of this study was to evaluate the use of LAH in a simulation of the transport of tambaqui (Colossoma macropomum) juveniles. A series of 17 hr simulations were performed using load densities of 30, 60 and 90 juveniles per L and four LAH concentrations of 0.4%, 1%, 2% and 5%, with a control of just water. Fish survival, temperature, dissolved oxygen, conductivity, pH, ammonia excretion, alkalinity water hardness and net Na+, K+, Cl‐ fluxes, were all evaluated before and after the transport simulations. Mucous cell densities in the gills were also determined microscopically, post simulation. The results showed that addition of 5% LAH achieved better survival during the simulation than the water control. Dissolved oxygen was significantly lower with an increased load density. Also, conductivity, NH4 excretion and net ionic fluxes (K+ and Cl?) were lower with 5% LAH, although mainly where higher load densities were tested. Mucous cell density was lower in LAH concentrations of 2% and 5%, again with higher load densities. In view of these findings, it is suggested that 5% LAH is an effective sedative for use in the transport of tambaqui, mainly with high load densities. African lion (Panthera leo) populations are in decline throughout most of Africa, but the problem is particularly acute in southern Kenya, where Maasai people are spearing and poisoning lions at a rate that will ensure near term local extinction. Lion killing is shaped by Maasai perception of livestock depredation, socio-economic factors, and the complex relationship between Maasai and conservation. These all affect tolerance for lions and consequently Maasai behavior towards conservation initiatives and carnivores in general. We used an in-depth quantitative questionnaire and participatory rural appraisals (PRAs) to identify the social and ecological predictors of lion killing and to investigate the effect of a compensation scheme on individual tolerance. Individuals who lose a greater proportion of their livestock to predators relative to their overall livestock loss, those affiliated with an evangelical church, and those who mainly sell rather than accumulate livestock all reported a higher propensity to kill carnivores. The future of carnivore conservation in this region depends on a better understanding of the nuances of human–carnivore conflict and a concerted effort to address appropriate cultural and community-level institutions, chiefly by providing economic benefits to local people who engage in positive conservation activities. 相似文献
